Respiratory Syncytial Virus (RSV) is an important cause of respiratory infection in humans. Severe cases are common in children ≤2 years old, immunocompromised individuals, and the elderly. In 2020, RSV infection reduced in Rio Grande do Sul (RS), southern Brazil; however, in 2021 resurgence of RSV was observed. This study analyzed epidemiological and genetic features of RSV infection cases reported in 2021 in RS. Nasopharyngeal samples collected from individuals with respiratory infection negative for SARS-CoV-2, Influenza A and B viruses were assessed for the presence of RSV by real time RT-qPCR. RSV-A and RSV-B genomic sequencing and phylogenetic reconstructions were performed for genotyping and clade characterization. Among 21,035 respiratory samples analyzed, 2,947 were positive for RSV, 947 of which were hospitalized patients. Positive cases were detected year-round, with the highest number in June-July (winter). Children <1 year comprised 56.28% (n = 533) of the hospitalized patients infected with RSV, whereas 14.46% (n = 137) were individuals >60 years. Of a total of 361 deaths, 14.68% (n = 53) were RSV positive, mostly patients >60 years old (73.58%, n = 39). Chronic kidney disease, cardiopathy, Down syndrome and neurological diseases were associated with RSV infection. RSV-A was identified in 58.5% (n = 117/200) of the patients, and RSV-B in 41.5% (n = 83/200). Of 95 RSV genomes recovered from SARI cases, 66 were RSV-A GA.2.3.5 genotype, while 29 were RSV-B GB.5.0.5a genotype. This study provides epidemiological and molecular data on RSV cases in RS during the COVID-19 pandemic and highlights that investigation of different respiratory viruses is essential for decision-making and disease prevention and control measures.

The Amazon has a range of species with high potential for sustainable timber harvesting, but for them to be utilized globally, the merchantable wood volume must be accurately quantified. However, since the 1950s, inadequate methods for estimating merchantable timber volumes have been employed in the Amazon, and Brazilian Government agencies still require some of them. The natural variability of the Amazon Forest provides an abundance of species of different sizes and shapes, conferring several peculiarities, which makes it necessary to use up-to-date and precise methods for timber quantification in Amazon Forest management. Given the employment of insufficient estimation methods for wood volume, this study scrutinizes the disparities between the actual harvested merchantable wood volume and the volume estimated by the forest inventory during the harvesting phase across five distinct public forest areas operating under sustainable forest management concessions. We used mixed-effect models to evaluate the relationships between inventory and harvested volume for genera and forest regions. We performed an equivalence test to assess the similarity between the volumes obtained during the pre-and post-harvest phases. We calculated root mean square error and percentage bias for merchantable volume as accuracy metrics. There was a strong tendency for the 100% forest inventory to overestimate merchantable wood volume, regardless of genus and managed area. There was a significant discrepancy between the volumes inventoried and harvested in different regions intended for sustainable forest management, in which only 22% of the groups evaluated were equivalent. The methods currently practiced by forest companies for determining pre-harvest merchantable volume are inaccurate enough to support sustainable forest management in the Amazon. They may even facilitate the region's illegal timber extraction and organized crime.

The Amazonian clearwater igapós are poorly studied floodplain ecosystems that are mainly covered by forests and are undergoing massive threats due to changes in land use and climate. Their hydrochemical characteristics and edaphic conditions fall between those of the eutrophic várzea floodplains on whitewater rivers and those of the oligotrophic igapós on blackwater rivers. Previous studies have indicated the management potential of timber species in the highly dynamic várzea floodplains due to the fast tree growth and high forest productivity. Timber resource management, however, is not recommended for the blackwater ecosystem because of its slow dynamics and high vulnerability to disturbances. For clearwater igapós, information on the potential for sustainable management of timber resources is lacking. In this study, we modeled the growth in diameter, height, and volume to derive species-specific minimum logging diameters (MLD) and felling cycles (FC) for eight merchantable species in the clearwater igapós of the Branco and Tapajós rivers in the northern and southern Amazon Basin, respectively. Diameter growth was modeled by analyzing the tree rings that are annually formed in the Amazonian floodplains as a consequence of the regular and predicable long-term flooding. Growth modeling followed the guidelines of the Growth-Oriented Logging (GOL) concept, with the adjustment of diameter growth improved by applying nonlinear mixed-effects regression. MLDs varied from 36 to 90 cm and FCs ranged from 6 to 21 years, which diverges from the standards of Brazilian logging regulations (MLD: 50 cm; FC: 25-35 years). This indicates the potential for timber resource management, which should be tested and introduced at small scales, integrated in protected areas to stepwise promote the sustainable management of these natural resources by traditional communities to increase their income and the conservation of this ecosystem.

