RESUMO

The present work focus on plant extracts mediated synthesis of silver chloride nanoparticles (AgCl-NPs). The AgCl-NPs were synthesized using the plant leaf extract of Origanum-majorana by one step green synthesis method. The characterization of as prepared AgCl-NPs were done by various analytical techniques such as UV-Vis spectroscopy, X-ray diffraction (XRD) and fourier transform infrared spectroscopy (FTIR). The morphology and composition of AgCl-NPs was confirmed by scanning electron microscopy analysis (SEM) and energy dispersive X-ray spectroscopy (EDX) analysis, respectively. Further, photocatalytic activity of as prepared AgCl-NPs observed by elimination of E. coli bacteria from contaminated water under solar light irradiation and it was observed that AgCl-NPs possess a good photocatalytic activity performance against E. coli bacteria.

Recently, graphitic carbon nitride has been investigated as a promising photocatalyst for organic dye degradation application. In this study, a facile strategy to synthesise silver nanoparticles (AgNPs) doped graphitic carbon nitride (GCN-Ag) has been reported. The characterisation study of the asprepared samples was performed using various analytical techniques. The X-ray diffraction (XRD) and fourier transform infrared spectroscopy (FTIR) revealed that the structure of pure graphitic carbon nitride (GCN-Pure) partly changed on the addition of the AgNPs. The diffused reflectance spectra (DRS) unveiled a significant red shift in the absorption edge of GCN-Ag. The scanning electron microscopy (SEM) analysis revealed that the morphological aspects of GCN-Pure changed on the addition of AgNPs. Further the as-prepared samples have been compared for their degradation activity towards organic dye pollutants including methylene blue, crystal violet and rose bengal. The phenomenon of the better separation of photogenerated charge carriers was attributed to the better photoactivity in the case of GCN-Ag than GCN-Pure. In addition to it the reusability experiment of GCN-Ag revealed that the catalyst remained highly stable after the three cyclic runs of photodegradation experiment.

A set of novel quinolone-triazole conjugates (12-31) were synthesized in three steps in good yields starting from 2-phenylquinoline-4-carboxylic acid. All the intermediates, as well as the final 1,2,4-triazolyl quinolines were fully characterized by their detailed spectral analysis utilizing different techniques such as IR, 1H NMR, 13C NMR, and finally mass spectrometry. All the synthesized compounds were evaluated in vitro for their potential antibacterial activity and their preliminary safety profile was assessed through cytotoxicity assay. Additionally, six selected conjugates were evaluated for their antioxidative properties on the basis of density functional theory calculations, using radical scavenging assay (DPPH) and cellular antioxidant assay. The reported results encourage further investigation of selected compounds and are shading light on their potential pharmacological use.

Lithium-sulfur batteries (LSBs) show promise for achieving a high energy density of 500 W h kg-1, despite challenges such as poor cycle life and low energy efficiency due to sluggish redox kinetics of lithium polysulfides (LiPSs) and sulfur's electronic insulating nature. We present a novel 2D Ti3C2 Mxene on a 2D graphitic carbon nitride (g-C3N4) heterostructure designed to enhance LiPS conversion kinetics and adsorption capacity. In a pouch cell configuration with lean electrolyte conditions (∼5 µL mg-1), the g-C3N4-Mx/S cathode exhibited excellent rate performance, delivering ∼1061 mA h g-1 at C/8 and retaining ∼773 mA h g-1 after 190 cycles with a Coulombic efficiency (CE) of 92.7%. The battery maintained a discharge capacity of 680 mA h g-1 even at 1.25 C. It operated reliably at an elevated sulfur loading of 5.9 mg cm-2, with an initial discharge capacity of ∼900 mA h g-1 and a sustained CE of over 83% throughout 190 cycles. Postmortem XPS and EIS analyses elucidated charge-discharge cycle-induced changes, highlighting the potential of this heterostructured cathode for commercial garnet LSB development.

