To determine whether exercise-induced increases in tissue plasminogen activator (t-PA) were related to plasma epinephrine concentration during exercise, 14 healthy men (aged 24 to 62 years) were studied during epinephrine infusions (10, 25 and 50 ng/kg per min) and graded supine bicycle exercise, beginning at 33 W and increasing in 33-W increments until exhaustion. Plasma epinephrine, active and total t-PA, active plasminogen activator inhibitor type 1 (PAI-1) and t-PA/PAI-1 complex concentrations were measured at each exercise and infusion level. During epinephrine infusion, active and total t-PA levels increased linearly with the plasma epinephrine concentration (respective slopes [+/- SEM] of 0.062 +/- 0.003 and 0.076 +/- 0.003 pmol/ng epinephrine). During exercise, t-PA levels did not increase until plasma epinephrine levels increased, after which both active and total t-PA levels again increased linearly with the plasma epinephrine concentration, but at twice the rate observed with epinephrine infusion (0.131 +/- 0.005 and 0.147 +/- 0.005 pmol/ng, respectively). The t-PA level in blood was directly proportional to the plasma epinephrine concentration during both exercise and epinephrine infusion, suggesting that epinephrine release during exercise stimulates t-PA secretion. In these healthy subjects, active plasminogen activator inhibitor type 1 and t-PA/PAI-1 complex levels were low (41 +/- 11 and 21 +/- 5 pmol/liter, respectively) and did not change significantly during exercise or epinephrine infusion. It is concluded that approximately 50% of the increase in t-PA during exercise is due to stimulated release of t-PA by epinephrine.(ABSTRACT TRUNCATED AT 250 WORDS)

Aging has been associated with glucose intolerance, insulin resistance, hyperinsulinemia, and diminished islet B-cell function. The relative contribution of these factors to the aging-associated changes in glucose tolerance has been difficult to discern, particularly so for B-cell function, since insulin sensitivity itself is a determinant of B-cell function and, therefore, comparisons of insulin levels and responses between old and young subjects are difficult. To reduce this effect, we compared B-cell function in 14 healthy older men (aged 61-82 yr; body mass index, 21-30 kg/m2), who were exercise trained for 6 months to improve insulin sensitivity, to that of 11 healthy young men (aged 24-31 yr; body mass index, 19-31 kg/m2), who were also trained. Insulin-glucose interactions were assessed by measuring indices of insulin sensitivity (SI) and glucose effectiveness at zero insulin (GEZI) using Bergman's minimal model. B-Cell function was assessed by determining the acute insulin responses (AIR) to glucose (AIRgluc) and arginine at 3 different glucose levels: fasting, approximately 14 mM, and greater than 28 mM (AIRmax). AIRmax provides a measure of B-cell secretory capacity, while the glucose level at which 50% of AIRmax occurs is termed PG50 and is used to estimate B-cell sensitivity to glucose. The insulin sensitivity and glucose effectiveness at zero insulin of the trained older subjects was similar to that of the trained young [SI: old, 5.1 +/- 0.6; young, 6.5 +/- 0.7 x 10(-5) min-1/pM (mean +/- SEM; P = NS); GEZI: old, 1.3 +/- 0.2; young, 1.7 +/- 0.2 x 10(-2) min (P = NS)]. Under these conditions, the fasting glucose levels (old, 5.4 +/- 0.2; young, 5.1 +/- 0.1 mM) and basal insulin levels (old, 49 +/- 6; young, 63 +/- 11 pM) were also similar in the two groups. AIRgluc values were lower in the exercised elderly (old, 253 +/- 50; young, 543 +/- 101 pM; P = 0.01). This decrease in stimulated insulin release was due solely to a reduction in the AIRmax (old, 1277 +/- 179; young, 2321 +/- 225 pM; P less than 0.005); the PG50 was not different (old, 8.9 +/- 0.4; young, 8.8 +/- 0.2 mM; P = NS). These differences in the older subjects were associated with a reduction in iv glucose tolerance (old, 1.49 +/- 0.15; young, 1.95 +/- 0.13%/min; P less than 0.05).(ABSTRACT TRUNCATED AT 400 WORDS)

