RESUMO

Recently, type H vessels were reported to couple angiogenesis and osteogenesis during osteoclastogenesis, and tartrate-resistant acid phosphatase (Trap)+ preosteoclasts were found to secrete increased PDGF-BB to promote type H vessel formation. Therefore, utilization of type H vessels may be a strategy to treat diseases involving bone loss. In the present study, we found that nuciferine, a natural bioactive compound, has various effects, including inhibiting osteoclastogenesis and promoting type H vessel formation. Nuciferine inhibited osteoclastogenesis and bone resorption but increased the relative number of Trap+ preosteoclasts. Nuciferine restrained the expression of osteoclast-specific genes and proteins, promoted PDGF-BB production and potentiated related angiogenic activities by inhibiting the MAPK and NF-κB signaling pathways in vitro. We confirmed the bone-protective effects of nuciferine in ovariectomized mice and found that nuciferine treatment increased the PDGF-BB concentration and the number of type H vessels in the femur. In conclusion, our results demonstrated that nuciferine can decrease multinucleated osteoclast formation and promote type H vessel formation through preservation of Trap+ preosteoclasts via inhibition of the MAPK and NF-κB signaling pathways and may be an excellent agent for the treatment of diseases involving bone loss.

Established RAW264.7 cell lines for osteoclastic differentiation has been widely engaged in bone homeostasis research, however, the efficacy of RANKL independently stimulating has rarely been defined, because protocols were usually developed and modified by various laboratories. Otherwise, problematic issues are also lie in the cell's seeding density, RANKL stimulating time point, and distinguishing osteoclastogenesis ability of RANKL-treated RAW264.7 cells. Therefore, in the current study, we examined the efficacy of various concentrations of RANKL-treated RAW264.7 for its osteoclastic differentiation with or without pretreated other costimulators such as: LPS and/or M-CSF. The oteoclastogenesis ability of RANKL-treated RAW264.7 cells was demonstrated by bone resorption pit, F-actin, and osteoclastogenesis specific marker studies. Besides that, through tartrate-resistant acid phosphatase (TRAP) staining, we clarified to start the treatment with 30 ng/ml RANKL at 12 hr after seeded RAW264.7 with the density of 6.25 × 10 3 cells/cm 2 manifested an significantly increased number of multinucleated osteoclastic cells. Overall, our results establishing an optimal method for RANKL independently inducing RAW 264.7 cell osteoclastic differentiation, which could efficiently generate osteoclasts in vitro for significant advances in our understanding of bone biology.

Different from five-membered rhodamine spirolactam (Rh-OH), a new six-membered spiro-rhodamine probe (SRh-OH) bearing urea structure has been developed in this paper. Compared with five-membered Rh-OH, six-membered SRh-OH exhibited higher selectivity and sensitivity to Cu2+ in aqueous solution. Upon addition of 10 equiv. Cu2+, the molar extinction coefficient of SRh-OH at 563 nm can be up to 4.73 × 104 Lmol-1cm-1. In the range of Cu2+ from 0 to 28 µM, there was an excellent linear relationship between the absorption or emission intensity and Cu2+ concentration. The detection limit of SRh-OH was as low as 26 nM (S/N = 3). The reversible binding mode of SRh-OH with Cu2+ was further confirmed by S2- and EDTA addition. Bio-imaging showed SRh-OH can map the distribution of Cu2+ not only in mitochondria but also in lysosomes of Hela cells.

RESUMO

Renal cell carcinoma (RCC) associated with Xp11.2 translocation/transcription factor E3 (TFE3) gene fusion is a rare and independent subtype of RCC included in the classification of MiT (microphthalmia-associated transcriptional factor) family translocation RCC. Herein, we report an adult case of Xp11.2 translocation RCC, and review the relevant literature to improve our understanding of the pathogenesis, epidemiology, clinical manifestations, diagnosis, differential diagnosis, treatment, and other aspects of the disease.

Research on coal desulfurization is very important for economic, social, and environmentally sustainable development. In this study, three batches of shake flask experiments were conducted for coal bio-desulfurization using Acidithiobacillus ferrooxidans to explore the relationship between microbial nutrients (iron-free M9 K medium) supply and coal bio-desulfurization efficiency. The results showed that the removal rates of pyritic sulfur and total sulfur from coal effectively increased following reintroduction of coal into the filtrate from previous batch. The removal rates of pyritic sulfur and total sulfur were 55.6% and 10.0%, 77.1% and 16.1%, and 86.5% and 28.2%, respectively, in the three batch experiments without iron-free M9 K medium addition. In contrast, the removal rates of pyritic sulfur and total sulfur reached 87.5% and 28.2%, 89.1% and 31.6%, and 92.0% and 29.1%, respectively, in the three batch experiments with 6.7% iron-free M9 K medium addition. However, addition of excessive iron-free M9 K medium was detrimental to coal bio-desulfurization because of the synthesis of jarosite (MFe3(SO4)2(OH)6, M = K+, NH4+) and gypsum (CaSO4·2H2O), which further declined the pyritic sulfur bio-oxidation efficiency and total sulfur removal efficiency.

RESUMO

To study the C21 steroids of the stems of Marsdenia tenacissima (Roxb.) Wight et Arn, various chromatography methods were used for the isolation of the constituents and their structures were identified by spectral analysis. Eight C21 steroids were isolated from the CHCl3 extract, which were identified as 11alpha-O-tigloyl-17beta-tenacigenin B (1), 17beta-tenacigenin B (2), tenacigenoside A (3), 11alpha-O-2-methylbutyryl-12beta-O-acetyl tenacigenin B (4), tenacissoside H (5), marsdenoside A (6), tenacissoside G (7), and tenacissoside I (8). Among them, compound 1 is a new compound.
