RESUMO

Perithemis mooma, Kirby, 1889 and Perithemis icteroptera (Selys in Sagra, 1857) live in sympatry from southern Brazil to central Argentina. The taxonomy of the genus Perithemis Hagen, 1861 has been hampered by the use of characters that are highly variable or show slight differences among species. Our objective was to assess the efficiency of traditional morphometrics (TM) and geometric morphometrics (GM) to discriminate between these species using wing size and shape and vulvar lamina contour, and to analyze the presence of sexual dimorphism in wing size and shape in both species. The TM and landmark-GM methods were applied on the fore and hind wings, while the outline-based GM method was applied on the vulvar lamina. GM allowed species delimitation using shape variables of either wing. The wing and vulvar lamina shapes were confirmed to be good diagnostic characters to separate these species and appear to be promising tools for distinguishing among other species of this genus. Centroid size failed to achieve species separation. Both species exhibited sexual size dimorphism (SSD). In contrast to what would be expected for Perithemis whose males are strongly territorial, P. icteroptera and P. mooma showed female-biased SSD suggesting a common pattern in Perithemis.

Background and Purpose: We aimed to determine the prevalence and predictors of delayed neurological improvement (DNI) after complete endovascular reperfusion in anterior circulation acute ischemic stroke (AIS). Methods: Retrospective analysis of an online multicenter prospective reperfusion registry of patients with consecutive anterior circulation AIS treated with endovascular thrombectomy (EVT) from January 2018 to June 2019 in tertiary stroke centers of the NORDICTUS (NORD-Spain Network for Research and Innovation in ICTUS) network. We included patients with AIS with a proximal occlusion in whom a modified Thrombolysis in Cerebral Infarction 3 reperfusion pattern was obtained. DNI was defined if, despite absence of early neurological improvement during the first 24 hours, patients achieved functional independence on day 90. Clinical and radiological variables obtained before EVT were analyzed as potential predictors of DNI. Results: Of 1565 patients with consecutive AIS treated with EVT, 1381 had proximal anterior circulation occlusions, 803 (58%) of whom achieved a modified Thrombolysis in Cerebral Infarction 3. Of these, 628 patients fulfilled all selection criteria and were included in the study. Mean age was 73.8 years, 323 (51.4%) were female, and median baseline National Institutes of Health Stroke Scale was 16. Absence of early neurological improvement was observed in 142 (22.6%) patients; 32 of these (22.5%) achieved good long-term outcome and constitute the DNI group. Predictors of DNI in multivariable-adjusted logistic regression were male sex (odds ratio, 6.4 [95% CI, 2.1­22.3] P=0.002), lower pre-EVT National Institutes of Health Stroke Scale score (odds ratio, 1.4 [95% CI, 1.2­1.5], P<0.001), and intravenous thrombolysis (odds ratio, 9.1 [95% CI, 2.7­30.90], P<0.001). Conclusions: One-quarter of patients with anterior circulation AIS who do not clinically improve within the first 24 hours after complete cerebral endovascular recanalization will achieve long-term functional independence, regardless of the poor early clinical course. Male sex, lower initial clinical severity, and use of intravenous thrombolysis before EVT predicted this clinical pattern.

The expansion of silver nanoparticle (AgNP) applications in industry as antibacterial agents has generated an increment of their presence in the environment. Once there, their behavior is not clear because they can undergo different transformation processes that affect their transport, mobility, bioavailability, and toxicity. Therefore, the characterization and quantification of these emerging contaminants are important to understand their behavior and the toxicity effects that can be exerted on living beings. Single particle inductively coupled plasma mass spectrometry (SP-ICPMS) has demonstrated its ability to characterize and give quantitative information on AgNPs in aqueous samples. However, sometimes, the discrimination of the signal corresponding to AgNPs from the signal of dissolved species (Ag(I)) is a challenge. In the present contribution, it is shown that the presence of high amounts of Ag(I) hamper silver nanoparticle size and nanoparticle concentration determination in aqueous samples by SP-ICPMS. To facilitate signal discrimination of both chemical forms, the combination of cloud point extraction (CPE) with SP-ICPMS was studied. CPE experimental conditions to separate AgNPs from Ag(I) were assessed and adapted taking into account the characteristics of the SP-ICPMS technique. CPE and soil matrix effects on particle size were evaluated, showing that particle size was not modified after being in contact with soil matrix and after being separated by CPE. Additionally, frequently used calculation methods for SP-ICPMS data treatment were assessed. Finally, the potential of the developed methodology CPE-SP-ICPMS was evaluated in aqueous soil leachates contaminated with mixtures of AgNPs/Ag(I).

