Understanding how soil microbiomes respond to management is essential to maximizing soil health. We contrasted microbiomes in bulk soil under long-term organic and conventional management in a grain production setting. Management category significantly impacted the relative abundances of 17% of the most abundant taxa. Both conventional and organic management favored particular taxa, but these effects were not reflected in summary richness and diversity indices. Management systems also lead to differences in soil edaphic properties, including pH and nutrient status; this may have been the mechanism by which change in the prokaryote community was enacted. Community change between years of sampling was less pronounced, with only 6 taxa differentially abundant among years. Management category also impacted the abundance of functional genes related to the production and consumption of greenhouse gases. Particulate methane monooxygenase genes were more frequent in soil under organic management, while soluble methane monooxygenase genes were more frequent in soil under conventional management in 1 of 2 years. Nitrous oxide reductase genes were significantly less abundant in soils under second-year alfalfa than in soils under corn. This work highlights the ability of agricultural management to enact broad rearrangements to the structure of bulk soil bacterial communities.

BACKGROUND: Light-dependent activities against enveloped viruses in St. John's Wort (Hypericum perforatum) extracts have been extensively studied. In contrast, light-independent antiviral activity from this species has not been investigated. RESULTS: Here, we identify the light-independent inhibition of human immunodeficiency virus-1 (HIV-1) by highly purified fractions of chloroform extracts of H. perforatum. Both cytotoxicity and antiviral activity were evident in initial chloroform extracts, but bioassay-guided fractionation produced fractions that inhibited HIV-1 with little to no cytotoxicity. Separation of these two biological activities has not been reported for constituents responsible for the light-dependent antiviral activities. Antiviral activity was associated with more polar subfractions. GC/MS analysis of the two most active subfractions identified 3-hydroxy lauric acid as predominant in one fraction and 3-hydroxy myristic acid as predominant in the other. Synthetic 3-hydroxy lauric acid inhibited HIV infectivity without cytotoxicity, suggesting that this modified fatty acid is likely responsible for observed antiviral activity present in that fraction. As production of 3-hydroxy fatty acids by plants remains controversial, H. perforatum seedlings were grown sterilely and evaluated for presence of 3-hydroxy fatty acids by GC/MS. Small quantities of some 3-hydroxy fatty acids were detected in sterile plants, whereas different 3-hydroxy fatty acids were detected in our chloroform extracts or field-grown material. CONCLUSION: Through bioguided fractionation, we have identified that 3-hydroxy lauric acid found in field grown Hypericum perforatum has anti-HIV activity. This novel anti-HIV activity can be potentially developed into inexpensive therapies, expanding the current arsenal of anti-retroviral agents.

Hypericum perforatum (Hp) is commonly known for its antiviral, antidepressant, and cytotoxic properties, but traditionally Hp was also used to treat inflammation. In this study, the anti-inflammatory activity and cytotoxicity of different Hp extractions and accessions and constituents present within Hp extracts were characterized. In contrast to the antiviral activity of Hp, the anti-inflammatory activity observed with all Hp extracts was light-independent. When pure constituents were tested, the flavonoids, amentoflavone, hyperforin, and light-activated pseudohypericin, displayed anti-inflammatory activity, albeit at concentrations generally higher than the amount present in the Hp extracts. Constituents that were present in the Hp extracts at concentrations that inhibited the production of prostaglandin E(2) (PGE(2)) were pseudohypericin and hyperforin, suggesting that they are the primary anti-inflammatory constituents along with the flavonoids, and perhaps the interactions of these constituents and other unidentified compounds are important for the anti-inflammatory activity of the Hp extracts.

