RESUMO

Salvia bowleyana is a traditional Chinese medicinal plant that is a source of nutritional supplements rich in salvianolic acid B and a potential experimental system for the exploration of salvianolic acid B biosynthesis in the Labiatae. Here, we report a high-quality chromosome-scale genome assembly of S. bowleyana covering 462.44 Mb, with a scaffold N50 value of 57.96 Mb and 44,044 annotated protein-coding genes. Evolutionary analysis revealed an estimated divergence time between S. bowleyana and its close relative S. miltiorrhiza of ~3.94 million years. We also observed evidence of a whole-genome duplication in the S. bowleyana genome. Transcriptome analysis showed that SbPAL1 (PHENYLALANINE AMMONIA-LYASE1) is highly expressed in roots relative to stem and leaves, paralleling the location of salvianolic acid B accumulation. The laccase gene family in S. bowleyana outnumbered their counterparts in both S. miltiorrhiza and Arabidopsis thaliana, suggesting that the gene family has undergone expansion in S. bowleyana. Several laccase genes were also highly expressed in roots, where their encoded proteins may catalyze the oxidative reaction from rosmarinic acid to salvianolic acid B. These findings provide an invaluable genomic resource for understanding salvianolic acid B biosynthesis and its regulation, and will be useful for exploring the evolution of the Labiatae.

We present a powerful method to enhance the magnetic plasmon (MP) resonances of metamaterials composed of periodic arrays of U-shaped metallic split-ring resonators (SRRs) for high-quality sensing. We show that by suspending the metamaterials to reduce the effect of the substrate, the strong diffraction coupling of MP resonances can be achieved, which leads to a narrow-band mixed MP mode with a large magnetic field enhancement. It is also shown that for such a diffraction coupling, the magnetic field component of the lattice resonance mode of periodic arrays must be parallel to the induced magnetic moment in the metallic SRRs. Importantly, the sensitivity and the figure of merit (FOM) of the suspended metamaterials can reach as high as 1300 nm/RIU and 40, respectively. These results suggest that the proposed metamaterials may find great potential applications in label-free biomedical sensing.

Due to the potential environment and health risks of tert-butylhydroquinone (TBHQ), rapid, portable, selective and sensitive quantification of TBHQ in food and the environment are strictly essential. With this in mind, a selective, sensitive and rapid colorimetric TBHQ biosensor was developed using rationally designed copper-crosslinked carbon dot hydrogel nanozyme (BC-CDs@Cu). The BC-CDs@Cu had a high peroxidase-like activity toward the chromogenic reaction of hydrogen peroxide with dopamine via the generation of hydroxyl radicals and electron transfer process. The Michaelis-Menten constants of BC-CDs@Cu for dopamine and hydrogen peroxide were determined to be 0.86 and 0.91 mM. The added TBHQ markedly inhibited the BC-CDs@Cu-catalyzed dopamine oxidation by hydrogen peroxide, ascribing to the highly effective and rapid scavenging of hydroxyl radicals and the suppression of electron transfer. The inhibitory extent was applied for well quantifying TBHQ in the range of 0.5 - 20.0 µM with a detection limit of 70 nM. The proposed biosensor had a negligible response to various interfering substances. Moreover, a smartphone-assisted visual ratiometric biosensor was fabricated, and used to accomplish portable quantification of TBHQ in edible oils and water samples. This work reveals the enormous potential of hydrogel nanozyme, which will open a new situation for the detection of hazardous substances.

A higher level of intelligence can improve adolescents' interactions with intelligent online services, although overemphasizing intelligent online services may nullify their sense of autonomy and in turn affect their acceptance intention. Enterprises have therefore focused on the best ways through which to provide intelligent online services. Based on the technology acceptance model, this study constructs a theoretical model of the impact of perceived control and power on adolescents' acceptance intention of intelligent services. Through a scenario experiment involving an intelligent online recommendation service, 195 participants were obtained to test the model. The results show that the adolescents' perceived control affects their acceptance intention of intelligent online services through their perceived usefulness. The adolescents' sense of power moderates the influence of perceived control on perceived usefulness. This study supplements the research on intelligent online services and provides a reference for online merchants seeking to design such service processes.

Customer engagement behavior is a critical success factor for community e-commerce. While many community e-commerce websites are currently improving service quality to enhance customer engagement behavior, little is known about how such e-services affect customer engagement behavior. Building upon the stimulus-organism-response (SOR) model, this study developed a research model to explain how e-service quality of community e-commerce platform affects customer engagement behavior through customer trust and perceived risk. The research model was empirically evaluated by surveying 326 customers who have shopped through the community e-commerce platforms. The results indicate that e-service quality (system design, intelligent fulfillment, security assurance, and interactive service) positively affects customer engagement behavior. Besides, customer trust and perceived risk play a mediating role between e-service quality and customer engagement behavior. This study offers recommendations to managers on how to build an attractive community e-commerce platform to stimulate customer engagement behavior.