BACKGROUND: In March 2020, the COVID-19 outbreak was declared a pandemic by the World Health Organization (WHO), generating stark economic and social repercussions that directly or indirectly affected families' wellbeing and health status. AIMS: This review aims at mapping the existing evidence on the impact of the COVID-19 pandemic on maternal mental health, early childhood development, and parental practices, worldwide, to identify evidence gaps and better inform future delivery of care and health policy measures. METHODS: Following the protocol defined by PRISMA-ScR, this scoping review has searched for relevant studies published between January 2020 and June 2021, selecting evidence sources based on pre-established criteria. From a total of 2,308 articles, data were extracted from 537 publications from 35 countries on all three health domains. RESULTS: The combined stressors brought forth by the pandemic have exerted a heavy burden on the mental health of mothers and the development of young children, partly mediated by its impact on parental practices. CONCLUSIONS: Despite remaining gaps, we have identified sufficient evidence pointing to an urgent need for more concerted global research efforts and rapid policy responses to timely address severe and pervasive negative impacts to the mental health of mothers and children at a key developmental stage.

The high demand for food and energy imposed by the increased life expectancy of the population has driven agricultural activity, which is reflected in the larger quantities of agro-industrial waste generated, and requires new forms of use. Brazil has the greatest biodiversity in the world, where corn is one of the main agricultural genres, and where over 40% of the waste generated is from cobs without an efficient destination. With the aim of the valorization of these residues, we proposed to study the immobilization of laccase from Aspergillus spp. (LAsp) in residual corn cob and its application in the degradation of Remazol Brilliant Blue R (RBBR) dye. The highest yields in immobilized protein (75%) and residual activity (40%) were obtained at pH 7.0 and an enzyme concentration of 0.1 g.mL-1, whose expressed enzyme activity was 1854 U.kg-1. At a temperature of 60 °C, more than 90% of the initial activity present in the immobilized biocatalyst was maintained. The immobilized enzyme showed higher efficiency in the degradation (64%) of RBBR dye in 48 h, with improvement in the process in 72 h (75%). The new biocatalyst showed operational efficiency during three cycles, and a higher degradation rate than the free enzyme, making it a competitive biocatalyst and amenable to industrial applications.

Gluconobacter oxydans is a well-known acetic acid bacterium that has long been applied in the biotechnological industry. Its extraordinary capacity to oxidize a variety of sugars, polyols, and alcohols into acids, aldehydes, and ketones is advantageous for the production of valuable compounds. Relevant G. oxydans industrial applications are in the manufacture of L-ascorbic acid (vitamin C), miglitol, gluconic acid and its derivatives, and dihydroxyacetone. Increasing efforts on improving these processes have been made in the last few years, especially by applying metabolic engineering. Thereby, a series of genes have been targeted to construct powerful recombinant strains to be used in optimized fermentation. Furthermore, low-cost feedstocks, mostly agro-industrial wastes or byproducts, have been investigated, to reduce processing costs and improve the sustainability of G. oxydans bioprocess. Nonetheless, further research is required mainly to make these raw materials feasible at the industrial scale. The current shortage of suitable genetic tools for metabolic engineering modifications in G. oxydans is another challenge to be overcome. This paper aims to give an overview of the most relevant industrial G. oxydans processes and the current strategies developed for their improvement.

