RATIONALE: Embedding resins are commonly used to facilitate high-resolution sampling for stable isotope analysis but anomalous δ13 C values have been observed in some cases. Here we compare the results of microsampling strategies for hand-drilled versus resin-embedded micromilled samples from the same marine shells to assess whether resin contamination is implicated in δ13 C spikes. The comparison allows assessment of the relative benefits for spatial resolution, seasonal range for both δ18 O and δ13 C, and sample failure rates. METHODS: Hand-drilled samples were obtained from two bivalve shells (Spisula sachalinensis), corresponding to micromilled samples on the same shells where high δ13 C spikes were observed. All carbonate powders were analysed using a dual-inlet Isoprime mass spectrometer and Multiprep device. Results from both sample sets were compared statistically. RESULTS: No anomalous high δ13 C values and no failures due to insufficient gas were observed in the hand-drilled samples in contrast to the embedded micromilled sequences. Spatial resolution was reduced (~2.5×) in the former compared with the latter, resulting in a small reduction in the total range observed in the micromilled δ13 C and δ18 O values. Reduced sampling resolution between the two datasets was only significant for δ18 O. CONCLUSIONS: For S. sachalinensis (as with other similar bivalves), rapid growth mitigates the reduced sampling resolution of hand drilling and does not significantly impact observed isotopic range and seasonal patterning. Occurrence of anomalous δ13 C values were eliminated and failure rates due to insufficient sample size greatly reduced in the hand-drilled dataset. We can find no other explanation for the occurrence of δ13 C spikes than contamination by the embedding agent. We conclude that the logistical and interpretational benefits of careful hand drilling may be preferable to resin embedding for micromilling in marine shells, corals or speleothems where growth rate is rapid and the highest resolution is not required.

OBJECTIVES: Stable isotope analysis of sequential dentine samples is a potentially powerful method to reveal insights into early life-histories of individuals in the past. Dentine incremental growth structures are complex, however, and current approaches that apply horizontal sectioning of demineralized tooth halves or quarters risk combining multiple growth layers and may include unwanted cementum or secondary dentine. They also require destruction of large parts of a tooth. Here, we present a less destructive and relatively straightforward protocol that reduces damage, increases temporal resolution, and improves the accuracy of age-alignment between individuals. MATERIAL AND METHODS: We outline a protocol that includes the sampling of small (1 mm diameter) cylindrical plug transects from a thin section, along with an age-alignment scheme predicated on average growth rates for dentine areas. RESULTS AND DISCUSSION: The proposed protocol is readily applicable and more anatomically sensitive than horizontal slicing. Micro-samples are smaller (in both length and depth), hence minimizing temporal overlap and avoid directions that may contravene growth pattern. They completely avoid areas where secondary and tertiary dentine or cementum can be deposited. Age-alignment is improved by using growth ratios of anatomical tooth zones. CONCLUSION: This method minimizes destruction, enables finer temporal resolution and facilitates data comparison. It can be readily combined with fluorescence imaging-based or other pre-screening methods of dentine collagen preservation.

This study assesses the seasonal scheduling of shellfish harvesting among hunter-gatherer populations along the southernmost coast of South Africa, based on a large number of serial oxygen isotope analyses of marine mollusk shells from four archaeological sites. The south coast of South Africa boasts an exceptional record of coastal hunter-gatherer occupation spanning the Holocene, the last glacial cycle and beyond. The significance of coastal adaptations, in this region in particular, for later modern human evolution has been prominently debated. Shellfishing behaviors are an important focus for investigation given the dietary and scheduling implications and the abundant archaeological shell remains in numerous sites. Key to better understanding coastal foraging is whether it was limited to one particular season, or year-round. Yet, this has proven very difficult to establish by conventional archaeological methods. This study reconstructs seasonal harvesting patterns by calculating water temperatures from the final growth increment of shells. Results from two Later Stone Age sites, Nelson Bay Cave (together with the nearby Hoffman's Robberg Cave) and Byneskranskop 1, show a pronounced cool season signal, which is unexpected given previous ethnographic documentation of summer as the optimal season for shellfishing activities and inferences about hunter-gatherer scheduling and mobility in the late Holocene. Results from two Middle Stone Age sites, Klasies River and Pinnacle Point 5-6, show distinct seasonal patterns that likely reflect the seasonal availability of resources in the two locations. The Pinnacle Point 5-6 assemblage, which spans the MIS5-4 transition, records a marked shift in shellfishing seasonality at c. 71 ka that aligns with other indications of archaeological and environmental change at this time. We conclude that the scheduling and intensity of shellfishing in this region is affected by a suite of factors, including environmental and cultural drivers, rather than a single variable, such as population growth.

