Highly unsaturated fatty acids of the omega-3 series (HUFA) are major constituents of cell membranes, yet are poorly synthesised de novo by consumers. Their production, mainly supported by aquatic microalgae, has been decreasing with global change. The consequences of such reductions may be profound for ectotherm consumers, as temperature tightly regulates the HUFA content in cell membranes, maintaining their functionality. Integrating individual, tissue and molecular approaches, we examined the consequences of the combined effects of temperature and HUFA depletion on the key cardio-respiratory functions of the golden grey mullet, an ectotherm grazer of high ecological importance. For 4 months, fish were exposed to two contrasting HUFA diets [4.8% eicosapentaenoic acid (EPA)+docosahexaenoic acid (DHA) on dry matter (DM) versus 0.2% EPA+DHA on DM] at 12 and 20°C. Ventricular force development coupled with gene expression profiles measured on cardiac muscle suggest that combining HUFA depletion with warmer temperatures leads to: (1) a proliferation of sarcolemmal and sarcoplasmic reticulum Ca2+ channels and (2) a higher force-generating ability by increasing extracellular Ca2+ influx via sarcolemmal channels when the heart has to sustain excessive effort due to stress and/or exercise. At the individual scale, these responses were associated with a greater aerobic scope, maximum metabolic rate and net cost of locomotion, suggesting the higher energy cost of this strategy. This impaired cardiac performance could have wider consequences for other physiological performance such as growth, reproduction or migration, all of which greatly depend on heart function.

UV filters (organic or mineral) present in sunscreen products are emerging contaminants of coastal aquatic environments. There is an urgent need to understand marine organisms responses to these compounds. In this study, we investigated the effect of exposure to dilutions of commercial sunscreen formulations on bacterial communities of mullet (Chelon sp.). The gut and skin mucus microbial communities were characterized using a metabarcoding approach targeting the 16S rRNA gene. Our results revealed that mullets had its own bacterial communities that differ from their surrounding habitats and specific to tissue. The dilutions of commercial sunscreens modified the relative abundance of Actinobacteroita, Bacteriodota and Proteobacteria for both gut and skin microbiota. They also allowed to bacteria affiliated to Mycobacterium, Nocardia and Tenacibaculum genera, known to house pathogenic species, to colonize the epithelium which may have implications for fish host health.

Microplastics (MPs) and rising marine seawater temperatures are one of the major environmental problems threatening the survival of marine organisms and biodiversity. However, interactions between such multiple stressors are virtually unexplored. This study aimed to assess the combined effect of two temperatures and polyethylene MPs on the Pacific oyster Crassostrea gigas, one of the most globalized mollusc species for aquaculture. Our work highlights the potential ecological risk posed by these two factors on marine bivalve molluscs. The experimental design was carried out following a 14-day exposure of oysters to environmental concentrations of polyethylene MPs (0.01 mg.L-1), and to two temperatures (15 °C and 22 °C). Sampling was performed on days 0, 7, and 14. The µ-FTIR analysis was applied to quantify MPs of interest and to check a potential environmental contamination. Tissue samples of digestive glands were collected from the oysters to evaluate the activity of biomarkers including superoxide dismutase, glutathione-s-transferase, malondialdehyde and laccase through protein levels. We note that the combination of MPs and high water temperature (HWT, 22 °C) had a significant impact both on the survival of animals and on stress markers, by modifying lipid peroxidation and immune responses. This original study gave the first innovative results on this topic and provides us with knowledge of the combined effects of MPs pollution and HWT (simulating marine heatwaves situation) on C. gigas. There remains a lack of information on the toxicity and the potential environmental hazard of plastics in the marine environment.

Organic UV filters are emerging contaminants in personal care products such as sunscreens. The toxicity of numerous of these UV filter compounds has been demonstrated in several marine taxa. However, whilst the biological impact has already been largely demonstrated, the anthropogenic drivers leading to UV filter contamination still need to be identified. In this work, a survey was conducted on a site of the French Atlantic Coast (i) to describe beachgoers' behaviours (sunscreen use and beach frequentation), (ii) provide an estimation of the UV filters released at sea and (iii) highlight the effect of air temperature on these behaviours and on the release of UV filters. In parallel with these estimations of the UV filters released at sea, in situ chemical measurements were performed. By comparing the results of both approaches, this interdisciplinary work provides an insight of how the observations of beachgoers' behaviour modulations and attendance level fluctuations could be used to prevent UV filter contaminations and ultimately manage the ecotoxicological risk.