The production of silver nanoparticles (AgNPs) has increased tremendously during recent years due to their antibacterial and physicochemical properties. As a consequence, these particles are released inevitably into the environment, with soil being the main sink of disposal. Soil interactions have an effect on AgNP mobility, transport and bioavailability. To understand AgNP adsorption processes, lab-controlled kinetic studies were performed. Batch tests performed with five different Mediterranean agricultural soils showed that cation exchange capacity and electrical conductivity are the main parameters controlling the adsorption processes. The adsorption kinetics of different sized (40, 75, 100 and 200 nm) and coated (citrate, polyvinylpyrrolidone and polyethyleneglycol (PEG)) AgNPs indicated that these nanoparticle properties have also an effect on the adsorption processes. To assess the mobility and bioavailability of AgNPs and to determine if their form is maintained during adsorption/desorption processes, loaded soils were submitted to leaching tests three weeks after batch adsorption studies. The DIN 38414-S4 extraction method indicated that AgNPs were strongly retained on soils, and single-particle inductively coupled plasma mass spectrometry confirmed that silver particles maintained their nanoform, except for 100 nm PEG-AgNPs and 40 nm citrate-coated AgNPs. The DTPA (diethylenetriaminepentaacetic acid) leaching test was more effective in extracting silver, but there was no presence of AgNPs in almost all of these leachates.

The aim of this study was to measure pain levels in noncommunicative patients with severe trauma who required tracheal suctioning and mobilization and to determine the utility of the Behavioral Indicators of Pain Scale (ESCID) in these cases. The pain scores for the procedures were recorded on Days 1, 3, and 6 of the patients' stay in the intensive care unit. These assessments were performed at 3 moments: before, during, and after the application of the procedures. Because of the longitudinal character of the study, data were fitted into a multivariate model using the Generalized Estimating Equations method. The sample of 124 patients comprised 77.4% males and 22.6% females with an average age of 45.93 (SD = 16.43) years. A significant increase (p < .01) in the ESCID score was observed during the application of the procedures that produced similar pain levels. Kappa coefficient value obtained for interobserver agreement of ESCID scale scores during the application of care procedures at the intervals being evaluated was greater than 0.84, which should be interpreted as almost perfect. The ESCID scores increased during 2 care procedures that are frequently carried out in intensive care units and indicated that they produced similar pain levels.