Sri Lanka has yielded some of the earliest dated fossil evidence for Homo sapiens (∼38-35,000 cal. years BP [calibrated years before present]) in South Asia, within a region that is today covered by tropical rainforest. Archaeozoological and archaeobotanical evidence indicates that these hunter-gatherers exploited tropical forest resources, yet the contribution of these resources to their overall subsistence strategies has, as in other Late Pleistocene rainforest settings, remained relatively unexplored. We build on previous work in this tropical region by applying both bulk and sequential stable carbon and oxygen isotope analysis to human and faunal tooth enamel from the sites of Batadomba-lena, Fa Hien-lena, and Balangoda Kuragala. Tooth enamel preservation was assessed by means of Fourier Transform Infrared Spectroscopy. We use these data to produce a detailed stable isotope ecology for Late Pleistocene-Holocene foragers in Sri Lanka from ∼36-29,000 to 3000 cal. years BP, allowing us to test the degree of human tropical forest resource reliance over a considerable time period. Given that non-human primates dominate the mammalian assemblages at these sites, we also focus on the stable isotope composition of three monkey species in order to study their ecological preferences and, indirectly, human hunting strategies. The results confirm a strong human reliance on tropical forest resources from ∼36-29,000 cal. years BP until the Iron Age ∼3 cal. years BP, while sequential tooth data show that forest resources were exploited year-round. This strategy was maintained through periods of evident environmental change at the Last Glacial Maximum and upon the arrival of agriculture. Long-term tropical forest reliance was supported by the specialised capture of non-human primates, although the isotopic data revealed no evidence for niche distinction between the hunted species. We conclude that humans rapidly developed a specialisation in the exploitation of South Asia's tropical forests following their arrival in this region.

Ranging and residence patterns among early hominins have been indirectly inferred from morphology, stone-tool sourcing, referential models and phylogenetic models. However, the highly uncertain nature of such reconstructions limits our understanding of early hominin ecology, biology, social structure and evolution. We investigated landscape use in Australopithecus africanus and Paranthropus robustus from the Sterkfontein and Swartkrans cave sites in South Africa using strontium isotope analysis, a method that can help to identify the geological substrate on which an animal lived during tooth mineralization. Here we show that a higher proportion of small hominins than large hominins had non-local strontium isotope compositions. Given the relatively high levels of sexual dimorphism in early hominins, the smaller teeth are likely to represent female individuals, thus indicating that females were more likely than males to disperse from their natal groups. This is similar to the dispersal pattern found in chimpanzees, bonobos and many human groups, but dissimilar from that of most gorillas and other primates. The small proportion of demonstrably non-local large hominin individuals could indicate that male australopiths had relatively small home ranges, or that they preferred dolomitic landscapes.

Stable isotope analysis of primate tissues in tropical forest contexts is an increasingly popular means of obtaining information about niche distinctions among sympatric species, including preferences in feeding height, forest canopy density, plant parts, and trophism. However, issues of equifinality mean that feeding height, canopy density, as well as the plant parts and plant species consumed, may produce similar or confounding effects. With a few exceptions, researchers have so far relied largely on general principles and/or limited plant data from the study area as references for deducing the predominant drivers of primate isotope variation. Here, we explore variation in the stable carbon (δ13 C), nitrogen (δ15 N), and oxygen (δ18 O) isotope ratios of 288 plant samples identified as important to the three primate species from the Polonnaruwa Nature Sanctuary, Sri Lanka, relative to plant part, season, and canopy height. Our results show that plant part and height have the greatest effect on the δ13 C and δ18 O measurements of plants of immediate relevance to the primates, Macaca sinica, Semnopithecus priam thersites, and Trachypithecus vetulus, living in this monsoonal tropical forest. We find no influence of plant part, height or season on the δ15 N of measured plants. While the plant part effect is particularly pronounced in δ13 C between fruits and leaves, differential feeding height, and plant taxonomy influence plant δ13 C and δ18 O differences in addition to plant organ. Given that species composition in different regions and forest types will differ, the results urge caution in extrapolating general isotopic trends without substantial local baselines studies.

