We investigated changes in concentrations of interleukin-1ß, interleukin-6, tumour necrosis factor-α and bradykinin in blood during passage through a cell salvage device and a leucocyte depletion filter, with or without application of subatmospheric pressure across the filter. Blood samples from 19 healthy women undergoing scheduled caesarean section showed concentrations of cytokines and bradykinin in blood filtered under gravity flow that were equal to or significantly lower than those of pre-operative venous blood samples. They were also significantly lower than that in postoperative orthopaedic shed blood, which is commonly reinfused after orthopaedic surgery. A minority of samples taken from blood that had been filtered using subatmospheric pressure showed raised interleukin-6 concentrations. We suggest that use of a leucocyte depletion filter for cell-salvaged blood with gravity flow is likely to be safe with regard to concentrations of cytokines and bradykinin. However, this may not hold true for the filter used with subatmospheric pressure. If transfusion of salvaged blood using a leucocyte depletion filter seems to induce hypotension, elevation of interleukin-6 should be suspected.

Attempts were made to examine the effect of paralytic shellfish poisoning toxins (PSP) on hepatic xenobiotic-metabolizing enzymes (XMEs) of tiger puffer (Takifugu rubripes). Two groups of nontoxic tiger fish were analyzed, and one group was fed with a PSP-containing diet (PSP group), and another with a PSP-free diet (control group). After 60 days of feeding, they were compared to each other mainly in terms of the activity of XMEs. Both groups did not differ from each other significantly in body weight gain, hepatosomatic index, and condition factor Hepatic level of cytochrome P450 was lower in PSP group than control group. NADPH-cytochrome P450 reductase, NADH-cytochrome b5 reductase, and ethoxyresorufin-O-deethylase (EROD) exhibited a reduced activity in PSP group than control group. Statistical analysis found that the activity or concentration of those enzymes correlated with the hepatic level of PSR with r2=0.497-0.611.

RESUMO

Vibrio sp. isolated from a xanthid crab, Atergatis floridus, was cultured, and tetrodotoxin (TTX) and anhydroTTX were indicated to be present in several fractions of the cell extract and the culture medium by reverse phase HPLC. The presence of the C9-base in alkaline hydrolyzates of these fractions was confirmed by GC-MS and UV spectrometry. These results showed the production of TTX and anhydroTTX in the Vibrio sp., thus indicating one of the origins of TTX in nature.

tetrodotoxin distribution in various tissues of the flatworm Planocera multitentaculata was examined. The oviduct was most toxic (2000-3240 MU/g), followed by digestive organs (900-1400 MU/g), oviduct-free genital organs (430-590 MU/g), etc. The mucus secreted also showed some lethal potency. On the other hand, the eggs laid, whether in laboratory aquarium or on coastal rocks, exhibited an extremely high potency up to 10,700 MU/g. It was suggested from these findings that flatworms possess tetrodotoxin as a defense or alarm substance against predators.

RESUMO

The paralytic toxin which occurs in a flatworm Planocera multitentaculata was partially purified by column chromatography using Bio-Gel P-2 and Bio-Rex 70 (H+ form). Thin-layer chromatographic, electrophoretic and gas chromatography-mass spectrometric analyses demonstrated that the flatworm toxin is tetrodotoxin.

Accumulation of tributyltin (TBT) was determined in liver of olive flounder exposed to TBT (3.65, 36.5, 365, 3,650, and 7,300 ng Sn/L) for 10 or 30 days, followed by 60 days depuration. Effect of TBT on hepatic cytochrome P450 content was also measured in liver of olive flounder. TBT was highly accumulated in liver of fish during the 10- to 30-day exposures, and hepatic cytochrome P450 content decreased with increasing TBT concentration. Hepatic cytochrome P450 contents were affected in olive flounder exposed to even environmentally relevant TBT concentrations, such as 3.65 ng Sn/L. In addition, the liver TBT levels demonstrated strong negative correlation to the hepatic cytochrome P450 content. The effects started to appear from 20 ng Sn/g dry weight of TBT in liver. Tributyltin concentrations and hepatic cytochrome P450 were also determined in feral fine-spotted flounder. The relationship between other organic pollutants known as cytochrome P450 inducers (e.g., polychlorinated biphenyls) as well as TBT and hepatic cytochrome P450 in the feral fish implied that TBT even at ppt level could impose antagonistic effects on hepatic cytochrome P450 induction.
