RESUMO

This study measured air exchange rates, indoor concentrations of aldehydes and volatile organic compounds (VOCs), and radioactivity levels at 19 temporary houses in different temporary housing estate constructed in Minamisoma City following the Great East Japan Earthquake. The 19 surveyed houses represented all of the companies assigned to construct temporary houses in that Minamisoma City. Data were collected shortly after construction and before occupation, from August 2011 to January 2012. Mean air exchange rates in the temporary houses were 0.28/h, with no variation according to housing types and construction date. Mean indoor concentrations of formaldehyde, acetaldehyde, toluene, ethylbenzene, m/p-xylene, o-xylene, styrene, p-dichlorobenzene, tetradecane, and total VOCs (TVOCs) were 29.2, 72.7, 14.6, 6.35, 3.05, 1.81, 7.29, 14.3, 8.32, and 901 µg/m(3), respectively. The levels of acetaldehyde and TVOCs exceeded the indoor guideline (48 µg/m(3)) and interim target (400 µg/m(3)) in more than half of the 31 rooms tested. In addition to guideline chemicals, terpenes (α-pinene and d-limonene) and acetic esters (butyl acetate and ethyl acetate) were often detected in these houses. The indoor radiation levels measured by a Geiger-Müller tube (Mean: 0.22 µSv/h) were lower than those recorded outdoors (Mean: 0.42 µSv/h), although the shielding effect of the houses was less than for other types of buildings.

AIMS: The aim of this work was to observe bacteria associated with the spores of Gigaspora margarita, an arbuscular mycorrhizal fungus (AMF). METHODS AND RESULTS: First, a direct analysis of DNA from sterilized spores indicated the bacteria belonging to the genus Janthinobacterium. In the second assay, two bacterial strains were isolated by osmosis from protoplasts, which were derived from spores by using two particular enzymes: lysing enzymes and yatalase. After isolation, cultivation and identification by their DNA as performed in the first experiment, the species with the closest relation were Janthinobacterium lividum (KCIGM01) and Paenibacillus polymyxa (KCIGM04) isolated with lysing enzymes and yatalase respectively. Morphologically, J. lividum was Gram negative and oval, while P. polymyxa was also oval, but Gram positive. Both strains had antagonistic effects to the pathogenic fungi Rosellimia necatrix, Pythium ultimum, Fusarium oxysporum and Rhizoctonia solani. In particular, J. lividum was much stronger in this role. However, in phosphorus (P) solubilization P. polymyxa functioned better than J. lividum. CONCLUSIONS: This experiment had revealed two new bacteria species (P. polymyxa and J. lividum), associated with AMF spores, which functioned to suppress diseases and to solubilize P. SIGNIFICANCE AND IMPACT OF THE STUDY: AMF spores could be a useful source for bacterial antagonists to soil-borne diseases and P solubilization.

PURPOSE: To report results of twin hook fixation for proximal femoral fractures in comparison to those fixed with the conventional lag screw. METHODS: Between August 2005 and July 2006, 2 men and 15 women aged 74 to 94 (mean, 85) years with proximal femoral fractures underwent open reduction and internal fixation using the twin hook system. The tip-apex distance was compared with that in 20 patients treated with the sliding hip screw between August 2004 and July 2005. RESULTS: In the 17 patients, the hook was inserted into the centre of the femoral head. Bone union was achieved and no intra- or post-operative cut-out or device failure was encountered. In patients using the twin hook and sliding hip screw respectively, the mean tip-apex distance was 22.3 mm and 14.6 mm (p<0.001). CONCLUSION: Using the twin hook system requires more surgical skill than using the sliding hip screw, because failure to insert the pin into the centre of the femoral head risks intra-articular perforation by the hooks.

Uridine diphosphogalactose:glycoprotein galactosyltransferases were examined in human lung adenocarcinoma and squamous cell carcinoma. The galactosyltransferase activities in tissue homogenates from both carcinomas were higher than in adjacent normal control with asialoagalactofetuin as a substrate. This activity in adenocarcinoma (27 cases) was two times higher than that in squamous cell carcinoma (19 cases) with statistical significance (p less than 0.001). Using Triton-solubilized enzymes from a particulate fraction, similar differences in the activity were observed with ovalbumin, asialoagalactofetuin, and its beta-eliminated derivative as acceptors but not with bovine submaxillary mucin. These observations mean that the higher activity of galactosyltransferase(s) in lung carcinomas (especially in adenocarcinoma) is mainly responsible for galactosylation of carbohydrate chains in N-glycoside-type but not O-glycoside-type glycoproteins.