Tropical forests constitute some of the most diverse and complex terrestrial ecosystems on the planet. From the Miocene onward, they have acted as a backdrop to the ongoing evolution of our closest living relatives, the great apes, and provided the cradle for the emergence of early hominins, who retained arboreal physiological adaptations at least into the Late Pliocene. There also now exists growing evidence, from the Late Pleistocene onward, for tool-assisted intensification of tropical forest occupation and resource extraction by our own species, Homo sapiens. However, between the Late Pliocene and Late Pleistocene there is an apparent gap in clear and convincing evidence for the use of tropical forests by hominins, including early members of our own genus. In discussions of Late Pliocene and Early Pleistocene hominin evolution, including the emergence and later expansion of Homo species across the globe, tropical forest adaptations tend to be eclipsed by open, savanna environments. Thus far, it is not clear whether this Early-Middle Pleistocene lacuna in Homo-rainforest interaction is real and representative of an adaptive shift with the emergence of our species or if it is simply reflective of preservation bias.

OBJECTIVES: As many individuals were cremated in Neolithic and Bronze Age Ireland, they have not featured in investigations of individual mobility using strontium isotope analysis. Here, we build on recent experiments demonstrating excellent preservation of biogenic (87) Sr/(86) Sr in calcined bone to explore mobility in prehistoric Northern Ireland. MATERIALS AND METHODS: A novel method of strontium isotope analysis is applied to calcined bone alongside measurements on tooth enamel to human remains from five Neolithic and Bronze Age sites in Northern Ireland. We systematically sampled modern vegetation around each site to characterize biologically available strontium, and from this calculated expected values for humans consuming foods taken from within 1, 5, 10 and 20 Km catchments. This provides a more nuanced way of assessing human use of the landscape and mobility than the 'local' vs. 'non-local' dichotomy that is often employed. RESULTS: The results of this study 1) provide further support for the reliability of strontium isotope analysis on calcined bone, and 2) demonstrate that it is possible to identify isotopic differences between individuals buried at the same site, with some consuming food grown locally (within 1-5 Km) while others clearly consumed food from up to 50 Km away from their burial place. DISCUSSION: Hints of patterning emerge in spite of small sample numbers. At Ballynahatty, for instance, those represented by unburnt remains appear to have consumed food growing locally, while those represented by cremated remains did not. Furthermore, it appears that some individuals from Ballynahatty, Annaghmare and Clontygora either moved in the last few years of their life or their cremated remains were brought to the site. RATIONALE: Strontium isotopes ((87) Sr/(86) Sr) are used in archaeological and forensic science as markers of residence or mobility because they reflect the local geological substrate. Currently, tooth enamel is considered to be the most reliable tissue, but it rarely survives heating so that in cremations only calcined bone fragments survive. We set out to test the proposition that calcined bone might prove resistant to diagenesis, given its relatively high crystallinity, as the ability to measure in vivo (87) Sr/(86) Sr from calcined bone would greatly extend application to places and periods in which cremation was the dominant mortuary practice, or where unburned bone and enamel do not survive. METHODS: Tooth enamel and calcined bone samples were exposed to a (87) Sr-spiked solution for up to 1 year. Samples were removed after various intervals, and attempts were made to remove the contamination using acetic acid washes and ultrasonication. (87) Sr/(86) Sr was measured before and after pre-treatment on a Nu Plasma multi-collector induced coupled plasma mass spectrometer using NBS987 as a standard. RESULTS: The strontium isotopic ratios of all samples immersed in the spiked solution were strongly modified showing that significant amounts of strontium had been adsorbed or incorporated. After pre-treatment the enamel samples still contained significant amounts of (87) Sr-enriched contamination while the calcined bone fragments did not. CONCLUSIONS: The results of the artificial enrichment experiment demonstrate that calcined bone is more resistant to post-mortem exchange than tooth enamel, and that in vivo strontium isotopic ratios are retained in calcined bone.