Marine heatwaves (MHWs) are becoming more frequent and intense due to climate change and have strong negative effects on ecosystem. Few studies have reproduced the complex nature of temperature changes of a MHW, while it is suggested that ectotherms may be more vulnerable to rapid changes such as during MHWs. Effects of an experimental MHW were investigated in the golden grey mullet Chelon auratus. Juveniles acclimated to 20 °C were exposed to a rapid 5 °C increase in temperature, followed by a five-day period at 25 °C, before quickly returning to 20°C. Metabolic variables (SMR-standard, MMR-maximum rate, AS-aerobic scope, EPOC-excess post­oxygen consumption) and critical swimming speed (Ucrit) were measured at different phases of this MHW and after a thermally stable recovery phase. Although the pattern was only significant for the SMR, the aerobic three variables describing aerobic metabolism (SMR, MMR and AS) immediately increased in fish exposed to the acute elevation of temperature, and remained elevated when fish stayed at 25 °C for five days. A similar increase of these metabolic variables was observed for fish that were progressively acclimated to 25 °C. This suggests that temperature increases contribute to increases in metabolism; however, the acute nature of the MHW had no influence. At the end of the MHW, the SMR remained elevated, suggesting an additional cost of obligatory activities due to the extreme event. In parallel, Ucrit did not vary regardless of the thermal conditions. Concerning EPOC, it significantly increased only when fish were acutely exposed to 25 °C. This strongly suggests that fish may buffer the effects of acute changes in temperature by shifting to anaerobic metabolism. Globally, this species appears able to cope with this MHW, but that's without taking into consideration future projections describing an increase in both intensity and frequency of such events, as well as other stressors like pollution or hypoxia.

With the omnipresence of plastic litter from oyster farming in marine coastal areas, the objective of this work was to better understand the weathering of plastics used in this field, focusing on oyster spat collectors. During their use, around fifteen years, collectors made of polypropylene (PP) undergo numerous degradations, alternatively submerged, emerged in seawater, and stored outdoor until the next cycle. They weaken, crack, break, end up fragmenting and disseminated in the environment as microplastics associated to persistent organic pollutants. In this work, a comparison of 55 months of in situ weathering with five months of artificial weathering in air or in artificial seawater in a homemade UV chamber was conducted to better understand the mechanisms involved. Chemical, thermal and surface characterizations of virgin and weathered samples were conducted using Fourier Transform Infrared Spectroscopy (FTIR), Differential Scanning Calorimetry (DSC) and Environmental Scanning Electron Microscopy (ESEM). After 55 months of in situ weathering, collectors were notably damaged with large fissures and loss of microplastics (MPs) associated with an increase of carbonyl index values and a decrease of melting temperatures and crystallinity rates. Considering only UV irradiation, five months of artificial weathering at 30 °C under continuous irradiation of 6.9 W/m2 under UV lamps (295-400 nm) reproduced approximately 4.4 months of natural sunlight. Artificial weathering confirmed that photooxidation by combined effects of UV rays and oxygen was the main weathering mechanism and was reduced in seawater. These results help to understand the mechanisms involved in the weathering of these collectors in the marine environment and provide valuable information for industrials and professionals. Our study suggests a better storage away from UV rays and a reduction of the duration of use compared to current practices.

Coastal areas are important interface environments between marine and terrestrial ecosystems and are also areas of high economic activity related, among others, to trade, fishing and tourism. The port areas of La Rochelle (France) are part of these areas with strong anthropic activities and are therefore subject to potential contamination, particularly with trace metals. Among the various sources of discharges, galvanic (or sacrificial) cathodic protection can be questioned. Indeed, few studies have been carried out on the potential impact of the degradation of sacrificial anodes in ports on the health of bivalves. A study was therefore carried out on the black scallop Mimachlamys varia in the commercial port and the marina of La Rochelle using the caging method. Several biomarkers of the health status of individuals were observed in the laboratory (SOD, GST, MDA, AChE, and LAC). Among the different results obtained over two exposure periods, it appears that the potential effects of sacrificial anodes, whether new or present on site for several years, are masked by meteorological conditions, as well as by harbor activities.