Film-forming lotions, precast films and adhesive patches containing testosterone (T) were prepared by compounding vinylic, acrylic and cellulosic polymers with a variety of excipients in order to achieve distribution of T in domains of heterogeneity within multicomponent matrices. The feasibility of this approach in achieving episodic transdermal delivery of testosterone (T) was investigated. Composition-dependent differences in extent of in vitro drug release and periodicity were observed. Representative formulations showing the most pronounced episodic T release in vitro were tested in female rats. Whereas intravenously administered T decayed exponentially, three maxima of T in serum were observed upon application of selected formulations. Thus, peak serum concentrations of 240, 36, and 29 ng/dL were observed at 0.2, 5, and 16.8 h after application of the preferred lotion formulation, and 89, 65, and 64 ng/dL at 1, 16.4, and 48.8 h after patches. Deconvolution, noncompartment pharmacokinetic analysis and multiple peak fitting also indicated episodicity. These results suggest the feasibility of using transdermal systems for pulsatile T delivery in a variety of clinical applications, including hormone supplementation and male contraception.

BACKGROUND: Varied reports suggest a contentious relationship of bladder malignancy with pioglitazone in patients with type 2 diabetes. AIM: To study an association (prevalence and predictors) of bladder malignancy with pioglitazone therapy in Asian-Indian type 2 diabetes patients. METHOD: In this observational multicenter study, type 2 diabetic patients attending out-patient diabetes-clinic were evaluated. A detailed history of anti-diabetic medication, dose, duration, pioglitazone usage, time since initiation of pioglitazone, physical examination, biochemical tests and details pertaining to prevalent neuropathy, retinopathy and nephropathy were recorded. Details of bladder cancer or any malignancy (if present), time since diagnosis, risk factors for bladder cancer and histopathology records were noted. The study cohort was divided into two groups-pioglitazone ever users (Group A) and never users (Group B). RESULTS: A total of 8000 patients were screened out of which 1560 were excluded. Among 6440 included patients, 1056 (16.3%) patients were in group A and 5384 (83.6%) group B. Patients on pioglitazone were older (59.1 vs 57.7 years, p < 0.001), had longer duration of diabetes (12.7 vs 10.6 years, p < 0.001) with poor glycemic control (HbA1c 8.5 vs 8.3%, p < 0.01). A total of 74 patients had prevalent bladder cancer [16 (1.5%) in Group A and 58 in Group B (1.0%)]. Prevalent bladder cancer was not significantly greater in ever-users (odds ratio OR = 1.29, 95% confidence interval CI, 0.83-2.00) compared to never-users (odds ratio OR = 0.94, 95% confidence interval CI, 0.834-1.061) of pioglitazone (p = 0.207). However, history of hematuria in pioglitazone-users; while older age (>58 year), history of smoking and hematuria in the whole cohort were significant associated with bladder cancer. In the entire study cohort, 254 patients; 3.5% of males (128 out of 3575) and 4.6% of females (126 out of 2713) developed any malignancy. Age was significantly associated with prevalent malignancy in people with diabetes (odds ratio OR 1.036, 95% confidence interval CI: 1.022-1.051, p = 0.00) on multivariate forward regression. CONCLUSION: Pioglitazone use in Asian-Indians is not associated with an increased bladder cancer risk. However, pioglitazone should be restricted in individuals with history of hematuria. Age more than 58 years is a significant risk factor for development of any malignancy, particularly bladder cancer.

Malaria is one of the most common parasitic infection in India. The diagnosis largely depends on peripheral blood smear examination. Newer diagnostic methods like various antigen detection assays are now in use for prompt diagnosis and treatment. This study was done to determine the effectiveness of Diagnos Malaria Stix (antigen detection) assay in diagnosis of malaria. This involves detection of PfHRP-2 antigen and P.V. specific pLDH antigen. 162 patients with signs and symptoms of malaria included in the study. Leishman stained blood smear examination was done for all patients. Commercially available Diagnos Malaria Stix assay was used. Diagnos Malaria Stix showed sensitivity, specificity positive and negative predictive values of 100% each while Sensitivity, specificity, positive and negative predictive values of Leishman stained blood smear examination were 45.45%, 100%, 100% and 92% respectively.

