RESUMO

Phosphorus (P) is a growth-limiting nutrient for plants, which is taken up by root tissue from the environment as inorganic phosphate (Pi). To maintain an appropriate status of cellular Pi, plants have developed sophisticated strategies to sense the Pi level and modulate their root system architecture (RSA) under the ever-changing growth conditions. However, the molecular basis underlying the mechanism remains elusive. Inositol polyphosphate kinase (IPK2) is a key enzyme in the inositol phosphate metabolism pathway, which catalyzes the phosphorylation of IP3 into IP5 by consuming ATP. In this study, the functions of a rice inositol polyphosphate kinase gene (OsIPK2) in plant Pi homeostasis and thus physiological response to Pi signal were characterized. As a biosynthetic gene for phytic acid in rice, overexpression of OsIPK2 led to distinct changes in inositol polyphosphate profiles and an excessive accumulation of Pi levels in transgenic rice under Pi-sufficient conditions. The inhibitory effects of OsIPK2 on root growth were alleviated by Pi-deficient treatment compared with wild-type plants, suggesting the involvement of OsIPK2 in the Pi-regulated reconstruction of RSA. In OsIPK2-overexpressing plants, the altered acid phosphatase (APase) activities and misregulation of Pi-starvation-induced (PSI) genes were observed in roots under different Pi supply conditions. Notably, the expression of OsIPK2 also altered the Pi homeostasis and RSA in transgenic Arabidopsis. Taken together, our findings demonstrate that OsIPK2 plays an important role in Pi homeostasis and RSA adjustment in response to different environmental Pi levels in plants.

RESUMO

The Complexity-to-Diversity (CtD) strategy was applied to synthesize a 23-member compound collection from the natural product drupacine, including 21 novel compounds. An unusual benzo [d] cyclopenta [b] azepin skeleton was constructed by Von Braun reaction to cleave C-N bond of drupacine. Moreover, compound 10 has potential cytotoxicity to human colon cancer cells with low toxicity to the normal human colon mucosal epithelial cell lines.

To analyse the value of the apparent diffusion coefficient (ADC) in diffusion-weighted imaging (DWI) and the choline (Cho)/creatine (Cr) ratio and Cho/N-acetyl-aspartate (NAA) ratio in magnetic resonance spectroscopy (MRS) in the differential diagnosis between recurrent glioma and radiation injury. Chinese and English studies related to the diagnosis of recurrent glioma and radiation injury using DWI and MRS and published before 15 October 2022 were retrieved from PubMed, Embase, the Cochrane Library, China National Knowledge Infrastructure, China Biomedical Literature Database, VIP Journal Database, and Wanfang Database for a meta-analysis. A total of 11 articles were included in this study. ADC was lower in the recurrent glioma group than in the radiation injury group (standardized mean difference = -1.29, 95% confidence interval (CI) (-1.87, -0.71), P < 0.001). The Cho/Cr ratio was higher in the recurrent glioma group than in the radiation injury group (weighted mean difference = 0.65, 95% CI (0.40, 0.90), and P < 0.001). The Cho/NAA ratio was higher in the recurrent glioma group than in the radiation injury group, as evidenced by the sensitivity analysis. The sensitivity and specificity of the Cho/Cr ratio were 0.85 (0.73-0.92) and 0.82 (0.67-0.91), respectively, and the area under the curve was 0.86. The sensitivity and specificity of the Cho/NAA ratio were 0.82 (0.66-0.91) and 0.94 (0.69-0.99), respectively, and the area under the curve was 0.93. This meta-analysis showed that ADC, Cho/Cr, and Cho/NAA ratios all had high sensitivity and specificity. Therefore, DWI combined with MRS can effectively improve the diagnosis of recurrent glioma and radiation injury.

In this work, hollow spherical Pt-loaded WO3/ZnO heterostructured composites were prepared by a chemical liquid phase synthesis method. The morphology, crystal structure and components of the composites were characterized by SEM, TEM, XRD, XPS, etc. The sensing performance for various gases was also tested. Compared with the pristine WO3 (S = 44@225 °C, 50 ppm) gas sensor, the gas sensor that is functionalized with 1 wt% Pt and 0.5 mmol ZnO (1Pt/WZ-2) has a high response of 842-50 ppm at a relatively low temperature of 100 °C for TEA, with a quick response/recovery time of 34/120 s, a lower detection limit of 50 ppb, and good selectivity and moisture resistance. This study provides a highly efficient synthesis method of composite materials for TEA gas detection and the sensitivity mechanism is also discussed in detail.

