RESUMO

The study area receives an average of 2840.0 mm of rainfall within four months every year. A portion of the rainwater is flown to the sea as surface water, and the other part is percolated into the bottom as groundwater. In coastal aquifers, the groundwater is transported to the sea due to a hydraulic gradient, and it contains a significant quantity of dissolved materials and nutrients. SGD processes impact the ocean productivity, mangrove and coral growth, local acidification and many. To isolate the SGD on the central west coast of India, different data was referred. The GWL concerning MSL contributed significantly to demarcating the SGD zones by considering the positive (>0 m) and negative (<0 m) values of GWL concerning above MSL. Thermal images for SST of pre-monsoon and post-monsoon periods of 2020 exhibit cooler surrounded by warmer, which might be the SGD buffering zones in the off-central west coast of India. By considering the results from GWL and SST, 8 SGD beach sites were identified for the further particularized study. The water samples were collected in March 2022, and analyzed using standard procedures and instruments. Fresh and mixing (recirculated) zones have been isolated by piper, hydrochemical facies evolution, and Ca2++Mg2+/K++Na + Vs log Cl- ionic ratio plots. The aquifer water chemical elements are converting possibly due to ionic exchange processes. The decrease in salinity and conductivity observed in the pore water just below the seawater might be due to the influence of freshwater inputs, helping to isolate the fresh SGD and recirculated SGD zones in the study area. Among 8 sites, 3 were found to be fresh SGD sites and 5 were noticed to be mixing/recirculated SGD sites. Most of these Beaches are bounded by hills, which helps to lead the SGD along the central west coast of India.

Citrus is an economically important fruit crop, belongs to family Rutaceae, cultivated commercially in over 130 countries, which holds a leading profitable position in the international market. The most important citrus varieties are mandarins, oranges, lemons, sweet limes, grapefruits and pomelos. Citrus yellow vein clearing virus (CYVCV) is an important graft transmissible plant pathogen known to reduce productivity of citrus fruits due to its predominant association and widespread occurrence. Requirement of fast, reliable, efficient & economical CYVCV indexing assay is a prerequisite for production of healthy planting material. Currently, nucleic acid isolation and thermal cycler-based assay available for CYVCV indexing is a cumbersome lab intensive method. The present study was undertaken to develop and validate reverse transcription-recombinase polymerase amplification (RT-RPA) assay requiring no tedious RNA isolation, separate cDNA synthesis and costlier instrument like thermo-cycler. Optimized RT-RPA assay was able to amplify CYVCV up to 10-7 dilution (equivalent to 0.1 pg/µl) with the prepared templates of both RNA and crude saps and showed higher sensitivity in detection of CYVCV infection in field samples as compared to the conventional RT-PCR. Developed RT-RPA assay showed high specificity without any cross-reaction with other citrus pathogens (Indian citrus ringspot virus, citrus yellow mosaic virus, citrus tristeza virus, citrus exocortis viroid and huanglongbing). RT-RPA using crude leaf sap as template is quite simple, robust, highly sensitive, time and cost effective; therefore, it can be used in resource constrained laboratories as screening tool, for field surveys and on-site testing programs in farms, nurseries and biosecurity. Present study, first time reports the development, optimization and validation of crude sap-based RT-RPA assay for the detection of CYVCV infection in citrus plants namely; Kinnow mandarin, Mosambi and Grape fruit.

RESUMO

Advancements in brain imaging techniques have emerged as a significant tool in detecting Alzheimer's disease (AD) progression. The complicated cascade of AD progression can be detected using radio imaging, especially with Positron emission tomography (PET). The review focus on recently introduced investigational PET tracers targeting neurofibrillary tau aggregates found typically in AD. Herein, we also address the use of different PET tracers and the clinical implementation of established and newer generation tracers. This review also intends to discuss the importance of several PET radiotracers and challenges in PET imaging.