Sodium dichloroacetate (DCA) is a metabolic regulator used to treat diabetes. Since DCA inhibits pyruvate dehydrogenase kinase, decreasing lactic acid formation, it can reverse the Warburg effect in cancer cells, promoting apoptosis. Therefore, this study aimed to investigate the potential of DCA as a drug repurposing candidate for the treatment of melanoma. For the in-vitro assay, murine B16-F10 melanoma cells were treated with 0.5, 1, 5, 10, 20 or 50 mM DCA for 3 days, analyzed with the crystal violet method. The in-vivo effect of DCA was evaluated in B16-F10 tumor-bearing C57BL/6 mice treated with different doses of DCA (0, 25, 75 or 150 mg/kg) by gavage for 10 days, followed by measurement of tumor volume. Upon necropsy, representative slices of lung, liver, kidney, spleen and intestine were collected, processed and submitted for histopathological examination. The DCA concentrations of 10, 20 and 50 mM reduced B16-F10 cell viability after 48 and 72 h of treatment, whereas 20 and 50 mM were effective after 24 h of treatment. A significant reduction in tumor growth was observed in B16-F10 melanoma bearing mice at all doses, with no change in body weight or histology. DCA attenuates the growth of B16-F10 melanoma in vitro and in vivo, without systemic toxic effects. Therefore, DCA is a candidate for drug repurposing against melanomas.

In tropical forests, the spatial distribution of trees may present random, uniform, or grouped patterns that can simultaneously be affected by site and species characteristics. In Central Amazon, topographic gradients and soil water levels drive differences in tree species distribution and in forest dynamics at local scales. Knowing this kind of information can be useful for a forest manager to plan harvesting operations considering the microhabitat preference of merchantable species to reduce the disturbances caused by logging activities. Thus, the spatial variation of tree species is an important information to be considered to support the planning process of forest logging. The present study aims to evaluate the spatial distribution pattern of six species and analyze the relationship between the topography and the population densities and stem size of those species. The study was carried out in a forest production compartment managed by a private company located in the municipality of Silves, state of Amazonas, Brazil. The spatial pattern of the six species was characterized by Ripley's K function. Spatial distribution of diameter at breast height (DBH) and tree density based on kernel incidence calculation were evaluated for topographic classes of slope, elevation, and distance from streams, which were mapped using geographic information systems (GIS). The means of DBH and density of each species were compared among topographic classes by ANOVA and Tukey's test. The results demonstrated the predominance of the aggregate distribution pattern for the six species up to 1105 m (p < 0.01). The tree species Minquartia guianensis Aubl., Protium puncticulatum J.F.Macbr, Manilkara elata (Allemão ex Miq.) Monach, and Caryocar glabrum Aubl. Pers showed an increase in the tree density as the distance from the streams and elevation increased, standing spatially incident on plateaus. Kernel densities of Dinizia excelsa Ducke and Goupia glabra Aubl. were higher closer to streams. The DBH averages followed similar trends of population density for M. guianensis, M. elata, and C. glabrum, and the opposite pattern for D. excelsa, which presented larger individuals in less densely populated areas. P. puncticulatum and G. glabra mean DBH distribution was not affected by the topographic variables analyzed. Topography-related variables showed effects on variations of density and tree size, suggesting that species may be spatially sensitive to the habitat variability available in the study area. In view of logging planning, spatial distribution must be considered in decisions related to cutting down trees and maintenance of remaining trees, especially because some species are more aggregated in smaller scales. Moreover, as topographic variations affect the spatial distribution of size and density, the timber yield will vary spatially in the area, bringing implications for planning logging intensities, roads, skid trails and forest operations. Finally, the procedures and information generated in this study can be reproduced and applied to other species and managed areas to support the planning toward minimizing impacts on the spatial structure of commercial species, as well as to increase the chances of future stock recovery of managed forests in the Amazon.