A green and efficient approach for efficient nanohybrid photocatalysts in extending the light response to the visible spectrum is a hot research topic in sustainable energy technologies. In this work, novel Au-TiO2@m-CN nanocomposite was synthesized using hard template of cubic ordered mesoporous KIT-6 via the nanocasting process. The as-prepared Au-TiO2@m-CN nanohybrids exhibit enhanced photocatalytic activities with improved stability and reusability using methyl orange dye. The enhanced photocatalytic performance is a result of the conjugated effect of catalytic active Au and TiO2 nanoparticles supported on highly efficient visible light sensitizer, graphitic carbon nitride (m-CN or g-C3N4), and ordered mesoporous morphology. Besides, the sensing performance of Au-TiO2@m-CN nanohybrids was also tested for the detection of amine gases, wherein a significant response was reported for triethylamine at low operating temperatures. This study reveals a simple and scalable methodology to design and develop next generation of layered mesoporous materials for multifunctional applications.

OBJECTIVE: To identify the predictors of pre-operative anxiety in school-going children in India. METHODS: A prospective observational study was done at a tertiary care teaching hospital. Children aged 7-12 y scheduled for elective surgery and their parents were enrolled. Pre-operative anxiety in children and parents was assessed using STAI-C and STAI-P questionnaires respectively. The child's anxiety in pre-op room, at the time of parental separation and induction of anesthesia were assessed using m-YPAS scale. Possible predictors of anxiety under 'individual', 'social' and 'healthcare' domains were analyzed using logistic regression. The quality of mask induction of anesthesia was assessed using Induction Compliance Checklist. RESULTS: Sixty children completed the study successfully. Receiver operating characteristic (ROC) curve indicted mYPAS score ≥ 30 (observed score) to identify children with high anxiety as identified by STAI-C ≥37 (self-reported score). The incidence of high anxiety escalated from pre-operative time (48%) to the time of parental separation (72%) and increased further at anesthetic induction (95%). Parental anxiety (p = 0.03) and socioeconomic background (p = 0.03) were significant predictors of the child's pre-operative anxiety. The presence of >4 people at induction (p = 0.002), but not pre-operative anxiety, was found significantly more in children with poor quality of mask induction. CONCLUSIONS: The incidence of peri-operative anxiety in Indian children is significant. Parental anxiety and socioeconomic background were found significant predictors of high pre-operative anxiety in our set-up.

Development of room-temperature gas sensors is a much sought-after aspect that has fostered research in realizing new two-dimensional materials with high surface area for rapid response and low-ppm detection of volatile organic compounds (VOCs). Herein, a fast-response and low-ppm ethanol gas sensor operating at near room temperature has been fabricated successfully by utilizing cubic mesoporous graphitic carbon nitride (g-CN, commonly known as g-C3N4), synthesized through template inversion of mesoporous silica, KIT-6. Upon exposure to 50 ppm ethanol at 250 °C, the optimized Ag/g-CN showed a significantly higher response (R a/R g = 49.2), fast response (11.5 s), and full recovery within 7 s in air. Results of sensing tests conducted at 40 °C show that the sensor exhibits not only a highly selective response to 50 ppm (R a/R g = 1.3) and 100 ppm (R a/R g = 3.2) of ethanol gas but also highly reversible and rapid response and recovery along with long-term stability. This outstanding response is due to its easily accessible three-dimensional mesoporous structure with higher surface area and unique planar morphology of Ag/g-CN. This study could provide new avenues for the design of next-generation room-temperature VOC sensors for effective and efficient monitoring of alarming concern over indoor environment.