Although there are considerable data concerning the effects of endurance exercise training (ET) on plasma lipoproteins, the results have been quite inconsistent. The observed variability of response may be related to the age, sex, adiposity, or diet of the subjects tested, or to the type and intensity of the exercise intervention. Furthermore, there is relatively little such data in older individuals. Therefore, in the present study, we investigated the effects of intensive ET on lipoprotein profiles in healthy young (n = 12; 28.2 +/- 2.4 years) and older (n = 15; 67.5 +/- 5.8 years) men. Unlike subjects in most similar studies, our subjects were weight-stabilized on a constant-composition diet for 21 days prior to determination of the lipoprotein profile before and after the ET program. At baseline, the two groups were not significantly different with respect to any individual component of their lipoprotein profiles, relative weight, or percent body fat, but the older men had a more central distribution of fat by both waist to hip ratio (WHR) and computed tomography (CT). Maximal aerobic power, expressed per kilogram of body weight (VO2 max), was 33% lower (P less than .001) in the older men at baseline. Following the 6-month, walk/jog/bike ET program (5 d/wk), both the young (+18%, P less than .001) and the older (+22%, P less than .001) men increased their VO2 max. This was associated with small, but significant, decrements in weight, percent body fat, and WHR only in the older men.(ABSTRACT TRUNCATED AT 250 WORDS)

Little is known about the effects of exercise interventions on the distribution of central and/or intra-abdominal (IA) fat, and until now there were no studies in the elderly. Therefore, in this study we investigated the effects of an intensive 6-month endurance training program on overall body composition (hydrostatic weighing), fat distribution (body circumferences), and specific fat depots (computed tomography [CT]), in healthy young (n = 13; age, 28.2 +/- 2.4 years) and older (n = 15; age, 67.5 +/- 5.8 years) men. At baseline, overall body composition was similar in the two groups, except for a 9% smaller fat free mass in the older men (P less than .05). The thigh and arm circumferences were smaller (P = .001 and P less than .05, respectively), while the waist to hip ratio (WHR) was slightly greater in the older men (0.92 +/- 0.04 v 0.97 +/- 0.04, P less than .01). Compared with the relatively small baseline differences in body composition and circumferences, CT showed the older men to have a twofold greater IA fat depot (P less than .001), 48% less thigh subcutaneous (SC) fat (P less than .01), and 21% less thigh muscle mass (P less than .001). Following endurance (jog/bike) training, both the young (+18%, P less than .001) and the older men (+22%, P less than .001) significantly increased their maximal aerobic power (VO2max). This was associated with small but significant decrements in weight, percent body fat, and fat mass (all P less than .001) only in the older men.(ABSTRACT TRUNCATED AT 250 WORDS)

Under laboratory conditions, most colony and field-collected Aedes bahamensis Berlin females developed eggs autogenously when they had access to sugar. However, significantly fewer starved females were autogenous, and they produced smaller egg clutches. Autogenous fecundity covaried with wing length, and smaller females generally failed to express autogeny. Mating had no effect on the maturation of the initial egg clutch. Most starved, nulliparous females blood fed from a restrained host. At a south Florida field site, both parous and nulliparous Ae. bahamensis were captured with a power aspirator, but concurrent sampling with dry ice-baited, light traps collected only parous females. Host-seeking females, taken either in chicken-baited traps or as they attempted to blood feed on humans, were also parous, with a single exception. Thus, at this field site, Ae. bahamensis females normally delayed blood feeding until after their first oviposition. Whether or not Ae. bahamensis females in other south Florida populations show a similar gonotrophic pattern probably will depend upon the availability of sugar sources and conditions in the mosquito's aquatic habitat that affect adult size.

Intracoronary streptokinase was offered and preliminary coronary angiography performed in 14 patients who were seen with the clinical diagnosis of acute myocardial infarction within 4 h of onset of symptoms. The procedure was performed in the Coronary Care Unit (CCU) of St. Peter's Medical Center with the use of a portable C-arm fluoroscope. Angiography was recorded on video tape. Service was provided by an "on-call" team consisting of two physicians, a CCU nurse, and a radiology technician, on a 24-h service basis. Adequate visualization of coronary anatomy was obtained in all patients. Patency of occluded vessels was achieved in 10 of 11 patients who received intracoronary streptokinase. The initial streptokinase bolus was administered at a mean interval of 4.1 h from onset of symptoms. It is concluded that speedy and effective coronary thrombolytic therapy can be provided in the CCU on a 24-h service basis by an on-call team. The use of CCU for this purpose will make this therapy widely available across the country, without the need for Cardiac Catheterization Laboratory.

