RESUMO

Fat has high energy density and is considered one of the primary energy sources for dogs, however, increasing fat level in dry dog food has been challenging due to the lubrication and limitation of the coating system. The objective was to determine the effect of whole soybeans (WSB) on nutrient digestibility, stool quality, and palatability by dogs. The corn gluten meal, chicken fat, and brewers rice were replaced by WSB at 10, 20, and 30% (WSB10, WSB20, and WSB30, respectively) in the base diet (WSB0). Twelve beagles were randomly assigned. The digestibility trial was duplicated 4 × 4 Latin square design where dogs were allowed a 9-d adaptation followed by a 5-d total fecal collection for each period. Least-square means were analyzed with a single degree of freedom contrasts and significance at α = 0.05. Palatability was determined with a 2-bowl test by 20 beagles for 2 d with each WSB diet compared to the WSB0. First choice preference between two diets and total food consumption were recorded. Individual intake ratios (IR) were calculated (intake of each diet/total intake) for each dog. First choice (FC) was analyzed by a Chi-square probability, and the diet consumption was compared by a Wilcoxon signed rank test and a 2-way analysis of variance. Fecal moisture, output, and defecation frequency increased linearly (P < 0.05) as WSB increased. Apparent total tract digestibility of dry matter, organic matter, crude protein, fat, and gross energy decreased linearly (P < 0.05) as dogs fed the increased level of WSB. The fresh fecal pH in dogs decreased linearly (P < 0.05) as WSB content increased. The acetate, propionate, and the total short-chain fatty acid concentration increased linearly (P < 0.05) while the total branched-chain fatty acid concentration decreased linearly (P < 0.05) as WSB increased. Dogs had greater (P < 0.05) FC for WSB diets than WSB0, but there was no difference among treatments for diet consumption and IR. In conclusion, additional thermal processing before extrusion may improve nutrient digestibility of WSB. The stool quality and palatability were not affected, and fermentation in hindgut increased by WSB by dogs.

Soybean use has been low in pet foods, even though they are an excellent source of protein, polyunsaturated fatty acids, and gut fermentable fibers. The purpose of this evaluation was to conduct a systematic review of the public literature to explore how soybeans have been researched for pet food applications since 2000 and to provide strengths, weaknesses, opportunities, and threats for soybeans in the pet food industry. The review covered a total of 44 articles related to soybean ingredients and their potential value in the pet food arena. The articles were categorized by their research contents and narratively summarized to demonstrate useful information to both the pet and soybean industries. When soybean-based products have been adequately processed to reduce the antinutritive factors, they are comparable to processed animal proteins in nutritional value, palatability, and functionality in pet food processing. We conclude that various food processing technologies and the versatility of soybean ingredients allow soybean to have considerable inclusion potential in pet foods. More research on dietary soybean ingredients regarding pet food processing, fermentation benefits on health, and consumer acceptance will be needed to understand soybean's position in the future pet food industry.

OBJECTIVES: This experiment was conducted to evaluate the effect of coat length and faecal hair removal on measured nutrient digestibility in longhair and shorthair cats. METHODS: A total of 14 adult domestic cats, with a mean ± SD body weight of 4.5 ± 1.21 kg and a mean ± SD age of 3.3 ± 1.38 years, were used for a nutrient digestibility trial. The nutrient digestibility of cats was measured by hair-included faeces and hair-removed faeces. Food was provided twice daily (09:00 h and 16:00 h) and water was provided ad libitum. Cats were adapted to a steel cage and diet for 16 days before a 10 day collection period. During the collection period, food offered, food refused and faecal output were measured daily and used for digestibility analysis. RESULTS: The digestibility of dry matter, crude protein and neutral detergent fibre (NDF) of longhair cats was underestimated by 8% ( P = 0.02), 9% ( P = 0.04) and 14% ( P = 0.04), respectively. In shorthair cats, the digestibility of dry matter, crude protein and crude ash increased by 4% ( P = 0.01), 5% ( P = 0.02) and 15% ( P <0.01), respectively, with hair-removed faeces. The nutrient digestibility between longhair and shorthair cats showed no difference when hair-included faeces were used. However, when using hair-removed faeces, the digestibility of NDF and amino acids in longhair cats were about 4-8% higher than in shorthair cats ( P <0.05). CONCLUSIONS AND RELEVANCE: This study showed that the nutrient digestibility values were affected by whether the hair in faeces was removed or not. Removing hair from faeces is a more precise method with which to evaluate the apparent nutrient digestibility of domestic cats.

