RESUMO

The Omicron BA.5 variant of SARS-CoV-2 is known for its high transmissibility and its capacity to evade immunity provided by vaccine protection against the (original) Wuhan strain. In our prior research, we successfully produced the receptor-binding domain (RBD) of the SARS-CoV-2 spike protein in an E. coli expression system. Extensive biophysical characterization indicated that, even without glycosylation, the RBD maintained native-like conformational and biophysical properties. The current study explores the immunogenicity and neutralization capacity of the E. coli-expressed Omicron BA.5 RBD using a mouse model. Administration of three doses of the RBD without any adjuvant elicited high titer antisera of up to 7.3 × 105 and up to 1.6 × 106 after a booster shot. Immunization with RBD notably enhanced the population of CD44+CD62L+ T cells, indicating the generation of T cell memory. The in vitro assays demonstrated the antisera's protective efficacy through significant inhibition of the interaction between SARS-CoV-2 and its human receptor, ACE2, and through potent neutralization of a pseudovirus. These findings underscore the potential of our E. coli-expressed RBD as a viable vaccine candidate against the Omicron variant of SARS-CoV-2.

The development of a dengue (DENV) vaccine remains challenging due to the heteroserotypic infection, which can result in a potentially deadly hemorrhagic fever or dengue shock syndrome, and only a tetravalent vaccine can overcome this issue. Here, we report the immunogenicity of DENV envelope protein domain 3 (ED3) from all four DENV serotypes (DENV1-4) in Swiss albino and BALB/c mice models. Firstly, we observed that despite having very similar sequences and structures, both the humoral and cellular immunogenicity of ED3s varied significantly, with strength ranging from DENV2 ED3 (2ED3)~3ED3 > 1ED3 > 4ED3, which was assessed through anti-ED3 IgG titers, and DENV1 ED3 (1ED3) > 2ED3~3ED3 > 4ED3 as determined by monitoring T-cell memory (CD44+CD62L+ T cells with IL-4 and IFN-γ expression). Secondly, anti-1ED3 sera cross-reacted with 2ED3 and 3ED3; anti-2ED3 and anti-3ED3 sera cross-reacted with each other, but anti-4ED3 was completely serotype-specific. The lack of reciprocity of anti-1ED3's cross-reaction was unanticipated. Such disparity in the ED3 responses and cross-reaction might underlie the appearance of hemorrhagic fever and dengue shock syndrome. Hence, the development of an ED3-based tetravalent subunit vaccine would require understanding the aforementioned disparities.

The concern over ensuing freshwater scarcity has forced the developing countries to delve for alternative water resources. In this study, we examined the potential of stagnant surface water bodies (SSWBs) as alternative freshwater resources in the densely populated Chittagong metropolitan area (CMPA) of Bangladesh--where there is an acute shortage of urban freshwater supply. Water samples were collected at 1-month intervals for a period of 1 year from 12 stations distributed over the whole metropolis. Samples were analyzed for pH, water temperature (WTemp), turbidity, electrical conductivity (EC), total dissolved solids, total solids, total hardness, dissolved oxygen (DO), chloride, orthophosphates, ammonia, total coliforms (TC), and trace metal (Cd, Cr, Cu, Pb, As, and Fe) concentrations. Based on these parameters, different types of water quality indices (WQIs) were deduced. WQIs showed most of CMPA-SSWBs as good or medium quality water bodies, while none were categorized as bad. Moreover, it was observed that the minimal water quality index (WQIm), computed using five parameters: WTemp, pH, DO, EC, and turbidity, gave a reliable estimate of water quality. The WQIm gave similar results in 72% of the cases compared with other WQIs that were based on larger set of parameters. Based on our finding, we suggest the wider use WQIm in developing countries for assessing health of SSWBs, as it will minimize the analytical cost to overcome the budget constraints involved in this kind of evaluations. It was observed that except turbidity and TC content, all other quality parameters fluctuated within the limit of the World Health Organization suggested standards for drinking water. From our findings, we concluded that if the turbidity and TC content of water from SSWBs in CMPA are taken care of, they will become good candidates as alternative water resources all round the year.