RESUMO

Presently, the traditional methods employed for detecting livestock and poultry meat predominantly involve sensory evaluation conducted by humans, chemical index detection, and microbial detection. While these methods demonstrate commendable accuracy in detection, their application becomes more challenging when applied to large-scale production by enterprises. Compared with traditional detection methods, machine vision and hyperspectral technology can realize real-time online detection of large throughput because of their advantages of high efficiency, accuracy, and non-contact measurement, so they have been widely concerned by researchers. Based on this, in order to further enhance the accuracy of online quality detection for livestock and poultry meat, this article presents a comprehensive overview of methods based on machine vision, hyperspectral, and multi-sensor information fusion technologies. This review encompasses an examination of the current research status and the latest advancements in these methodologies while also deliberating on potential future development trends. The ultimate objective is to provide pertinent information and serve as a valuable research resource for the non-destructive online quality detection of livestock and poultry meat.

ETHNOPHARMACOLOGICAL RELEVANCE: In traditional Chinese folk medicine, Artemisia argyi H.Lév. & Vaniot (A. argyi) has been used for thousands of years, and it is clinically used to treat bronchitis and asthma. However, the mechanism of action of A. argyi on respiratory tract inflammation is not clear. Accumulating evidence that phosphofructo-2-kinase/fructose-2,6-bisphosphatase 3 (PFKFB3) is actively expressed in inflammation. Here, we found that iso-seco-tanapartholide (IST), a sesquiterpene isolated from A. argyi, exhibited potent anti-inflammatory activity and significant inhibition of PFKFB3 expression. Therefore, we evaluated the effect of IST on airway inflammation and revealed its possible mechanisms. AIM OF THE STUDY: This study aimed to investigate the protective effect and possible mechanism of IST in lipopolysaccharide (LPS)-induced acute lung injury in mice. MATERIALS AND METHODS: In vitro, RAW264.7 cells and BMDMs were stimulated with LPS, and the level of NO and inflammatory factors TNF-α, IL-1ß, and IL-6 were detected by Griess reagent and ELISA, respectively. The effect of IST on the levels of PFKFB3 and its downstream proteins (p-STAT3, p-p65) in cells was assayed by western blotting. Lactate and glycolytic phenotypes were detected by lactate kit and Seahorse assay. In vivo, a mouse model of acute lung injury was induced by LPS, and the levels of inflammatory factors were measured by ELISA. Expression of PFKFB3 and its downstream proteins (p-STAT3, p-p65) in mouse alveolar macrophages by western blotting analysis. Lung permeability assessment by Evans Blue dye assay. H&E staining and Immunocytochemistry were used to observe the protection of IST against lung injury. RESULTS: IST significantly reduced LPS-induced expression of PFKFB3 and its downstream proteins (p-STAT3, p-p65). The inhibition of PFKFB3 has an impact on the glycolytic phenotype, such as a reduction in the rate of extracellular acidification (ECAR) and elevated lactate levels, and an increase in the rate of cellular oxygen consumption (OCR). Furthermore, IST inhibited LPS-induced NO release and increased the expression of pro-inflammatory factors TNF-α, IL-1ß, and IL-6. In vivo, IST reduced pulmonary edema in LPS-induced acute lung injury, improved lung function, and reduced levels of inflammatory factors and lactate secretion. CONCLUSIONS: These results suggest that IST improves the characteristics of ALI by inhibiting the expression of the PFKFB3-mediated glycolytic pathway and may be a potential anti-inflammatory agent for inflammation-related lung diseases.

In this study, a straightforward two-step hydrothermal process was used to synthesize Fe-doped NiO nanomaterials. A number of characterization approaches were employed to explore the structure and morphology of the synthesized Fe-doped NiO. The as-prepared samples were multi-layered flower-like structures formed by nanoparticles, according to scanning electron microscopy and transmission electron microscopy studies. The findings of the study on gas sensing performance showed that the response of the 1.5 at % Fe-NiO sensor was nearly 100 times greater than that of the pure NiO sensor, and the lower limit of detection was greatly decreased (50 ppb). The 1.5 at % Fe-NiO sensor exhibited superior sensing performance for n-butanol. The incorporation of an appropriate amount of Fe into the NiO lattice modified the carrier concentration, which is the primary cause of the increased sensor performance of an appropriate amount of Fe-doped NiO. In addition, the density functional theory calculation method based on the first-principles theory was used to study the adsorption performance and electronic behavior of pure NiO and 1.5 at % Fe-NiO for n-butanol. The calculated results were consistent with the experimental results.

