RESUMO

It is believed that increased livestock production is limited by tropical climate. Thermal imbalance in bulls can lead to hyperthermia and alter testicular metabolism, causing subfertility or infertility. Therefore, the thermoregulation of composite Canchim bulls (5/8 Charolais × 3/8 Zebu) raised in tropical climate as well as their consequences in the physiological, hematological, hormonal, and andrological parameters were evaluated monthly. The bulls (n = 18; 30.0 ± 1.5 months; 503.8 ± 23.0 kg) were kept on pasture, in a single group, from August 2015 to March 2016, comprising the winter, spring, and summer seasons. Biometeorological variables were continuously monitored, and the Temperature and Humidity Index (THI) was calculated. A greater thermal challenge occurred in spring and summer (THI ≥ 72.0). Nevertheless, the bulls exhibited normothermia (38.6 to 38.9 °C) in these seasons. The cortisol did not vary between seasons (7.0 vs. 8.7 vs. 6.8 ng/mL; P > 0.05) and remained within the physiological patterns. Independent of the seasons, stress leukogram was also not observed, refuting the incidence of acute or chronic thermal stress. It is noteworthy that T3 and testosterone increased (P < 0.0001, P < 0.05) in spring and summer, the time that coincides with the breeding season, when there is increased metabolic requirement from the bulls. The progressive thermal challenge increase did not affect the scrotal thermoregulatory capacity, and in general, scrotal temperature remained at 5.2 °C below the internal body temperature. In summer, there was a 5% reduction in the minor sperm defects (P < 0.05) and DNA fragmentation in 2.4% of spermatozoa, a compatible value for high fertility bulls. The results show that the studied composite bulls can be considered as climatically adapted and constitute a viable alternative to be used in production systems in a tropical climate, even if the breeding seasons occur during the most critical thermal condition periods of the year.

RESUMO

Two mutations in the CD4 bovine gene (G>T/Q306H; A>C/K310N) were identified as causative for altered staining with anti-CD4 mAb #CC8. We developed a HRM qPCR for genotyping these mutations and compare with immunophenotyping in different cattle breeds. The assay distinguished five genotypes, B (homozygous, G/A) and C (heterozygous, G/A and T/C), found in taurine, A (homozygous, G/C) and D (heterozygous, T/C and G/C), found in zebu. The E genotype (homozygous, T/C) was not observed in tested animals. As expected, B and C presented high/very high and intermediate CD4 staining, respectively. The lack/low CD4 staining was mainly related to the A, while the intermediate staining was mainly related to D genotype. The developed HRM qPCR assay accurately identified the altered phenotypes associated with CC8 staining in taurine. However, the assay cannot be applicable in zebu or hybrid breeds, probably due to additional mutations in the CD4 gene from zebu descendant animals.

RESUMO

Analyses of livestock genomes have been used to detect selection signatures, which are genomic regions associated with traits under selection leading to a change in allele frequency. The objective of the present study was to characterize selection signatures in Canchim composite beef cattle using cross-population analyses with the founder Nelore and Charolais breeds. High-density single nucleotide polymorphism genotypes were available on 395 Canchim representing the target population, along with genotypes from 809 Nelore and 897 Charolais animals representing the reference populations. Most of the selection signatures were co-located with genes whose functions agree with the expectations of the breeding programs; these genes have previously been reported to associate with meat quality, as well as reproductive traits. Identified genes were related to immunity, adaptation, morphology, as well as behavior, could give new perspectives for understanding the genetic architecture of Canchim. Some selection signatures identified genes that were recently introduced in Canchim, such as the loci related to the polled trait.