A sensitive, multi-residue method using solid-phase extraction followed by liquid chromatography-tandem mass spectrometry (LC-MS/MS) was developed to determine a representative group of 35 analytes, including corrosion inhibitors, pesticides and pharmaceuticals such as analgesic and anti-inflammatory drugs, five iodinated contrast media, ß-blockers and some of their metabolites and transformation products in water samples. Few other methods are capable of determining such a broad range of contrast media together with other analytes. We studied the parameters affecting the extraction of the target analytes, including sorbent selection and extraction conditions, their chromatographic separation (mobile phase composition and column) and detection conditions using two ionisation sources: electrospray ionisation (ESI) and atmospheric pressure chemical ionisation (APCI). In order to correct matrix effects, a total of 20 surrogate/internal standards were used. ESI was found to have better sensitivity than APCI. Recoveries ranging from 79 to 134 % for tap water and 66 to 144 % for surface water were obtained. Intra-day precision, calculated as relative standard deviation, was below 34 % for tap water and below 21 % for surface water, groundwater and effluent wastewater. Method quantification limits (MQL) were in the low ng L(-1) range, except for the contrast agents iomeprol, amidotrizoic acid and iohexol (22, 25.5 and 17.9 ng L(-1), respectively). Finally, the method was applied to the analysis of 56 real water samples as part of the validation procedure. All of the compounds were detected in at least some of the water samples analysed. Communities in many parts of the world are unintentionally exposed to arsenic (As) and other toxic metals through ingestion of local drinking water and foods. The concentrations of individual toxic metals often exceed their guidelines in drinking water but the health risks associated with such multiple-metal exposures have yet to receive much attention. This study examines the co-occurrence of toxic metals in groundwater samples collected from As-rich areas of Nadia district, West Bengal, India. Arsenic in groundwater (range: 12-1064 µg L(-1); mean ± S.D: 329±294 µg L(-1)) was the most important contaminant with concentrations well above the WHO guideline of 10 µg L(-1). Another important toxic metal in the study area was manganese (Mn) with average concentration of 202±153 µg L(-1), range of 18-604 µg L(-1). The average concentrations (µg L(-1)) of other elements in groundwater were: Cr (5.6±5.9), Mo (3.5±2.1), Ni (8.3±8.7), Pb (2.9±1.3), Ba (119±43), Zn (56±40), Se (0.60±0.33), U (0.50±0.74). Saliva collected from the male participants of the area had mean concentrations of 6.3±7.0 µg As L(-1) (0.70-29 µg L(-1)), 5.4±5.5 µg Mn L(-1) (0.69-22 µg L(-1)), 2.6±3.1 µg Ni L(-1) (0.15-13 µg L(-1)), 0.78±1.0µg Cr L(-1) (

Haematophagous insects suffer big changes in water needs under different levels of starvation. Rhodnius prolixus is the most important haematophagous vector of Chagas disease in the north of South America and a model organism in insect physiology. Although there have been some studies on patterns of gas exchange and metabolic rates, there is little information regarding water loss in R. prolixus. We investigated whether there is any modulation of water loss and metabolic rate under different requirements for saving water. We measured simultaneously CO2 production, water emission and activity in individual insects in real time by open-flow respirometry at different temperatures (15, 25 and 35°C) and post-feeding days (0, 5, 13 and 29). We found: (1) a clear drop in metabolic rate between 5 and 13 days after feeding that cannot be explained by activity and (2) a decrease in water loss rate with increasing starvation level, by a decrease in cuticular water loss during the first 5 days after feeding and a drop in the respiratory component thereafter. We calculated the surface area of the insects and estimated cuticular permeability. In addition, we analysed the pattern of gas exchange; the change from a cyclic to a continuous pattern was affected by temperature and activity, but it was not affected by the level of starvation. Modulation of metabolic and water loss rates with temperature and starvation could help R. prolixus to be more flexible in tolerating different periods of starvation, which is adaptive in a changing environment with the uncertainty of finding a suitable host.

Saliva, an easily accessible biofluid, is validated as biomarker of arsenic (As) exposure in several villages of West Bengal, India. Pentavalent arsenic [As(V)] was found to be the predominant species in saliva, with the amount of inorganic As [As(V) and trivalent form, As(III)] being more than half of the total As in the samples. Significant association was found between total daily ingestion of As and As(V) (r = 0.59; p = 0.000), As(III) (r = 0.60; p = 0.000), dimethylarsinous acid (DMA(V)) (r = 0.40; p = 0.000), and monomethylarsonous acid (MMA(V)) (r = 0.44; p = 0.000), implying that these species have mainly been derived from the methylation of the inorganic As in the water that study participants drank and the food they ate. Analysis of confounding effects of age, sex, smoking, body mass index and the prevalence of skin lesion suggests that women and controls with no skin lesion had a higher capacity to methylate the ingested As compared to the rest of the population. Thus, our study demonstrates that As species in saliva can be an useful tool to predict the individual susceptibility where higher As exposure and a lower methylation capacity are implicated in the development of As-induced health effects.