Aß cascade hypothesis being considered most evident event in AD pathology and even today it holds good. Dysregulation of catalytic events of Aß regulating enzymes can possibly cause faulty Aß trafficking; inequity of Aß formation and clearance resulting in misfolded protein accumulation, neurodegeneration and cognitive impairment. Many novel approaches have been made on this pathway to discover new molecules, unfortunately couldn't reach the terminal phases of clinical trials. Over decades, studies have been more focused on enzyme chemistry and explored the relationship between structural features and catalytic function of Aß regulating enzymes. However, the modulations of catalytic mechanisms of those enzymes have not been imposed so far to reduce the Aß load. Hence, in this review, we have critically detailed the knowledge of basic structural dynamics and possible catalytic modulations of enzymes responsible for Aß formation and clearance that will impart new perspectives in drug discovery process.

OBJECTIVE: Fabrication and analyses of mucoadhesive patches made from chitosan oligosaccharide for the purpose of oromucosal drug delivery. SIGNIFICANCE: The mucosal epithelium in the oral cavity, consisting of buccal and sublingual epithelium, has gained significant attention in the last decade as an alternative anatomical site for systemic drug delivery that could potentially minimize the challenges of solid oral dosage and parenteral delivery. In this study, we have fabricated and tested drug-loaded chitosan oligosaccharide-based patches for the oromucosal drug delivery. METHODS: The chitosan oligosaccharide (with and without alginate) based patches were fabricated using the conventional solvent casting method and were analyzed for their swelling capacity, hydrophilicity, anti-cancer activity, in vitro drug release, and in vivo drug release activity. The in-house developed artificial saliva was used for the swelling study. RESULTS: Alginate-containing patches showed lesser swelling ability compared to the bare chitosan oligosaccharide-based patches. The former was also found to be more hydrophobic compared to the latter one. Both the unloaded patches restricted the growth of epithelial cancer cells indicating their anti-cancer behavior. In vitro drug release indicated a super case II release pattern while in vivo study demonstrated the release of drug from the patch into the plasma indicating the purpose of the fabricated patch. CONCLUSIONS: The chitosan oligosaccharide-based mucoadhesive hydrogel patch fabricated in this study can be highly suitable for possible translational purposes.

Background: Ethyl glucuronide (EtG) is a phase II metabolite of ethanol and is an upcoming biomarker for recent alcohol intake. Monitoring of alcohol intake in case of alcohol-dependent syndrome is very useful for early intervention and preventing harmful effects. EtG has also been identified as a very useful marker in differentiating antemortem ingestion of alcohol from postmortem production of alcohol. This study was undertaken with an objective of developing a sensitive and specific method for determination of EtG in urine. Methods: Triple quadruple Liquid Chromatography (LC)-Mass Spectrometry (MS) with Electrospray Ionization (ESI) negative mode has been used for developing the multiple reaction monitoring method by using the Polaris 3 C 18-Ether analytical column. A simple sample preparation method was adopted using the Bond Elute Plexa PAX SPE cartridge. The developed method was also tested on actual urine samples from 15 individuals after consumption of 60 and 90 ml of whiskey at different time intervals. Results: A simple method was developed for determination of EtG in urine, with a sensitivity of 100 ppb and a recovery of 75%. Validation of the method on urine samples revealed that EtG could be detected for up to 18 h in individuals who ingested 60 ml of whiskey and up to 24 h in those who ingested 90 ml of whiskey. Conclusion: The simple method was developed for determination of EtG in urine and validated on actual urine samples. This method can now be used in aircraft accident investigation to differentiate postmortem production of alcohol, and the method is also a very useful tool to monitor Alcohol dependent Syndrome (ADS) cases.