Taking advantage of the unique system of doubly uniparental inheritance (DUI) of mitochondria, we developed a reliable molecular method to sex individuals of the marine bivalve Macoma balthica rubra. In species with DUI (~100 known bivalves), both sexes transmit their mitochondria: males bear both a male- and female-type mitogenome, while females bear only the female type. Male and female mitotypes are sufficiently divergent to reliably PCR-amplify them specifically. Loop-mediated isothermal amplification (LAMP) is a precise, economical and portable alternative to PCR for molecular sexing and we demonstrate its application in this context. We used 154 individuals sampled along the Atlantic coast of France and sexed microscopically by gonad examination to test for the congruence among gamete type, PCR sexing and LAMP sexing. We show an exact match among the sexing results from these three methods using the male and female mt-cox1 genes. DUI can be disrupted in inter-specific hybrids, causing unexpected distribution of mitogenomes, such as homoplasmic males or heteroplasmic females. To our knowledge, DUI disruption at the intra-specific scale has never been tested. We applied our sexing protocol to control for unexpected heteroplasmy caused by hybridization between divergent genetic lineages and found no evidence of disruption in the mode of mitochondrial inheritance in M. balthica rubra. We propose LAMP as a useful tool to accelerate eco-evolutionary studies of DUI. It offers the opportunity to investigate the potential role of, previously unaccounted-for, sex-specific patterns such as sexual selection or sex-specific dispersal bias in the evolution of free-spawning benthic species.

This study focused on the impacts of aged aquaculture microplastics (MPs) on oysters (Crassostrea gigas). Adult oysters were exposed for two months to a cocktail of MPs representative of the contamination of the Pertuis Charentais area (Bay of Biscay, France) and issuing from oyster framing material. The MPs mixture included 28% of polyethylene, 40% of polypropylene and 32% of PVC (polyvinyl chloride). During the exposure, tissues were sampled for various analyzes (MP quantification, toxicity biomarkers). Although no effect on the growth of adult oysters was noted, the mortality rate of bivalves exposed to MPs (0.1 and 10 mg. L-1 MP) increased significantly (respectively 13.3 and 23.3% of mortalities cumulative). On the one hand, the responses of biomarkers revealed impacts on oxidative stress, lipid peroxidation and environmental stress. At 56 days of exposure, significant increases were noted for Glutathione S-Transferase (GST, 10 mg. L-1 MP), Malondialdehyde (MDA, 10 mg. L-1 MP) and Laccase (LAC, 0.1 and 10 mg. L-1 MP). No variations were observed for Superoxyde Dismutase (SOD). Besides, ingestion of MPs in oyster tissues and the presence in biodeposits was highlighted. In addition, in vitro fertilisations were performed to characterize MPs effects on the offspring. Swimming behavior, development and growth of D-larvae were analysed at 24-, 48- and 72-h after fertilisation. D-larvae, from exposed parents, demonstrated reduced locomotor activity. Developmental abnormalities and arrest as well as growth retardation were also noted. This study highlighted direct and intergenerational effects of MPs from aged plastic materials on Pacific oysters.

BACKGROUND: Scallops (Bivalvia: Pectinidae) present extraordinary variance in both mitochondrial genome size, structure and content, even when compared to the extreme diversity documented within Mollusca and Bivalvia. In pectinids, mitogenome rearrangements involve protein coding and rRNA genes along with tRNAs, and different genome organization patterns can be observed even at the level of Tribes. Existing pectinid phylogenies fail to resolve some relationships in the family, Chlamydinae being an especially problematic group. RESULTS: In our study, we sequenced, annotated and characterized the mitochondrial genome of a member of Chlamydinae, Mimachlamys varia-a species of commercial interest and an effective bioindicator-revealing yet another novel gene arrangement in the Pectinidae. The phylogeny based on all mitochondrial protein coding and rRNA genes suggests the paraphyly of the Mimachlamys genus, further commending the taxonomic revision of the classification within the Chlamydinae subfamily. At the scale of the Pectinidae, we found that 15 sequence blocks are involved in mitogenome rearrangements, which behave as separate units. CONCLUSIONS: Our study reveals incongruities between phylogenies based on mitochondrial protein-coding versus rRNA genes within the Pectinidae, suggesting that locus sampling affects phylogenetic inference at the scale of the family. We also conclude that the available taxon sampling does not allow for understanding of the mechanisms responsible for the high variability of mitogenome architecture observed in the Pectinidae, and that unraveling these processes will require denser taxon sampling.