The sorting and processing of food raw materials is an important step in the food production process, and the quality of the sorting operation can directly or indirectly affect the quality of the product. In order to improve production efficiency and reduce damage to food raw materials, some food production enterprises currently use robots for sorting operations of food raw materials. In the process of robot grasping, some food raw materials such as fruits, vegetables and meat have a soft appearance, complex and changeable shape, and are easily damaged by the robot gripper. Therefore, higher requirements have been put forward for robot grippers, and the research and development of robot grippers that can reduce damage to food raw materials and ensure stable grasping has been a major focus. In addition, in order to grasp food raw materials with various shapes and sizes with low damage, a variety of sensors and control strategies are required. Based on this, this paper summarizes the low damage grasp principle and characteristics of electric grippers, pneumatic grippers, vacuum grippers and magnetic grippers used in automated sorting production lines of fruit, vegetable and meat products, as well as gripper design methods to reduce grasp damage. Then, a grasping control strategy based on visual sensors and tactile sensors was introduced. Finally, the challenges and potential future trends faced by food robot grippers were summarized.

BACKGROUND: Intracranial solitary fibrous tumor(SFT)/hemangiopericytoma (HPC) is an aggressive malignant tumor originating from the intracranial vasculature. Angiomatous meningioma (AM) is a benign tumor with a good prognosis. The imaging manifestations of the two are very similar. Thus, novel noninvasive diagnostic method is urgently needed in clinical practice. Texture analysis and model building through machine learning may have good prospects. AIM: To evaluate whether a 3D-MRI texture feature model could be used to differentiate malignant intracranial SFT/HPC from AM. METHOD: A total of 97 patients with SFT/HPC and 95 with AM were included in this study. Patients from each group were randomly divided into the train (70%) and test (30%) sets. ROIs were drawn along the edge of the tumor on each section of T1WI, T2WI, and contrasted T1WI using ITK-SNAP software. The segmented image was imported into the AK software for texture feature extraction, and the 3D ROI signal intensity histograms of T1WI, T2WI, and contrasted T1WI were automatically obtained along with all the parameters. Modeling was performed using the language R. Confusion matrix was used to analyze the accuracy of the model. ROC curve was constructed to assess the grading ability of the logistic regression model. RESULTS: After Lasso dimension reduction, 5, 9, and 7 texture features were extracted from T1WI, T2WI, and contrasted T1WI, respectively; additional 8 texture features were extracted from the combined sequence for modeling. The ROC analyses on four models resulted in an area under the curve (AUC) of 0.885 (sensitivity 76.1%, specificity 87.9%) for T1WI model, 0.918 (73.1%, 95.5%) for T2WI model, 0.815 (55.2%, 93.9%) for contrasted T1WI model, and 0.959 (92.5%, 84.8%) for the combined sequence model and were enough to correctly distinguish the two groups in 71.2%, 81.4%, 69.5%, and 83.1% of cases in test set, respectively. CONCLUSIONS: The radiological model based on texture features could be used to differentiate SFT/HPC from AM.

PURPOSE: To quantitatively assess the blood oxygen levels of the cerebral vein using quantitative susceptibility mapping (QSM), and to analyze the correlation between magnetic susceptibility value (MSV) and clinical laboratory indicators/cognitive scores in patients with Alzheimer's disease (AD). MATERIALS AND METHODS: Fifty-nine patients (21 males and 38 females) with clinically confirmed AD (AD group) and 22 control subjects (12 males, 10 females; CON group) were recruited. Clinical data and laboratory examination indexes were collected. All patients underwent Mini-mental State Examination, Montreal Cognitive Assessment, Clock Drawing Task, and Activity of Daily Living Scale test, as well as a routine MRI and enhanced gradient echo T2 star weighted angiography (ESWAN). RESULTS: Higher cerebral venous MSV was observed in AD group compared to CON group, significant differences were observed for bilateral thalamus veins and left dentate nucleus veins. The MSV of bilateral thalamus veins, bilateral internal cerebral veins, and bilateral dentate nucleus veins had significant negative correlation with Mini-mental State Examination score; the MSV of bilateral thalamus veins, bilateral dentate nucleus veins, right septal vein had a significant negative correlation with Montreal Cognitive Assessment scores; a significant negative correlation between the MSV of bilateral thalamus veins, left dentate nucleus vein, right septal vein and the Clock Drawing Task score; the MSV of bilateral thalamus veins, left dentate nucleus vein had a significant negative correlation with Activity of Daily Living Scale score. The MSV of left dentate nucleus vein was positively correlated with the course of the disease, the MSV of bilateral septal vein were positively correlated with the total cholesterol, and the MSV of left septal vein had a positive correlation with LDL. CONCLUSION: Decreasing cerebral venous oxygen level in AD patients may affect cognitive status, and associated with the deterioration of the disease in AD patients.