RESUMO

Different allelic forms of bovine CD4 were previously described in cattle and were also observed in Canchim calves examined in the present experiment. However, the functional relevance of these different CD4 phenotypes has not yet been investigated. CD4 + T helper cells are known to play a central role in immune control against Babesia bovis infection. Thus, our study aimed to compare the profiles of immune cells, specific antibody titers and blood infection levels measured by qPCR (quantitative polymerase chain reaction) in calves naturally infected with B. bovis, phenotyped as CD4- (absence of anti-CD4 staining), CD4 + (intermediate staining) or CD4 ++ (high staining). The CD4 mRNA precursor was also measured in these animals. Calves with the CD4- phenotype showed higher amounts of B. bovis DNA in blood samples, compared to the other CD4 phenotypes. It was also observed that these calves with higher levels of infection had lower amounts of natural killer cells and higher expression of the CD4 gene, which can be interpreted as a compensation for the failure of the altered CD4 receptor to recognize relevant B. bovis epitopes.

RESUMO

Rhipicephalus microplus is a vector of cattle tick fever, a disease caused by the protozoans Babesia bovisand B. bigemina, and also anaplasmosis, produced by the Rickettsiales Anaplasma marginale. These tick-borne pathogens cause considerable losses to Brazilian livestock breeders and represent an obstacle to the expanded use of taurine breeds due to their higher sensitivity to ticks and hemoparasites compared to zebu breeds. Differences in the susceptibility to hemoparasites were also verified within breeds, suggesting that may be possible to select a most resistant phenotype. Therefore, repeatability of R. microplus counts and copy number of hemoparasites DNA were estimated, along with correlations between themselves, aiming to verify if those measures can be used as parameters to classify animals according to their parasite resistance degrees. Forty-two Canchim females kept on pastures naturally infested by ticks were evaluated for the level of infestation by R. microplus and infection by B. bovis, B. bigemina, and A. marginale. Twenty-four evaluations were performed once a month, for adult female ticks counts and blood samplings. The experimental period was divided into four phases, according to the animals age range: Phase 1: 8 to 13 months (collections 1 to 6); phase 2: 14 to 19 months (collections 7 to 12); phase 3: 20 to 25 months (collections 13 to 18), and phase 4: 26 to 31 months (collections 19 to 24). Blood samples were submitted to absolute quantification of hemoparasites DNA sequences using qPCR. The hemoparasite and tick counts data were transformed for normalization and were analyzed using mixed models. Among three species of hemoparasites studied, A. marginale presented the highest level of infection. During phase 3, B. bigemina presented higher infection levels (p < 0.05) compared to B. bovis, whereas no differences were observed in other phases. Estimated repeatabilities for parasite infection levels varied from low to moderate during our experiment. There were low correlations between tick counts and parasite infection levels, and between parasite infection levels from different species by themselves. Based on these results, under conditions of the present study, we suggest that it is possible to identify animals presenting a most resistant phenotype against infection by both hemoparasites and ticks. Moreover, the animal age may be an important factor related to resistance against these pathogens. The data obtained shed more light on the resistance to hemoparasites studied.

RESUMO

Birth weight is a performance parameter of great zootechnical importance for both meat and dairy production, as well as for breeding animals, mainly due to its relation to survival rate at weaning, and the weight of animals throughout their developmental and growth phases. Therefore, the objective of this work was to establish heritability estimates, as well as phenotypic and genetic trends of birth weight of water buffaloes from State of Pará, Brazil. Descriptive statistics were calculated and the Shapiro-Wilk Normality test was performed with a Statistical Analysis System package. Heritability estimates were established by Bayesian inference. BW was 36.6 kg in average. The statistical model considered sex, year of birth and breed composition of the animals as fixed effects, and animal, maternal and residual as random effects. Direct heritability was platykurtic (flattened) and with higher asymmetry, presenting a bimodal distribution with the first mode close to 0.10, and the second mode close to 0.30; the maternal heritability was trimodal, with peaks very close to 0.15, and another, less evident, close to 0.20. The direct genetic trend of birth weight was negative (-0.03kg year-1) and maternal genetic trend was close to zero (0.001kg ano-1), even though the phenotypic trend had been positive (0.156 kg year-1). There is genetic variability to be addressed in a breeding program, however, very little was done as far as selection for growth of water buffaloes in the State of Pará.