PurposeTo investigate the prevalence of microcystic macular edema (MME) in patients with glaucoma and the relationship between glaucomatous visual field defects and MME.Patients and methodsWe analyzed 636 eyes of 341 glaucoma patients who underwent spectral domain optical coherence tomography (SD-OCT). MME was defined as vacuoles observed in the inner nuclear layer (INL) on SD-OCT. Quantitative assessment of MME area was performed using en-face imaging obtained swept-source OCT (SS-OCT) and Adobe Photoshop CS6 Extended software. These values were compared with the visual field results with the Humphrey field analyzer.ResultsMME was observed in 1.6% of eyes. The visual field mean deviation (MD), pattern standard deviation (PSD) and visual acuity was significantly worse (P= 0.023, P=0.037, and P=0.018, respectively) in eyes with MME. The average MME area was 2.38±1.43%. There was no significant correlation between visual field deficits and MME area.ConclusionsThe MME detection rate based on general inspection was 1.6%. MME in glaucomatous eyes were associated with worse MD, PSD, and visual acuity. Further research is needed to increase the number of cases to allow for more detailed analysis.

The Forssman antigenicity of a chemically synthesized globopentaose was studied. Globopentaose at 40 ng showed strong inhibitory activity for the formation of a precipitin line between globopentaosylceramide (Forssman glycolipid) and anti-Forssman rabbit antiserum, while much more pentasaccharide (7 and 100 micrograms, respectively) was required to inhibit a 50% quantitative precipitin reaction and a hemolysis reaction. An immune complex of the 3H-labeled globopentaose with anti-Forssman antibody was hardly formed. Thus, the chemically synthesized globopentaose possesses the same antigenic specificity as globopentaosylceramide but it is difficult to achieve a stable complex with Forssman antibody.

The accumulation of accident-derived radiocesium was investigated in nine water bodies located 300-700 km from the Fukushima Dai-ichi Nuclear Power Plant (FDNPP). (134)Cs from the accident was detected in surface sediment of five water bodies. The (134)Cs concentration, corrected to the time of the accident in 2011, was generally lower than that of (137)Cs, and its spatial pattern does not fully correspond to that of (137)Cs. These results suggest that radiocesium derived from both FDNPP and past global fallout can be separately observed and that the contributions of both sources are non-uniform within these sites. The (134)Cs inventory in surface sediments is smaller than its deposition, suggesting that almost all deposited (134)Cs remains within the catchment and/or a part has been discharged from the saline and brackish water bodies.

The change of acid cholesteryl ester hydrolase activity in mononuclear leukocyte following treatment of diabetes mellitus was studied in 21 patients with non-insulin-dependent diabetes mellitus (NIDDM). Enzyme activity before treatment in the patients was significantly lower than that in 14 age-matched healthy subjects (1.20 +/- 0.15; mean +/- S.E. vs. 2.20 +/- 0.17 nmol/mg protein/h, P less than 0.01). Enzyme activity before treatment in the patients was significantly increased (P less than 0.05) after 4-8 weeks of treatment. However, enzyme activity of 1.43 +/- 0.14 nmol/mg protein/h observed after treatment in the patients was significantly lower (P less than 0.01) than that in the healthy subjects. There was a significant negative correlation between enzyme activity before treatment and the increase in enzyme activity following treatment (rs = -0.555, P less than 0.01, n = 21). These results indicate that low level of enzyme activity may be insufficiently improved by the treatment of diabetes, and the risk for the development of atherosclerosis as viewed from the enzyme activity may persist even after the treatment in NIDDM.

The CD23 molecule is a low-affinity receptor for IgE and has a marked homology in amino acid sequence with C-type animal lectins, including asialoglycoprotein receptor. We tested whether the CD23 antigen can indeed interact with the sugar chain of glycoproteins. Detergent extract of the membrane component from Epstein-Barr Virus (EBV)-transformed human B-cell line, L-KT9 cells, was incubated with asialofetuin (ASF)-coupled Sepharose, and bound proteins were effectively eluted by 0.3 M lactose or galactose which were among the competitive sugars tested. In this eluate, the CD23 molecule was detected by an immunoblotting technique. Because fetuin has both an N- and O-type sugar chain on the molecule, we tested which type of sugar chain can interact with CD23. The CD23 molecule interacted with asialocasein having a sugar chain with the Gal-GalNAc structure with asialobovine submaxillary mucin having the GalNAc structure, and also with ASF; however, it faintly interacted with ASF after removal of the O-type sugar chain by beta-elimination. These results showed that the CD23 molecule can, indeed, interact with the galactose residue, especially with the Gal-GalNAc rather than the Gal-GlcNAc structure of the terminal sugar chain of glycoproteins.