Plastic is currently used in aquaculture as a material for settlement and magnification of oyster spats. Plastic weathering and fragmentation under natural conditions can lead to the production of micro and nanoparticles and additive leakage, with potential toxic effects on marine life. This study investigates the effects of the exposure to microplastic (MPs) cocktail derived from aged aquaculture material on oyster pediveliger larvae (Crassostrea gigas). The cocktail was made of high-density polyethylene (HDPE), polypropylene (PP) and polyvinyl chloride (PVC). The concentrations tested were 0, 0.1, and 10 mg MP·L-1. During the 7-day fixation phase, pediveliger larvae (17 days) were exposed to the MP cocktail in laboratory-controlled conditions. After exposure, the success of settlement was significantly lower for larvae exposed to 10 mg MP·L-1 (49 ± 0.9%) compared to control ones (61.8 ± 1.6%). No malformations or metamorphosis abnormalities were observed. Growth of pediveliger and spat stages was monitored up to 11 months. During the first twenty-eight days of development, spat growth was significantly lower for the two MPs exposure conditions (0.1 and 10 mg MP·L-1; respectively -51.8% and -44.4%) compared to control group. Subsequently, the previously exposed oysters grew faster than the control condition, resulting in a significantly greater growth (0.1 and 10 mg MP·L-1: +18.3% and +19.7%) than the control group at the end of follow-up. The nearly one-year follow-up highlighted the potential effects of MPs from aquaculture on larvae and spat of C. gigas.

This study aims to assess the toxic effects and the potential risk of widely used agricultural pesticides on the development (malformations and developmental arrest), growth and swimming activity of oyster D-larvae (Crassostrea gigas). Freshly fertilized oyster embryos were exposed for 24 h at 24 °C to different concentrations (0, 0.01, 0.1, 1 and 10 µg.L-1) of six different pesticides: Glyphosate and its commercial solution (Roundup), Isoproturon, Nicosulfuron, Chlortoluron and Boscalid. The six pesticides tested induced a significant increase in larval malformations and developmental arrests. All pesticides except Glyphosate and Isoproturon affected larval growth. Roundup, Nicosulfuron, Chlortoluron and Boscalid also affected the swimming behaviour of the D-larvae, with a significant decrease recorded in their maximum swimming speed. Comparison of the LOEC (Lowest-Observed-Effect Concentration) of each compound led to the following toxicity classification: Boscalid > Chlortoluron = Nicosulfuron > Glyphosate > Roundup > Isoproturon, with respectively LOEC of 0.0028; 0.015; 0.017; 0.11; 0.3 and 0.78 µg.L-1. By comparison of the maximum concentrations in the Pertuis Charentais (South West, France) and LOEC of each pesticide, the following risk scale was obtained: Chlortoluron > Boscalid > Glyphosate > Roundup > Nicosulfuron > Isoproturon. Our results revealed that Chlortoluron, Boscalid and to a lesser extent Glyphosate represent a potential threat to early life stages of oyster living in the Pertuis Charentais marine area.

Nowadays, pesticides and microplastics (MPs) are commonly found in coastal waters worldwide. Due to their widespread use, their persistence and toxicity, they may induce adverse effects on physiology and behaviour of marine organisms such as the Pacific oyster (Crassostrea gigas). This study explored the growth and valve activity of juvenile oysters exposed for 24 days to two frequently detected pollutants in the Pertuis Charentais (South West, France): a herbicide (chlortoluron, 85 µg.L-1) and high-density polyethylene microparticles (HDPE 20-25 µm, 112 MP.mL-1) alone or in combination (cocktail condition; 97 µg.L-1 of chlortoluron + 108 MP. mL-1). The valve activity of juvenile oysters recorded by using a High Frequency and Non-Invasive valvometer (HFNI) was characterized by three parameters: the number of valve micro-closures (VMC), the Valve Opening Amplitude (VOA), and the Valve Opening Duration (VOD). Additionally, daily shell growth and the oyster daily rhythm were assessed. The exposure to MPs of oysters led to a significant increase of VMC and a decrease of VOD and shell growth. The exposure to chlortoluron showed a significant increase of VOA and a decrease of VMC. In combination with MPs, chlortoluron still increased VOA and decreased VMC but also reduced the shell growth. Chronobiological analysis did not reveal any effects on the daily rhythm of both contaminants. This work highlighted significant effects of high environmental concentrations of MPs and Chlortoluron on the behaviour and growth of the Pacific oyster.