PURPOSE: To evaluate the application of conventional MRI histogram analysis based on the whole tumor measurement on assessing meningioma grading. MATERIALS AND METHODS: This retrospective study was approved by the institutional review board. A total amount of 90 patients with meningioma were enrolled and the preoperative MRI of them were analyzed. To be specific, the patient group were consisted of 45 patients with grade I, 38 with grade II, and 7 with grade III meningioma. Grade I meningioma is classified as low grade meningioma (LGM), whereas Grade II and III meningioma were combined and classified as high grade meningioma (HGM). ROIs were drawn along the edge of the tumor on each section of T1WI, T2WI, and contrasted T1WI. 3D ROI signal intensity histogram and all its parameters were obtained. Independent t-test and Kruskal-Wallis test were used for comparison between two groups. Univariate logistic regression analysis and Spearman's correlation analysis were used to screen for the parameters with high predictive efficiency, while multivariate logistic regression analysis was used to determine the optimal model for the classification of meningioma. RESULTS: There were significant differences observed between HGM and LGM groups regarding to histogram volume count, uniformity of three sequences, range of T1WI and T2WI, kurtosis, standard deviation, variance, max intensity of T2WI, skewness, mean deviation, minimum intensity, mean value, the 5th percentile, the 10th percentile, the 25th percentile, the 50th percentile, the 75th percentile, and the 90th percentile of contrasted T1WI. Volume count and uniformity were high predictive parameters in distinguishing HGM from LGM. Logistic regression model included contrasted T1WI histogram parameters (i.e. minimum intensity, volume count, skewness, uniformity, and the 75th percentile) showed the best diagnostic efficiency for meningioma grade, with a sensitivity and specificity of 83.9% and 77.4% (AUC = 0.834, cutoff value = 0.413), respectively. The optimal model was achieved with a sensitivity of 71.4% and a specificity of 78.6% in the test set (AUC = 0.791, cutoff value = 0.413). CONCLUSIONS: Histogram analysis of conventional MRI based on 3D tumor measurement can be applied in the assessment of meningioma grading in clinical.

OBJECTIVE: To evaluate the differentiation efficiency of texture analysis of T1WI, T2WI and contrasted-enhanced T1WI MRI sequences in gliomas with and without IDH1 mutation based on entire tumor region. MATERIALS AND METHODS: A total of 42 patients with histopathologically confirmed gliomas, including 21 patients carrying IDH1 mutation (IDH1mutation group) and 21 with wild-type IDH1 (IDH1wt group) were included in this study. The preoperative MRI and clinical data were collected. The regions of interest (ROIs) covering the entire tumor and edema were manually delineated on axial slices using O.K. (Omni Kinetics, GE Healthcare, China) software; and the histogram and GLCM features based on T1WI, T2WI and contrasted-enhanced T1WI sequences were automatically generated. RESULTS: Based on contrasted-enhanced T1WI features, the inertia resulted as the best feature for diagnosis, with the AUC of 0.844. Furthermore, the AUC for gliomas prediction with IDH1mutation was 0.800 for cluster prominence. IDH1-mutation was differentiated on T2WI with the highest AUC of 0.848, which corresponded to GLCM Entropy. After modeling, the accuracy of the contrasted-enhanced T1WI, T1WI, and T2WI features model was 0.952, 0.857, and 0.738, respectively. The AUC of Joint VariableT1WI+C for predicting IDH1mutation was 0.984, while the AUC of Joint VariableT1WI for predicting the same mutation was 0.927. The diagnostic efficiency of Joint VariableT2WI was also desirable. CONCLUSION: MRI texture analysis could be used as a new noninvasive method for identification of gliomas with IDH1 mutation. The present results show that the Joint Variable derived from conventional MR imaging histogram and GLCM features is suitable for precise detection of IDH1-mutated gliomas.
