RESUMO

Objective To study the effect and mechanism of pearl hydrolysate on hepatic sinusoidal capillarization in liver fibrosis. Methods Hepatic sinusoidal endothelial cells (HSEC) and hepatic stellate cells (HSC-LX2) were incubated with Hepu pearl hydrolysate.The proliferation of HSEC and HSC-LX2 was examined by MTT colorimetry.The cell cycle and apoptosis of HSC-LX2 were measured by flow cytometry.The changes of the microstructures such as fenestra and basement membrane of HSEC were observed by transmission electron microscopy. Results The intervention with leptin increased the viability of HSC-LX2 (P=0.041),decreased the viability of HSEC (P=0.004),and caused capillarization signs such as decreased number and diameter of fenestrae and formation of continuous basement membrane.The treatment with pearl hydrolysate at different doses increased and expanded the fenestrae of HSEC (low dose:P=0.020;medium dose:P=0.028;high dose:P=0.032),disintegrated the extracellular basement membrane of HSEC (low dose:P=0.020;medium dose:P=0.028;high dose:P=0.032),decreased the viability of HSC-LX2 (low dose:P=0.018;medium dose:P=0.013;high dose:P=0.009),and induced the apoptosis of HSC-LX2 (low dose:P=0.012;medium dose:P=0.006;high dose:P=0.005).Pearl hydrolysate exerted therapeutic effect on capillarization in a dose-dependent manner (low dose:P=0.020;medium dose:P=0.028;high dose:P=0.032).Moreover,high-dose pearl hydrolysate showed stronger effect on capillarization of hepatic sinuses than colchicine (P=0.034) and salvianolic acid B (P=0.038). Conclusion Hepu pearl hydrolysate can increase the viability of HSEC,restore the area of fenestrae,disintegrate the basement membrane,and decrease the viability and induce the apoptosis of HSC-LX2,demonstrating significant pharmacological effects on the capillarization of HSEC and HSC-LX2.

RESUMO

We propose a design of a retinal-projection-based near-eye display for achieving ultra-large field of view, vision correction, and occlusion. Our solution is highlighted by a contact lens combo, a transparent organic light-emitting diode panel, and a twisted nematic liquid crystal panel. Its design rules are set forth in detail, followed by the results and discussion regarding the field of view, angular resolution, modulation transfer function, contrast ratio, distortion, and simulated imaging.

Lithium garnets are considered as promising solid-state electrolytes for next-generation solid-state Li metal batteries (SSLBs). However, the Li intrusion driven by external stack pressure triggers premature of Li metal batteries. Herein, for the first time, an in situ constructed interfacial shield is reported to efficiently inhibit the pressure-induced Li intrusion in SSLBs. Theoretical modeling and experimental investigations reveal that high-hardness metallic Mo nanocrystals inside the shield effectively suppress Li dendrite growth without alloy hardening-derived interfacial contact deterioration. Meanwhile the electrically insulated Li2 S as a shield component considerably promotes interfacial wettability and hinders Li dendrite penetration into the bulk of garnet electrolyte. Interfacial shield-protected Li6.4 La3 Zr1.4 Ta0.6 O12 (LLZTO)-based cells exhibit significantly enhanced cyclability without short circuits under conventional pressures of ≈0.2 MPa and even at high pressure of up to 70 MPa; which is the highest endurable stack pressure reported for SSLBs using garnet electrolytes. These key findings are expected to promote the wide-pressure-range applications of SSLBs.

