Opuntia ficus-indica Mill. (forage cactus) is farmed with relative success in the semi-arid region of the Brazilian northeast for commercial purposes, particularly as forage and food. Endophytic microorganisms are those that can be isolated inside plant tissues and can be a new source to production of enzymes with different potentialities. The objective of this study was to describe the richness of endophytic fungi from O. ficus-indica and to detect the capacity of these species to produce extracellular hydrolytic enzymes. Forty-four endophytic fungi species were isolated. Among them, the most commonly found were Cladosporium cladosporioides (20.43%) and C. sphaerospermum (15.99%). Acremonium terricola, Monodictys castaneae, Penicillium glandicola, Phoma tropica and Tetraploa aristata are being reported for the first time as endophytic fungi for Brazil. The majority of isolated fungi exhibited enzymatic potential. Aspergillus japonicus and P. glandicola presented pectinolytic activity. Xylaria sp. was the most important among the other 14 species with positive cellulase activity. All 24 isolates analysed were xylanase-positive. Protease was best produced by isolate PF103. The results indicate that there is a significant richness of endophytic fungi in O. ficus-indica, and that these isolates indicate promising potential for deployment in biotechnological processes involving production of pectinases, cellulases, xylanases and proteases.

OBJECTIVE: A measles outbreak occurred in S. Paulo state, during 1996 and 1997, resulting in 20,921 cases. Forty seven percent of the cases occurred in people between 20 and 29 years of age, and one of the control strategies of the Department of Health was the vaccination of health care workers. The prevalence of antibodies against measles among the hospital pediatricians was investigated. METHODS: One hundred and fifty samples were taken from volunteer pediatricians to test for measles antibodies using ELISA. A questionnaire about their having had measles and the vaccine was filled out. RESULTS: Of the 150 doctors, 122 (81.4%) were female and 28 (18.6%) male, of between 23 and 46 years of age (mean and median 27 years). The majority (98%-147/150) had protective levels of antibodies against measles (>100 UI/ml); 118 (80.3%-118/147) without and 29 (19.7%-29/147) with a history of measles. Only 3 pediatricians (2%-3/150), had negative serology, 2 without and 1 with a history of measles. Out of the 118 without history of measles, 79 (67%-39/118) in spite of the protective level of antibodies against measles, did not know if they, had been vaccinated. Out of the 79 vaccinated pediatricians, 64 (81%-64/79), had been vaccinated 25 years before, and still maintained protective levels of antibodies. Of the 3 doctors with negative serologies only one declared that he had been vaccinated. CONCLUSIONS: Measles seroprevalence among pediatricians of this hospital is high, especially due to preceding vaccination. On the other hand, the 2% of pediatricians with negative serology, in an epidemic situation could constitute a significant population for the acquisition and dissemination of the disease.

Objetivou-se avaliar Urochloa spp. submetida a duas situações de estresse hídrico: déficit e alagamento. As forrageiras estudadas foram: U. humidicola cv. Tully, U. humidicola cv. Llanero, U. brizantha cv. BRS Piatã, U. brizantha cv. Xaraés, U. decumbens cv. Basilisk e U. ruziziensis cv. Kenedy, e os parâmetros avaliados foram relacionados às características estruturais e morfogênicas. O trabalho foi desenvolvido em casa de vegetação pertencente à Unidade Universitária de Aquidauana. Os tratamentos consistiram em diferentes condições de umidade do solo: 50 e 80% da capacidade de retenção de umidade do solo, e lâminas de 0cm, 1cm e 5cm de água acima do solo, com quatro repetições, em delineamento inteiramente ao acaso. O modelo considerou o efeito de umidade do solo para cada espécie forrageira a 5% de significância. As características estruturais e morfogênicas do capim-tully não foram afetadas pelas diferentes condições hídricas aqui avaliadas. O capim-llanero não reagiu ao alagamento. O capim-piatã foi tolerante às condições de déficit hídrico e apresentou mecanismos de adaptação ao alagamento. A U. decumbens cv. Basilisk apresentou respostas adaptativas que melhoram sua tolerância ao alagamento. A U. ruziziensis cv. Kenedy apresentou-se inapta às condições de alagamento e de déficit hídrico.(AU)