The decline of the Tiwanaku state saw the emergence of two new cultures-Pica-Tarapacá and Atacama-during the Late Intermediate Period in northern Chile. Archeological evidence suggests that both groups practised maize agriculture and pastoralism, but that their interaction zones differed significantly. Marine resources are common at Pica-Tarapacá sites, even those far from coast, while Atacama sites in the desert oases and precordilleran area seem to have directed their networks towards the highlands. Here we apply stable carbon, nitrogen, and oxygen isotope analysis on human bone and enamel to test dietary patterns and residential mobility at two sites, Pica 8 and Quitor 6, representing the Pica-Tarapacá and Atacama cultures, respectively. Our results show that diet at the two sites indeed differed: significant but variable consumption of marine resources and maize is indicated at Pica 8, despite being an inland site, while diet at Quitor 6 was based mainly on terrestrial resources. The use of seabird guano and llama dung as fertilizers and extreme aridity may have contributed to the high nitrogen isotope values observed in Pica 8 humans. The δ(18) O values in Pica 8 individuals are generally lower than for Quitor in spite of its greater distance from the Andes. All three isotopes suggest the presence of at least five nonlocals in the 30 measured at Pica 8. This evidence for human mobility is consistent with the high levels of trade and interaction observed in the archeological record, and begins to quantify the degree of movement of specific individuals.

Foods derived from C(4) plants were important in the dietary ecology of early Pleistocene hominins in southern and eastern Africa, but the origins and geographic variability of this relationship remain unknown. Carbon isotope data show that Australopithecus bahrelghazali individuals from Koro Toro in Chad are significantly enriched in (13)C, indicating a dependence on C(4) resources. As these sites are over 3 million years in age, the results extend the pattern of C(4) dependence seen in Paranthropus boisei in East Africa by more than 1.5 million years. The Koro Toro hominin fossils were found in argillaceous sandstone levels along with abundant grazing and aquatic faunal elements that, in combination, indicate the presence of open to wooded grasslands and stream channels associated with a greatly enlarged Lake Chad. In such an environment, the most abundant C(4) plant resources available to A. bahrelghazali were grasses and sedges, neither of which is usually considered as standard great ape fare. The results suggest an early and fundamental shift in hominin dietary ecology that facilitated the exploitation of new habitats.

RATIONALE: Stable isotope analysis of archaeological charred plants has become a useful tool for interpreting past agricultural practices and refining ancient dietary reconstruction. Charred material that lay buried in soil for millennia, however, is susceptible to various kinds of contamination, whose impact on the grain/seed isotopic composition is poorly understood. Pre-treatment protocols have been adapted in distinct forms from radiocarbon dating, but insufficient research has been carried out on evaluating their effectiveness and necessity for stable carbon and nitrogen isotope analysis. METHODS: The effects of previously used pre-treatment protocols on the isotopic composition of archaeological and modern sets of samples were investigated. An archaeological sample was also artificially contaminated with carbonates, nitrates and humic acid and subjected to treatment aimed at removing the introduced contamination. The presence and removal of the contamination were investigated using Fourier transform infrared spectroscopy (FTIR) and δ(13)C and δ(15)N values. RESULTS: The results show a ca 1‰ decrease in the δ(15)N values of archaeological charred plant material caused by harsh acid treatments and ultra-sonication. This change is interpreted as being caused by mechanical distortion of the grains/seeds rather than by the removal of contamination. Furthermore, specific infrared peaks have been identified that can be used to detect the three types of contaminants studied. We argue that it is not necessary to try to remove humic acid contamination for stable isotope analysis. The advantages and disadvantages of crushing the grains/seeds before pre-treatment are discussed. CONCLUSIONS: We recommend the use of an acid-only procedure (0.5 M HCl for 30 min at 80 °C followed by three rinses in distilled water) for cleaning charred plant remains. This study fills an important gap in plant stable isotope research that will enable future researchers to evaluate potential sources of isotopic change and pre-treat their samples with methods that have been demonstrated to be effective.