RESUMO

The cytologic findings in voided urine specimens from prostatic duct carcinoma are presented. Neoplastic cells from these unusual tumors were identified as well as the occasional red blood cell and histiocyte. The adenocarcinoma cells features irregular nuclei and their cytoplasm was occupied almost entirely by large secretory vacuoles. The differential diagnosis of this tumor from some other neoplasms and its association with transitional cell carcinoma as well as its histogenesis are discussed.

RESUMO

Multi-frequency (multi-component) acoustic impedance measurements may evolve into a sensitive technique for the remote detection of aural pathologies. Such data are also relevant to models used in hearing aid design and could be an asset to the hearing aid prescription and fitting process. This report describes the development and use of a broad-band procedure which acquires impedance data in 20 Hz intervals and describes a comparison of data collected at two sites by different investigators. Mean data were in excellent agreement, and an explanation for a single case of extreme normal variability is presented.

Differential cell counts were done on bronchial lavage specimens from 166 horses. Postmortem gross and histologic examinations were done to determine the pathologic diagnosis of the lungs from these horses. Ninety-two (55%) were normal, 18 (11%) had interstitial pneumonia, 13 (8%) had bronchopneumonia, 13 (8%) had focal eosinophilia, 11 (7%) had diffuse eosinophilia, 11 (7%) had chronic bronchitis, and 8 (5%) had suppurative bronchitis. Little relationship was found between cellular features of the lavage specimens and the pathologic status of the lungs of individual horses.

Oxytetracycline (OTC) was administered IV to 3 clinically normal horses at a dosage of 10 mg of OTC/kg of body weight. Plasma OTC concentrations were determined at 30-minute intervals until postinjection minute (PIM) 240. At PIM 240, the mean OTC concentration in pulmonary tissue was 3.96 microgram/g of tissue (wet weight) and in renal tissue was 25.47 micrograms/g. diluted bronchial fluid had a mean concentration of 0.288 microgram of OTC/ml at PIM 240. The data demonstrated that OTC has adequate tissue distribution in horses.

Statistical parameters necessary for the comparison of herd blood beta hydroxybutyrate (HB) concentration means with HB concentrations in the general population were determined in 11 clinically normal herds of Holstein-Friesian cows. These parameters were then used to compare clinically normal cows from 2 Holstein-Friesian herds experiencing a high prevalence of clinical ketosis with the normal population. Blood HB concentration means were compared with plasma glucose concentration means for their relative value as predictors of clinical ketosis prevalence within a herd. Blood HB concentration values were distributed approximately log normal and were directly related to amount of milk production. Herds with a high prevalence of clinical ketosis had blood HB log means higher (P less than 0.01) and glucose means lower (P less than 0.05) than did the normal population. Herd blood HB log means should be determined on 7 or more animals within specified production groups. When these means exceed the normal population log mean by more than 1 SD, susceptible cows in the herd may be at increased risk for the development of clinical ketosis. Blood HB log means may be of greater value than are plasma glucose concentration means for the prediction of herd ketosis prevalence.

The effect of ration composition, with respect to concentrate, crude protein, and methionine hydroxyanalog content, on blood beta-hydroxybutyrate and plasma glucose concentrations was assessed in Holstein cows every 2 weeks over the first 6 weeks of lactation. The correlation of these metabolites with estimated energy balance, and the effects of these ration variables on this correlation were studied. High concentrate diets (60% of dry matter) compared with low concentrate diets (40% of dry matter) increased mean plasma glucose values and reduced mean blood beta-hydroxybutyrate concentration. Variation in crude protein and methionine hydroxyanalog supplementation did not affect metabolite concentration. The correlations between blood beta-hydroxybutyrate and energy balance and between plasma glucose and energy balance were weak and subject to the influence of variation in ration composition. It was concluded that plasma glucose and blood beta-hydroxybutyrate concentrations cannot be used as valid indicators of energy balance. However, it did appear that blood beta-hydroxybutyrate might be used as an indicator of the relative glucogenic potential of dairy rations and that blood concentrations of this metabolite could potentially be used to adjust factors in the ration which influence glucose availability to the cow.