BACKGROUND: Time and cost constraints lead to majority of clinical laboratories deviating away from an ideal practice of checking for antinuclear antibodies (ANAs) by indirect immunofluorescence (IIF) at multiple dilutions. Usage of screening dilution of 1:40 recommended by most manufacturers of commercial ANA kits results in numerous false positive-tests and misdiagnosis of connective tissue disorders (CTDs). We sought to study the ideal screening dilution for ANA by IIF for a diagnosis of ANA-related CTDs. METHODS: Serum samples of patients with ANA-related conditions (n = 233) and healthy controls (n = 154) were evaluated by IIF using Immuno Concepts Hep-2000 ® ANA kits at dilutions from 1:40 to 1:640. Accuracy for diagnosis of CTDs for each serum dilution was assessed by receiver operating curve (ROC) analysis. RESULTS: Antinuclear antibodies (ANA) positivity was observed in 19.5%, 10.4%, 4.55%, 0.65%, and 0% of healthy controls at dilutions of 1:40, 1:80, 1:160, 1:320, and 1:640, respectively. ANA positivity at 1:40 dilution was observed among 26.4% cases with mimics of CTDs. Prevalence of ANA positivity in ANA-related CTDs was 97.3%, 96.4%, 89.3%, 83.9%, and 71.4% at dilutions of 1:40, 1:80, 1:160, 1:320, and 1:640, respectively. ROC analysis revealed best test performance for distinction between healthy and ANA-related CTD populations at a serum dilution of 1 in 80. CONCLUSIONS: Antinuclear antibodies (ANA) positivity at low titers (1:40) is highly prevalent in healthy population (19.5%) as well as amongst mimics of CTD (26.4%). Our study suggests a higher screening dilution of 1:80 for ANA by IIF for diagnosis of CTD maybe better. Combination of 1:80 and 1:160 dilutions provides optimum sensitivity and specificity for diagnosis of ANA-related disorders.

BACKGROUND: Social media has become an integral part in the life of every individual in the 21st century. Social media addiction in the younger age group is a major problem. The objective of this study was to find a correlation between academic performance and social media use. METHODS: This was a cross-sectional questionnaire-based study conducted in a medical school over a period of 3 months (Nov 2018-Jan 2019), where 400 medical undergraduates who use social media participated in the study. Data collected from the questionnaire included the academic performance in terms of university examination marks, the duration of social media use per day and the social media addiction score. Data correlation was done using the Pearson's correlation factor. RESULTS: 41.5% of students used social media for upto 3 h per day. Whatsapp (98.25%) and Youtube (91.75%) were the most commonly used social media applications. 73.5% used social media to read health-related news, 71.5% used it to complete assignments and more than 50% used it for seminar preparation, test preparation and research-related purposes. Academic performance of female students was better than male students. There was a significantly higher use of social media among academically low-performing medical students compared with high-performing medical students. There was a weak negative correlation between academic performance and social media usage and a strong positive correlation between social media usage and the social media addiction score. CONCLUSIONS: Social media has a negative impact on the academic performance of 21st-century undergraduate medical students.

The temperature-dependent spin-reorientation transition (SRT) and spin interaction mechanism of bulk TmFeO3 were studied by the electron paramagnetic resonance (EPR) method. The combined experimental results of magnetic curves and EPR spectra confirmed that there is an antiferromagnetic transition at 85 K with a reentering ferromagnetic state due to the spin-reorientation behavior. In the high-temperature region of T > 90 K, there are three distinct resonance peaks in the EPR spectrum, which indicates the presence of multiple magnetic phases (canted antiferromagnetic, weak ferromagnetic, and paramagnetic phases). In the low-temperature region (T < 85 K), the temperature dependence of the EPR linewidth, effective g-factor, and intensity can be used to infer a strong spin-lattice correlation. Different magnetic interactions such as Fe3+-Fe3+, Fe3+-Tm3+, and Tm3+-Tm3+ lead to a paramagnetic-canted antiferromagnetic phase at T > 85 K, with SRT between 85-65 K and ferromagnetic interaction at the lower temperature, respectively. Above 90 K, we find that the spin relaxation mechanism is determined by the mixture of spin-spin and spin-lattice interactions. Below 85 K, the transverse relaxation rate increases with the decrease in temperature, which is consistent with the weakening of the fluctuating internal field in this temperature region. This EPR detection provides a new method to clarify the strong spin coupling in antiferromagnetic materials.

We report the physical properties of Eu-doped bulk TmFeO3 through X-ray diffraction, magnetic susceptibility (χ), Raman scattering and X-ray absorption spectroscopy (XAS) study, which shows a similar orthorhombic structure with the Pbnm space group as TmFeO3. Magnetic measurement on Eu-doped TmFeO3 provides evidence for spin reorientations of Fe3+. Further, the Raman spectra of Eu3+ doped TmFeO3 show significant changes in Raman modes as a function of temperature, which are evidence for strong spin-lattice interaction. From the XAS spectra, the L-edge of Fe provides information on the valence state of Fe, whereas the K-edge of oxygen shows that the compound has a strong influence on the hybridization of the O(2p) state with the 3d states of Fe.

