RESUMO

Life molecules' distributions in live systems construct the complex dynamic reaction networks, whereas it is still challenging to demonstrate the dynamic distributions of biomolecules in live systems. Herein, we proposed a dynamic analysis strategy via sequence-structure bispecific RNA with state-adjustable molecules to monitor the dynamic concentration and spatiotemporal localization of these biomolecules in live cells based on the new insight of fluorescent RNA (FLRNA) interactions and their mechanism of fluorescence enhancement. Typically, computer-based nucleic acid-molecular docking simulation and molecular theoretical calculation have been proposed to provide a simple and straightforward method for guiding the custom-design of FLRNA. Impressively, a novel FLRNA with sequence and structure bispecific RNA named as a structure-switching aptamer (SSA) was introduced to monitor the real-time concentration and spatiotemporal localization of biomolecules, contributing to a deeper insight of the dynamic monitoring and visualization of biomolecules in live systems.

To evaluate the effects of nitrite on the oxidative damage of blood cells of Grass Carp Ctenopharyngodon idella, the isolated hemocytes were exposed to nitrite (0, 1, 10, or 100 mg/L) for up to 24 h. Hemoglobin (Hb) and methemoglobin (MetHb) concentrations, reactive oxygen species (ROS) and malondialdehyde (MDA) levels, mitochondrial membrane potential (∆Ψm), and antioxidant enzyme activity were assayed to assess hematological parameters and the antioxidant defense mechanism. Results showed a remarkable decrease in Hb concentration with increasing nitrite concentration after a 24-h exposure, while the MetHb concentration increased significantly in nitrite exposure groups. The levels of ROS, ∆Ψm, and MDA increased to varying degrees with increases in nitrite exposure concentration and time. The total antioxidant capacity, catalase (CAT) activity, glutathione peroxidase (GPx) activity, and glutathione content showed a trend of rising initially and then decreasing with prolonged exposure time. Superoxide dismutase (SOD) activity was higher in the 1-mg/L nitrite exposure group and lower in the 100-mg/L group than in the control. The relative messenger RNA expression ratios of cat, sod1, and gpx were up-regulated significantly in the 1- and 10-mg/L groups and then declined in the 100-mg/L group. Therefore, it can be concluded that nitrite exposure activates the antioxidant defense mechanism of Grass Carp hemocytes and that the balance of oxidant-antioxidant homeostasis will be undermined by higher nitrite doses or longer exposure periods.

Selenium (Se), an essential component of deiodinases (DIOs), regulates the contents of thyroid hormones and thus improves animal growth. To explore the influences of selenium supplementation on fish growth metabolism, a total of 270 healthy grass carp (Ctenopharyngodon idella) were divided into three groups and feed three graded dietary selenium (0.141, 0.562, and 1.044 mg Se/kg) levels. The results showed that after 60-day feeding, dietary selenium improved the final body weight and specific growth rate (SGR) of grass carp. The hepatic DIO activities in selenium-supplemented groups were higher than those in control group. A significant increase in triiodothyronine (T3), free triiodothyronine (FT3), and thyroid-stimulating hormone (TSH) levels was accompanied by a decrease in the contents of thyroxine (T4) and free thyroxine (FT4) in selenium-supplemented groups. The histopathological observation of thyroid suggested that selenium deficiency resulted in hypertrophy of follicular epithelial cells. Moreover, the gene relative expression levels of dio1, dio2, and dio3 showed an increasing trend with the rising concentration of dietary selenium. The transcription levels of HPT axis-related genes (crh, tsh-ß, ttr, tr-s, tpo, nis) and GH/IGF1-related genes (gh, ghr, igf1, igf1r) were significantly upregulated in selenium-supplemented groups. No significant differences in the above indicators were observed between 0.562 and 1.044 mg Se/kg diet group except T3 content and dio1 relative expression ratio. These results indicate that dietary selenium supplementation improves the hepatic DIO activities and thyroid hormone metabolism and regulates the transcription levels of HPT and GH/IGF axis-related genes, which may be responsible for the growth promotion in grass carp.