Liposome-microbubble conjugates are considered as better targeted drug delivery vehicles compared to microbubbles alone. The microbubble in the integrated drug delivery system delivers the drug intracellularly on the target, whereas the liposome component allows loading of high drug dose and extravasation through leaky vasculature. In this work, a new high yielding microbubble production method was used to prepare microbubbles for formulation of the liposome-conjugated drug delivery system. In formulation process, the prepared liposome of 200 nm diameter was attached to the microbubble surface using the avidin-biotin interaction. The analysis of the confocal scanning laser microscope images showed that approximately 8 × 108 microbubbles per millilitre (range: 2-7 µm, mean size 5 ± 0.5 µm) can be efficiently conjugated to the liposomes. The method of conjugation was found to be effective in attaching liposome to microbubbles.

Cushing's syndrome is characterized by any cause of excess cortisol in the blood and produces many physiologic consequences. Left untreated, Cushing's is associated with significant morbidity and mortality. Seventy percent of endogenous cases of Cushing's syndrome are secondary to a pituitary tumor; because of this, the primary mode of management is surgical resection of the tumor. Should hypercortisolism persist following surgical resection, further treatment options are limited. Metyrapone is an orphan medication that is often used in the diagnosis of the disease and occasionally for short-term treatment prior to surgery. Long-term treatment with metyrapone is usually discouraged due to the contradictory increase in ACTH production, acne, hirsutism, hyperkalemia, edema, and other mineralocorticoid effects. We present a patient with refractory Cushing's syndrome successfully treated for nearly 6 years with metyrapone with minimal adverse effects. This orphan medication may be a viable long-term treatment option for this difficult disease.

The aim of this research was to investigate zinc chromium ferrite (ZnCrFeO4) nanoparticles, synthesized using the sol gel technique with nanoparticle size controlled through a two-stage annealing process. Stage one was a low temperature firing which produced low quality nanocrystals with an average size of 15 nm. This was followed by a second firing stage at high temperature which enhanced the crystal quality. The nanoparticles were then coated with a bio-compatible shell to form a stable suspension in the ferrofluid carrier. The resulting nanoparticles were found by electron microscopy, atomic force microscopy and X-ray diffraction studies to have excellent crystal quality. The average size was 8.5 nm. Preliminary cell culture studies indicated the ZnCrFeO4 nanoparticles were non-toxic. The relatively high measured value of the relaxivity r2 showed that the nanoparticle coating was effective in substantially reducing aggregation and enhancing the properties of the nanoparticles associated with contrast enhancement in MRI.

OBJECTIVE: Because approximately 40% of patients with type 1 diabetes have the metabolic syndrome, we tested the hypothesis that addition of liraglutide to insulin in obese patients with type 1 diabetes will result in an improvement in plasma glucose concentrations, a reduction in hemoglobin A1c (HbA1c), a fall in systolic blood pressure, and weight loss. METHODS: This is a retrospective analysis of data obtained from 27 obese patients with type 1 diabetes treated with liraglutide in addition to insulin. Patients were also treated for hypertension. Paired t tests were used to compare the changes in HbA1c, insulin doses, body weight, body mass index, 4-week mean blood glucose concentrations (28-day insulin pump mean blood glucose), blood pressure, and lipid parameters prior to and 180 ± 14 days after liraglutide therapy. RESULTS: Mean glucose concentrations fell from 191 ± 6 to 170 ± 6 mg/dL (P = .002). HbA1c fell from 7.89 ± 0.13% to 7.46 ± 0.13% (P = .001), without an increase in frequency of hypoglycemia. Mean body weight fell from 96.20 ± 3.68 kg to 91.56 ± 3.78 kg (P<.0001). Daily total and bolus doses of insulin fell from 73 ± 6 to 60 ± 4 (P = .008) units and from 40 ± 5 to 29 ± 3 units (P = .011), respectively. Mean systolic blood pressure fell from 130 ± 3 to 120 ± 4 mm Hg (P = .020). CONCLUSION: Addition of liraglutide to insulin in obese patients with type 1 diabetes mellitus leads to improvements in glycemic control and HbA1c and to reductions in insulin dose, systolic blood pressure, and body weight.