In this paper, we report on the facile synthesis of graphitic carbon nitride (g-C3N4)-tin oxide (SnO2) nanohybrid as an efficient photocatalyst prepared via sol-gel method. SnO2 nanoparticles are pointcontacted with g-C3N4. The results of physio-chemical characterizations such as SEM-EDAX, XRD, BET, FT-IR and UV-DRS spectra reveal the successful formation and integration of nanohybrid. The photocatalytic activity has been studied by using methylene-blue as a model dye for degradation. It has been observed that the pseudo-first order rate constant was increased up to 1.78 times compared with pure SnO2. The enhanced photocatalytic activity was ascribed from the inhibition of electron-hole recombination where g-C3N4 nanosheets acts as an electron receiver from SnO2 via point contact. This mechanism is further verified via photoluminescence spectra. Our results prominently show new insights and potential applications of g-C3N4-SnO2 nanohybrids in the waste water treatment and environmental remediation sectors.

Amitraz is an acaricide and insecticide used to treat ticks, which infest domestic animals in developing countries. Because of its widespread use, it is one of the common poisons unintentionally consumed by infants and children when left unsupervised. A 3-year-old boy was brought with unintentional consumption of Amitraz. On examination, he was found to be progressively drowsy, with an irregular pulse, bradycardia, and hypotension. He was treated with atropine, intravenous fluids, and dopamine infusion; hemodynamic stability was achieved within 36 hours after ingestion. Amitraz is an unusual but deadly poison unintentionally consumed by children. It can be suspected in the setting of rural households in developing countries having pets. There is no antidote available, and treatment is mainly supportive.

BACKGROUND: Repeated blood transfusions is the mainstay of treatment for beta thalassemia major patients. Multiple blood transfusions lead to significant iron overload in these patients. Iron overload causes liberation of oxygen free radicals and peroxidative lipid injury. This study has been designed to study whether thalassemics suffer from oxidative injury. It also aims to study the quantum of oxidative injury. METHODS: It is a cross sectional study using cases and controls. Thirty thalassemic patients receiving multiple blood transfusions were included in this study and thirty healthy age and sex matched controls were recruited for the study. Serum ferritin levels, malondialdehyde, nitric oxide levels were estimated. RESULTS: Levels of all the three parameters were significantly increased (p < 0.05) in the cases compared to controls. Mean levels of all three parameters were correlated with serum ferritin levels and number of blood transfusions in increasing order. All the parameters showed fair degree of correlation (r ≥ 0.25, p ≤ 0.05). CONCLUSION: Thalassemic patients receiving multiple blood transfusion suffer from iron overload which results in increased oxidative stress.

Motivated from our previous studies on the upconversion properties of BaMgF4:Yb3+,Tb3+ phosphor, here we investigated the upconversion properties of BaMgF4:Yb3+,Er3+ phosphor. We demonstrate a two-way versatile approach for the fine-tuning of emission from green to the red region, by varying the dopant concentration and adjusting the pulse width of an infrared laser. The mechanism involved in tuning the emission color by laser power and pulse width variation was illustrated in detail. The temperature dependent upconversion spectra were studied by analyzing the fluorescence intensity ratio of the thermally coupled levels. The maximum sensitivity obtained is 83.29 × 10-4 K-1 at 583 K, which is much higher than the temperature sensitivity reported for other fluoride based materials. Moreover, the influence of the excitation power density on the ability of the phosphor for temperature sensing was also investigated. We obtained a maximum (∼415 K) temperature detection at 2563 mW laser power. The obtained results illustrate the potential use of BaMgF4:Yb3+,Er3+ phosphor in an optical thermometer due to its highly sensitive temperature detection ability.