PURPOSE: Studies have shown that laser therapy is a recommended therapy for improving the postoperative period in patients undergoing extraction of the third molars; however, there is still no definition regarding the best protocol to be used. The aim of this study was to measure and compare periodontal tissue healing using 2 different laser protocols. MATERIALS AND METHODS: A double-blinded, randomized, prospective study with patients submitted to inferior third molar extraction was performed, with the sample divided into 3 groups according to the laser application protocol and followed for 6 months: group I, 10 J/cm2; group II, 30 J/cm2; and group III, sham. The primary variable was probing depth, and the secondary variables were trismus, facial edema, and pain. RESULTS: The sample was composed of 57 patients: 19 in group I, 20 in group II, and 18 in group III. Analysis of the variables showed statistically significant differences between both groups that received laser therapy, with values of 1.46 for edema control on the third day and 0.54 on the seventh day in group I (P = .017) and 1.26 and 0.52, respectively, in group II (P = .001) compared with 0.59 and 0.49, respectively, in the sham group (P = .702), as well as a statistically significant difference for the 10-J/cm2 laser protocol for probing depth, with values of 7.58 mm preoperatively and 9.09 mm after 6 months (P = .013). CONCLUSIONS: The use of the low-intensity laser as adjuvant therapy after third molar extraction was more effective in the group undergoing the 10-J/cm2 laser protocol for improving periodontal tissue healing and in both laser therapy groups for reducing facial edema.

INTRODUCTION: Vesicoureteral Reflux (VUR) is characterized by a retrograde flow of urine from the bladder into the ureters and kidneys. It is one of the most common urinary tract anomalies and the major cause of urinary tract infection (UTI) in the first years of life. If not properly diagnosed and treated can lead to recurrent UTI, renal scar and, in severe cases, to end stage renal disease. Despite recent advances in scientific and technological knowledge, evaluation and treatment of VUR is still controversial and there is still considerable heterogeneity in evaluation methods and therapeutic approaches. The aim of the present consensus is to give a practical orientation on how to evaluate and treat VUR. METHODS: The board of Pediatric Urology of the Brazilian Society of Urology joined a group of experts and reviewed all important issues on Vesicoureteral Refl ux evaluation and treatment and elaborated a draft of the document. On November 2017 the panel met to review, discuss and write a consensus document. RESULTS AND DISCUSSION: Vesicoureteral Reflux is a common and challenging problem in children. Children presenting with Vesicoureteral Reflux require careful evaluation and treatment to avoid future urinary tract infections and kidney scars. The panel addressed recommendations on up to date choice of diagnosis evaluation and therapies.

The present study describes a clinical case in which the buccal fat pad (BFP) was used to improve the contour of the peri-implant mucosa. To our knowledge, this is the first case report of such an application in the literature. A 58-year-old woman presented with teeth #2 and #3 missing and an indication for extraction of tooth #4. After clinical examination and CBCT analysis, immediate implants were placed in the region of the extracted tooth and that of tooth #2 for prosthetic rehabilitation using a three-unit fixed partial denture. There was an extensive mucosal defect in the region of tooth #3, with vertical and horizontal changes in the contour of the mucosa. As an alternative to the use of a subepithelial connective tissue graft, we opted for displacement of the BFP and its accommodation on the alveolar ridge of tooth #3 to improve the buccal tissue contour. After 5 years of follow-up, satisfactory gain and maintenance of mucosal volume were observed in the treated area, as well as improvement of buccal tissue contour. The use of BFP seems to be a feasible alternative for filling and treating peri-implant mucosal defects.

PURPOSE: The objective of the present study was to compare 3 sedation protocols using diazepam, midazolam, and nitrous oxide. PATIENTS AND METHODS: A total of 120 patients with an indication for extraction of third molars were selected. All 120 patients had had moderate to severe levels of anxiety according to the Corah Dental Anxiety Scale. The patients were randomly divided into 3 groups. The patients' vital signs were measured, and the results analyzed by descriptive statistical analysis and statistical tests of comparison. RESULTS: No statistically significant differences were found in the patients' heart rate. However, the differences in the systolic and diastolic blood pressure were statistically significant after 15 minutes of nitrous oxide sedation. The oximetry data showed no differences among the 3 sedation protocols. We also found no statistically significant differences in the retrograde amnesia test. The differences in anxiety from preoperatively to postoperatively were statistically significant for all techniques, demonstrating their effectiveness in anxiety control. CONCLUSIONS: All 3 preoperative sedation techniques for anxious patients undergoing extraction of third molars used in the present study were effective in controlling the anxiety, with little effect on the patients' vital signs and retrograde amnesia.