The aim of this study was to evaluate braquiarias submitted to two situations of water stress: deficit and flood. The studied forages were: U. humidicola cv. Tully, U. humidicola cv. Llanero, U. brizantha cv. BRS Piatã, U. brizantha cv. Xaraés, U. decumbens cv. Basilisk and U. ruziziensis cv. Kenedy, and the parameters evaluated were related to the structural and morphogenic characteristics. The work was carried out in a greenhouse at the University Unit of Aquidauana. The treatments consisted of different soil moisture conditions: 50 and 80% of the moisture retention capacity of the soil, and slides of 0cm, 1cm and 5cm of water above the soil, with four replications, in a completely randomized design. The model considered the effect of soil moisture for each forage species at 5% of significance. The structural and morphogenic characteristics of grass-tully were not affected by the different water conditions evaluated here. The grass-llanero did not react to flooding. The grass-piatã was tolerant to water deficit conditions and presented adaptation to flooding. The U. decumbens cv. Basilisk presented adaptive responses that improved its tolerance to flooding. The U. ruziziensis cv. Kenedy was unfit for flood and water deficit conditions.(AU)

The objectives of this study were to analyze body measurements of 40 crossbred bulls grazing low quality forage with different supplementation strategies, to estimate interrelationships among those measurements and carcass and body compositions, and to develop systems of equations to predict body fat using body and carcass measurements. Eight animals were slaughtered at the beginning of the experiment, and the remaining animals were slaughtered at 90 or 220 d. The biometric measures (BM) were obtained the day before the slaughter and included hook width, pin width, pelvic girdle length, rump depth, rump height, abdomen width, body length, height at withers, rib depth, girth, and body diagonal length. Other measurements included full, shrunk, and empty BW; internal physical and chemical fats; body volume; body area; carcass weight; 9th- to 11th-rib section weight and composition; fat thickness; subcutaneous fat; intermuscular fat; carcass chemical fat; and empty body physical and chemical fats. The relationships between BM and body components were evaluated, and equations to predict body area, body volume, subcutaneous fat, and carcass and body physical and chemical fat were developed. Biological interpretations of the parameter estimates of equations were similar to those found in the literature such as a ratio of 1 kg of subcutaneous fat to 1.6 kg of intermuscular fat and a deposit of 72 to 76% of body fat in the carcass. The first system used to predict carcass and empty body physical and chemical fat was devised using in vivo information, whereas the second system used BW and the 9th- to 11th-rib fat weight. Our results indicated the combination of BW, carcass traits, and BM was precise and accurate in estimating carcass and body fat composition of backgrounding bulls. The second system had better adequacy statistics [r(2) > 0.92, concordance correlation coefficient (CCC) > 0.957, and root mean square error (RMSE) < 14.4% of the average observed value] compared with the first system. The first system had acceptable adequacy statistics (r(2) > 0.767, CCC > 0.866, and RMSE varying from 15.8 to 22.3% of the average observed value). For both systems, the simultaneous F-test of the linear regression of observed on model-predicted values indicated intercepts were equal to zero, and slopes were equal to 1 (P > 0.246). We concluded that BM can improve the accuracy and precision of the predictions of body composition of grazing animals.

The development of piatã grass fertilized with phosphorus sources with different solubilities in water was evaluated. The experiment was developed in the unit of Aquidauana of UEMS, MS, from February to August, 2012. Four treatments were evaluated: control (without phosphorus fertilization), slow solubility, fast solubility (readily available), and mixed solubility sources of phosphorus (FH Pasture(r)). It was adopted a randomized blocks design with four blocks and four replicates in a total of 16 pastures (0.5 ha each one). Measurements were performed weekly to evaluate structural characteristics and morphogenic variables of the plants. Data were submitted to a variance analysis according to the model. When necessary, the means of treatments with different phosphorus fertilizer sources were compared with the control using the Dunnett test and with other treatments by using student's t test. The significance level of 5% was adopted in all analysis. No significant effect was observed (P 0.05) in the evaluated traits in the first year of the experiment.

