High ocean temperatures caused by global warming induce oxidative stress in aquatic organisms. Melatonin treatment and irradiation using red light-emitting diodes (LEDs) have been reported to reduce oxidative stress in a few aquatic organisms. However, the effects of red LED irradiation and melatonin injection on the antioxidant capacity and degree of apoptosis in abalones, which are nocturnal organisms, have not yet been reported. In this study, we compared the expression levels of antioxidant enzymes, total antioxidant capacity, and the degree of apoptosis in abalones subjected to red LED irradiation and melatonin treatment. The results revealed that at high water temperatures (25 °C), the mRNA expression levels of the superoxide dismutase (SOD) and glutathione peroxidase (GPx) genes and the antioxidant activity of SOD decreased in abalones in the red-LED irradiated and melatonin-treated groups compared with those in abalones in the control group. Although high water temperatures induced DNA damage in the abalone samples, the degree of apoptosis was lower in the red-LED irradiated and melatonin-treated groups than in the control group. Overall, the abalones in the melatonin-treated and red-LED irradiated groups showed reduced oxidative stress and increased antioxidant enzyme levels under thermal stress compared with those in the control group. Therefore, red LED irradiation is a promising alternative to melatonin treatment, which is difficult to administer continuously for a long time, for protecting abalones from oxidative stress.

Sirtuins (SIRTs), which are nicotinamide adenine dinucleotide-dependent class III histone deacetylases, regulate cell division, survival, and senescence. Although sirtinol, a synthetic SIRT inhibitor, is known to exhibit antitumor effects, its mechanism of action is not well understood. Therefore, we aimed to assess the anticancer effects and underlying mechanism of MHY2245, a derivative of sirtinol, in HCT116 human colorectal cancer cells in vitro. Treatment with MHY2245 decreased SIRT1 activity and caused DNA damage, leading to the upregulation of p53 acetylation, and increased levels of p53, phosphorylation of H2A histone family member X, ataxia telangiectasia and Rad3-related kinase, checkpoint kinase 1 (Chk1), and Chk2. The level of the breast cancer type 1 susceptibility protein was also found to decrease. MHY2245 induced G2/M phase cell cycle arrest via the downregulation of cyclin B1, cell division cycle protein 2 (Cdc2), and Cdc25c. Further, MHY2245 induced HCT116 cell death via apoptosis, which was accompanied by internucleosomal DNA fragmentation, decreased B-cell lymphoma 2 (Bcl-2) levels, increased Bcl-2-asscociated X protein levels, cleavage of poly(ADP-ribose) polymerase, and activation of caspases -3, -8, and -9. Overall, MHY2245 induces cell cycle arrest, triggers apoptosis through caspase activation, and exhibits DNA damage response-associated anticancer effects.

Sirtuins (SIRTs) belong to the nicotinamide adenine dinucleotide (NAD+)-dependent class III histone deacetylase family. They are key regulators of cellular and physiological processes, such as cell survival, senescence, differentiation, DNA damage and stress response, cellular metabolism, and aging. SIRTs also influence carcinogenesis, making them potential targets for anticancer therapeutic strategies. In this study, we investigated the anticancer properties and underlying molecular mechanisms of a novel SIRT1 inhibitor, MHY2251, in human colorectal cancer (CRC) cells. MHY2251 reduced the viability of various human CRC cell lines, especially those with wild-type TP53. MHY2251 inhibited SIRT1 activity and SIRT1/2 protein expression, while promoting p53 acetylation, which is a target of SIRT1 in HCT116 cells. MHY2251 treatment triggered apoptosis in HCT116 cells. It increased the percentage of late apoptotic cells and the sub-G1 fraction (as detected by flow cytometric analysis) and induced DNA fragmentation. In addition, MHY2251 upregulated the expression of FasL and Fas, altered the ratio of Bax/Bcl-2, downregulated the levels of pro-caspase-8, -9, and -3 proteins, and induced subsequent poly(ADP-ribose) polymerase cleavage. The induction of apoptosis by MHY2251 was related to the activation of the caspase cascade, which was significantly attenuated by pre-treatment with Z-VAD-FMK, a pan-caspase inhibitor. Furthermore, MHY2251 stimulated the phosphorylation of c-Jun N-terminal kinase (JNK), and MHY2251-triggered apoptosis was blocked by pre-treatment with SP600125, a JNK inhibitor. This finding indicated the specific involvement of JNK in MHY2251-induced apoptosis. MHY2251 shows considerable potential as a therapeutic agent for targeting human CRC via the inhibition of SIRT1 and activation of JNK/p53 pathway.