Meat from Holstein and crossbred organic dairy steers finished on winter rye and winter wheat pastures was evaluated and compared for meat quality, fatty acid and amino acid profiles, and consumer acceptability. Two adjacent 4-ha plots were established with winter rye or winter wheat cover crops in September 2015 at the University of Minnesota West Central Research and Outreach Center (Morris, MN). During spring of 2015, 30 steers were assigned to one of three replicate breed groups at birth. Breed groups were comprised of: Holstein (HOL; n = 10), crossbreds comprised of Montbéliarde, Viking Red, and HOL (MVH; n = 10), and crossbreds comprised of Normande, Jersey, and Viking Red (NJV; n = 10). Dairy steers were maintained in their respective replicate breed group from three days of age until harvest. After weaning, steers were fed an organic total mixed ration of organic corn silage, alfalfa silage, corn, soybean meal, and minerals until spring 2016. Breed groups were randomly assigned to winter rye or winter wheat and rotationally grazed from spring until early summer of 2016. For statistical analysis, independent variables were fixed effects of breed, forage, and the interaction of breed and forage, with replicated group as a random effect. Specific contrast statements were used to compare HOL versus crossbred steers. Fat from crossbreds had 13% greater omega-3 fatty acids than HOL steers. Furthermore, the omega-6/3 ratio was 14% lower in fat from crossbreds than HOL steers. For consumer acceptability, steaks from steers grazed on winter wheat had greater overall liking than steers grazed on winter rye. Steak from crossbreeds had greater overall liking than HOL steers. The results suggest improvement in fatty acids and sensory attributes of beef from crossbred dairy steers compared to HOL steers, as well as those finished on winter wheat compared to winter rye.

Organic production of one of the most popular botanical supplements, Echinacea, continues to expand in the U.S. Echinacea seeds typically show a high degree of dormancy that can be broken by ethephon or gibberelic acid (GA), but these methods are currently disallowed in organic production. In order to determine the efficacy of non-chemical seed treatments, we evaluated the effect of varying seed source and supplying light, with and without cold-moist stratification, on seed germination of the three most important medicinal species of Echinacea, E. angustifolia DC, E. purpurea (L) Moench, and E. pallida (Nutt.) Nutt. Treatments included cold-moist stratification under 24 h light, 24 h dark, and 16/8 h light/dark to break seed dormancy. We found that germination was greater in the E. purpurea and E. pallida seeds from a commercial organic seed source compared to a public germplasm source. When seeds were not cold-moist stratified, 16-24 h light increased germination in E. angustifolia only. Echinacea angustifolia, E. purpurea, and E. pallida seeds that were cold-moist stratified under 16-24 h of light for 4 wk had a significantly greater percentage and rate of germination compared to seeds germinated in the dark. Therefore, cold-moist stratification under light conditions is recommended as a method to break seed dormancy and increase germination rates in organic production of Echinacea.

Ivy gourd, Coccinia grandis (L.) Voigt, patches throughout Kailua-Kona, Hawaii Island, HI, were identified as persistent sources of melon fly, Bactrocera cucurbitae (Coquillett). These patches had a low incidence of Psyttalia fletcheri (Silvestri), its major braconid parasitoid natural enemy in Hawaii, and were used to evaluate augmentative releases of P. fletcheri against melon fly. In field cage studies of releases, numbers of melon flies emerging from ivy gourd fruit placed inside treatment cages were reduced up to 21-fold, and numbers of parasitoids were increased 11-fold. In open field releases of P. fletcheri into ivy gourd patches, parasitization rates were increased 4.7 times in release plots compared with those in control plots. However, there was no significant reduction in emergence of melon flies from fruit. In subsequent cage tests with sterile melon flies and P. fletcheri, combinations of sterile flies and P. fletcheri produced the greatest reduction (9-fold) in melon fly emergence from zucchini, Cucurbita pepo L. Reductions obtained with sterile flies alone or in combination with parasitoids were significantly greater than those in the control, whereas those for parasitoids alone were not. Although these results suggest that the effects of sterile flies were greater than those for parasitoids, from a multitactic melon fly management strategy, sterile flies would complement the effects of P. fletcheri. Cost and sustainability of these nonchemical approaches will be examined further in an ongoing areawide pest management program for melon fly in Hawaii.