The endogenous hydrogen sulfide (H2S) can be adopted as an indicator for the indirect detection of sulphate-reducing bacteria (SRB), which considered to be closely related to pipeline corrosion and human intestinal health. Unfortunately, the in-situ detection of endogenous H2S from SRB in the complex culture medium still faces huge challenges. Besides nonspecific adsorption from the culture medium of SRB, the problem of electrode passivation by produced elemental sulfur during electrochemical detection processes of H2S cannot be ignored. To address these challenges, herein a synergistic sensing platform based on self-cleaning electrode interface and indirect detection strategy (specific H2S-induced chemical reaction) is developed. This indirect sensing strategy-assisted self-cleaning electrochemical platform showed a relatively good linear response toward H2S in the range of 0.5 - 5 µM, and the corresponding limit of detection (LOD) was calculated to be 5.09 nM. More importantly, the satisfactory self-cleaning electrode interface in indirect detection system (with only a 4.10% decrease in signal over 50 electrochemical repeated cycles) showed the electrode surface not being disturbed by elemental sulfur. Furthermore, this good selectivity of the indirect detection strategy in combination with the reproducibility, stability, and antifouling activity of the self-cleaning interface, enabled a synergistic sensing platform to detect H2S directly in the complex culture medium of SRB without time-consuming sample pretreatments. Moreover, this proposed construction strategy of synergetic sensing platform could be explored to other endogenous molecules in complex environment based on different antifouling materials and specific reactions.

Objective: To identify differentially expressed genes via bioinformatical analysis for nasopharyngeal carcinoma (NPC) and explore potential biomarkers for NPC. Methods: We downloaded the NPC gene expression datasets (GSE40290, GSE53819) and obtained differentially expressed genes (DEGs) via GEO2R. Functional analysis of DEGs was performed by Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis. In order to explore the interaction of DEGs and screen the core genes, we established protein-protein interaction (PPI) network. Then the expression level, prognostic and diagnostic analysis of the core genes in NPC were performed to reveal their potential effects on NPC. Furthermore, we obtained the transcription factors (TF) and microRNAs of core genes to construct the coregulatory network. Results: We obtained 124 up-regulated genes and 190 down-regulated genes in total. These genes were found to be related to signal transduction, extracellular matrix organization and cell adhesion based on GO analysis. KEGG analysis revealed that the NF-kappa B (NF-κB) signaling pathway, pathways in cancer were mainly enriched signaling pathways. 25 core genes were obtained by constructing PPI network. Then the high expression of 10 core genes in NPC were verified via GEPIA, Oncomine databases and laboratory experiments. The TF-microRNA coregulatory network of the 10 core genes was built. Survival and diagnostic analysis indicated that SPP1 had negative influence on the prognosis of NPC patients based on two datasets and nine up-regulated core genes (FN1, MMP1, MMP3, PLAU, PLAUR, SERPINE1, SPP1, COL8A1, COL10A1) might be diagnostic markers for NPC. Conclusions: Core genes of NPC were screened out by bioinformatical analysis in the present study and these genes may serve as prognostic and diagnostic biomarkers for NPC.

PURPOSE: This study aimed to investigate the prediction of early response to concurrent chemoradiotherapy (CCRT) through a combination of pretreatment multi-parametric magnetic resonance imaging (MRI) with clinical prognostic factors (CPF) in cervical cancer patients. METHODS: Eighty-five patients with pathologically confirmed cervical cancer underwent conventional MRI, intravoxel incoherent motion diffusion weighted imaging (IVIM-DWI), and dynamic contrast-enhanced MRI (DCE-MRI) before CCRT. The patients were divided into non- and residual tumor groups according to post-treatment MRI. Univariable and multivariable analyses were performed to pretreatment MRI parameters and CPF between the two groups, and optimal thresholds and predictive performance for post-treatment residual tumor occurrence were estimated by drawing the receiver operating characteristic (ROC) curve. RESULTS: There were 52 patients in non- and 33 in residual group. The residual group showed a lower perfusion fraction (f) value and volume transfer constant (Ktrans) value, a higher apparent diffusion coefficient (ADC) value, diffusion coefficient (D) value and volume fraction of extravascular extracellular space (Ve) value, and a higher stage than the non-residual tumor group (all P < .05). D, Ktrans, Ve and stage were independent prognostic factors. The combination of D, Ktrans and Ve improved the diagnostic performance compared with individual MRI parameters. A further combination of these three MRI parameters with stage exhibited the highest predictive performance. CONCLUSIONS: Pretreatment D, Ktrans, Ve and stage were independent prognostic factors for cervical cancer. The predictive capacity of multi-parametric MRI was superior to individual MRI parameters. The combination of multi-parametric MRI with CPF further improved the predictive performance.