OBJECTIVES: This experiment was conducted to evaluate the behavioural time budget for grooming and grooming patterns for shorthair and longhair cats, and to assess the effect of grooming behaviour on apparent digestibility of nutrients in domestic cats ( Felis catus) by comparing hair-included faeces and hair-removed faeces. METHODS: A total of 10 adult domestic cats, with a mean ± SD body weight of 4.3 ± 0.89 kg and a mean ± SD age of 3.5 ± 1.38 years, were used for behavioural observation. Cats were housed individually in stainless steel cages at the animal hospital. The cats' behaviour was recorded on a webcam videotaping system for one 24 h period; then, faecal samples were collected and analysed to measure apparent digestibility. RESULTS: There was no significant difference between longhair and shorthair cats in behavioural time budget for grooming and grooming patterns. The apparent digestibility of dry matter, crude protein, crude ash, acid detergent fibre (ADF) and neutral detergent fibre (NDF) of hair-removed faeces was significantly higher than that of hair-included faeces: about 6% ( P <0.01), 7% ( P <0.01), 14% ( P <0.01), 12% ( P = 0.01) and 10% ( P <0.01), respectively. CONCLUSIONS AND RELEVANCE: There was no difference in grooming patterns between longhair cats and shorthair cats. Also, the digestibility of dry matter, crude protein, crude ash, ADF and NDF has been underestimated by approximately 6%, 7%, 14%, 12%, and 10%, respectively, when they have been calculated using the conventional digestibility method for domestic cats.

Several signals have been suggested in maintaining skin barrier homeostasis, but epidermal calcium ions are currently thought to be a main signaling factor. It is not clear, however, exactly how an intracellular calcium level decreases in response to the loss of an extracellular calcium gradient. In this study, we investigated the effects of several broad-type and isozyme-specific protein kinase C (PKC) inhibitors on epidermal permeability barrier recovery. Topical application of chelerythrine chloride, a broad-type PKC inhibitor, and rottlerin, a PKCdelta-specific inhibitor, significantly impeded the barrier recovery rate at 3 and 6 hours after barrier disruption. A significant decrease in the number and secretion of lamellar bodies was also observed at the inhibitor-treated site. Calcium ion-capture cytochemistry showed that the epidermal calcium gradient was rapidly reformed in inhibitor-treated skin, though recovery of the corresponding barrier function was not observed. In cultured keratinocytes treated with either inhibitor, increased intracellular calcium did not return to the baseline concentration after extracellular calcium decreased. These results suggest that PKC inhibitors, especially a PKCdelta-specific inhibitor, delay barrier recovery by affecting the intracellular calcium concentration after a loss of the extracellular calcium gradient. Furthermore, PKCdelta is important in controlling a decrease in intracellular calcium concentration.

RESUMO

Mitochondrial outer membrane permeabilization (MOMP) is the ultimate step in dozens of lethal apoptotic signal transduction pathways which converge on mitochondria. One of the representative systems proposed to be responsible for the MOMP is the mitochondrial permeability transition pore (MPTP). Although the molecular composition of the MPTP is not clearly understood, the MPTP attracts much interest as a promising target for resolving two conundrums regarding cancer treatment: tumor selectivity and resistance to treatment. The regulation of the MPTP is closely related to metabolic reprogramming in cancer cells including mitochondrial alterations. Restoration of deregulated apoptotic machinery in cancer cells by tumor-specific modulation of the MPTP could therefore be a promising anti-cancer strategy. Currently, a number of MPTP-targeting agents are under pre-clinical and clinical studies. Here, we reviewed the structure and regulation of the MPTP as well as the current status of the development of promising MPTP-targeting drugs.

We analyzed insulin-like growth factor receptor I (IGF-IR) and insulin growth factor binding protein-3 (IGFBP-3) expression with respect to overall survival and relapse-free survival (RFS) in 460 patients with primary invasive breast cancer using immunohistochemistry. IGF-IR expression had a significant positive correlation with positive estrogen receptor (ER), positive progesterone receptor (PR) and Bcl-2 expression. Univariate analysis showed significantly better overall survival (p=0.000) and RFS (p=0.004), in the IGF-IR-positive group. Multivariate analysis showed a significant reduction in relative risk for overall survival (p=0.019, HR=0.221, 95% CI=0.062-0.780) and RFS (p=0.026, HR=0.462, 95% CI=0.234-0.913) in the IGF-IR-positive group. IGF-IR expression in primary breast cancer is an independent favorable prognostic factor. IGFBP-3 expression in breast cancer is associated with poor outcome.