BACKGROUND: RNA sequencing (RNA-seq) has become an indispensable tool to identify disease associated transcriptional profiles and determine the molecular underpinnings of diseases. However, the broad adaptation of the methodology into the clinic is still hampered by inconsistent results from different RNA-seq protocols and involves further evaluation of its analytical reliability using patient samples. Here, we applied two commonly used RNA-seq library preparation protocols to samples from acute leukemia patients to understand how poly-A-tailed mRNA selection (PA) and ribo-depletion (RD) based RNA-seq library preparation protocols affect gene fusion detection, variant calling, and gene expression profiling. RESULTS: Overall, the protocols produced similar results with consistent outcomes. Nevertheless, the PA protocol was more efficient in quantifying expression of leukemia marker genes and showed better performance in the expression-based classification of leukemia. Independent qRT-PCR experiments verified that the PA protocol better represented total RNA compared to the RD protocol. In contrast, the RD protocol detected a higher number of non-coding RNA features and had better alignment efficiency. The RD protocol also recovered more known fusion-gene events, although variability was seen in fusion gene predictions. CONCLUSION: The overall findings provide a framework for the use of RNA-seq in a precision medicine setting with limited number of samples and suggest that selection of the library preparation protocol should be based on the objectives of the analysis.

Flavonoids represent an important bioactive component in plants. Accumulation of flavonoids often occurs in plants subjected to abiotic stresses, including the adaptation of plants to the environment and in overcoming their stress conditions. This fact makes their analysis and determination an attractive field in food science since they can give interesting information on the quality and safety of foods. In this study, we discuss reports on plants flavonoids biosynthesis against abiotic stresses and advances in analytical capillary electrophoresis used for their identification and quantification in plants.

In this work we have comprehensively studied the up-conversion (UC) properties of BaMgF4:Yb3+,Tb3+ phosphor for the first time. BaMgF4:Yb3+,Tb3+ phosphors were prepared by a simple and low cost precipitation method. To determine the influence of dopant concentration on luminescence properties, the corresponding UC luminescence spectra of BaMgF4:Yb3+,Tb3+ phosphors were studied under NIR excitation. Emission spectra under NIR excitation reveal the vital role of Tb3+ concentration in spectral tuning from the blue to green region. The UC decay curves were also studied to explore the possible energy transfer (ET) mechanisms between Yb3+ and Tb3+. The results reported here are expected to provide an approach for better understanding ET mechanisms in many Yb3+/Tb3+ codoped UC phosphors. This study will be helpful in applications where precisely defined optical transitions is an essential criterion.

Sulfur is easy to be incorporated into ZnO nanoparticles by the solution-combustion method. Herein, the magnetic and adsorption properties of a series of ZnO1-xSx (x = 0, 0.05, 0.1, 0.15, and 0.2) nanoparticles were systematically investigated. The X-ray diffraction patterns show that the as-prepared ZnO1-xSx nanoparticles have the hexagonal wurtzite structure of ZnO with a low sulfur content that gradually transforms into the zinc blende structure of ZnS when the x value is greater than 0.1. PL spectra show several bands due to different transitions, which have been explained by the recombination of free excitons or defect-induced transitions. The introduction of sulfur not only modifies the bandgap of ZnO, but also impacts the concentration of Zn vacancies. The as-prepared ZnO shows weak room-temperature ferromagnetism, and the incorporation of sulfur improves the ferromagnetism owing to the increased concentration of Zn vacancies, which may be stabilized by the doped sulfur ions. The adsorption capability of ZnO1-xSx nanoparticles has been significantly improved, and the process can be well described by the pseudo-first-order kinetic model and the Freundlich isotherm model. The mechanism has been confirmed to be due to the active sulfate groups existing in zinc oxysulfide nanoparticles.

Thalassemia is a congenital hemolytic disease which is treated by repeated blood transfusion. Chronic iron overload is currently considered to be the primary cause of mortality in ß-thalassemia, mainly due to the induction of left-sided cardiac failure. Iron overload results from a number of mechanisms associated with the disease itself. In addition to chronic iron overload thalassemic patients are more prone for procoagulant status which in turn lead to clinical thrombotic events. The hypercoagulable state in thalassemia is due to multiple elements, a combination of which is often the drive behind a clinical thromboembolic events. PAI-1 study was done in thalassemia major patients receiving multiple blood transfusion as a marker for procoagulant status. Total of 30 thalassemic patients on repeated blood transfusion was included in the study and total of 30 healthy age and sex matched controls were included in the study. It was also found that there was significant differences between cases and controls. The mean level of PAI 1 in controls was 3047 ± 414 pg/ml, the value in cases was 3683 ± 358 pg/ml. The level was significantly increased (p < 0.05) in the cases compared to controls. PAI-1 levels were also compared with the total number of blood transfusion which correlates well.
