RESUMO

Herein, an all-solid-state sequential self-organization and self-assembly process is reported for the in situ construction of a color tunable luminous inorganic/polymer hybrid with high direct piezoresponse. The primary inorganic self-organization in solid polymer and the subsequent polymer self-assembly are achieved at high pressure with the first utilization of piezo-copolymer (PVDF-TrFE) as the host matrix of guest carbon quantum dots (CQDs). This process induces the spontaneous formation of a highly ordered, microscale, polygonal, and hierarchically structured CQDs/PVDF-TrFE hybrid with multicolor photoluminescence, consisting of very thermodynamic stable polar crystalline nanowire arrays. The electrical polarization-free CQDs/PVDF-TrFE hybrids can efficiently harvest the environmental available kinetic mechanical energy with a new large-scale group-cooperation mechanism. The open-circuit voltage and short-circuit current outputs reach up to 29.6 V cm-2 and 550 nA cm-2 , respectively. The CQDs/PVDF-TrFE-based hybrid nanogenerator demonstrates drastically improved durable and reliable features during the real-time demonstration of powering commercial light emitting diodes. No attenuation/fluctuation of the electrical signals is observed for ≈10 000 continuous working cycles. This study may offer a new design concept for progressively but spontaneously constructing novel multiple self-adaptive complex inorganic/polymer hybrids that promise applications in the next generation of self-powered autonomous optoelectronic devices.

Limited information is available on the balance state of pro- and anti-inflammatory cytokines in patients with recurrent implantation failure (RIF). This study assessed the pro- and anti-inflammatory cytokines in plasma of 34 patients with RIF, compared with those of 25 women with a successful pregnancy in the first IVF/intracytoplasmic sperm injection-embryo transfer (IVF/ICSI-ET) cycle. The IFN-γ, IL-1ß, IL-6 and IL-4 concentrations were higher, whereas the TGF-ß1 concentration was lower in the RIF group compared with the control group. Furthermore, the ratios of pro-inflammatory and anti-inflammatory cytokines IFN-γ/IL-4, IFN-γ/IL-10, IFN-γ/TGF-ß1, IL-6/IL-10, IL-6/TGF-ß1, IL-1ß/TGF-ß1 and TNF-α/TGF-ß1 were higher in the RIF group (all P < 0.01). The results suggested a shift toward a pro-inflammatory state in peripheral blood of the patients with RIF.

Staphylococcus aureus (S. aureus), particularly methicillin-resistant S. aureus (MRSA), and its biofilms are great threats in the food industry. Bacteriophage-encoded endolysins are promising tools to inhibit pathogens and to eliminate their biofilms. In this work, a virulent phage qdsa002 against S. aureus ATCC43300 (MRSA) was isolated, and the phage's endolysin (Lys84) and its domains were expressed and purified. Morphological and genome analyses demonstrated that qdsa002 is a Twort-like phage from Myoviridae. Lys84 contains two catalytic domains (CHAP and Amidase_2) and one cell binding domain (SH3b). This endolysin exhibits a strong lytic activity against S. aureus and has a wider bactericidal spectrum than qdsa002. Moreover, Lys84 exceed 10 µM effectively removed around 90 % of the biofilms of S. aureus. Besides, CHAP and Amidase_2 domains remained 61.20 % and 59.46 % of lytic activity as well as 84.31 % and 70.11 % of anti-biofilm activity of Lys84, respectively. The lytic and anti-biofilm activities of the combination of CHAP and Amidase_2 were close to 90 % of those of Lys84. These results indicated that Lys84 and its domains might be alternative antimicrobials for controlling S. aureus and its biofilms.

PROBLEM: The efficacy of lymphocyte immunotherapy (LIT) for unexplained recurrent miscarriage (uRM) remains indefinite. The objective of the present study is to comprehensively evaluate the alterations in the proportions and functions of peripheral blood lymphocytes in patients with uRM before and after LIT. METHOD OF STUDY: Seventy-six women with uRM were included in the present study. Immunophenotype of peripheral blood lymphocytes (76 patients), NK cytotoxicity (62 patients) and Th1 (IFN-γ and TNF-α)/Th2 (IL-10) ratios (73 patients) were assessed during the luteal phase before LIT and after the third LIT by the flow cytometer. RESULTS: To date, 29 patients had already delivered or been pregnant more than 20 weeks (successful pregnancy group) and 5 patients experienced subsequent abortion (abortion group). The percentages of CD3(+) T cell, CD3(+) CD4(+) T cell and IL-10-producing Th1 cell were significantly increased, whereas the percentages of CD3(+) HLA-DR(+) T cell, CD56(+) CD3(-) NK cell and CD56(dim ) CD16(+) NK cell, the NK cytotoxicity and the ratios of Th1/Th2 were significantly decreased after LIT in the total patients and in the successful pregnant group. The percentages of CD56(+ ) CD3(-) NK cell and IFN-γ-producing cell, and the ratio of IFN-γ/IL-10 were significantly lower after LIT in the successful pregnant group compared to those in the abortion group. CONCLUSIONS: LIT alters the proportions and functions of most peripheral blood lymphocyte subsets. Some of these alterations may be beneficial for pregnancy maintenance, whereas some may be potential markers for predicting subsequent abortion.