The beta chain of human histocompatibility complex class II antigen, HLA-DR, showed 4 to 5 microheterogeneous spots on a gel obtained by two-dimensional polyacrylamide gel electrophoresis. The types of oligosaccharide chains on the beta chains were analyzed by the lectin-nitrocellulose sheet method for each microheterogeneous spot with 3 cell lines of two haplotypes (HLA-DR 4,4, and 3,3). Two kinds of oligosaccharide chains were observed and were essentially the same in the microheterogeneous spots from all three cell lines. One, the oligosaccharide chain on the most basic spot (beta 1), was stained with peroxidase-coupled concanavalin A (Con A-P.O.) but not with peroxidase-coupled wheat germ agglutinin and was sensitive to endo-beta-N-acetylglucosaminidase H (endo H), indicating that it was a high-mannose type. The oligosaccharide chains on other spots that were not stained with Con A-P.O. but were stained with peroxidase-coupled Ricinus communis agglutinin were resistant to endo H. beta 2 and beta 3 were stained with E-PHA. Thus, they probably had bisected biantennary and others probably had multiantennary complex-type oligosaccharides. Sialidase experiments showed that the charge heterogeneity was due to post-translational sialylation of the oligosaccharide chains. In pulse-chase experiments, the most basic spot of beta chain (beta 1) was labeled first, beta 2 and beta 3 were labeled next, and beta 4 was labeled last. These labeling characters accorded well with the results on the oligosaccharide types mentioned above.

In order to examine the effect of spectinomycin on outbreaks of swine dysentery, experimental infection of piglets with Brachyspira hyodysenteriae was carried out. Feed with and without spectinomycin (SP) was given to each piglet ad libitum and the susceptibility of the piglets to infection with B. hyodysenteriae was compared between SP-treated and untreated piglets. The results showed that the SP-treated piglets did not display clinical signs of swine dysentery unlike the untreated piglets. The sera obtained from these piglets were examined by the microscopic agglutination test and antibodies to B. hyodysenteriae in both groups of experimentally infected piglets were detected and the reaction was serogroup-specific. The agglutination titers were very high in the untreated piglets with dysentery while the titers in the SP-treated piglets were lower than those in the untreated piglets. In addition, the immunoblotting technique was applied and the results demonstrated that 22- and 17-kDa proteins in strain ATCC 31212 (serogroup B) reacted strongly with the sera from the untreated piglets but not with the sera from the SP-treated piglets. The 22- and 17-kDa proteins also reacted with strain ATCC 27164 (serogroup A) which belongs to a different serogroup. The 22- and 17-kDa proteins were also confirmed in six other strains of B. hyodysenteriae which belong to six different serogroups. These proteins were sensitive to proteinase K. These results indicate that the 22- and 17-kDa proteins are common to eight strains of B. hyodysenteriae which differ serologically from each other.

Rhodococcus equi is an emerging opportunistic pathogen of human immunodeficiency virus-infected patients. However, little is known about the distribution of virulent and intermediately virulent R. equi in human environment. In the present study, R. equi was isolated from 173 of 234 (73.9%) samples collected from soil and sand on 115 parks and 49 yards in Japan. The numbers of R. equi from soil and sand ranged from 2.5 x 10(1) to 1.2 x 10(5) per gram of sample. None of 1,294 isolates from those samples showed virulence-associated 15- to 17-kDa antigens and a 20-kDa antigen. These results suggest that avirulent R. equi is widespread in parks and yards, but the human environment has not been contaminated with virulent and intermediately virulent R. equi strains yet.

The aim of this study was to elucidate the distribution and morphological changes of tight junctions during secretion in parotid gland acinar cells. Localization of tight junction-associated polypeptide ZO-1, and of tight junction transmembrane protein Occludin, was examined in rat parotid gland by immunofluorescence and immunogold labelling of ultrathin sections. Adult male Sprague-Dawley rats were intraperitoneally injected with IPR and, after 10 and 30 minutes, parotid glands were extirpated. In control specimens, positive immunoreaction for ZO-1 and Occludin was observed on the adluminal side between adjacent cells in the form of narrow elongated profiles corresponding to intercellular canaliculi. After IPR injection, canaliculi became dilated and fluorescence was no longer seen as a continuous line but appeared as an aggregation of separate bright particles. ZO-1 was more widely distributed and was recognized in other areas of the cytoplasm as well. Concurrently, omega-shaped concavities, marked by actin fluorescence, appeared along the intercellular canaliculi. We concluded that, during exocytosis, the selective permeability barrier to the paracellular pathway, based on tight junctions, becomes more leaky, owing to segregation of Occludin caused by intracellular ZO-1 distributional changes associated with actin filaments.