Constructing nanostructures with high structural stability and ultrafast electrochemical reaction kinetics as anodes for sodium-ion batteries (SIBs) is a big challenge. Herein, the robust 2D VS2 / Ti3 C2 Tx MXene nanostructures with the strong Ti─S covalent bond synthesized by a one-pot self-assembly approach are developed. The strong interfacial interaction renders the material of good structural durability and enhanced reaction kinetics. Meanwhile, the enlarged and few-layered MXene nanosheets can be easily obtained according to this interaction, providing a conductive network for sufficient electrolyte penetration and rapid charge transfer. As predicted, the VS2 /MXene nanostructures exhibit an extremely low sodium diffusion barrier confirmed by DFT calculations and small charge transfer impedance evidenced by electrochemical impedance spectroscopy (EIS) analysis. Therefore, the SIBs based on the VS2 /MXene electrode present first-class electrochemical performance with the ultrahigh average initial columbic efficiency of 95.08% and excellent sodium-ion storage capacity of 424.6 mAh g-1 even at 10 A g-1 . It also shows an outstanding sodium-ion storage capacity of 514.2 mAh g-1 at 1 A g-1 with a capacity retention of nearly 100% within 500 times high-rate cycling. Such impressive performance demonstrates the successful synthesis strategy and the great potential of interfacial interactions for high-performance energy storage devices.

Sodium-ion batteries (SIBs) suffer from limited ion diffusion and structural expansion, generating the urgent demand for Na+ accommodable materials with promising architectures. In this work, the rational exploration for Co4S3 nanoparticles confined in an MnS nanorod-grafted N, S-codoped carbon polyhedron (Co-Mn-S@N-S-C) is achieved by the in situ growth of MOF on MnO2 nanorod along with the subsequent carbonization and sulfurization. Benefiting from the distinctive nanostructure, the Co-Mn-S@N-S-C anode delivers excellent structural stability, resulting in prolonged cycling stability with a capacity retention of 90.2% after 1000 cycles at 2 A g-1. Moreover, the reaction storage mechanism is clarified by the in situ X-ray diffraction (XRD) and transmission electron microscopy (TEM) measurements. The results indicate that properly designed electrode materials have huge potential applications for highly efficient energy storage devices.

RESUMO

Advanced architecture and rational design of electrode materials for electrochemical sodium-ion storage are well developed by researchers worldwide. MXene-based materials are considered as one of the most potential electrode materials for sodium-ion-based devices, such as sodium-ion batteries (SIBs), sodium-sulfur batteries (SSBs), and sodium-ion capacitors (SICs), because of the excellent physicochemical characteristics of MXenes. Here, in this review, the recent research work and progress, both theoretical and experimental, on MXene-based materials including pure MXenes and MXene-based composites in application of SIBs, SSBs, and SICs are comprehensively summarized. The sodium storage mechanisms and the effective methods to enhance the electrochemical performance are also discussed. Finally, the current critical challenges and future research directions on the development of these MXene-based materials for electrochemical sodium-ion storage are presented.

The microstructure and mechanical properties of a Mg-6Al-1Zn-0.9Sn alloy processed by equal channel angular pressing (ECAP) at temperatures of 250 °C and 300 °C were investigated. It was found that the refinement of the microstructure was very dependent on the processing temperature. The main reason for the difference in grain refinement was the precipitation of secondary-phase particles. Texture information obtained by electron back-scatter diffraction (EBSD) showed the gradual formation of a 45° texture during the ECAP process, while the maximum intensity was different for processing temperatures at 250 °C and 300 °C. By calculating the contribution from different strengthening mechanisms, it was found that a 45° texture had a huge influence on grain boundary strengthening and thus the yield strength.

