RESUMO

With the advent of long-duration space explorations, ionizing radiation (IR) may pose a constant threat to astronauts without the protection of Earth's magnetic field, or hypomagnetic field (HMF). However, the potential biological effects of a HMF on the cellular response to IR have not been well characterized so far. In this study, immortalized human bronchial epithelial cells were exposed to X-rays under either a geomagnetic field (GMF, ~50 uT) or HMF (<50 nT) culture condition. A significant increase of the cell survival rate in HMF after radiation was observed by colony formation analysis. The kinetics of DNA double-strand breaks (DSBs), determined by γH2AX foci formation and disappearance, presented a faster decrease of foci-positive cells and a significantly lower mean number of γH2AX foci per nucleus in HMF-cultured cells than in GMF-cultured cells after radiation. In addition, a γH2AX/53BP1 colocalization assay showed an upregulated DSB recovery rate in HMF cultured cells. These findings provided the first evidence that HMF exposure may enhance the cellular DSB repair efficiency upon radiation, and consequently modulate the genotoxic effects of IR.

OBJECTIVE: To investigate whether the apurinic/apyrimidinic endonuclease 1 (APE1) 1349 T>G and -656 T>G polymorphisms were associated with the risk of noise-induced hearing loss (NIHL) in a Chinese population. METHODS: The two APE1 polymorphisms were analyzed among 613 NIHL workers and 613 normal hearing workers using the minor groove binder TaqMan probe assay. RESULTS: We found that the APE1 -656 TT genotype was associated with a increased risk of NIHL [adjusted odds ratio (OR) 1.46, 95% confidence interval (CI) 1.05-2.06]. This increased risk was more pronounced in the stratification analysis. Furthermore, we found that subjects with two risk genotypes (hOGG1 Cys/Cys, APE1 -656 TT) had a significantly increased risk of NIHL (adjusted OR 1.91, 95% CI 1.27-2.88). CONCLUSION: Our study identified that the APE1 -656 T>G polymorphism may contribute to the susceptibility of NIHL.

Coffee consumption has been linked to risk of colorectal cancer theoretically, but the findings were conflicting from observational studies. Results from the recent meta-analysis suggested a moderate favorable effect of coffee consumption on colorectal cancer risk, especially for colon cancer. However, the relationship, if exists, between coffee consumption and colorectal cancer risk is unclear. Thus, the dose-response relationship was assessed by restricted cubic spline model and multivariate random-effect meta-regression. The results suggested that a significant association was found between coffee consumption and decreased risk of colorectal and colon cancer among subjects consuming ≥4 cups of coffee per day. A potential nonlinear relationship should be assessed before assuming a linear relationship.

A meta-analysis was conducted to assess the effect of glutathione peroxidase1 (GPX1) gene Pro200Leu (rs1050450) polymorphism on cancer risk. A comprehensive search was performed to identify all studies on the association of GPX1 gene Pro200Leu polymorphism with cancer risk. The fixed or random effect pooled measure was selected based on homogeneity test among studies. Heterogeneity among studies was evaluated using the I (2). Potential sources of between-study heterogeneity were explored by meta-regression and the sensitivity analysis. Publication bias was estimated using Egger's linear regression test. 35 published articles with 36 results were identified involving 16,920 cases and 19,946 controls. Results from the articles that both obeyed Hardy-Weinberg equilibrium in controls and met high quality design, showed no significant association of GPX1 gene Pro200Leu polymorphism with cancer risk in any of dominant (OR = 1.05, 95 %CI = 0.98-1.12), recessive (OR = 1.04 (0.95-1.13), and TT versus CC (OR = 1.05, 95 %CI = 0.97-1.15) models, and the findings were consistent considering the stratified analysis by cancer type. However, multivariate-adjusted ORs from articles that both obeyed Hardy-Weinberg equilibrium in controls and met high quality design, showed a significant association considering dominant (OR = 1.22, 95 %CI = 1.06-1.41), TT versus CC (OR = 1.16, 95 %CI = 1.02-1.32) models, and a marginally significant association was found considering TC versus CC (OR = 1.11, 95 %CI = 0.99-1.25) model. And compared with the CC genotype, the erythrocyte GPX activity was significantly lower for TT genotype: the standardized mean difference (SMD) = -0.37, 95 %CI = (-0.624, -0.118), and CT genotype: SMD = -0.19, 95 %CI = (-0.37, -0.002). The association of GPX1 gene Pro200Leu polymorphism with cancer risk might be influenced by confounders.

Results from the recent meta-analysis suggested a favorable effect of dietary calcium and vitamin D levels on breast cancer risk. However, the relationship of dietary calcium and vitamin D levels with breast cancer risk is unclear. Thus, the dose-response relationship was assessed by restricted cubic spline model and multivariate random-effect meta-regression. Results suggested that women might suffer from the lowest risk of breast cancer with dietary calcium intake of about 600 mg/day, dietary vitamin D intake of about 400 IU/day, and serum vitamin D levels of about 30 ng/ml.

RESUMO

A large number of studies have reported that tumor cells are often out of sync with the surrounding healthy tissue. Exploiting this misalignment may be a way to obtain a substantial gain in the therapeutic window. Specifically, based on reports to date, we will assess whether radiotherapy outcomes differ depending on the administration time. Collectively, 24 studies met the inclusion criteria, out of which 12 at least reported that radiation therapy is less toxic when administered at a particular time, probably because there is less collateral damage to healthy cells. However, discrepancies exist across studies and urge further investigation. Mechanistic studies elucidating the relationship between radiotherapy, circadian rhythms, and cell cycle, combined with either our "digital" or "biological" chronodata, would help oncologists successfully chronotype individual patients and strategize treatment plans accordingly.