The present pilot study aimed to provide an overview of organic contaminant concentration levels in the littoral ecosystems of the Pertuis seas. The study determined the concentrations of twenty-nine pesticides, six nonylphenols and seven polybrominated diphenyl ethers (PBDEs) in sediments, seawater, Pacific oysters and blue mussels. Oysters accumulated a higher number of pesticides than blue mussels. Indeed, alpha BHC (0.60-0.72 ng/g, ww), chlorfenvinphos (1.65-2.12 ng/g, ww), chlorpyrifos (0.79-0.93 ng/g, ww), chlortoluron (2.50-4.31 ng/g, ww), metolachlor (up to 0.38 ng/g, ww) and parathion (0.56-0.69 ng/g, ww) were quantified in oysters whereas only alpha BHC (0.24-0.31 ng/g, ww), was quantified in mussels. The present results also revealed that the POPs detected in water or sediments were not ultimately found accumulated in bivalves. Other molecules such as methylparathion and BDE47 were quantified in sediments. These molecules, BDE99 and one nonylphenol (OP2OE) were quantified in seawater. Finally, the comparison with the available environmental guidelines showed that the values measured were at concentrations not considered to cause adverse effects at the populations' level except for chlortoluron in seawater (15-50 ng/L).

An important, but underappreciated, consequence of climate change is the reduction in crucial nutrient production at the base of the marine food chain: the long-chain omega-3 highly unsaturated fatty acids (n-3 HUFA). This can have dramatic consequences on consumers, such as fish as they have limited capacity to synthesise n-3 HUFA de novo. The n-3 HUFA, such as docosahexaenoic acid (DHA, 22:6n-3) and eicosapentaenoic acid (EPA, 20:5n-3), are critical for the structure and function of all biological membranes. There is increasing evidence that fish will be badly affected by reductions in n-3 HUFA dietary availability, however the underlying mechanisms remain obscure. Hypotheses for how mitochondrial function should change with dietary n-3 HUFA availability have generally ignored ATP production, despite its importance to a cell's total energetics capacity, and in turn, whole-animal performance. Here we (i) quantified individual variation in mitochondrial efficiency (ATP/O ratio) of muscle and (ii) examined its relationship with content in EPA and DHA in muscle membrane of a primary consumer fish, the golden grey mullet Chelon auratus, receiving either a high or low n-3 HUFA diet. Mitochondria of fish fed on the low n-3 HUFA diet had higher ATP/O ratio than those of fish maintained on the high n-3 HUFA diet. Yet, mitochondrial efficiency varied up about 2-fold among individuals on the same dietary treatment, resulting in some fish consuming half the oxygen and energy substrate to produce the similar amount of ATP than conspecific on similar diet. This variation in mitochondrial efficiency among individuals from the same diet treatment was related to individual differences in fatty acid composition of the membranes: a high ATP/O ratio was associated with a high content in EPA and DHA in biological membranes. Our results highlight the existence of interindividual differences in mitochondrial efficiency and its potential importance in explaining intraspecific variation in response to food chain changes.

Plastics are persistent synthetic polymers that accumulate as waste in the marine environment. Microplastics (MPs, <5 mm) can be found either as microbeads in body care and some industrial products or as plastic debris through degradation. Plastic microbeads (1-5 µm, fluorescent, Cospheric) were used to characterise the MP ingestion and determine their potential harmful effects on both the swimming behaviour and development of oyster D-larvae (Crassostrea gigas). For 24 h, embryos were first exposed to MPs at a temperature of 24 °C. In addition, 3 day-old D-larvae were exposed to the same temperature for 1, 3 and 5 h. Three concentrations of MPs were used: 0.1, 1 and 10 mg MP. L-1. After a 24-h period of embryonic exposure, we noted that MP agglomerates were stuck to the D-larvae coat and locomotor eyelashes. We also observed a significant increase in severe malformations and developmental arrests for larvae exposed to MPs ranging from 1 mg MP. L-1. In terms of swimming behaviour, the maximum speed recorded was lower for larvae exposed at 0.1 and 1 mg MP. L-1. After an acute exposure to MPs, particles were found in the digestive tract of 3 dpf (days post fertilisation) D-larvae. After 1-h exposure, the concentrations tested (0.1, 1 and 10 mg MP. L-1) resulted in respectively 38%, 86% and 98%. The larvae swimming behaviour was recorded and analysed. Unlike the results observed at the embryo-larval stage, 3-dpf larvae showed significant impacts with no dose-response effect.

This study aims at (1) experimentally estimating first sexual maturation of the European sardine S. pilchardus, (2) using the results to calibrate existing bioenergetic models. During the 183 days-experiment, fish growth and body condition were assessed by biometry, and gonads were weighed when present. Age, wet weight and total length at first maturity were estimated at 262 days, 10.79 ± 0.75 g, and 11.26 ± 0.21 cm, respectively. Including these traits in biphasic Von Bertalanffy models did not significantly improve simulations for either length or weight data, meaning that energy allocation was not impacted by these traits. The implementation of the results in the Dynamic Energy Budget (DEB) calibration procedure strengthened the parameter set of the existing model, but resulted in significant changes in the energy allocation. Our results are a first step that will allow the design of new experiments to further quantify maturation and reproduction rates in diverse environmental conditions, consolidating DEB model calibration.

