RESUMO

Dual-frequency comb spectroscopy has emerged as a disruptive technique for measuring wide-spanning spectra with high resolution, yielding a particularly powerful technique for sensitive multi-component gas analysis. We present a spectrometer based on two electro-optical combs with subsequent conversion to the mid-infrared via tunable difference frequency generation, operating in the range from 3 to 4.7 µm. The repetition rate of the combs can be tuned from 250 to 500 MHz. For 500 MHz, the number of detected comb modes is 440 with a signal-to-noise ratio exceeding 105 in 1 s. The conversion preserves the coherence of the combs within 3 s measurement time. Concentration measurements of 5 ppm methane at 3.3 µm, 100 ppm nitrous oxide at 3.9 µm and a mixture of 15 ppm carbon monoxide and 5% carbon dioxide at 4.5 µm are demonstrated with a noise-equivalent absorption coefficient of 6.4(3) x 10-6 cm-1 Hz-1/2.

RESUMO

Nonlinear interferometers allow for mid-infrared spectroscopy with near-infrared detection using correlated photons. Previous implementations have demonstrated a spectral resolution limited by spectrally selective detection. In our work, we demonstrate mid-infrared transmission spectroscopy in a nonlinear interferometer using single-pixel near-infrared detection and Fourier-transform analysis. A sub-wavenumber spectral resolution allows for rotational-line-resolving spectroscopy of gaseous samples in a spectral bandwidth of over 700 cm-1. We use methane transmission spectra around 3.3 µm wavelength to characterize the spectral resolution, noise limitations and transmission accuracy of our device. The combination of nonlinear interferometry and Fourier-transform analysis paves the way towards performant and efficient mid-infrared spectroscopy with near-infrared detection.

Infrared attenuated total reflection (ATR) spectroscopy is a common laboratory technique for the analysis of highly absorbing liquids or solid samples. However, ATR spectroscopy is rarely found in industrial processes, where inline measurement, continuous operation, and minimal maintenance are important issues. Most materials for mid-infrared (MIR) spectroscopy and specifically for ATR elements do not have either high enough infrared transmission or sufficient mechanical and chemical stability to be exposed to process fluids, abrasive components, and aggressive cleaning agents. Sapphire is the usual choice for infrared wavelengths below 5 µm, and beyond that, only diamond is an established material. The use of diamond coatings on other ATR materials such as silicon will increase the stability of the sensor and will enable the use of larger ATR elements with increased sensitivity at lower cost for wavelengths above 5 µm. Theoretical and experimental investigations of the dependence of ATR absorbances on the incidence angle and thickness of nanocrystalline diamond (NCD) coatings on silicon were performed. By optimizing the coating thickness, a substantial amplification of the ATR absorbance can be achieved compared to an uncoated silicon element. Using a compact FTIR instrument, ATR spectra of water, acetonitrile, and propylene carbonate were measured with planar ATR elements made of coated and uncoated silicon. Compared to sapphire, the long wavelength extreme of the spectral range is extended to approximately 8 µm. With effectively nine ATR reflections, the sensitivity is expected to exceed the performance of typical diamond tip probes.

Nonlinear interferometers allow spectroscopy in the mid-infrared range by detecting correlated visible light, for which non-cooled detectors with higher specific detectivity and lower dark count rates are available. We present a new approach for the registration of spectral information, which combines a nonlinear interferometer using non-degenerate spontaneous parametric down-conversion (SPDC) with a Fourier-transform spectroscopy concept. In order to increase the spectral coverage, we use broadband non-collinear SPDC in periodically poled LiNbO3. Without the need for spectrally selective detection, continuous spectra with a spectral bandwidth of more than 100 cm-1 are achieved. We demonstrate transmission spectra of a polypropylene sample measured with 6 cm-1 resolution in the spectral range between 3.2 µm to 3.9 µm.

Mid-infrared spectroscopy is an essential analytical method in science and industry. Unlike in the near-infrared range, grating spectrometers for the mid-infrared are rarely employed, mostly due to the limited availability and performance of suitable line array detectors. In this work, continuous-wave nonlinear-optical upconversion is used to enable mid-infrared spectroscopy. A broad spectral window between 3.7 and 4.7 µm is upconverted to 825 - 867 nm for detection on a silicon-camera-based near-infrared grating spectrometer with a high sensitivity down to sub-picowatt of input power. A theoretical model is presented that accurately describes the upconversion process and the total system behavior. Spectroscopic flame emission measurements demonstrate the applicability towards the analysis of highly dynamic processes.

Pulsed nonlinear-optical upconversion is used for mid-infrared signal detection. A setup for both mid-infrared generation and upconversion based on a single pump laser enables sensitive light detection and is utilized for gas spectroscopy. With the demonstrated pulsed setup, quantum efficiencies above 80 % for the upconversion of a Gaussian beam signal and 25 % for the upconversion of backscattered radiation are achieved and in agreement with theoretical predictions. Combined with efficient background suppression due to spectral and temporal gating, this results in highly sensitive detection of the infrared signals. As a demonstration of application, the presented system is used for methane sensing in an open path geometry, highlighting the potential for stand-off leak detection with a concentration resolution better than 1.5 ppm·m.

