The interrelationship was investigated between the contractile function of the myocardium, the activity of some lipolytic enzymes in the body of rabbits and the blood content of free fatty acids under conditions of the hemodynamic heart overstrain in winter and summer. It was shown that winter is a less favourable season for the course of the above pathological process in the cardiac muscle as compared to summer.

The effect of large doses (700 U/kg daily for 6 days) of heparin on the heart contractile function, myocardial morphology, content of free fatty acids in the blood, and the activity of tri- and monoglycerin lipase in blood plasma and heart tissue of intact rabbits was studied 24, 72 hours and 6 days after the beginning of heparin administration. It was established that the activity of lipolytic enzymes intensified, the concentration of free fatty acids in blood plasma grows, and the myocardial contractility increases after heparin administration. The phase structure of the cardiac cycle is simultaneously disturbed slightly. It is concluded that the intensification of lipolysis plays a role in increasing the myocardial contractility.

Left-ventricular contractility and arterial blood levels of free fatty acids were studied experimentally in rabbits with acute focal myocardial ischemia at different hours of the day (12 a. m., 6 p. m., 12 p. m.). All parameters were measured before a myocardial acute-ischemia focus was created, and 15, 20 and 40 minutes after coronary artery ligation. Heart contractility and blood free fatty acid levels were shown to vary throughout the day. In acute focal myocardial ischemia, these parameters also show different patterns of variation. A relationship was demonstrated between the pattern of changes in some contractility indices under acute focal ischemia and blood baseline levels of free fatty acids, which was particularly manifest at 12 p. m., and grew very weak at 12 a. m.

A relationship between the production of interleukin 1 (IL-1) by macrophages from adjuvant-induced arthritic rats and cytochrome P-450-dependent hepatic microsomal monooxygenase was studied. The synthesis of IL-1 by splenic and peritoneal macrophages on day 17 postadjuvant treatment was not altered, but the hepatic cytochrome P-450 levels and monooxygenase activity were significantly decreased. Beta-carotene treatment of arthritic rats reduced hind paw swelling and concurrently stimulated the ability of macrophages to secrete IL-1 and increased the cytochrome P-450 levels and the activity of hepatic monooxygenase. The findings did not establish a definite relationship between the production of IL-1 by systemic macrophages on the one hand, and the hepatic cytochrome P-450 levels a and monooxygenase activity on the other hand. It thus appears that IL1 is unable to play a role of a mediator between the immune system and the hepatic cytochrome P-450-dependent monooxygenase system of rats with adjuvant-induced arthritis.

Alkaline phospholipases A were studied in rabbit myocardium under normal conditions and in heart impairment of various etiology. In mitochondrial fraction activity of phospholipase A was increased with elevation of Ca2+ concentration, while the enzyme was Ca2(+)-independent in lysosomes. Hemodynamic overloading of heart, acute local myocardial ischemia and diphtheritic intoxication led to an increase (2-4-fold) of mitochondrial phospholipases A activity with simultaneous decrease of the enzymatic activity in lysosomal membranes.

Dynamics of lipolytic enzymes activity was studied in pre- and postheparin blood plasma of rabbits with hemodynamic heart overloading, with acute local ischemia of myocardium and with diphtheria intoxication. Development of all these pathological processes, impairing myocardium, was accompanied by appearance of lipolytic activity specific for intralipid and for activated intralipid in preheparin blood plasma, by an increase in activity of lipolytic enzymes in postheparin blood plasma as well as by alteration in the spectrum of blood lipoproteins. These alterations depended on the character of disease, impairing the heart muscle.

Fatty emulsion intralipid, activated by rabbit blood plasma, intralipid, micellar emulsion of monooleine and tributyryne emulsion were used, respectively, as substrates for estimation of asctivities of lipoprotein lipase (EC 3.1.1.34), triacyl glycerolipase (EC 3.1.1.3), monoacyl glycerolipase (EC 3.1.1.23) and tributyrynase (EC 3.1.1.13) by modified Dol's method in rabbit heart tissue, heart fatty tissue and blood plasma. Effect of heparin on extraction of lipolytic enzymes from tissues was studied.

The paper discusses changes in the activity of lipolytic enzymes in blood plasma of rats before and after heparin administration in the course of experimental atherosclerosis development. This development was shown to be characterized by an abrupt rise of preheparin plasma level (comparatively to normal) and of monoacylglycerolipase and tributyrinase activity, with this rise being unchanged throughout the experiment. An increase in the postheparin plasma activity of heparin-dependent lipolytic enzymes, lipoprotein lipase, triacylglycerolipase, monoacylglycerolipase, and tributyrinase was seen only in the initial stage of the disease. After 30-40 weeks the activity of the enzymes was below normal. It is concluded that the increased activity of lipoproteid lipase and triacylglycerolipase is related to their activation in the blood channel.

Twenty-eight physiological, biochemical and morphological parameters that characterize intact heart activity were studied during different seasons. The factor analysis made it possible to define two groups of the characteristics with a pronounced internal correlation. Analysis of the characteristics has demonstrated that the lysosomal apparatus of cardiomyocytes is involved in the maintenance of heart muscle contractility.

It has been shown that prophylactic administration of ubiquinone protects rats liver from the toxic damage by D-galactosamine both on ultrastructural and on cell levels. Ubiquinone administration prevents necrosis in hepatocytes and preserves their ability for compensatory reactions expressed in activation of protein-synthesis regulating structures in the cell. Ubiquinone decreases hyperfermentemia and hyperbilirubinemia as well as prevents the decrease in liver protein content caused by galactosamine. Ubiquinone exerts an antioxidant effect, blocking the induction of lipid peroxidation both in intact and hepatic rats.
