RESUMO

Numerous efforts have been undertaken to mitigate the Debye screening effect of FET biosensors for achieving higher sensitivity. There are few reports that show sub-femtomolar detection of biomolecules by FET mechanisms but they either suffer from significant background noise or lack robust control. In this aspect, deformed/crumpled graphene has been recently deployed by other researchers for various biomolecule detection like DNA, COVID-19 spike proteins and immunity markers like IL-6 at sub-femtomolar levels. However, the chemical vapor deposition (CVD) approach for graphene fabrication suffers from various surface contamination while the transfer process induces structural defects. In this paper, an alternative fabrication methodology has been proposed where glass substrate has been initially texturized by wet chemical etching through the sacrificial layer of synthesized silver nanoparticles, obtained by annealing of thin silver films leading to solid state dewetting. Graphene has been subsequently deposited by thermal reduction technique from graphene oxide solution. The resulting deformed graphene structure exhibits higher sensor response towards glial fibrillary acidic protein (GFAP) detection with respect to flat graphene owing to the combined effect of reduced Debye screening and higher surface area for receptor immobilization. Additionally, another interesting aspect of the reported work lies in the biomolecule capture by dielectrophoretic (DEP) transport on the crests of the convex surfaces of graphene in a coplanar gated topology structure which has resulted in 10 aM and 28 aM detection limits of GFAP in buffer and undiluted plasma respectively, within 15 min of application of analyte. The detection limit in buffer is almost four decades lower than that documented for GFAP using biosensors which is is expected to pave way for advancing graphene FET based sensors towards ultrasensitive point-of-care diagnosis of GFAP, a biomarker for traumatic brain injury.

The tumor microenvironment (TME) is like the Referee of a soccer match who has constant eyes on the activity of all players, such as cells, acellular stroma components, and signaling molecules for the successful completion of the game, that is, tumorigenesis. The cooperation among all the "team members" determines the characteristics of tumor, such as the hypoxic and acidic niche, stiffer mechanical properties, or dilated vasculature. Like in soccer, each TME is different. This heterogeneity makes it challenging to fully understand the intratumor dynamics, particularly among different tumor subpopulations and their role in therapeutic response or resistance. Further, during metastasis, tumor cells can disseminate to a secondary organ, a critical event responsible for approximately 90% of the deaths in cancer patients. The recapitulation of the rapidly changing TME in the laboratory is crucial to improve patients' prognosis for unraveling key mechanisms of tumorigenesis and developing better drugs. Hence, in this chapter, we provide an overview of the characteristic features of the TME and how to model them, followed by a brief description of the limitations of existing in vitro platforms. Finally, various attempts at simulating the TME using microfluidic platforms are highlighted. The chapter ends with the concerns that need to be addressed for designing more realistic and predictive tumor-on-a-chip platforms.

OBJECTIVE: To describe and compare the characteristics of ectopic pregnancies (EPs) in the year prior to vs during the coronavirus disease 2019 (COVID-19) pandemic. METHODS: This was a retrospective analysis of women diagnosed with an EP on transvaginal sonography conducted at a center in London, UK, providing early-pregnancy assessment, between 1 January 2019 and 31 December 2020. Women were identified via the Astraia ultrasound reporting system using coded and non-coded outcomes of EP or pregnancy outside the uterine cavity. Data related to predefined outcomes were collected using Astraia and Cerner electronic reporting systems. Main outcome measures included clinical, ultrasound and biochemical features of EP, in addition to reported complications and management. RESULTS: There were 22 683 consultations over the 2-year period. Following consultation, a similar number and proportion of EPs were diagnosed in 2019 (141/12 657 (1%)) and 2020 (134/10 026 (1%)). Both cohorts were comparable in age, ethnicity, weight and method of conception. Gestational age at the first transvaginal sonography scan and at diagnosis were similar, and no difference in location, size or morphology of EP was found between the two cohorts. Serum human chorionic gonadotropin (hCG) levels at the time of EP diagnosis were higher in 2020 than in 2019 (1005 IU/L vs 665 IU/L; P = 0.03). The proportions of women according to type of final EP management were similar, but the rate of failed first-line management was higher during vs before the pandemic (16% vs 6%; P = 0.01). The rates of blood detected in the pelvis (hemoperitoneum) on ultrasound (23% vs 26%; P = 0.58) and of ruptured EP confirmed surgically (9% vs 3%; P = 0.07) were similar in 2019 vs 2020. CONCLUSIONS: No difference was observed in the location, size, morphology or gestational age at the first ultrasound examination or at diagnosis of EP between women diagnosed before vs during the COVID-19 pandemic. Complication rates and final management strategy were also unchanged. However, hCG levels and the failure rate of first-line conservative management measures were higher during the pandemic. Our findings suggest that women continued to access appropriate care for EP during the COVID-19 pandemic, with no evidence of diagnostic delay or an increase in adverse outcome in our population. © 2021 International Society of Ultrasound in Obstetrics and Gynecology.