The cleansing efficiencies of laundry detergents depend on composition and variation of ingredients such as surfactants, phosphate, and co-builders. Among these ingredients, surfactants and phosphate are considered as hazardous materials. Knowledge on compositions and micellar behavior is very useful for understanding their cleansing efficiencies and environmental impact. With this view, composition, critical micelle concentration, and dissolved oxygen level in aqueous solution of some laundry detergents available in Bangladesh such as keya, Wheel Power White, Tibet, Surf Excel, and Chaka were determined. Surfactant and phosphate were found to be maximum in Surf Excel and Wheel Power White, respectively, while both of the ingredients were found to be minimum in Tibet. The critical micelle concentration decreased with increasing surfactant content. The amount of laundry detergents required for efficient cleansing was found to be minimum for Surf Excel and maximum for Chaka; however, cleansing cost was the highest for Surf Excel and the lowest for Tibet. The maximum amount of surfactants and phosphate was discharged by Surf Excel and Wheel Power White, respectively, while discharges of both of the ingredients were minimum for Tibet. The maximum decrease of dissolved oxygen level was caused by Surf Excel and the minimum by Tibet. Therefore, it can be concluded that Tibet is cost-effective and environment friendly, whereas Surf Excel and Wheel Power White are expensive and pose a threat to water environment.

Musa sapientum (M.sapientum) commonly known as 'banana' is widely used in Bangladeshi folk medicine for the treatment of various ailments including diarrhea. Hence, the present study was designed to investigate antidiarrheal, antioxidant and antibacterial potential of the methanolic extract of M.sapientum seed (MMSS). The extract was studied for antidiarrheal property using castor oil and magnesium sulfate induced diarrheal model and charcoal induced gastrointestinal motility test in mice. Total phenolic and flavonoids content, total antioxidant activity, scavenging of 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical, as well as nitric oxide (NO) and assessment of reducing power were used to evaluate antioxidant potential of MMSS. In addition, disc diffusion methods were used for antibacterial assay using various diarrheal induced bacterial strains. At the doses of 100 and 200 mg/kg body weight, the extract reduced the frequency and severity of diarrhea in test animals throughout the study period. At the same doses, the extracts significantly (p<0.001) delayed the intestinal transit of charcoal meal in test animals as compared to the control. In DPPH and NO scavenging method, MMSS showed good antioxidant potentiality in a dose dependent manner with the IC(50) value of 12.32±0.33 µg/ml and 18.96±1.01 µg/ml, respectively with a significant (p<0.001) good reducing power. The extract also displayed strong anti-bacterial effect against when tested against Escherichia coli, Shigella dysenteriae, and Pseudomonas aeruginosa. Altogether, these results suggest that the MMSS could be used as a potential antidiarrheal agent along with its antioxidant and antibacterial potentiality.

Gut-derived lymphocytes transiently migrate through the peripheral circulation before homing back to mucosal sites and can be detected using an ELISPOT-based antibody secreting cell (ASC) assay. Alternatively, transiently circulating lymphocytes may be cultured in vitro, and culture supernatants may be assayed for antigen-specific responses (antibody in lymphocyte supernatant [ALS] assay). The ALS assay has not been validated extensively in natural mucosal infection, nor has the ALS response been compared to the ASC assay and other cholera-specific immunological responses. Accordingly, we examined immune responses in 30 adult patients with acute cholera in Bangladesh, compared with 10 healthy controls, measuring ALS-immunoglobulin A (IgA), ASC-IgA, and serum and fecal IgA responses to two potent Vibrio cholerae immunogens, the nontoxic B subunit of cholera toxin (CtxB) and lipopolysaccharide (LPS) and a weaker V. cholerae immunogen, the mannose-sensitive hemagglutinin (MSHA). We found significant increases of anti-CtxB, anti-LPS, and anti-MSHA IgA in supernatants of lymphocytes cultured 7 days after onset of cholera using the ALS assay. We found that ALS and ASC responses correlated extremely well; both had comparable sensitivities as the vibriocidal responses, and both procedures were more sensitive than fecal IgA measurements. An advantage of the ALS assay for studying mucosal immune responses is the ability to freeze antibodies in supernatants for subsequent evaluation; like the ASC assay, the ALS assay can distinguish recent from remote mucosal infection, a distinction that may be difficult to make in endemic settings using other procedures.