Understanding the effects of plastic debris on marine ecosystems is essential in encouraging decision-makers to take action. The present study investigates the effect of a 24 h experimental exposure to high density polyethylene (HDPE) microplastics (MPs) of different sizes (4-6, 11-13 and 20-25 µm) and at three concentrations (0.1, 1 and 10 mg MP.L-1) on the development and locomotor activity of early stages of Pacific oyster, Crassostrea gigas. The bivalve embryo-larval assay (NF ISO 17244, 2015) was used in this study but with additional toxicity criteria: developmental arrests, abnormal D-larvae, maximum speed and swimming trajectory. Copper (Cu), was used as a positive control. Our results show that smaller MPs (4-6 and 11-13 µm) induced higher rates of malformations and developmental arrests than the larger ones (20-25 µm). In addition, a dose-dependent decrease of maximum swimming speed was observed for larvae exposed to MPs of 4-6 and 11-13 µm. On the other hand, there was no significant difference in swimming speed with the largest MPs size tested (20-25 µm). For all three sizes of MPs, there was a decrease in straight-line swimming trajectories, and an increase in circular trajectories. This abnormal swimming behaviour could affect larvae survival as well as colonization of new habitats.

The development of human activities on French Atlantic coastlines (La Rochelle) lead to chronic pollution of the environment by organic (pesticides, hydrocarbons, agrochemicals) and inorganic (heavy metals) contaminants. These past years, several regulations have been implemented to preserve coastal environments. The purpose of this study was to perform biomonitoring of bivalve species using an outdoor caging technique. The goal of our work was to assess the impact of harbour's trace elements on the state of health of the marine bivalve Mimachlamys varia. First, various molecular defence biomarkers were measured: SOD (oxidative stress), GST (detoxification process), MDA (lipid peroxidation), and Laccase (immune reaction). Thus, in April 2016, scallops were collected at three caging sites, which differ by their levels of pollution, after transplantation into port areas (fairing, rainwater) and a control site (marsh). Bivalve samples were taken at three sampling dates (D0, D07, D21). Biomarker assays were performed in the digestive glands due to their bioaccumulation properties. The second aim was to explore the impacts of inorganic pollutants placed in environmental harbour's sites. After 21 days, the biomarker response of transplanted bivalves revealed a SOD decrease, Laccase and GST stimulations, higher concentrations in Cu, Fe, As, Co, Mn, Zn, Sn and no significant variation of MDA concentration. Our ecological relevance of biomarker approaches opens interesting perspectives to identify M. varia such as a pertinent marine sentinel species. The several selected biomarkers determined could confirm their ability to appraise the water quality of hydro-systems located in French coastlines, such as port areas.

Mimachlamys varia is a sub-littoral bivalve encountered from Norway to the Mediterranean Sea, which lives mostly byssally attached to rocks. During the low tide period, M. varia individuals, located highest on the shore, may experience short time of aerial exposure and face a low availability of oxygen. Here we report a comparative metabolomic profiling of gill samples of M. varia obtained by both LC-QToF and APGC-QToF mass spectrometry, to analyze metabolic changes occurring during emersion in comparison with immersion. Scallops were grown in aquaria with a simulated intertidal environment mimicking short-duration air exposure that they might experience during extreme tides: alternating 2 h emersion and 10 h immersion. Our results show a switch from aerobic to anaerobic metabolism after only 2 h of emersion, with the resort to different pathways: glucose-lactate, glucose-succinate and aspartate-succinate pathways. Furthermore, carnitine-conjugated metabolites were found to accumulate during emersion, as well as urate. The level of tyrosine on the contrary was found to decrease. These findings indicate a complex metabolic reprogramming that occurs after a two hour emersion period and upon re-immersion. Furthermore, M. varia is used as sentinel species in pollution biomonitoring, through the assay of biomarkers to evaluate the effects of pollutants. Here we show that emersion induces a significant decrease of superoxide dismutase activity, an enzyme developed by bivalves to face oxidative stress and used as biomarker. These findings have to be taken into account to normalize sampling during campaigns of environmental monitoring, by taking in situ, as far as possible only immersed individuals.
