RESUMO

A novel avian alphaherpesvirus, preliminarily designated sphenicid alphaherpesvirus 1 (SpAHV-1), has been independently isolated from juvenile Humboldt and African penguins (Spheniscus humboldti and Spheniscus demersus) kept in German zoos suffering from diphtheroid oropharyngitis/laryngotracheitis and necrotizing enteritis (collectively designated as penguin-diphtheria-like disease). High-throughput sequencing was used to determine the complete genome sequences of the first two SpAHV-1 isolates. SpAHV-1 comprises a class D genome with a length of about 164 kbp, a G+C content of 45.6 mol% and encodes 86 predicted ORFs. Taxonomic association of SpAHV-1 to the genus Mardivirus was supported by gene content clustering and phylogenetic analysis of herpesvirus core genes. The presented results imply that SpAHV-1 could be the primary causative agent of penguin-diphtheria-like fatal diseases in banded penguins. These results may serve as a basis for the development of diagnostic tools in order to investigate similar cases of penguin diphtheria in wild and captive penguins.

Two Red-breasted Mergansers (Mergus serrator), one Hooded Merganser (Lophodytes cucullatus), and one Common Eider (Somateria mollissima) from a German zoological collection died of necrotizing typhlitis/typhlohepatitis within 2 years. Using a newly established chromogenic in situ hybridization assay, numerous intralesional trophozoites of Tetratrichomonas gallinarum could be detected in formalin-fixed and paraffin-embedded tissues from the caeca and livers of the affected birds. Partial sequencing of the 18S rRNA-gene revealed two unique nucleotide sequences very similar to T. gallinarum strains isolated from avian and human hosts. One turkey kept in the same zoological collection succumbed to histomonosis (blackhead disease) confirmed with chromogenic in situ hybridization at the time of the first duck fatalities. This turkey also harboured T. gallinarum trophozoites within necrotic cell debris in the caecal lumen, which might be epidemiologically related to the T. gallinarum infections in the ducks.

Two cases of acute babesiosis in captive reindeer (Rangifer tarandus) in two German zoos in 2009 and 2012 triggered this study to investigate the occurrence and species diversity of Babesia parasites infecting reindeer in different zoos and deer parks in Germany. Between June and December 2013, blood samples were taken from 123 clinically inapparent reindeer from 16 different facilities. Samples were tested for the presence of Babesia species DNA by conventional PCR and sequence analysis of part of the 18S rRNA gene. Also, Giemsa-stained smears of reindeer blood samples were examined for parasitaemia by light microscopy. The overall PCR-prevalence in blood samples was 23.6% (n=29). Comparison of sequenced amplicons with GenBank entries possibly revealed up to five different Babesia species: B. venatorum (n=19), B. capreoli (n=2) and B. capreoli-like (n=4), B. odocoilei-like (n=2) and B. divergens (n=1), while one sample turned out to be a Theileria sp. Out of the 16 facilities in the study, 12 housed at least one positive animal. In Giemsa-stained blood smears, intra-erythrocytic Babesia parasites were detected in samples of three reindeer from three locations. The high prevalence of Babesia infections implicates babesiosis to be a relevant infectious disease threat for captive reindeer in Germany. Consequently, reindeer with clinical signs compatible to those of acute babesiosis should either be tested for the presence of Babesia spp. DNA or blood smears should be examined for parasitaemia.

An undersized, 16-mo-old, captive wood bison (Bison bison athabascae) was observed with dysphagia and postprandial regurgitation. The animal died shortly after anesthesia was initiated for diagnostic purposes. At necropsy, a persistent right aortic arch and aberrant left subclavian artery were diagnosed. The vascular ring structure around the esophagus and the trachea caused a megaesophagus with complete esophageal obstipation and ulcerative esophagitis. The aberrant left subclavian artery did not significantly compromise the esophagus. Persistence of the right aortic arch is relatively rare in ruminants and has not previously been reported in a bison.

Infection of the Bursa of Fabricius with Cryptosporidium baileyi was diagnosed in a group of hand reared Red-breasted Merganser (Mergus serrator) ducklings during one breeding season in a German zoological garden.The birds had died in an emaciated and anaemic state after problems with spontaneous feeding. The bursae were infected with moderate to high numbers of cryptosporidia, which were associated with hyperplasia, degeneration and sloughing of the affected epithelial cells and mild heterophilic bursitis, lesions typically seen in bursal cryptosporidiosis in other avian species. In addition, lymphatic tissue was nearly absent, which was probably caused by chronic stress and malnutrition related to the rearing of these highly stress-sensitive birds in an artificial environment. Companion ducklings from stress-resistant, spontaneously feeding species from the zoological collection were used to calm the Mergansers, but may have introduced the cryptosporidia into the rearing boxes. Another possible source for the introduction of C. baileyi were adult mallards (Anas platyrhynchos), which were used to increase the hatching rate by a phase of natural breeding in the middle third of the incubation period of the Merganser eggs.