It was envisaged to develop surface modified Guar Gum Nanoparticles (GGNP) with Folic acid (FA) charged with methotrexate (MTX) to target the colon specifically. The MTX loaded FA functionalized GGNP (MTX-FA-GGNP) have been prepared by emulsion crosslinking method. These surface modified nanoparticles were compared with unmodified MTX loaded GGNP (MTX-GGNP). The developed formulations were evaluated for size and size distribution, zeta potential, Differential Scanning Calorimetry (DSC), release profile and uptake studies. The nanoparticles have been found to have average size of 325 nm in diameter having polydispersity index (PDI) 0.177 indicating mono-disperse particles. The zeta potential of the particles was found to be -36.9 mV. The percent growth inhibition of Caco 2 cells with MTX-FA-GGNP was found to be better than MTX-GGNP indicating folate receptor mediated uptake. The MTX-GGNP protects the release of MTX in upper gastrointestinal tract while maximum release of MTX occurred in simulated colonic fluids of pH 6.8. The in vivo uptake studies revealed preferential uptake of nanoparticles formulation in the colon. These studies provide evidences that MTX-FA-GGNP holds promise to address colorectal cancer over-expressing folate receptors. This prototype formulation enjoys dual advantage of having propensity to release the drug in the colon and in the conditions of colorectal carcinoma; it could be better localized and targeted with improved therapy due to over-expression of folate receptors.

RESUMO

Biodegradable nanoparticles (NP) of average size 75 nm and composed of poly(lactic acid, PLA) were prepared by single emulsion. Upon instillation into the vagina of mice in estrus, these particles undergo retrograde transport across the cervix to the uterus. Uterus lavage conducted after instillation of NP into the vagina indicated that proinflammatory signals such as RANTES and TNF were induced in the uterine environment, which is inimical to establishment of pregnancy. These NP are under investigation for contraceptive potential.

BACKGROUND: The present study aimed to determine the occurrence of human disease-causing enteric bacteria on raw vegetables, fruits, meats, and milk products sold in Indian markets. The study further aimed to analyze antibiotic resistance rates and the presence of blaCTX-M, blaTEM, blaSHV, and blaAmpC. METHODOLOGY: Twenty-three food-borne and 23 clinical isolates were compared for antibiotic resistance rates and the presence of blaCTX-M, blaTEM, blaSHV, and blaAmpC. Swabs were taken from unwashed and washed food items, as well as from some chopped food specimens, and inoculated on appropriate culture medium. Bacterial isolates were identified, antibiotic susceptibility was performed, and bla genes were detected by PCR. RESULTS: Thirty-eight bacterial isolates were obtained from the food specimens, of which 36 (94.7%) were Gram-negative and two (5.3%) were Gram-positive bacterial species. Klebsiella pneumoniae was the most prevalent (52.6%; 20/38) bacterial species isolated, followed by Citrobacter koseri (18.4%; 7/38). In food isolates, the majority of the isolates were resistant to gentamicin (33.3%) followed by amikacin (11.1%). Resistance to a third-generation cephalosporin was noticed in only 5.6% isolates. However, in clinical isolates, maximal resistance was noticed against third-generation cephalosporins followed by ofloxacin in 91.3% and 86.9% isolates, respectively, and resistance to gentamicin and amikacin was noticed in 78.3% and 52.2% isolates, respectively. The presence of blaCTX-M, blaTEM, blaSHV, and blaAmpC in clinical isolates was noticed in 52.2%, 60.9%, 21.7%, and 43.5%, respectively. None of the isolates from food showed the presence of any of the above-cited genes. CONCLUSIONS: Probably bla genes have not yet disseminated to raw-food vegetation in India.