We present an all solid state, transportable photoacoustic spectrometer for highly sensitive mid-infrared trace gas detection. A complete spectral coverage between 3.1 and 3.9 microm is obtained using a PPLN-based continuous-wave optical parametric oscillator pumped by a Nd:YAG laser at 1064 nm. A low threshold is achieved by resonating the pump, and spectral agility by employing a dual-cavity setup. An etalon suppresses mode-hops. Active signal cavity stabilization yields a frequency stability better than +/- 30 MHz over 45 minutes. Output idler power is 2 x 100 mW. The frequency tuning qualities of the OPO allow reliable scan over gas absorption structures. A detection limit of 110 ppt for ethane is achieved.

A sensitive photoacoustic absorption spectrometer for highly transparent solids has been built and tested. As the light source an optical parametrical oscillator pumped by a nanosecond pulse laser with 10 Hz repetition rate is employed, covering the complete wavelength range from 407 to 2600 nm. A second-harmonic-generation unit extends the range of accessible wavelengths down to 212 nm. A lead-zirconate-titanate piezo transducer, directly coupled to the sample, detects the photoacoustically generated sound waves. Absorption spectra of lithium triborate, lithium niobate, and alpha barium borate crystals with absorption coefficients down to 10(-5) cm(-1) are presented.

Caterpillar-induced ethylene emissions play an important role in plant-herbivore interactions. The ethylene burst that ensues after attack exceeds wound-induced ethylene emissions, but the mechanisms responsible remain unknown. Adding larval oral secretions (OS) to wounds mimics this ethylene burst. We demonstrate that fatty acid-amino acid conjugates are the responsible elicitors in Manduca sexta OS, and identify genes that are important in OS-elicited ethylene biosynthesis and perception in the larvae's host, Nicotiana attenuata, by examining the consequences of gene silencing on OS-elicited ethylene emissions, as quantified by photo-acoustic spectroscopy. OS elicitation increased transcript accumulation of ACC synthase (ACS), virus-induced gene silencing of ACS halved the OS-elicited ethylene release, and ACC supplementation to ACS-silenced plants restored ethylene emissions, demonstrating that ACS activity limits the rate of release. Silencing three wound- or OS-elicited ACC oxidase (ACO) genes with an ACO consensus fragment abolished the OS-elicited ethylene release. Virus-induced gene silencing of each ACO individually revealed that only NaACO2a and NaACO3 regulate the OS-elicited ethylene release. Transforming plants with various etr1-1 constructs rendered them differentially 'deaf' to ethylene, and dramatically increased the OS-elicited ethylene burst, largely without regulating the transcripts of biosynthetic genes. The volume of the OS-elicited ethylene 'scream' was proportional to the plant's deafness, as determined by 1-MCP treatments. We conclude that the OS-elicited ethylene burst is tuned by a tag-team of transcriptional responses and ethylene perception. Ethylene signaling is shown to be essential in regulating two traits that are important in the N. attenuata-M. sexta interaction: OS-induced nicotine levels and floral longevity.

The method of laser induced reaction is used to obtain high-resolution IR spectra of H2D+ and D2H+ in collision with n-H2 at a nominal temperature of 17 K. For this purpose three cw-laser systems have been coupled to a 22-pole ion trap apparatus, two commercial diode laser systems in the ranges of 6100-6600 cm(-1) and 6760-7300 cm(-1), respectively, and a high-power optical parametric oscillator tunable in the range of 2600-3200 cm(-1). In total, 27 new overtone and combination transitions have been detected for H2D+ and D2H+, as well as a weak line in the nu1 vibrational band of H2D+ (2(20)<--1(01)) at 3164.118 cm(-1). The line positions are compared to high accuracy ab initio calculations, showing small but mode-dependent differences, being largest for three vibrational quanta in the nu2 symmetric bending of H2D+. Within the experimental accuracy, the relative values of the ab initio predicted Einstein B coefficients are confirmed.

A recent study demonstrated that gravi-incompetent coleoptile-less seedlings of rye exhibit gravi-competence after exogenous application of ethylene. Treatments and conditions which induce and interfere with this phenomenon were analysed in more detail. Aminocyclopropane-1-carboxylic acid (ACC) as a precursor of ethylene has similar gravicompetence-inducing effects and also appropriate conditions of light, which strongly enhances ethylene synthesis. Both effects can be inhibited by the ethylene-perception blocking agent methylcyclopropene (MCP) or inhibitors of ethylene synthesis such as aminovinylglycine (AVG), indicating that light exerts its gravicompetence-generating effect via induced/enhanced ethylene synthesis. Gain in gravicompetence is accompanied by the induced/enhanced occurrence of calreticulin and lipoxygenase as detected by 2D-gels and Q-TOFF-analyses. Previously gravicompetent, light-grown coleoptile-less seedlings are characterized by gravi-incompetent growth during subsequent horizontal gravistimulation when perception of ethylene is inhibited by MCP. The results demonstrate that continuous perception of ethylene is an indispensable step, permanently required for the regulation of gravitropic growth in germinating primary shoots of rye, either within the process of graviperception and/or of the transduction of the gravi-signal.