A peroxisome proliferator-activated receptor gamma (PPARgamma) ligand, 15-deoxy-Delta(12,14)-prostaglandin J(2) (15d-PGJ(2)), has been reported to possess anti-inflammatory activity in activated monocytes/macrophages. In this study, we investigated the effect of 15d-PGJ(2) on the lipopolysaccharide (LPS)-induced expression of chemokine mRNAs, especially macrophage inhibitory protein (MIP)-2 (CXCL2), in mouse peritoneal macrophages. The inhibitory actions of the natural PPARgamma ligands, 15d-PGJ(2) and prostaglandin A1 (PGA1), on the expression of RANTES (regulated upon activation, normal T expressed and secreted; CCL5), MIP-1beta (CCL4), MIP-1alpha (CCL3), IFN-gamma-inducible protein 10 kilodaltons (IP-10; CXCL10) and monocyte chemoattractant protein-1 (MCP-1; CCL2) mRNA in LPS-treated cells were stronger than those of the synthetic PPARgamma ligands troglitazone and ciglitazone. However, 15d-PGJ(2) enhanced the expression of LPS-induced MIP-2 (CXCL2) mRNA. A specific PPARgamma antagonist (GW9662) had no effect on the inhibitory action of 15d-PGJ(2) and PGA1 in LPS-induced chemokine mRNA expression and on the synergistic action of 15d-PGJ(2) in LPS-induced MIP-2 (CXCL2) expression. Moreover, LPS itself reduced the expression of PPARgamma. Although the synergistic effect of 15d-PGJ(2) on LPS-induced MIP-2 (CXCL2) mRNA expression was remarkable, the production of MIP-2 (CXCL2) in cells treated with 15d-PGJ(2) and LPS did not increase compared to the production in cells treated with LPS alone. The synergistic action of 15d-PGJ(2) on LPS-induced MIP-2 (CXCL2) mRNA expression was dependent on the activation of nuclear factor-kappaB (NF-kappaB), and 15d-PGJ(2) increased the phosphorylation of p38 and stress-activated protein kinase/c-Jun N-terminal kinase (SAPK/JNK) in cells stimulated with LPS. These results suggest that the synergistic effect of 15d-PGJ(2) on LPS-induced MIP-2 (CXCL2) expression is PPARgamma-independent, and is mediated by the p38 and SAPK/JNK pathway in mitogen-activated protein kinase signaling pathways, which activates NF-kappaB. Our data may give more insights into the different mechanisms contrary to the anti-inflammatory effect of 15d-PGJ(2) on the expression of chemokine genes.

15-Deoxy-Delta(12,14)-prostaglandin J(2) (15d-PGJ(2)) was initially identified as a high affinity natural ligand for the peroxisome proliferator-activated receptor (PPAR)-gamma. Recent studies have shown that it has a potent anti-inflammatory effect by attenuating the expression of proinflammatory mediators in activated macrophages, mainly through the inhibition of nuclear factor (NF)-kappaB-dependent transcription of inflammatory genes. In this study, we investigated the synergistic effect of 15d-PGJ(2) on the expression of LPS-induced chemokine KC mRNA in mouse peritoneal macrophages. The time course of KC mRNA expression in cells stimulated with 15d-PGJ(2) plus LPS simultaneously (15d-PGJ(2)/LPS) showed similar patterns to the cells treated with LPS alone, and 15d-PGJ(2) had no effect on the stability of LPS-induced KC mRNA expression. Although NF-kappaB activity in cells treated with LPS was augmented by 15d-PGJ(2), pyrrolidone dithiocarbamate (PDTC) did not block the synergistic effect of 15d-PGJ(2) on LPS-induced KC mRNA expression. However, the synergistic effect of 15d-PGJ(2) was markedly inhibited when the macrophages were treated with a inhibitor of the mitogen-activated protein kinase (MAPK) signalling pathway, 2'-amino-3'-methoxyflavine (PD98059). Therefore, the mechanism of synergistic action of 15d-PGJ(2) on the expression of LPS-induced KC mRNA in mouse peritoneal macrophages is possibly related to the MAPK signalling pathway, not to NF-kappaB activation. These data may contribute to unravelling some of the different mechanisms contrary to the anti-inflammatory effect of 15d-PGJ(2).