High resolution incremental isotopic analysis of the dentine from early forming teeth, especially first molars (M1s), provides a means to assess the effects of poor childhood nutrition and healthcare on individuals in an assemblage where there are no infants to study. This approach is applied to an 18th and 19th century cemetery population associated with St Saviour's Almshouse burial ground in Southwark, London, to assess whether, or how, early dietary history, including weaning age, influenced health and nutritional status. The results show a general pattern in which non-breast milk foods were introduced before or by 6 months of age, as indicated by elevated δ(15) N during this period. Almost all individuals for which we also have second molar (M2) records, showed lower δ(15) N values from a very young age (>1 year) until approximately 8-10 years, compared to adult values. The overall results show a significant difference in δ(1) (3) C (p = 0 to 4sf, F = 17.327) and a weaker statistical difference in δ(15) N between males and females (p = 0.019, F = 5.581). One possible cause of this is a difference in the diet of males and females early in life, or alternatively, a greater susceptibility of males to nutritional deprivation compared to females. The latter argument is strengthened by a significant difference in the incidence of enamel hypoplasia between the males and females, with 7.7% of male teeth showing defects, compared to 3.9% of females.

Intra-tooth stable isotope analysis of dentine provides a more sensitive means to examine infant and childhood life history in the past than conventional cross-sectional analyses that rely on age-at-death. In addition, reconstructions of early diet and life history using an intra-tooth approach circumvent potential problems associated with mortality bias, which may operate strongly during infancy and childhood. We present new intra-tooth stable carbon and nitrogen isotope profiles in dentine collagen of early forming permanent teeth in a sample of adults from the Medieval Nubian site of Kulubnarti. We interpret the profiles in terms of weaning behavior and dietary history, and we compare profiles generated from first molars and canines to explore the degree to which these tooth types correspond. We then compare the profiles to the occurrence of linear enamel hypoplasia to assess the relationship between the timing of the weaning process and stress events. Finally, we compare the longitudinal profiles to cross-sectional stable isotope data obtained from rib collagen to investigate how life histories might differ between those that survived into adulthood and those that did not. Results suggest that canine and first molar profiles are in broad agreement, that hypoplastic stress events occurred during rather than before or after the weaning process in our sample, and that survivors appear to have weaned earlier than the average non-survivor. We suggest that this approach may be useful for addressing the effects of selective mortality on reconstructions of early life history and the relationship between early life history and morbidity.

Historical evidence documents mass migration from Ireland to London during the period of the Great Irish Famine of 1845-52. The rural Irish were reliant on a restricted diet based on potatoes but maize, a C(4) plant, was imported from the United States of America in 1846-47 to mitigate against Famine. In London, Irish migrants joined a population with a more varied diet. To investigate and characterize their diet, carbon and nitrogen isotope ratios were obtained from bone collagen of 119 and hair keratin of six individuals from Lukin Street cemetery, Tower Hamlets (1843-54), and bone collagen of 20 individuals from the cemetery at Kilkenny Union Workhouse in Ireland (1847-51). A comparison of the results with other contemporaneous English populations suggests that Londoners may have elevated δ(15) N compared with their contemporaries in other cities. In comparison, the Irish group have lower δ(15) N. Hair analysis combined with bone collagen allows the reconstruction of perimortem dietary changes. Three children aged 5-15 years from Kilkenny have bone collagen δ(13) C values that indicate consumption of maize (C(4)). As maize was only imported into Ireland in quantity from late 1846 and 1847, these results demonstrate relatively rapid bone collagen turnover in children and highlight the importance of age-related bone turnover rates, and the impact the age of the individual can have on studies of short-term dietary change or recent migration. Stable light isotope data in this study are consistent with the epigraphic and documentary evidence for the presence of migrants within the London cemetery.