Pneumonia was induced in a group of calves that were then treated with oxytetracycline (OTC). When the plasma pharmacokinetics and tissue concentrations of this group were compared with those of healthy controls, obvious differences were observed. The volume of distribution and the half-life of OTC were increased in the group with pneumonia. Tissue concentrations of OTC (1.48 +/- 0.29 microgram/ml) were greater in the lungs of calves with pneumonia than that in the lungs (1.09 +/- 0.09 microgram/ml) of healthy controls. For the dose of OTC used, the correlation between serum concentration and tissue concentrations at 24 hours was poor. Seemingly, current concepts of antibiotic dosages and dosage intervals may be inadequate because they are based on blood and tissue values of healthy animals.

Pituitary-dependent hyperadrenocorticism was diagnosed in a 14-year-old Arabian mare with chronic weight loss, hirsutism, polyuria, and polydipsia. The mare had a stress leukogram, glucosuria, and consistent hyperglycemia. Plasma glucose concentrations were resistant to suppression by insulin. Plasma cortisol concentrations were within normal limits, but did not respond to dexamethasone suppression and had an exaggerated response to ACTH stimulation. At necropsy, a chromophobe adenoma of the pars intermedia of the pituitary gland was found. The zona fasciculata of the adrenal cortex and the pancreatic islets of Langerhans were hypertrophied. An immunohistologic staining technique was used to demonstrate ACTH-containing neoplastic cells in the pituitary mass. These cells released ACTH and other peptides that initiated the chain of endocrinologic events leading to clinical disease.

Bovine respiratory syncytial virus (BRSV) was the viral agent most commonly identified in 14 epizootics of pneumonia in dairy calves. A microtiter serum-virus neutralization test proved to be the best means of identifying involvement of BRSV; seroconversion (fourfold or greater rise in titer) was demonstrated in 10 of the 14 epizootics. Only limited involvement of bovine viral diarrhea virus, infectious bovine rhinotracheitis virus, parainfluenza 3 virus, and bovine adenovirus type 3 was recognized. Pasteurella multocida was isolated in 12 of 14 epizootics, and Pasteurella haemolytica in 4 of 14 epizootics. Mycoplasmal and ureaplasmal agents were isolated in all 14 epizootics.

Seasonal trends in reproductive performance and size were examined among female Mansonia dyari. The incidence of parity ranged between 6.9 and 58.1% and averaged 22.5% during an 8-month interval, with 4 peaks in parous rate believed to follow periods of emergence. Parous rates were positively correlated with wing lengths, indicating size-dependent survivorship. Wing lengths were highest in the winter and early spring, decreased to a minimum at the end of summer, and increased in the autumn, showing negative correlations with ambient temperatures recorded prior to adult capture. Female biomasses and fecundities were positively correlated with wing lengths, and their mean values varied significantly among sample dates. Egg development in Ma. dyari was completed by 5.4 days post blood meal at 26 degrees C and 7.6 days at 20 degrees C. No evidence for autogeny was observed.

Simple and economic methods were developed for control programs to demonstrate the movement of mosquitoes from a breeding source to residential areas. Using mark-release-recapture methods and examples, mean, median and maximum distances traveled were estimated or observed and compared for 11 species produced in a wastewater treatment facility near Lakeland, FL. The applicability of these methods and data interpretation for operational mosquito control programs are discussed.

The exotic mosquito, Aedes bahamensis, is now well-established in south Florida, where it is widely distributed throughout Dade and southern Broward Counties in both urban and rural areas east of the Everglades. When discarded automobile tires were sampled in areas near human habitation, larvae and pupae of Ae. bahamensis were frequently found in association with immature Ae. aegypti. Elsewhere, however, Ae. bahamensis generally occurred in the absence of Ae. aegypti. The persistence of Ae. bahamensis populations at specific sites was documented in egg collections from ovitraps and in larval samples from the water retained in discarded tires.