AIMS: In the age where bacterial resistance to conventional antibiotics is increasing at an alarming rate, the use of the traditional plant, herb extracts or other bioactive constituents is gradually becoming popular as an anti-virulence agent to treat pathogenic diseases. Carvacrol, a major essential oil fraction of Oregano, possesses a wide range of bioactivities. Therefore, we aimed to study the effect of sub-inhibitory concentrations of carvacrol on major virulence traits of Vibrio cholerae. METHODS AND RESULTS: We have used in vitro as well as ex vivo models to access the anti-pathogenic role of carvacrol. We found that the sub-inhibitory concentration of carvacrol significantly repressed bacterial mucin penetrating ability. Carvacrol also reduced the adherence and fluid accumulation in the rabbit ileal loop model. Reduction in virulence is associated with the downregulated expression of tcpA, ctxB, hlyA and toxT. Furthermore, carvacrol inhibits flagellar synthesis by downregulating the expression of flrC and most of the class III genes. CONCLUSIONS: Carvacrol exhibited anti-virulence activity against V. cholerae, which involved many events including the inhibition of mucin penetration, adhesion, reduced expression of virulence-associated genes culminating in reduced fluid accumulation. SIGNIFICANCE AND IMPACT OF THE STUDY: These findings indicate that carvacrol possesses inhibitory activity against V. cholerae pathogenesis and might be considered as a potential bio-active therapeutic alternative to combat cholera.

We report gapless quantum spin liquid behavior in the layered triangular Sr_{3}CuSb_{2}O_{9} system. X-ray diffraction shows superlattice reflections associated with atomic site ordering into triangular Cu planes well separated by Sb planes. Muon spin relaxation measurements show that the S=1/2 moments at the magnetically active Cu sites remain dynamic down to 65 mK in spite of a large antiferromagnetic exchange scale evidenced by a large Curie-Weiss temperature θ_{CW}≃-143 K as extracted from the bulk susceptibility. Specific heat measurements also show no sign of long-range order down to 0.35 K. The magnetic specific heat (C_{m}) below 5 K reveals a C_{m}=γT+αT^{2} behavior. The significant T^{2} contribution to the magnetic specific heat invites a phenomenology in terms of the so-called Dirac spinon excitations with a linear dispersion. From the low-T specific heat data, we estimate the dominant exchange scale to be ∼36 K using a Dirac spin liquid ansatz which is not far from the values inferred from microscopic density functional theory calculations (∼45 K) as well as high-temperature susceptibility analysis (∼70 K). The linear specific heat coefficient is about 18 mJ/mol K^{2} which is somewhat larger than for typical Fermi liquids.

We present a combination of thermodynamic and dynamic experimental signatures of a disorder driven dynamic cooperative paramagnet in a 50% site diluted triangular lattice spin-1/2 system: Y_{2}CuTiO_{6}. Magnetic ordering and spin freezing are absent down to 50 mK, far below the Curie-Weiss scale (-θ_{CW}) of ∼134 K. We observe scaling collapses of the magnetic field and temperature dependent magnetic heat capacity and magnetization data, respectively, in conformity with expectations from the random singlet physics. Our experiments establish the suppression of any freezing scale, if at all present, by more than 3 orders of magnitude, opening a plethora of interesting possibilities such as disorder stabilized long range quantum entangled ground states.