Simple, precise, and low-cost methods for the simultaneous determination of phenolic endocrine disrupting compounds such as bisphenol A, trichlorophenol, pentachlorophenol, 4-nonylphenol, and 4-octylphenol in water samples were developed. The Direct, in situ derivatization methods are based on polydimethylsiloxane rod extraction followed by liquid desorption and chromatographic analysis by liquid chromatography and diode array detection. Several parameters affecting the extraction and desorption of the phenolic compounds and their acetylated derivates were studied, as well as the chromatographic and detection conditions. For the direct method, determination coefficients (r(2) ) > 0.990 and LODs in the 0.6-2 µg/L range were obtained for all compounds except bisphenol A (9.5 µg/L). With the derivatization-based method, based on in situ acetylation, lower limits of detection (0.3-0.9 µg/L) were obtained for all the compounds with r(2) > 0.988 and RSDs in the 2-9% range. The developed methods were applied to the analysis of spiked water samples obtaining recoveries of between 60.2 and 131.7% for the direct method, and of between 76.6 and 108.2% for the derivatization-based method. The results demonstrate the feasibility of using these two methods for determining bisphenol A, trichlorophenol, pentachlorophenol, 4-nonylphenol, and 4-octylphenol in water.

BACKGROUND: Persons infected with human immunodeficiency virus (HIV) have increased rates of coronary artery disease (CAD). The relative contribution of genetic background, HIV-related factors, antiretroviral medications, and traditional risk factors to CAD has not been fully evaluated in the setting of HIV infection. METHODS: In the general population, 23 common single-nucleotide polymorphisms (SNPs) were shown to be associated with CAD through genome-wide association analysis. Using the Metabochip, we genotyped 1875 HIV-positive, white individuals enrolled in 24 HIV observational studies, including 571 participants with a first CAD event during the 9-year study period and 1304 controls matched on sex and cohort. RESULTS: A genetic risk score built from 23 CAD-associated SNPs contributed significantly to CAD (P = 2.9 × 10(-4)). In the final multivariable model, participants with an unfavorable genetic background (top genetic score quartile) had a CAD odds ratio (OR) of 1.47 (95% confidence interval [CI], 1.05-2.04). This effect was similar to hypertension (OR = 1.36; 95% CI, 1.06-1.73), hypercholesterolemia (OR = 1.51; 95% CI, 1.16-1.96), diabetes (OR = 1.66; 95% CI, 1.10-2.49), ≥ 1 year lopinavir exposure (OR = 1.36; 95% CI, 1.06-1.73), and current abacavir treatment (OR = 1.56; 95% CI, 1.17-2.07). The effect of the genetic risk score was additive to the effect of nongenetic CAD risk factors, and did not change after adjustment for family history of CAD. CONCLUSIONS: In the setting of HIV infection, the effect of an unfavorable genetic background was similar to traditional CAD risk factors and certain adverse antiretroviral exposures. Genetic testing may provide prognostic information complementary to family history of CAD.

Saliva is a biological fluid that has not been used extensively as a biomonitoring tool in epidemiological studies. This study presents the arsenic (As) concentrations in saliva and urine samples collected from populations of West Bengal, India who had been previously exposed to high As levels in their drinking water. We found a significant (p < 0.05) association between the Log transformed Daily Ingestion of As (µg day(-1)) and the As concentration in saliva (r = 0.68). Additionally, As concentration of saliva and urine also had a significant positive correlation (r = 0.60, p < 0.05). Male participants, smokers, and cases of skin lesion were independently and significantly associated with an increase in salivary As. Thus our findings show that saliva is a useful biomarker of As exposure in the study population. The study also advocates that measurement of the forms of As in saliva may additionally provide insight into the internal dose and any individual differences in susceptibility to As exposure.