Fundamental understanding of the transport properties within nanoparticle composite electrolytes is necessary for the development of next-generation electrochemical devices. Herein, the effect of surface-modified silica nanoparticles with aminophenyl, amide, and sulfonate functional groups (AP-SiO2, AM-SiO2, and SU-SiO2) on the ion transport properties of composite electrolytes is systematically investigated. The competition between surface repulsive and attractive interactions of nanoparticles is reflected in the nature of the morphology and particle network in electrolytes, further affecting the ionic conductivity of electrolytes and diffusion coefficient of ions. The obvious decrease is observed in the AP-SiO2-based system because of the severe agglomeration of nanoparticles. By contrast, the AM-SiO2 and SU-SiO2 form the regular particle network structure and accelerate the salt dissociation in electrolytes, thereby providing an effective ion transport pathway and more mobile ions for conduction, respectively. Consequently, the composite systems with AM-SiO2 and SU-SiO2 deliver remarkable enhancement in the ion transport properties.

It has been well known that electric pulse can be utilized to enhance the plasticity of metals, which is attributed to the change of dislocation dynamics, e.g., localized planar slip to homogeneous wavy slip. Here, we show another effect of pulse current, which facilitates texture weakening through room-temperature dynamic recrystallization and additionally improve the plasticity of a polycrystalline Mg-3Al-1Sn-1Zn alloy. By conducting a tensile test under electrical pulse, we found that the peak flow stress and fracture strain depend strongly on current density. As peak current densities increases, the flow stress drops and the fracture strain increases. Our Electron Backscatter Diffraction results suggest that dynamic recrystallization occurs at room temperature, which develops a weakened texture. Our work provides a new insight into electroplasticity mechanism in Mg alloys.

Gel electrolytes are promising candidates for dye-sensitized solar cells (DSSCs) and other devices, but the ways to obtain stable gels always result in sacrifice of their ionic conductivity. This contradiction seriously limits the practical application of gel electrolytes. Herein, a new design strategy using rich carboxylic group-modified silica nanoparticles (COOH-SiO2) with a branched, well-organized framework to develop ionic liquid-based gel electrolytes possessing high conductivity is demonstrated. The branched network of COOH-SiO2 and the strong interaction in electrolytes result in the effective solidification of ionic liquids. Moreover, adding COOH-SiO2 to ionic liquid electrolytes contributes to salt dissociation, decreases the activation energy, and improves the charge transport and recombination characteristics at the electrolyte/electrode interface. DSSCs fabricated with COOH-SiO2 nanoparticles deliver a higher short-circuit photocurrent density (Jsc) than the reference cell. A maximum efficiency of 8.02% with the highest Jsc value of 16.60 mA cm-2 is obtained for solar cells containing 6 wt% COOH-SiO2.

The high-temperature superplastic deformation behavior of rolled Mg-8Al-2Sn (AT82) and Mg-8Al-1Sn-1Zn (ATZ811) alloys were investigated in this study. During tensile deformation at 573 K, no obvious grain growth occurred in both alloys, because of the high-volume fraction of second phases located at grain boundaries. Meanwhile, texture weakening was observed, suggesting that grain boundary sliding (GBS) is the dominant superplastic deformation mechanism, which agreed well with the strain rate sensitivity (m) and the activation energy (Q) calculations. The microstructural evolution during tensile deformation manifested that there were more and larger cavities in AT82 than ATZ811 during high-temperature tensile deformation. Therefore, superior superplasticity was found in the ATZ811 alloy that presented a tensile elongation of ~510% under a strain rate of 10-3 s-1 at 573 K, in contrast to the relatively inferior elongation of ~380% for the AT82 alloy. Meanwhile, good tensile properties at ambient temperature were also obtained in ATZ811 alloy, showing the ultimate tensile strength (UTS) of ~355 MPa, yield strength (YS) of ~250 MPa and elongation of ~18%. Excellent mechanical performance at both ambient and elevated temperatures can be realized by using economical elements and conventional rolling process, which is desirable for the industrial application of Mg alloy sheets.