OBJECTIVE: The current study was carried out to examine the response of different levels of fat and protein in calf starter on nutrient utilisation, nitrogen metabolism, weight gain, blood parameters and immunity level in pre-ruminant calves. METHODS: Twenty four calves (5 days old) were divided into six groups in a 2x3 factorial design, with two levels of fat (10% and 14 %) and three levels of protein (18%, 21% and 24%). The calves were kept in individual pens for 120 days and fed with whole milk (1/10th of body weight) and calf starter ad-libitum. Daily dry matter intake (DMI) was recorded; whereas body weight was taken on fortnightly basis to calculate average daily gain (ADG). During the growth trial blood samples were collected at 30 days interval to estimate blood glucose, albumin, total protein, total leucocyte count, total immunoglobulin and IgG levels. A metabolic trial of seven days was carried out to find out the digestibility of different nutrients. RESULTS: The DMI was reduced (p<0.05) with higher fat and protein levels whereas feed conversion efficiency (FCE) was improved (p<0.05) with higher protein level. Different levels of fat and protein in calf ration did not affect average daily gain in calves.The dry matter (DM), organic matter (OM) and crude protein (CP) digestibility were significantly (p<0.01) higher with increased level of protein. The nitrogen retention was also significantly higher (p<0.05) at 24 % protein level, similarly the total immunoglobulin was significantly (p< 0.05) high in higher protein fed groups, showed better immunity. CONCLUSION: The present finding suggested that 10 % fat and 18 % protein level of calf starter could be used in Sahiwal calves for optimum performance in terms of weight gain and immunity.

OBJECTIVES: The contribution of depression to mortality in adults with and without HIV infection is unclear. We hypothesized that depression increases mortality risk and that this association is stronger among those with HIV infection. METHODS: Veterans Aging Cohort Study (VACS) data were analysed from the first clinic visit on or after 1 April 2003 (baseline) to 30 September 2015. Depression definitions were: (1) major depressive disorder defined using International Classification of Diseases, Ninth Revision (ICD-9) codes; (2) depressive symptoms defined as Patient Health Questionnaire (PHQ)-9 scores ≥ 10. The outcome was all-cause mortality. Covariates were demographics, comorbid conditions and health behaviours. RESULTS: Among 129 140 eligible participants, 30% had HIV infection, 16% had a major depressive disorder diagnosis, and 24% died over a median follow-up time of 11 years. The death rate was 25.3 [95% confidence interval (CI) 25.0-25.6] deaths per 1000 person-years. Major depressive disorder was associated with mortality [hazard ratio (HR) 1.04; 95% CI 1.01, 1.07]. This association was modified by HIV status (interaction P-value = 0.02). In HIV-stratified analyses, depression was significantly associated with mortality among HIV-uninfected veterans but not among those with HIV infection. Among those with PHQ-9 data (n = 7372), 50% had HIV infection, 22% had PHQ-9 scores ≥ 10, and 28% died over a median follow-up time of 12 years. The death rate was 27.3 (95% CI 26.1-28.5) per 1000 person-years. Depressive symptoms were associated with mortality (HR 1.16; 95% CI 1.04, 1.28). This association was modified by HIV status (interaction P-value = 0.05). In HIV-stratified analyses, depressive symptoms were significantly associated with mortality among veterans with HIV infection but not among those without HIV infection. CONCLUSIONS: Depression was associated with all-cause mortality. This association was modified by HIV status and method of depression ascertainment.

Radiation damage and stimulated desorption of nucleotides 2'-deoxyadenosine 5'-monophosphate (dAMP), adenosine 5'-monophosphate (rAMP), 2'-deoxycytidine 5'-monophosphate (dCMP), and cytidine 5'-monophosphate (rCMP) deposited on Au have been measured using x-rays as both the probe and source of low energy secondary electrons. The fluence dependent behavior of the O-1s, C-1s, and N-1s photoelectron transitions was analyzed to obtain phosphate, sugar, and nucleobase damage cross sections. Although x-ray induced reactions in nucleotides involve both direct ionization and excitation, the observed bonding changes were likely dominated by the inelastic energy-loss channels associated with secondary electron capture and transient negative ion decay. Growth of the integrated peak area for the O-1s component at 531.3 eV, corresponding to cleavage of the C-O-P phosphodiester bond, yielded effective damage cross sections of about 23 Mb and 32 Mb (1 Mb = 10-18 cm2) for AMP and CMP molecules, respectively. The cross sections for sugar damage, as determined from the decay of the C-1s component at 286.4 eV and the glycosidic carbon at 289.0 eV, were slightly lower (about 20 Mb) and statistically similar for the r- and d- forms of the nucleotides. The C-1s component at 287.6 eV, corresponding to carbons in the nucleobase ring, showed a small initial increase and then decayed slowly, yielding a low damage cross section (∼5 Mb). Although there is no statistical difference between the sugar forms, changing the nucleobase from adenine to cytidine has a slight effect on the damage cross section, possibly due to differing electron capture and transfer probabilities.