RESUMO

The study of 27 illegal users of marijuana, through psychoanalysis and analytical group psychotherapy and through psychiatric examinations, made it possible to discover that these patients felt their parents to be aggressive or indifferent or unaffectionate or absent (among the parents of different temperaments, and confirmed by statistical data). These feelings were related to the errors committed by their parents during the molding periods (lactation and family environment).

The activity of horse liver alcohol dehydrogenase was inhibited by phenylhydrazine. Kinetic experiments showed that this compound produced linear competitive inhibition with respect to NAD+ and linear noncompetitive inhibition with respect to ethanol. These results suggested that the inhibitor competes with NAD+ for the coenzyme binding site of alcohol dehydrogenase, forming a dead-end complex with the free form of the enzyme. A Ki value of 393 +/- 51 microM was estimated for the enzyme-inhibitor complex. Further evidence for this mechanism of inhibition arose from the fact that the same kind of inhibition was found for rabbit muscle lactate dehydrogenase. The Ki value for the lactate dehydrogenase-phenylhydrazine complex was 43.41 +/- 2.10 mM. The significant difference between these Ki values is explained in terms of known differences in hydrophobicity of the nicotinamide binding region in the two enzymes.

OBJECTIVE: To present an analysis of the occurrence of gastrointestinal bleeding in children and emphasize: i) diagnostic methods; ii) the organized use of different therapeutic approaches in upper gastrointestinal bleeding; iii) the review of concepts, classifications and techniques used in endoscopy, which are important to the practice of clinical pediatrics. METHODS: Literary review of chapters selected from textbooks, pertinent articles obtained through the Medline system and active search, as well as personal archives belonging to the authors. RESULTS: The differential diagnosis of gastrointestinal bleeding in children varies according to the age. The causes of upper gastrointestinal bleeding are subdivided into variceal and nonvariceal. Nonselective beta-blockers are recommended to prevent variceal bleeding. Vasoactive drugs, such as somatostatin, octreotide, and glypressin may be used, showing good results in both variceal and nonvariceal acute bleeding. Both sclerotherapy and variceal ligation can be used in children to achieve variceal eradication. Cyanoacrylate is effective and presents the lowest complication rate related to gastric variceal bleeding. The presence of hemorrhage stigmata, such as active bleeding and visible vessel in ulcers is indicative of a higher risk for recurrent bleeding, suggesting the need for endoscopic hemostasis. Proton pump inhibitors are more efficacious than H2-receptor antagonists to promote the peptic ulcer healing. CONCLUSION: The correct etiologic diagnosis of gastrointestinal bleeding in children is fundamentally important to adopt the adequate therapeutic approach, whose main advances concern the pharmacological and the endoscopic treatment.

1The Enantiomeric Ratio (E) of the enzyme, acting as specific catalysts in resolution of enantiomers, is an important parameter in the quantitative description of these chiral resolution processes. In the present work, two novel methods hereby called Method I and II, for estimating E and the kinetic parameters Km and Vm of enantiomers were developed. These methods are based upon initial rate (v) measurements using different concentrations of enantiomeric mixtures (C) with several molar fractions of the substrate (x). Both methods were tested using simulated "experimental data" and actual experimental data. Method I is easier to use than Method II but requires that one of the enantiomers is available in pure form. Method II, besides not requiring the enantiomers in pure form shown better results, as indicated by the magnitude of the standard errors of estimates. The theoretical predictions were experimentally confirmed by using the oxidation of 2-butanol and 2-pentanol catalyzed by Thermoanaerobium brockii alcohol dehydrogenase as reaction models. The parameters E, Km and Vm were estimated by Methods I and II with precision and were not significantly different from those obtained experimentally by direct estimation of E from the kinetic parameters of each enantiomer available in pure form.

RESUMO

The authors discuss the microscopic changes in gastrocnemius muscle in Leptospirosis in 63 patients. The hyaline necrosis frequently associated to a hemorrhagic infiltrate represented the most important feature in the lesional picture of the disease. Other findings were swelling, loss of cross striations, vacuolation and regenerative changes of muscle fibres. This last one was represented by multinucleated sarcoblasts and the presence of thin and strongly basophilic new-formed fibres. Interstitial hemorrhage was frequently seen and an inflammatory afflux was only rarely observed. This histological picture was considered as characteristic and highly significant for the diagnosis of Leptospirosis.