Two types of ionic liquids (ILs), 1-(3-hexenyl)-3-methyl imidazolium iodide and 1-(3-butenyl)-3-methyl imidazolium iodide, are synthesized by introducing an unsaturated bond into the side alkyl chain of the imidazolium cation. These new ionic liquids exhibit high thermal stability and low viscosity (104 cP and 80 cP, respectively). The molecular dynamics simulation shows that the double bond introduced in the alkane chain greatly changes the molecular system space arrangement and diminishes the packing efficiency, leading to low viscosity. The low viscosity of the synthesized ionic liquids would enhance the diffusion of redox couples. This enhancement is detected by fabricating dye-sensitized solar cells (DSSCs) with electrolytes containing the two ILs and I2 . The highest efficiency of DSSCs is 6.85 % for 1-(3-hexenyl)-3-methyl imidazolium iodide and 5.93 % for 1-(3-butenyl)-3-methyl imidazolium iodide electrolyte, which is much higher than that of 5.17 % with the counterpart 1-hexyl-3-methyl imidazolium iodide electrolyte.

Strategies for environmentally friendly reutilization of waste biomass are highly desirable nowadays. Meanwhile, seeking Pt-free electrocatalysts for triiodide reduction with both high catalytic activity and low cost is critical for the development of dye-sensitized solar cells (DSCs). In the study, heteroatom tri-doped porous carbons (TPCs) were prepared via carbonization of a typical food waste (fish waste) and explored as a counter electrode (CE) in a DSC. The as-prepared carbon materials possessed a porous structure with a large BET surface area of 2933 m2 g-1, while being simultaneously naturally doped by three heteroatoms (N, P and S). More importantly, the resultant N, P, S-tri-doped porous carbon exhibited outstanding electrochemical activity towards triiodide reduction with good stability. Moreover, the DSC with the optimized TPC electrode showed a power conversion efficiency of 7.83%, which is comparable to the device with a costly Pt-based CE (8.34%), measured under one sun illumination (AM 1.5G). This work demonstrates that carbonization of fish waste offers a cost-effective approach to prepare multifunctional carbon materials for advanced energy applications.

In this work, a unique approach is developed to generate photoswitchable and water-soluble fluorescent nano-aggregates. Initially, a new light-controlled diarylethene-dansyl dyad DAE 1 is formed by linking two dansyl fluorophores covalently to a symmetrical dithienylethene backbone, whose photophysical properties can be reversibly switched by optical stimuli. Subsequently, the water insolubility of the molecular switch 1 is overcome by incorporating it into the bilayer of liposome DPPC (1,2-dihexadecanoyl-sn-glycero-3-phosphocholine) in water. This strategy creates stable fluorescent nano-aggregates OF-1@DPPC (≈25 nm diameter) that are soluble in an aqueous medium. The nano-aggregates OF-1@DPPC retain and even improve the photoswitchable fluorescence properties of DAE 1. More importantly, OF-1@DPPC exhibits a remarkable photostability and fatigue resistance after 5 cycles of irradiation with UV and visible light, which is crucial for its practical application.

A simple yet general swelling-deswelling microencapsulation strategy has been developed to achieve well dispersed and intimately passivated crystalline organic-inorganic perovskites nanoparticles within polymer matrixes and results in a series of highly luminescent CH3 NH3 PbBr3 (MAPbBr3 )-polymer composite films with unprecedented water and thermal stabilities and superior color purity.

OBJECTIVE: To screen and identify zebrafish mutants with erythropoiesis defects by N-ethyl-N-nitrosourea (ENU) mutagenesis and large-scale forward genetic screening using beta e 1 as the marker. METHODS: The chemical mutagen ENU was used to treat healthy wild-type male fish (AB strain, F0). The surviving ENU-treated fish were mated with wild-type female fish to generate F1, and further F2 family was generated by F1 family intercross. The adult F2 fish were intercrossed within each F2 family and the resulting F3 embryos from each crossing were subjected to whole mount in situ hybridization (WISH) with the beta e 1 probe. Mutagenesis was performed by treating the male zebrafish with ENU to induce mutations in pre-meiotic germ cells to generate the founders, which were outcrossed to obtained the F1 fish. The F1 fish from different founders were mated to generate the F2 families. F3 embryos from the sibling cross in the F2 family were examined by whole mount in situ hybridization using beta e 1-globin probe. The putative mutants were then characterized with different hematopoiesis markers. RESULTS AND CONCLUSION: We identified 4 beta e 1-deficient mutants with erythropoiesis defects, including two with specific erythiod lineage defects and two with concurrent lymphopoiesis defects.