Longitudinal studies have revealed how variation in resource use within consumer populations can impact their dynamics and functional significance in communities. Here, we investigate multi-decadal diet variations within individuals of a keystone megaherbivore species, the African elephant (Loxodonta africana), using serial stable isotope analysis of tusks from the Kruger National Park, South Africa. These records, representing the longest continuous diet histories documented for any extant species, reveal extensive seasonal and annual variations in isotopic--and hence dietary--niches of individuals, but little variation between them. Lack of niche distinction across individuals contrasts several recent studies, which found relatively high levels of individual niche specialization in various taxa. Our result is consistent with theory that individual mammal herbivores are nutritionally constrained to maintain broad diet niches. Individual diet specialization would also be a costly strategy for large-bodied taxa foraging over wide areas in spatio-temporally heterogeneous environments. High levels of within-individual diet variability occurred within and across seasons, and persisted despite an overall increase in inferred C(4) grass consumption through the twentieth century. We suggest that switching between C(3) browsing and C(4) grazing over extended time scales facilitates elephant survival through environmental change, and could even allow recovery of overused resources.

Determining the diet of an extinct species is paramount in any attempt to reconstruct its paleoecology. Because the distribution and mechanical properties of food items may impact postcranial, cranial, mandibular, and dental morphologies related to their procurement, ingestion, and mastication, these anatomical attributes have been studied intensively. However, while mechanical environments influence skeletal and dental features, it is not clear to what extent they dictate particular morphologies. Although biomechanical explanations have been widely applied to extinct hominins in attempts to retrodict dietary proclivities, morphology may say as much about what they were capable of eating, and perhaps more about phylogenetic history, than about the nature of the diet. Anatomical attributes may establish boundary limits, but direct evidence left by the foods that were actually (rather than hypothetically) consumed is required to reconstruct diet. Dental microwear and the stable light isotope chemistry of tooth enamel provide such evidence, and are especially powerful when used in tandem. We review the foundations for microwear and biogeochemistry in diet reconstruction, and discuss this evidence for six early hominin species (Ardipithecus ramidus, Australopithecus anamensis, Au. afarensis, Au. africanus, Paranthropus robustus, and P. boisei). The dietary signals derived from microwear and isotope chemistry are sometimes at odds with inferences from biomechanical approaches, a potentially disquieting conundrum that is particularly evident for several species.

The African elephant (Loxodonta africana) is a large-bodied, generalist herbivore that eats both browse and grass. The proportions of browse and grass consumed are largely expected to reflect the relative availability of these resources. We investigated variations in browse (C(3) biomass) and grass (C(4)) intake of the African elephant across seasons and habitats by stable carbon isotope analysis of elephant feces collected from Kruger National Park, South Africa. The results reflect a shift in diet from higher C(4) grass intake during wet season months to more C(3) browse-dominated diets in the dry season. Seasonal trends were correlated with changes in rainfall and with nitrogen (%N) content of available grasses, supporting predictions that grass is favored when its availability and nutritional value increase. However, switches to dry season browsing were significantly smaller in woodland and grassland habitats where tree communities are dominated by mopane (Colophospermum mopane), suggesting that grasses were favored here even in the dry season. Regional differences in diet did not reflect differences in grass biomass, tree density, or canopy cover. There was a consistent relationship between %C(4) intake and tree species diversity, implying that extensive browsing is avoided in habitats characterized by low tree species diversity and strong dominance patterns, i.e., mopane-dominated habitats. Although mopane is known to be a preferred species, maintaining dietary diversity appears to be a constraint to elephants, which they can overcome by supplementing their diets with less abundant resources (dry season grass). Such variations in feeding behavior likely influence the degree of impact on plant communities and can therefore provide key information for managing elephants over large, spatially diverse, areas.

It is well-known that pigs (Sus scrofa) were domesticated very early in Neolithic China, but far less is known about the processes by which pig husbandry intensified so that pork became the most important animal protein for humans are less clear. Here, we explore pig feeding practices using the carbon and nitrogen isotope composition of bone collagen, focusing on developments in pig husbandry during the Yangshao period (7000-5000 BP) in the middle Yellow River region of China, and at the site of Xipo (5800-5000 BP) in particular. The results show that the diets of domestic pigs at Xipo were dominated by millet foods. Comparisons with other Yangshao sites in the region show a trend of increasing millet foddering for pigs throughout the Yangshao period. These results, and comparisons of the isotopic data for pigs against those for humans from the Xipo cemetery (5300-5000 BP), suggest that pigs were closely managed by humans. The evidence points to an intensification of Neolithic pig husbandry in the middle Yellow River region from this period.