Insight is greatly impaired in Bipolar Disorder (BD), especially during mania. Cognitive impairment is also present in BD. Despite that, few studies have investigated a possible association between these two aspects. The main goal of the current study is to compare BD affective states regarding performance in cognitive testing and investigate clinical and cognitive predictors for insight loss in BD. The study investigated a sample of 65 patients who were evaluated in one of the BD phases (mania, euthymia or depression). All the subjects underwent neuropsychological evaluation and completed the Insight Scale for Affective Disorders (ISAD). The relationship between level of insight and clinical/cognitive variables was analyzed through multiple regression models. No significant differences were found among BD phases regarding performance on cognitive testing. Insight was more impaired in mania then in depression or euthymia. Predictors for loss of insight were: severity of manic symptoms and impairments in selective attention (Symbol search test), divided attention (Trail making test) and inhibition (Stroop test). The sample size is a potential limitation of the current study. Nevertheless, the results suggest this had limited impact, with group differences being detected for a number of variables. The results found have important clinical importance, suggesting, for example, that rehabilitation of specific cognitive skills may improve insight in BD.

Woody debris, defined as standing and downed deadwood, consists in an essential component of the forest carbon stock. However, few studies have been carried out to get an efficient and accurate sampling procedure for estimating it. This work proposes two methodologies to estimate the woody debris volume in a Brazilian mixed tropical forest: 1) two-stage systematic sampling, using a mixed methodology, in which the Strand's method is applied to standing dead trees and stumps, and line intercept sampling is used to fallen trees and branches; and 2) ratio estimate of the sum of cross-sectional areas of deadwood pieces and forest basal area, aiming to obtain the total woody debris volume indirectly in the natural forest. Conversions for biomass and carbon stocks were made applying the mean basic density on the estimates of deadwood volumes. Both methodologies are accurate for woody debris volume estimates, with a sampling error equal to 16.1% (methodology 1) and 5.7% (methodology 2), at a 95% probability level. Thus, the methodology 2 has potential to be used in strategic forest inventories of woody debris, such as in National Forest Inventories, due to increasing importance of its quantification in all forest ecosystems.

This study aimed to evaluate the behavior of human osteoblastic cells cultured on dense and porous Titanium (Ti) samples comparing to dense and porous Ti-35 Niobium (Ti-35Nb) samples, using gene expression analysis. Scanning electronic microscopy confirmed surface porosity and pore interconnectivity and X-ray diffraction showed titanium beta-phase stabilization in Ti-35Nb alloy. There were no differences in expression of transforming growth factor-ß, integrin-ß1, alkaline phosphatase, osteopontin, macrophage colony stimulating factor, prostaglandin E synthase, and apolipoprotein E regarding the type of alloy, porosity and experimental period. The experimental period was a significant factor for the markers: bone sialoprotein II and interleukin 6, with expression increasing over time. Porosity diminished Runt-related transcription factor-2 (Runx-2) expression. Cells adhering to the Ti-35Nb alloy showed statistically similar expression to those adhering to commercially pure Ti grade II, for all the markers tested. In conclusion, the molecular mechanisms of interaction between human osteoblasts and the Ti-35Nb alloy follow the principal routes of osseointegration of commercially pure Ti grade II. Porosity impaired the route of transcription factor Runx-2.