Isochrysis galbana is considered an ideal bait for functional foods and nutraceuticals of humans because of its high fucoxanthin (Fx) content. However, multi-omics analysis of the regulatory networks for Fx biosynthesis in I. galbana has not been reported. In this study, we report a high-quality genome assembly of I. galbana LG007, which has a genome size of 92.73 Mb, with a contig N50 of 6.99 Mb and 14,900 protein-coding genes. Phylogenetic analysis confirmed the monophyly of Haptophyta, with I. galbana sister to Emiliania huxleyi and Chrysochromulina tobinii. Evolutionary analysis revealed an estimated divergence time between I. galbana and E. huxleyi of âˆ¼ 133 million years ago. Gene family analysis indicated that lipid metabolism-related genes exhibited significant expansion, including IgPLMT, IgOAR1, and IgDEGS1. Metabolome analysis showed that the content of carotenoids in I. galbana cultured under green light for 7 days was higher than that under white light, and ß-carotene was the main carotenoid, accounting for 79.09% of the total carotenoids. Comprehensive multi-omics analysis revealed that the content of ß-carotene, antheraxanthin, zeaxanthin, and Fx was increased by green light induction, which was significantly correlated with the expression of IgMYB98, IgZDS, IgPDS, IgLHCX2, IgZEP, IgLCYb, and IgNSY. These findings contribute to the understanding of Fx biosynthesis and its regulation, providing a valuable reference for food and pharmaceutical applications.

Schizochytrium sp. is the best natural resource for omega-3 long-chain polyunsaturated fatty acids. We report a high-quality genome sequence of Schizochytrium limacinum SR21, which has a 63 Mb genome size, with a contig N50 of 2.67 Mb and 6,838 protein-coding genes. Phylogenomic and comparative genomic analyses revealed that DHA-producing Schizochytrium and Aurantiochytrium strains were highly similar and possessed similar genes. Analysis of the fatty acid synthase (FAS) for LC-PUFAs production results in the annotation of all genes in map00062 and map01212. A gene cluster and 10 ORFs related to PKS pathway were found in the genome. 1,402 differentially expressed genes (DEGs) of the treated groups (0.5 g/L yeast extract) were identified by comparing with the control groups (1.0 g/L yeast extract) at 36 h. A weighted gene coexpression network analysis revealed that 2 of 7 modules correlated highly with the fatty acid and DHA contents. The DEGs and transcription factors were significantly correlated with fatty acid biosynthesis, including MYB, Zinc Finger and ACOX. The results showed that these hub genes are regulated by genes involved in fatty acid biosynthesis pathways. The results providing an important reference for further research on promoting fatty acid and DHA accumulation in S. limacinum SR21.

The Podostemaceae are ecologically and morphologically unusual aquatic angiosperms that survive only in rivers with pristine hydrology and high water quality and are at a relatively high risk of extinction. The taxonomic status of Podostemaceae has always been controversial. Here, we report the first high-quality genome assembly for Cladopus chinensis of Podostemaceae, obtained by incorporating Hi-C, Illumina and PacBio sequencing. We generated an 827.92 Mb genome with a contig N50 of 1.42 Mb and 27,370 annotated protein-coding genes. The assembled genome size was close to the estimated size, and 659.42 Mb of the assembly was assigned to 29 superscaffolds (scaffold N50 21.22 Mb). A total of 59.20% repetitive sequences were identified, among which long terminal repeats (LTRs) were the most abundant class (28.97% of the genome). Genome evolution analysis suggested that the divergence time of Cladopus chinensis (106 Mya) was earlier than that of Malpighiales (82 Mya) and that this taxon diverged into an independent branch of Podestemales. A recent whole-genome duplication (WGD) event occurred 4.43 million years ago. Comparative genomic analysis revealed that the expansion and contraction of oxidative phosphorylation, photosynthesis and isoflavonoid metabolism genes in Cladopus chinensis are probably related to the genomic characteristics of this growing submerged species. Transcriptome analysis revealed that upregulated genes in the shoot group compared to the root group were enriched in the NAC gene family and transcription factors associated with shoot development and defense responses, including WUSCHEL (WUS), ASYMMETRIC LEAVES (ASL), SHOOT MERISTEMLESS (STM), NAC2, NAC8, NAC29, NAC47, NAC73, NAC83 and NAC102. These findings provide new insights into the genomic diversity of unusual aquatic angiosperms and serve as a valuable reference for the taxonomic status and unusual shoot apical meristem of Podostemaceae.

A laser-triggered vacuum switch (LTVS) is an advanced closing switch with nanosecond delay and jitter. In order to enhance hold-off voltage and extend the service lifetime of an LTVS, we designed a multichannel laser-triggered vacuum switch (MLTVS) utilizing a cone-shaped target electrode placed on the cathode platform. The fabrication and testing of the MLTVS is described in this paper. Experimental results show that the working voltage of the MLTVS with a gap distance of 12 mm is from 30 V to 20 kV. The threshold energy for triggering the switch is 0.4 mJ corresponding to a peak power density of 27.9 MW/cm(2). The triggering lifetime of a spot can reach up to 18,000 shots. In addition, the relationship between triggering lifetime and target materials is analyzed using a field emission scanning electron microscope. A hypothesis of the vacuum gap's triggering mechanism is discussed based on the measured results.
