RESUMO

Regulatory single nucleotide polymorphisms (rSNPs) are the least-studied group of SNP; however, they play an essential role in the development of human pathology by altering the level of candidate genes expression. In this work, we analyzed 29 rSNPs in 17 new candidate genes associated with preeclampsia (PE) according to the analysis of the transcriptome in placental tissue. Three ethnic groups have been studied (yakut, russian, and buryat). We have detected significant associations of PE with eight rSNPs in six differentially expressed genes, i.e., rs10423795 in the LHB gene; rs3771787 in the HK2 gene; rs72959687 in the INHA gene; rs12678229, rs2227262, and rs3802252 in the NDRG1 gene; rs34845949 in the SASH1 gene; and rs66707428 in the PPP1R12C gene. We used a new approach to detecting genetic markers of multifactorial diseases in the case of PE based on a combination of genomic, transcriptomic, and bioinformatic approaches. This approach proved its efficiency and may be applied to detecting new potential genetic markers in genes involved in disease pathogenesis, which reduces missing heritability in multifactorial diseases.

Sex ratio in first trimester of pregnancy is skewed due to preferential elimination of female embryos. It could be resulted from aberrant X-chromosome inactivation. X-chromosome inactivation was analyzed in extraembryonic tissues of miscarriages and induced abortions with 46, XX karyotype. In chorion cytotrophoblast of both miscarriages and induced abortions observed either random or skewed X-chromosome inactivation. In extraembryonic mesoderm of the control group, random inactivation was observed, whereas 15% of miscarriages had skewed X-chromosome inactivation. The highest frequency of skewed inactivation of one of the parental homologues was observed in the groups of blighted ovum pregnancy and embryos from women with recurrent pregnancy loss. It was suggested that in these cases compartmentalization of cells in the blastocyst probably leads to predominance of cell with mutant active X-chromosome among the cells of inner cell mass carrying the aberrations that are incompatible with normal embryonic development.

RESUMO

The pregnancy course, labor, delivery and early neonatal period were studied in pregnant women with Chlamydia trachomatis infection as well as in their children. The levels of specific antibodies to Chlamydia trachomatis pgp 3 plasmid protein in maternal sera as well as perinatal complications were comparatively analyzed. According to the results, examination of anti-pgp3 antibodies is a highly specific and useful tool for the detection of Chlamydia trachomatis infection at its active stage in pregnant women.

Ultrastructural changes of blood vessels of ovaries and oviducts, destruction of the part of follicular epitheliocytes and disruption of other structures of blood-follicular barrier occur at the early period of experimental inflammations of appendage uterus on the background of traditional antibiotic therapy. All changes lead to atresia of growing follicules. Fibrotic-sclerotic changes develop on the 30th day and during later period. Course of eplir phonophoresis decreases ultrastructural disruptions of vessels, follicular apparatus of ovaries and oviducts epithelium. Effect of physiotherapy is manifested though activity of fibroclasts and macrophages, that regulate collagen production/resorption ratio thus preventing the development of fibrotic changes of ovary stroma and oviduct wall.

RESUMO

The levels of azithromycin in the chorion tissue within the first 3 months of the pregnancy term (42 to 56 days post coitus) were determined. Three days after the use of azithromycin (Sumamed) the antibiotic concentration in the villi tissue was 0.065 to 0.26 mcg/mg (the average of 0.129 mcg/mg). The azithromycin concentration above 0.125 mcg/mg was detected in 38.5% of the women. The results confirmed the fact of significant azithromycin accumulation in the chorion tissue. The percentage of the azithromycin transplacental transfer within the above mentioned pregnancy term varied from 2.3 to 9.3 (the average of 4.6).

RESUMO

Experimental and clinical investigations have demonstrated that the addition of abdominal-sacral electrophoresis of esobel 1% solution to combined therapy of acute inflammation of the uterine appendages improves treatment results. Bioantioxidants bring about the antiinflammatory action of the mud preparation esobel while its high-polar lipids prevent structural-metabolic impairment of the appendages' tissue elements as shown by a pronounced antiexudative effects and weak secondary alteration.

RESUMO

On the original experimental model of chronic inflammation of the ovaries and oviducts in white rats it was established that electrophoresis of 1% esobel solution prevents an increase in the specific volume of interstitial connective tissue, development of fibrosclerotic alterations and adhesive process. Peloidophysiotherapy normalizes growth and atresia of the ovarian follicular system. Thus, electrophoresis of 1% esobel solution is highly effective in combined treatment of chronic inflammatory diseases of the uterine appendages.

RESUMO

The paper presents the results of the action of underwater exercises and adaptational respiratory exercise under the hypoventilation mode on health of pregnant women with threatened abortion at different terms of gestation and of their babies. The exercises led to physiological development of pregnancy, diminished intrauterine fetal hypoxia, less frequent hospitalizations of the pregnant women, good pregnancy outcomes, delivery of healthy babies. Moreover, maternal morbidity decreased 1.5-fold and perinatal morbidity 2-fold.

Clinical study of the efficiency of Poetam (affinity-purified antibodies to recombinant human erythropoietin) in the treatment of anemia in patients with pubertal uterine hemorrhages proved that combined therapy with Poetam and iron preparation normalized erythron parameters, structural and metabolic status of erythrocytes, and ferrokinetic parameters of the peripheral blood sooner than monotherapy with Poetam or sorbifer.