This paper reports the bioavailability of the metals (cadmium, copper, zinc, lead, and nickel) in sediment samples collected in seven stations from the São Paulo Estuary, Todos os Santos Bay, Brazil. The bioavailability was determined by employing the technique "acid-volatile sulfide (AVS) and simultaneously extracted metal (SEM)". The elements cadmium, copper, lead, and zinc were determined using differential pulse anodic stripping voltammetry (DPASV), while nickel was quantified utilizing electrothermal atomic absorption spectrometry (ET AAS). The accuracy of these methods was confirmed using a certified reference material of estuarine sediment (NIST 1646). The sulfide was quantified using potentiometry with selective electrode and the organic matter determination employing an indirect volumetric method using potassium dichromate and iron(II) sulfate solutions. The bioavailability of the metals was estimated by relationship between the concentration of AVS and the sum of the concentrations of the simultaneously extracted metals (ΣSEM), considering a significant toxicity when (ΣSEM)/(AVS) is higher than 1. The bioavailability values in the seven stations studied varied from 0.93 to 1.31 (June, 2014) and from 0.34 to 0.58 (September, 2014). These results demonstrated a critical condition of toxicity (bioavailability >1) in six of the seven sediment samples collected during the rainy season (June, 2014). In the other period (September, 2014), the bioavailability was always lower than 1 for all sediment samples collected in the seven stations. The individual values of the concentrations of the five metals were compared with the parameters PEL (probable effects level) and TEL (threshold effects level), which are commonly employed for characterization of ecological risk in environmental systems. This comparison revealed that all metals have concentrations lower than the PEL and only zinc and lead in some stations have contents higher than the TEL. The bioavailability evaluation and the concentrations achieved for the five elements in the sediments samples analyzed demonstrated that the ecosystem studied does not present an environmental risk.

This article presents the case of an 8-year-old patient who presented with a large radiolucency associated with the maxillary left canine and a supernumerary tooth. A computed tomography scan showed the radiolucency was in close proximity to the roots of the anterior teeth, with no areas of root resorption, and expansion into the left maxillary sinus. The boundaries of the maxillary sinus floor were still preserved. After positive aspiration of intralesional liquid and due to the large size of the radiolucency, a decompression technique was selected to preserve the permanent canine. Surgery was performed to remove the supernumerary tooth, followed by orthodontic treatment and surgery to allow access for orthodontic traction of the permanent canine. More than 5 years post-treatment, no recurrence was observed and the therapeutic option to position and preserve the permanent canine was successful.

With the aim of contributing to the discussion on stomatognathic system dysfunction after surgical procedures, this study compared the electromyographic activity of the superficial masseter and temporal masticatory muscles before, during, and after impacted mandibular third molar extractions. Muscular activity was recorded presurgery, transsurgery, immediately postoperatively, and on postoperative days 7, 15, and 30. Twenty patients requiring extraction of impacted mandibular third molars were selected and evaluated. In 20 patients who underwent mandibular third molar extractions, electromyography showed no alterations in muscle tone, and no statistically significant differences were observed in the left and right temporal and masseter muscles at any of the experimental periods at either mandibular rest or isometric contraction position. However, the degree of mouth opening increased 11.76% from pretreatment to 30 days after surgery. These results may reflect the shorter, careful extraction procedure performed by the surgeon.

Cuniculus paca, commonly known as the paca, is a rodent of the Cuniculidae family that is widely distributed throughout the Americas, including all Brazilian territories, and is abundant in the Amazon region. It is one of the most hunted species and faces significant predation in the extreme western Amazon region of Brazil because it constitutes a staple in the diet of local communities, for subsistence and commercial purposes. Understanding the helminthic fauna of these animals is of paramount importance, given that some nematodes have zoonotic potential and may pose risks to consumer health. This study aimed to contribute to the records of the nemtodes of this highly consumed species in the Amazon region, highlighting the occurrence of gastrointestinal parasites in free-living pacas intended for human subsistence consumption. The study was conducted in the Paranã da Floresta community, located in the municipality of Guajará, Amazonas from 2022 to 2023. The community members hunted this rodent for consumption and voluntarily provided the viscera for analysis. Nematodes were extracted from each organ. The organs were opened, and the contents were processed using a sieve (0.15 mm) and subsequently evaluated separately. In total, 10,157 nematodes were found in the 14 pacas. Based on morphological analyses, the nematodes were identified as Heligmostrongylus sedecimradiatus (n = 10,068), Trichuris sp. (n = 85), and Physaloptera sp. (n = 4). This study provides insights into the nematodes diversity of free-living pacas in the extreme Western Amazon region, emphasizing the importance of sanitary surveillance and public awareness of the risks associated with bushmeat consumption.