The objective of this study was to estimate genetic parameters using Bayesian inference for the estimates of individual parameters of mature weight (Â) and growth rate (K), obtained by Brody growth function. The file consisted of 14,563 records relating to weights and ages of 1,158 Nelore females, participants in the Genetic Improvement Program of the Nellore. For the estimates analysis of the curve parameters via Bayesian inference, it was used an univariated animal model that included as fixed effect of contemporary group (animals born on the same state, in the same quarter of the year, the same year and feedlot) and random as the direct genetic and residual. In this analysis it was used two different sizes for the chains generated by Gibbs sampling algorithm, 550 and 1.1 million cycles, with initial discarding of 50 000 and sample of 100 000 cycles, respectively, and sampled every 500 cycles and 1000, respectively. The mean posterior and residual additive genetic variance showed a small variation to both  and K, even when implemented in different sizes for the chains generated by Gibbs sampling algorithm. The heritability estimates for Â, ranged from 0.44 to 0.46, similar to the range 0.46 to 0.48 obtained for the estimates of K. These magnitudes indicate that the selection can be used as a tool to change the shape of the growth curve of these animals. However, the use of the information to amend the growth curve of animals, must be done with great care, since the traits to be worked in the modification of the shape of the growth curve according to the literature results are negatively correlated.

RESUMO

With the objective of to adjust nonlinear models for the growth curves for a buffaloes herd raised in floodable lands in Rio Grande do Sul state, monthly records measured from birth to two years-old of 64 males and 63 females born between 1982 and 1989 were used. The models used were: Von Bertalanffy, Brody, Gompertz and Logistic. The parameters were estimated by NLIN procedure and the criteria used to evaluate the adjustment given by the models were: asymptotic standard deviation; coefficient of determination; average absolute deviation of residues and asymptotic index. Von Bertalanffy and Brody models overestimated the male asymptotic weight (A) in 15.9 and 171.3kg, respectively, and the Gompertz and Logistic models underestimated it in 4.5 and 13.4kg, respectively. For females, the Logistic model underestimated the asymptotic weight (-2.09kg), and Gompertz, Von Bertalanffy and Brody overestimated this parameter in 8.04, 17.7, and 280.33kg, respectively. The biggest average deviation was estimated by Brody model for both sexes, characterizing the biggest index. Considering the criteria, it is recommended the Gompertz and Logistic models for adjust females and males Murrah buffaloes breed growth curves.

Parameters based on the probability of gene origin were used to describe the genetic variability in strains of Nellore and polled Nellore (Bos indicus) cattle that participated in the Program for Genetic Improvement of the Nellore Breed (Programa de Melhoramento Genético da Raça Nelore). The effective number of founders was 87.2 for Nellore and 107.9 for polled Nellore, while the number of ancestors was 59.8 for Nellore and 61.5 for polled Nellore and the remaining genomes were 39.4 for Nellore and 34.5 for polled Nellore cattle. The results indicate an intense use (by artificial insemination) of some sires and the absence of subdivisions in the population. The family structure in the two breeds was mainly caused by the genetic contribution of the same sires and only mating preferences for descendants of some founders are recorded in either breed. The results suggest that genetic variability needs monitoring in order to avoid the compromise of genetic improvement in economically important traits in the breeding program.

RESUMO

There are few studies on weight covering the full life cycle of Zebu cattle, and there is no entire growth description or mean growth pattern for animals belonging to this breed. In order to provide such data, 1,158 Nelore females born between 1985 and 1995 were weighed 14,563 times from birth to full growth maturity, in ten herds spread over seven Brazilian states. The Von Bertalanffy, Brody, logistic and Gompertz non-linear models were used to obtain the asymptotic weights (A) and the maturation rates (K). The (co)variance and breeding value components for A and K were obtained by using the multiple trait derivative free restricted maximum likelihood method under the animal model. Genetic trends were calculated in function of the mean expected progeny differences (EPD) for the trait (A or K) divided by the number of animals according to their year of birth. The genetic trends of the expected progeny difference with reference to the date of birth of the cows were, on average, -6.5g y-1 for A and 2.0g y-1 for K, close to zero as confirmed by the low (0.0023 to 0.003) coefficient of regression values. The curve parameters are recommended as a selection criterion to reach precocity and avoid adult weight increase in the female herd.