This qualitative study explored the experiences of new nurses with less than one year of clinical experience in caring for COVID-19 patients in a military hospital. In-depth interviews were conducted with six new nurses working in a negative-pressure isolation unit of the Armed Forces Capital Hospital. Data were analyzed using the phenomenological method proposed by Colaizzi, and 12 themes were derived and classified into four clusters: burden of nursing in isolation units; hardship of nursing critically ill patients; efforts to perform nursing tasks; positive changes through patient care. The participants were anxious while caring for COVID-19 patients with severe illness due to a lack of clinical experience. Furthermore, the wearing of heavy personal protective equipment impeded communication with patients, leading to physical and psychological exhaustion. However, they tried to utilize their own know-how and provide the best nursing care, resulting in them gaining confidence. Participants were able to think critically and took pride in being military nursing professionals. This study is meaningful as it provides insight into the experiences of new military nurses who were rapidly dispatched during a national medical crisis. The results can be applied to develop future strategies aimed at improving new nurses' competency in military hospitals.

The purpose of this study was to investigate leisure satisfaction among outdoor sports participants in golf and yachting. Influence was also measured of recreation specialization on leisure satisfaction, and the effect of the relationship between leisure facilitation and leisure satisfaction on golf and yacht participation was investigated as well. Frequency, reliability, confirmatory, and correlation analysis, as well as structural equation modeling results, indicate that leisure facilitation had no influence on outdoor sports participants' recreation specialization. Leisure facilitation had a positive influence on leisure satisfaction among the golf and yachting participants, and their recreation specialization had a positive influence on their leisure satisfaction. The effects of the COVID-19 pandemic are addressed, specifically the constraints that the disease has imposed on outdoor sports and leisure, and strategies are presented for addressing these constraints and promoting outdoor sports participation.

BACKGROUND/PURPOSE: Previous studies have suggested that functional dominance in one part of the body can be correlated with functional dominance in another part. Thus, the present research aimed to determine whether brain laterality (handedness, footedness, earedness, and eyedness) was related to mixing ability and chewing side preference. MATERIALS AND METHODS: Fifty-four volunteers who were not undergoing any form of dental treatment took part in this study. Self-defined brain laterality was determined through a questionnaire. The volunteers performed five tasks related to brain laterality, which was identified by the side used to perform three or more of the five tasks. Chewing side preference was determined by observing the main gum location on the occlusal area when volunteers chewed for 30 strokes. Mixing Ability Index (MAI) was measured by analyzing the degree of mixing of two differently colored waxes (height, 3 mm; diameter, 20 mm). Occlusion contact area was measured by taking the maximum intercuspation bite with polysiloxane. RESULTS: Thirty-nine volunteers (72%) showed significant agreement between brain dominance and chewing preference side. The association between brain dominance and MAI was not significant. The occlusal contact area of the dominant side (mean = 48.2 mm2) was significantly wider than that of the nondominant side (25.7 mm2). CONCLUSION: Brain laterality can be explained by the side of functional (preference of the hands, eyes, ears, and feet, and survey) has a positive correlation with chewing preference side. MAI between the brain dominant and nondominant sides was not significant. This shows that mastication efficiency does not differ between dominant and nondominant sides. So, this study suggests that brain dominance is correlated with chewing preference, but it does not affect efficiency of mastication.

The purpose of this study were to evaluate the discoloration of a silorane-based resin and two methacrylated-based resin composites upon exposure to different staining solutions coffee, red wine, porcine liver esterase and distilled water for 7 days. The colors of all specimens before and after storage in the solutions were measured by a spectrophotometer based on CIE Lab system, and the color differences thereby calculated. Data were statistically analyzed by ANOVA and Scheffe's test. For coffee and red wine, the mean color change in silorane-based resin was significantly lower than that in methacylate-based resin composites (p<0.05). For porcine liver esterase and distilled water, there was no significant difference in the mean values of color change between silorane- and methacrylate-based resin composites (p>0.05). In conclusion, the silorane-based resin composites exhibited better color stability (less ΔE) after exposure to the colored staining solutions.

OBJECTIVE: This study aimed to measure the disability weights for the Korean Burden of Disease study, and to compare them with those adopted in the Australian study to examine the validity and describe the distinctive features. METHODS: The standardized valuation protocol was developed from the Global Burden of Disease (GBD) study and the Dutch Disability Weights study. Disability weights were measured for 123 diseases of the Korean version of Disease Classification by three panels of 10 medical doctors each. Then, overall distribution, correlation coefficients, difference by each disease, and mean of differences by disease group were analyzed for comparison of disability weights between the Korean and Australian studies. RESULTS: Korean disability weights ranged from 0.037 to 0.927. While the rank correlation coefficient was moderate to high (rs=0.68), Korean disability weights were higher than the corresponding Australian ones in 79.7% of the 118 diseases. Of these, war, leprosy, and most injuries showed the biggest differences. On the contrary, many infectious and parasitic diseases comprised the greater part of diseases of which Korean disability weights were lower. The mean of the differences was the highest in injuries of GBD disease groups, and in cardiovascular disease, injuries, and malignant neoplasm of the Korean disease category. CONCLUSIONS: Korean disability weights were found to be valid on the basis of overall distribution pattern and correlation, and are expected to be used as basic data for broadening the scope of burden of disease study. However, some distinctive features still remain to be explored in following studies.