RESUMO

Two new low alloyed steels were developed with different fracture toughness values but at similar level of hardness with same composition and microstructural phase. The steels were subjected to impact-abrasion wear test. This work examines specifically the additional role of toughness during impact-abrasion wear, using a newly developed high toughness steel. Microstructural characterisation of the damaged samples revealed that better toughness helps resist both impact and abrasion damage.

Chromium (Cr) is one of the toxic metals adversely affecting organisms including humans in the ecosystems, and it is present in considerable concentration in the tannery industrial effluent. Toxicity expression of Cr is suspected to be influenced considerably by other accompanying ions present in the effluent used for irrigation. In a screen house experiment, interactive effects of chloride and sulfate ions in a Vertisol on uptake of Cr by spinach crop were investigated and treatments imposed were three levels each of Cr (0, 50, 100 mg kg-1), chloride (Cl-) (0, 25, 50 mM kg-1), and sulfur (S) (0, 4, 8 mM kg-1) in possible combinations. Plant growth parameters and leaf Cr concentrations were recorded to find out the effect of anions on Cr dynamics in the plant. Increasing the concentration of Cl- ions in soil reduced the Cr concentration in both root and shoot. Similarly, increasing the concentration of S from 4 to 8 mM kg-1 also reduced the concentration and uptake of Cr. Application of sulfate ions augmented the plant growth and counters the negative effect of Cl- ions and Cr. Thus, the study revealed that the addition of S fertilizers could minimize the Cr toxicity in high Cr contaminated soils.

Unstable trochanteric fractures and fractures with reverse obliquity pose difficulty in fixation. In recent years, intramedullary nails, for the treatment of comminuted and unstable intertrochanteric hip fractures, are becoming more popular relative to conventional, sliding hip screws. The purpose of our study was to evaluate the result of Trochanteric femoral nailing in comminuted, unstable Trochanteric femur fracture in terms of anatomical restoration and functional outcome. It is a prospective and without control study. Trochanteric femoral nailing has been done in comminuted unstable inter-trochanteric fracture femur (AO A2.2 to A3.3) of 25 patients and they are followed up postoperatively for at least 12 months. Pre-operative and post-operative clinical and radio-logical parameters are compared accordingly. Union in all cases. Overall complication rate 12% including some implant related complications. Functional outcome on Harris Hip Score is comparable with standard literature. For treatment of intertrochanteric hip fractures, particularly with comminuted fracture fragments, intramedullary devices offer beneficial features, such as closed insertion, a shorter lever arm, and controlled telescoping of the head-neck fragment. Insertion of the nail through the tip of the greater trochanter requires less dissection and may lead to less blood loss and fewer wound complications, as well as earlier postoperative mobility. Further biomechanical and clinical studies are necessary to validate the efficacy of the trochanteric femoral nail. Level of Evidence : Level III therapeutic study.

Tamoxifen administration enhanced overall disease-free survival and diminished mortality rates in cancer patients. However, patients with breast cancer often fail to respond for tamoxifen therapy due to the development of a drug-resistant phenotype. Functional analysis and molecular studies suggest that protein mutation and dysregulation of survival signaling molecules such as epidermal growth factor receptor, vascular endothelial growth factor receptor 2, and Akt contribute to tamoxifen resistance. Various strategies, including combinatorial therapies, show chemosensitize tamoxifen-resistant cancers. Based on chemotoxicity issues, researchers are actively investigating alternative therapeutic strategies. In the current study, we fabricate a mesoporous silica gold cluster nanodrug delivery system that displays exceptional tumor-targeting capability, thus promoting accretion of drug indices at the tumor site. We employ dual drugs, ZD6474, and epigallocatechin gallate (EGCG) that inhibit EGFR2, VEGFR2, and Akt signaling pathways since changes in these signaling pathways confer tamoxifen resistance in MCF 7 and T-47D cells. Mesoporous silica gold cluster nanodrug delivery of ZD6474 and EGCG sensitize tamoxifen-resistant cells to apoptosis. Western and immune-histochemical analyses confirmed the apoptotic inducing properties of the nanoformulation. Overall, results with these silica gold nanoclusters suggest that they may be a potent nanoformulation against chemoresistant cancers.