The Smicrideabrasiliana species group includes five species distributed in northeastern Argentina and Brazil: Smicridea (Rhyacophylax) brasiliana (Ulmer), S. (R.) weidneri Flint, S. (R.) vermiculata Flint, S. (R.) arobasis Flint, and S. (R.) nanda Flint. The original descriptions of these species and their placement in the brasiliana species group were mainly based on the morphology of the male genitalia. However, the fine structure of the internal sclerites of the phallus, which proved to be useful for species delimitation, was not analyzed at the time. In this contribution, we provide a detailed description of the male genitalia and the morphology of the head, and analyze the shape of the wings using geometric morphometrics. The analyzed species can be easily differentiated by the shape of the phallus, especially by the structure of the internal sclerites, the shape of the head in dorsal view, and the shape of the cephalic setose warts. Furthermore, the geometric morphometric approach allowed their separation through the wing shape. The preliminary analysis of these features suggests that the brasiliana species group is not natural but its monophyly should be further tested within the framework of a phylogenetic analysis of all the species of the subgenus Rhyacophylax.

The RNA polymerase (RNAP) omega factor (ω) forms a complex with the α2ßß' core of this enzyme in bacteria. We have characterized the rpoZ gene of Streptomyces coelicolor, which encodes a small protein (90 amino acids) identified as the omega factor. Deletion of the rpoZ gene resulted in strains with a slightly reduced growth rate, although they were still able to sporulate. The biosynthesis of actinorhodin and, particularly, that of undecylprodigiosin were drastically reduced in the ΔrpoZ strain, suggesting that expression of these secondary metabolite biosynthetic genes is dependent upon the presence of RpoZ in the RNAP complex. Complementation of the ΔrpoZ mutant with the wild-type rpoZ allele restored both phenotype and antibiotic production. Interestingly, the rpoZ gene contains a PHO box in its promoter region. DNA binding assays showed that the phosphate response regulator PhoP binds to such a region. Since luciferase reporter studies showed that rpoZ promoter activity was increased in a ΔphoP background, it can be concluded that rpoZ is controlled negatively by PhoP, thus connecting phosphate depletion regulation with antibiotic production and morphological differentiation in Streptomyces.

Silver nanoparticles (AgNPs) are frequently found in everyday products and, as a consequence, their release into the environment cannot be avoided. Once in aquatic systems, AgNPs interact with natural constituents and undergo different transformation processes. Therefore, it is important to characterize and quantify AgNPs in environmental waters in order to understand their behavior, their transformation, and their associated toxicological risks. However, the coexistence of ionic silver (Ag+) with AgNPs in aquatic systems is one of the greatest challenges for the determination of nanosilver. Ion-exchange resins can be used to separate Ag+ from AgNPs, taking advantage of the different charges of the species. In this work, Dowex 50W-X8 was used to separate Ag+ and AgNPs in order to easily determine AgNP concentrations using inductively coupled plasma optical emission spectroscopy. The separation methodology was successfully applied to river water samples with different ratios of Ag+ and AgNPs. However, the methodology is not useful for wastewater samples. The described methodology also demonstrated an improvement in the determination of the particle size of AgNPs present in river waters by single particle inductively coupled plasma mass spectrometry when a significant amount of Ag+ is also present.