The authors report a case of radiation-induced calvarial osteosarcoma. A 58-year-old female received subtotal removal of the pituitary adenoma and 5000 rads postoperative irradiation. Seven years later, an osteoblastic osteosarcoma occurred in the frontotemporal region. She received total tumor removal and chemotherapy. However, computed tomography subsequently revealed multiple small lesions at the margin of the bone flap. A chest x-ray film demonstrated lung metastasis. Local recurrence and lung metastasis require careful attention in radiation-induced osteosarcoma patients.

The three-dimensional structure of the collagen fibers in the outer membrane of recurrent chronic subdural hematoma was studied by scanning electron microscopy (SEM). Specimens obtained at surgery were treated with NaOH at room temperature to digest away all cellular components and expose the collagen fibers. SEM observation of the dural side of the outer membrane showed the collagen fibers were woven into a compact feltwork with a dense arrangement. The fiber bundles had a honeycomb structure framed by the collagen fibers. Observation of the hematoma side found the collagen bundles had a sparse wavy appearance. The arrangement of the collagen fibers on the dural side is different from that on the hematoma side. The thick outer membrane may be formed by granulation resulting from inflammatory reaction. Collagen fibrillar networks are not fragile, and may reinforce the outer membrane of the recurrent hematoma.

Two patients with Tolosa-Hunt syndrome (THS) who had atypical lesions in the intrasellar and juxtasellar regions are reported. They manifested with painful ophthalmoplegia. Magnetic resonance imaging (MRI) commonly showed an increase in the volume of the cavernous sinus occupied by homogeneously well-enhanced lesions in both cases. These lesions extended to the intrasellar and juxtasellar regions with meningela enhancement. Follow-up MRI after steroid treatment demonstrated normalized or decreased size of the cavernous sinus. These findings suggested nonspecific inflammatory granulomatosis with atypical extension.

Lower lung field tuberculosis has been considered as rare disease, but it was observed about ten percent in the literature. It seemed that the important problems in lower lung field tuberculosis are delay and difficulty of diagnosis. The materials for this report was accumulated over a period of 10 years, from January 1983 to December 1992 at the National Sanatorium Fukuoka-Higashi Hospital. The criteria for lower lung field tuberculosis are: (1) disease confined to lower lobe except for S6a, middle lobe and lingular segment, (2) bacteriologically and/or histologically proved diagnosis of tuberculosis, (3) clinically considered as tuberculosis for which improved by only anti-tuberculous drugs. During this period, 672 new patients suffering from active pulmonary tuberculosis were admitted to the sanatorium. Of the total of 672 patients, 57 patients were selected as the criteria for lower lung field tuberculosis. The incidence of them was 8.5 percent of the total new admissions. Of these 57 patients, 30 were females and 27 males. The mean age of these 57 patients was 59.4 years ranging between 19 and 86 years. The incidence of them obtained through ten years did not show any tendency to increase or decrease. The respiratory and general symtoms most commonly observed were cough and chest pain. The average duration of used for diagnosis of the lower lung field tuberculosis over 65 years old was 71.7 days and it seemed to show the delay and difficulty of diagnosis.

It is very difficult to treat pulmonary infection with MAC, because we have few effective drugs against this organism. In this situation, an early diagnosis and treatment are very important to manage this disease. We evaluated chest CT scans of the primary pulmonary MAC infection which had no underlying lung diseases and no immunocompromised diseases such as HIV infection. We defined suspected cases of pulmonary MAC infection as cases in which abnormal features of chest CT scans were recognized but frequency of detection of organisms of MAC did not fulfil the diagnostic criteria for atypical mycobacteriosis according to Japanese Mycobacteriosis Research Group of the National Chest Hospitals. CT scans of suspected cases were compared with the definite cases. Results obtained were as follows: 1. In classification by CT scans of primary pulmonary MAC infection, the proportion of localized type and diffuse type was the same both in suspected and definite cases. In localized type, more tuberculosis-like pattern was seen in definite cases. 2. In suspected cases, characteristic features of CT scans of primary pulmonary MAC infection were recognized in the same frequency as in definite cases. 3. In pulmonary tuberculosis-like type, definite cases showed more cavitary lesions than suspected cases. These results showed that a careful long term follow-up of suspected cases with frequent bacteriological tests of sputum and chest CT scannings was important for early diagnosis of primary pulmonary MAC infection.