This study was conducted to evaluate the utilisation of the residual feed intake (RFI) as a feed efficiency selection tool and its relationship with methane emissions. Eighteen Murrah buffalo (Bubalus bubalis) heifers were fed ad libitum with total mixed ration (TMR) for 120 days. Based on linear regression models involving dry matter intake (DMI), average daily gain (ADG) and mid-test metabolic body size (MBW0.75 ), heifers were assigned into low and high RFI groups. The RFI varied from -0.09 to +0.12 kg DM/day with average RFI of -0.05 and 0.05 kg DM/day in low and high RFI heifers respectively. Low RFI heifers ate 11.6% less DM each day, yet average daily gain (ADG) and feed utilisation were comparable among low and high RFI groups. Low RFI heifers required significantly (p < .05) less metabolizable energy for maintenance (MEm) compared to high RFI heifers. Apparent nutrient digestibility showed non-significant difference (p > .05) among low and high RFI groups. Although the nitrogen balance was similar among heifers of low and high RFI groups, nitrogen metabolism was significantly higher (p > .05) in high RFI heifers. Comparison of data from heifers exhibiting the low (n = 9) and high (n = 9) RFI showed that the low RFI heifers have lower enteric methane production and methane losses than high RFI heifers. In conclusion, results of this study revealed that selection of more efficient buffalo heifers has multiple benefits, such as decreased feed intake and less emission of methane.

Shortfall of rain and the decreased groundwater level force farmers to use poor quality water for crop production in developing countries. In this study, the quality of agricultural produce and soil health affected by sewage water (Patranala) irrigation was evaluated. Sewage sediment, soil and crop samples were analyzed for physico-chemical properties. Sewage water found to contain trace concentration of heavy metals. However, long-term application of this water for crop production could build up a significant amount of trace metals in both soil and sediments. The DTPA extractable heavy metals ranged Cu 2.7-7.59, Cd 0.04-0.06, Pb 1.29-2.05, Cr 0.01-0.04, Ni 0.24-1.03 and Zn 0.63-2.59 mg kg-1 soil. The heavy metal risk assessment (hazard quotient, HQ) was calculated and found that HQ for these metals in the crops under study was in safe limits. From the study, it is concluded that growing vegetables using sewage water of Patranala is safe, but periodic monitoring to be carried out to avoid food chain contamination.

Folate, natural form of water soluble vitamin folic acid, is significant for humans as involved in most important metabolic reactions i.e. nucleotide synthesis and amino acid inter conversions. Thus its deficiency causes neural tube defects in newborns and cardiovascular diseases, and cancers. Humans cannot synthesize folate de novo so consumption through diet is essential. Natural food sources, supplements and fortified food products are the choices available to complete the Daily recommended intake. However microbial fortification using probiotics recently gained wide attention due to dual advantage of natural food matrix with enhanced folate content along with the probiotics benefits. Current study was focused on the microbial fortification of fruit juices and their efficacy and stability studies. Freshly filtered orange and tomato juice was prepared and inoculated with Streptococcus thermophilus NCIM 2904. Incubation was done at 40°C and samples were collected at different time interval. Folate extraction was done using human plasma and content was measured by microbiological assay using Lactobacillus casei NCIM No. 2364. Efficacy and stability studies were carried out to ensure the quality of juices to be consumed in terms of folate content, viable cell count and pH after 4 weeks of storage at low temperature. Positive results were observed as folate content was quite stable whereas viable cell count was also found to be significant till some time without adding any preservatives. The results indicated that fortified fruit juices could be used as probiotic beverages with enhanced folate content.