BACKGROUND: The province of Formosa, Argentina, is endemic for leprosy. In the present paper, we assessed the trend (T, 2002-2016 time series) and the forecast for 2022 of new case detection rate (NCDR) and determined the spatial distribution of new cases detected (NCD) of leprosy. METHODOLOGY/PRINCIPAL FINDINGS: This is a descriptive observational study of 713 NCD of leprosy from provincial medical records between January 2002 and December 2016. The whole dataset from the provincial medical record was used to independently estimate the NCDR trends of the general population, age groups, sexes and Departments. This same database was used to estimate the NCDR forecast of the general population for 2022, applying a dynamic linear model with a local linear trend, using the MCMC algorithm. The NCDR was higher in men (p<0.05), increased with age (0.20, 8.17, 21.04, and 29.49 for the 0-14, 15-44, 45-64 and over 65-year-old age groups, respectively; p<0.05) and showed a downward trend (negative values) of estimated slopes for the whole province and each Department. Bermejo Department showed the highest (T:-1.02, 95%CI: [-1.42, -0.66]) and Patiño the lowest decreasing trend (T:-0.45, 95%CI: [-0.74, -0.11]). The NCDR trend for both sexes was similar (T:-0.55, 95%CI: [-0.64, -0.46]), and age groups showed a decreasing trend (S15-44:-103, S45-64:-81, S>65:-61, p<0.05), except for the 0-14 age group (S:-3, p>0.05), which showed no trend. Forecasts predicted that leprosy will not be eliminated by 2022 (3.64, 95%CI: [1.22, 10.25]). CONCLUSIONS/SIGNIFICANCE: Our results highlight the status of leprosy in Formosa and provide information to the provincial public health authorities on high-risk populations, stressing the importance of timely detection of new cases for further elimination of the disease in the province.

The broad use of silver nanoparticles (AgNPs) in daily life products enhances their possibilities to reach the environment. Therefore, it is important to understand the uptake, translocation and biotransformation in plants and the toxicological impacts derived from these biological processes. In this work, Lactuca sativa (lettuce) was exposed during 9 days to different coated (citrate, polyvinylpyrrolidone, polyethylene glycol) and sized (60, 75, 100 nm) AgNPs at different concentrations (1, 3, 5, 7, 10, 15 mg L-1). Total silver measurements in lettuce roots indicated that accumulation of AgNPs is influenced by size and concentration, but not by nanoparticle coating. On the other hand, nanosilver translocation to shoots was more pronounced for neutral charged and large sized NPs at higher NP concentrations. Single particle inductively coupled plasma mass spectrometry analysis, after an enzymatic digestion of lettuce tissues indicated the dissolution of some NPs. Ag K-edge X-ray absorption spectroscopy analysis corroborated the AgNPs dissolution due to the presence of less Ag-Ag bonds and appearance of Ag-O and/or Ag-S bonds in lettuce roots. Toxicological effects on lettuces were observed after exposure to nanosilver, especially for transpiration and stomatal conductance. These findings indicated that AgNPs can enter to edible plants, exerting toxicological effects on them.

Introduction: We aimed to evaluate if prior oral anticoagulation (OAC) and its type determines a greater risk of symptomatic hemorrhagic transformation in patients with acute ischemic stroke (AIS) subjected to mechanical thrombectomy. Materials and Methods: Consecutive patients with AIS included in the prospective reperfusion registry NORDICTUS, a network of tertiary stroke centers in Northern Spain, from January 2017 to December 2019 were included. Prior use of oral anticoagulants, baseline variables, and international normalized ratio (INR) on admission were recorded. Symptomatic intracranial hemorrhage (sICH) was the primary outcome measure. Secondary outcome was the relation between INR and sICH, and we evaluated mortality and functional outcome at 3 months by modified Rankin scale. We compared patients with and without previous OAC and also considered the type of oral anticoagulants. Results: About 1.455 AIS patients were included, of whom 274 (19%) were on OAC, 193 (70%) on vitamin K antagonists (VKA), and 81 (30%) on direct oral anticoagulants (DOACs). Anticoagulated patients were older and had more comorbidities. Eighty-one (5.6%) developed sICH, which was more frequent in the VKA group, but not in DOAC group. OAC with VKA emerged as a predictor of sICH in a multivariate regression model (OR, 1.89 [95% CI, 1.01-3.51], p = 0.04) and was not related to INR level on admission. Prior VKA use was not associated with worse outcome in the multivariate regression model nor with mortality at 3 months. Conclusions: OAC with VKA, but not with DOACs, was an independent predictor of sICH after mechanical thrombectomy. This excess risk was associated neither with INR value by the time thrombectomy was performed, nor with a worse functional outcome or mortality at 3 months.