OBJECTIVE: To establish a method for detection of M. tuberculosis by molecular beacon real-time PCR and evaluate its application in the clinical detection of M. tuberculosis. METHODS: The primers and molecular probe were designed to M. tuberculosis specific DNA sequence for Ag85B, and constitute a standards. 158 sputum specimens from patients with pulmonary tuberculosis were detected using molecular beacon real-time PCR, acid-fast stain and cultivation, and the detection rates of the three methods were compared. RESULTS: The lowest detection limit of the detected method is 2 genome copies per reaction, the linear standard curve was obtained between 2 x 10(2) and 2 x 10(8) DNA copies/mL. The method detected 5 strains of non-mycobacterium, 6 strains of non-tuberculosis mycobacterium and M. tuberculosis H37Rv, only M. tuberculosis H37Rv had 75 bp specific band. The positive rates of detection using molecular beacon real-time PCR, acid-fast stain and cultivation were 60.13%, 16.46%, 31.01% respectively, the positive rates of detection of real-time PCR was significantly higher than that of acid-fast stain and cultivation. CONCLUSION: Molecular beacon real-time PCR has high sensitivity and specificity in the detection of M. tuberculosis, which is clinical significance for the diagnosis of tuberculosis.

DNA damage to cochlear hair cells caused by 8-oxoguanine (8-oxoG) is essential for the development of noise-induced hearing loss (NIHL). Human 8-oxoG DNA glycosylase1 (hOGG1) is a key enzyme in the base excision repair (BER) pathway that eliminates 8-oxoG. Many epidemiological and functional studies have suggested that the hOGG1 Ser326Cys polymorphism (rs1052133) is associated with many diseases. The purpose of this investigation was to investigate whether the hOGG1 Ser326Cys polymorphism in the human BER pathway is associated with genetic susceptibility to NIHL in a Chinese population. This polymorphism was genotyped among 612 workers with NIHL and 615 workers with normal hearing. We found that individuals with the hOGG1 Cys/Cys genotype had a statistically significantly increased risk of NIHL compared with those who carried the hOGG1 Ser/Ser genotype (adjusted OR=1.59, 95% CI=1.13-2.25) and this increased risk was more pronounced among the workers in the 15- to 25- and >25-year noise exposure time, 85-92 dB(A) noise exposure level, ever smoking, and ever drinking groups, similar effects were also observed in a recessive model. In summary, our data suggested that the hOGG1 Cys/Cys genotype may be a genetic susceptibility marker for NIHL in the Chinese Han population.

A novel kind of high-impedance cable attenuator for measurement of high-voltage ns-range pulses is investigated in this paper. The input and output ports of the proposed attenuator were both high-impedance ports, and good pulse response characteristics of the proposed attenuator were obtained with pulse response time less than 1 ns. According to the requirement of measurement, two attenuators with lengths at 14 m and 0.7 m were developed with response time of 1 ns and 20 ns, and the attenuation coefficient of 96 and 33.5, respectively. The attenuator with the length of 14 m was used as a secondary-stage attenuator of a capacitive divider to measure the high-voltage pulses at several hundred ns range. The waveform was improved by the proposed attenuator in contrast to the result only measured by the same capacitive divider and a long cable line directly. The 0.7 m attenuator was also used as a secondary-stage attenuator of a standard resistant divider for an accurate measurement of high-voltage pulses at 100 ns range. The proposed cable attenuator can be used to substitute the traditional secondary-stage attenuators for the measurement of high-voltage pulses.

A new trigger program is proposed to realize the synchronization of high speed framing camera (HSFC) and intense electron-beam accelerator (IEBA). The trigger program which include light signal acquisition radiated from main switch of IEBA and signal processing circuit could provide a trigger signal with rise time of 17 ns and amplitude of about 5 V. First, the light signal was collected by an avalanche photodiode (APD) module, and the delay time between the output voltage of APD and load voltage of IEBA was tested, it was about 35 ns. Subsequently, the output voltage of APD was processed further by the signal processing circuit to obtain the trigger signal. At last, by combining the trigger program with an IEBA, the trigger program operated stably, and a delay time of 30 ns between the trigger signal of HSFC and output voltage of IEBA was obtained. Meanwhile, when surface flashover occurred at the high density polyethylene sample, the delay time between the trigger signal of HSFC and flashover current was up to 150 ns, which satisfied the need of synchronization of HSFC and IEBA. So the experiment results proved that the trigger program could compensate the time (called compensated time) of the trigger signal processing time and the inherent delay time of the HSFC.

A radial-current aqueous resistive solution load was applied to characterize a laser triggered transformer-type accelerator. The current direction in the dummy load is radial and is different from the traditional load in the axial. Therefore, this type of dummy load has smaller inductance and fast response characteristic. The load was designed to accommodate both the resistance requirement of accelerator and to allow optical access for the laser. Theoretical and numerical calculations of the load's inductance and capacitance are given. The equivalent circuit of the dummy load is calculated in theory and analyzed with a PSPICE code. The simulation results agree well with the theoretical analysis. At last, experiments of the dummy load applied to the high power spiral pulse forming line were performed; a quasisquare pulse voltage is obtained at the dummy load.