Due to an increase in the occurrence of multi drug resistant microorganisms a need for the development of alternative drugs comes in light. This alternative drug should be such that the microorganisms should not be able to develop resistance against them easily. Antimicrobial peptides are the most potential candidates to be developed as alternative drug. In the present study the three toxins ETA, ETB and PVL of Staphylococcus aureus were docked with four antimicrobial peptides, Ib-AMP1, JCPep7, Snakin2, Sesquin, derived from plants. The docking studies predict that Ib-AMP1 shows significant interactions with all these three toxins. Hence, further studies can be carried out for developing Ib-AMP1 as an alternative drug against the toxins of Staphylococcus aureus.

Bioethanol is an excellent alternative for petrol and has long-term economic advantages over non-renewable liquid biofuels. Bioethanol can be produced from different biomass materials, and it is categorized into three generations by biomass. First generation bioethanol directly competes with food items while second generation bioethanol requires more land area and fertilizers. Bioethanol produced from algae comes under third generation bioethanol and has many advantages over first and second generation bioethanol. Algae have higher growth rates than plants and require less land area also they do not need additional fertilizers for their growth. Microalgae can fix atmospheric CO2 from the environment and assimilate it into lipid and carbohydrates which can be used as a substrate for biofuel production.  In this work, microalgae Chlorella sp. was cultivated at 28oC and light intensity of 25µmol m-2 s-1 and used as a source of starch for bioethanol production. Chlorella starch was hydrolyzed into fermentable sugars by amylases produced from Aspergillus niger NCIM 616 for bioethanol production by yeast Saccharomyces cerevisiae NCIM 3494. Mathematical modelling gave the insight to predict the experimental profiles of bioethanol production from algal biomass.  Microbial growth, reducing sugar consumption, and bioethanol production were described quantitatively by using Logistic, Pirt, and Luedeking-Piret equations respectively.  The equations used in modelling of the fermentation kinetics matched very well with the experimental profiles, thus concluding that ethanol production from algal starch hydrolysate was growth-associated under the evaluated conditions.

PURPOSE: The aim of this study was to analyze whether the umbilical artery pH value can be estimated throughout CTG assessment 60 min prior to delivery and if the estimated umbilical artery pH value correlates with the actual one. This includes analysis of correlation between CTG trace classification and actual umbilical artery pH value. Intra-and interobserver agreement and the impact of professional experience on visual analysis of fetal heart rate tracing were evaluated. METHODS: This was a retrospective study. 300 CTG records of the last 60 min before delivery were picked randomly from the computer database with the following inclusion criteria; singleton pregnancy >37 weeks, no fetal anomalies, vaginal delivery either spontaneous or instrumental-assisted. Five obstetricians and two midwives of different professional experience classified 300 CTG traces according to the FIGO criteria and estimated the postnatal umbilical artery pH. RESULTS: The results showed a significant difference (p < 0.05) in estimated and actual pH value, independent of professional experience. Analysis and correlation of CTG assessment and actual umbilical artery pH value showed significantly (p < 0.05) diverging results. Intra- and interobserver variability was high. Intraobserver variability was significantly higher for the resident (p = 0.001). No significant differences were detected regarding interobserver variability. CONCLUSION: An estimation of the pH value and consequently of neonatal outcome on the basis of a present CTG seems to be difficult. Therefore, not only CTG training but also clinical experience and the collaboration and consultation within the whole team is important.

Soil and groundwater from long-term (>50 years) tannery effluent irrigated areas of Kanpur were analyzed and significant buildup of heavy metals such as Cr, Ni, Cd, Pb, Zn, and As in the range of 252-972, 23-30, 2.3-14.1, 23.7-58.8, 138-338 and 6.8-11 mg kg-1, respectively in soil was found. Few groundwater samples in the effluent irrigated areas also exhibited high Cr concentration above the permissible limit of United States Environmental Protection Agency. The tannery effluents contained 1.53-57.3 ppm Cr, 0-0.12 ppm Ni, 0-0.02 ppm Cd, 0-0.07 ppm Pb, 0-0.48 ppm Zn and 0-0.03 ppm As. The Geo-accumulation index (Igeo) revealed that soil samples were unpolluted to moderately polluted with Cu, Ni, Zn, Pb and As; moderately polluted in case of Cd; and heavily to extremely polluted by Cr.