More volatile organic carbon is lost from plants as isoprene than any other molecule. This flux of carbon to the atmosphere affects atmospheric chemistry and can serve as a substrate for ozone production in polluted air. Isoprene synthesis may help leaves cope with heatflecks and active oxygen species. Isoprene synthase, an enzyme related to monoterpene synthases, converts dimethylallyl diphosphate derived from the methylerythritol 4-phosphate pathway to isoprene. We used dideuterated deoxyxylulose (DOX-d(2)) to study the regulation of the isoprene biosynthetic pathway. Exogenous DOX-d(2) displaced endogenous sources of carbon for isoprene synthesis without increasing the overall rate of isoprene synthesis. However, at higher concentrations, DOX-d(2) completely suppressed isoprene synthesis from endogenous sources and increased the overall rate of isoprene synthesis. We interpret these results to indicate strong feedback control of deoxyxylulose-5-phosphate synthase. We related the emission of labeled isoprene to the concentration of labeled dimethylallyl diphosphate in order to estimate the in situ K(m) of isoprene synthase. The results confirm that isoprene synthase has a K(m) 10- to 100-fold higher for its allylic diphosphate substrate than related monoterpene synthases for geranyl diphosphate.

The orientation growth of coleoptiles of dark-grown seedlings of rye (Secale cereale L. cv. Marder II), when grown under various conditions, was analysed with respect to the gravivector ('gravitropic setpoint angle', GSA). Coleoptiles growing through moist vermiculite attain and maintain a GSA with an average of about 180 degrees, i.e. a vertical orientation. Seedlings growing uncovered either on the surface of vermiculite or positionally fixed on filter paper attain and maintain a GSA of 140-150 degrees (i.e. deviating from the vertical by an average of 30-40 degrees ). Changing the position of the embryo relative to the horizontally fixed seed kernel or of the angle of the seed with respect to gravity during germination (+/-40 degrees relative to the horizontal) had no significant effect on the subsequent GSA of both covered and uncovered seedlings. The GSA of uncovered coleoptiles could be restored close to 180 degrees by treatment of the seedlings with ethylene, either applied via ethephon or 1-aminocyclopropane-1-carboxylic acid (ACC) as well as by fruit-released ethylene. The results are discussed with respect to the mechanism of the regulation of gravitropic growth of grass seedlings.

Spectroscopic detection of ethane in the 3-microm wavelength region was performed by means of a cw optical parametric oscillator and cavity leak-out. We achieved a minimum detectable absorption coefficient of 1.6 x 10(-10) cm 1/square root of Hz, corresponding to an ethane detection limit of 6 parts per trillion/square root of Hz. For 3-min integration time the detection limit was 0.5 parts per trillion. The levels are to our knowledge the best demonstrated so far. These frequency-tuning capabilities facilitated multigas analysis with simultaneous monitoring of ethane, methane, and water vapor in human breath.

Herbivore attacks induce leaves to emit a specific blend of volatiles. Here we show that exposure to Tetranychus urticae-induced volatiles, as well as T. urticae infestation and artificial wounding, activates the transcription of the genes involved in the biosynthesis of ethylene [S-adenosylmethionine (SAM) synthetase and 1-aminocyclopropane-1-carboxylic acid oxidase] and a gene involved in the biosynthesis of polyamines from SAM (SAM decarboxylase) in lima bean leaves. Moreover, exposure of leaves to any one of the seven major chemical components of T. urticae-induced volatiles also induces expression of these genes. Furthermore, we found that, when lima bean plants were exposed to T. urticae-induced volatiles, they emitted ethylene. Lima bean plants infested by T. urticae and artificially wounded plants also emitted ethylene. Endogenous polyamine levels were not increased in the exposed leaves or the infested leaves, suggesting that polyamine production from SAM was only slightly promoted at the metabolic levels present in the leaves. We found that jasmonate (JA) accumulated in leaves exposed to T. urticae-induced volatiles, and that both JA and salicylate (SA) accumulated in leaves infested by T. urticae. These findings, as well as results of pharmacological analyses, suggest that, in leaves exposed to T. urticae-induced volatiles, ethylene biosynthesis might be regulated by pathways involving JA and the ethylene positive feedback loop. They also suggest that ethylene biosynthesis might be regulated by signaling pathways involving JA, SA and ethylene in T. urticae-infested leaves.
