MAIN CONCLUSION: Transcription of PagMYB147 was induced in poplar infected by Melampsora magnusiana, and a decline in its expression levels increases the host's susceptibility, whereas its overexpression promotes resistance to rust disease. Poplars are valuable tree species with diverse industrial and silvicultural applications. The R2R3-MYB subfamily of transcription factors plays a crucial role in response to biotic stresses. However, the functional studies on poplar R2R3-MYB genes in resistance to leaf rust disease are still insufficient. We identified 191 putative R2R3-MYB genes in the Populus trichocarpa genome. A phylogenetic analysis grouped poplar R2R3-MYBs and Arabidopsis R2R3-MYBs into 33 subgroups. We detected 12 tandem duplication events and 148 segmental duplication events, with the latter likely being the main contributor to the expansion of poplar R2R3-MYB genes. The promoter regions of these genes contained numerous cis-acting regulatory elements associated with response to stress and phytohormones. Analyses of RNA-Seq data identified a multiple R2R3-MYB genes response to Melampsora magnusiana (Mmag). Among them, PagMYB147 was significantly up-regulated under Mmag inoculation, salicylic acid (SA) and methyl jasmonate (MeJA) treatment, and its encoded product was primarily localized to the cell nucleus. Silencing of PagMYB147 exacerbated the severity of Mmag infection, likely because of decreased reactive oxygen species (ROS) production and phenylalanine ammonia-lyase (PAL) enzyme activity, and up-regulation of genes related to ROS scavenging and down-regulation of genes related to PAL, SA and JA signaling pathway. In contrast, plants overexpressing PagMYB147 showed the opposite ROS accumulation, PAL enzyme activity, SA and JA-related gene expressions, and improved Mmag resistance. Our findings suggest that PagMYB147 acts as a positive regulatory factor, affecting resistance in poplar to Mmag by its involvement in the regulation of ROS homeostasis, SA and JA signaling pathway.

Rice (Oryza sativa) production consumes a huge amount of fresh water, and improvement of drought tolerance in rice is important to conserve water resources and minimize yield loss under drought. However, processes to improve drought tolerance in rice have not been fully explored, and a comparative study between rice and wheat (Triticum aestivum) is an effective method to understand the mechanisms determining drought tolerance capacity. In the present study, we applied short-term drought stress to Shanyou 63 rice and Yannong 19 wheat to create a range of water potentials and investigated the responses of gas exchange, plant hydraulic conductance, and root morphological and anatomical traits to soil drought. We found that photosynthesis in rice was more sensitive to drought stress than that in wheat, which was related to differences in the decline of stomatal conductance and plant hydraulic conductance (Kplant). The decline of Kplant under drought was mainly driven by the decrease of soil-root interface hydraulic conductance (Ki) because Ki was more sensitive to drought than root and shoot hydraulic conductance and the soil-root interface contributed to >40% of whole-plant hydraulic resistance in both crops. Root shrinkage in response to drought was more severe in rice than that in wheat, which explains the larger depression of Ki and Kplant under drought stress in rice. We concluded that the decline of Ki drives the depression of Kplant and photosynthesis in both crops, and the plasticity of root morphology and anatomy is important in determining drought tolerance capacity.

The grain yield is closely associated with spikelet fertility in rice (Oryza sativa L.) under high temperatures, and nitrogen (N) plays a crucial role in yield formation. To investigate the effect of panicle N application on yield formation under high temperatures at the panicle initiation stage, two rice varieties [Liangyoupeijiu (LYPJ, heat susceptible) and Shanyou63 (SY63, heat tolerant)] were grown and exposed to high daytime temperature (HT) and control temperature (Control) during the panicle initiation stage. Low (LPN) and high (HPN) panicle N applications were conducted. HT markedly decreased the yields by 87% at LPN and 48% at HPN in LYPJ and 31% at LPN and 36% at HPN in SY63. The decrease in grain yield under HT was primarily attributed to the decline in spikelet fertility, HPN increased spikelet fertility. HT resulted in the abnormal development of anthers, which included disordered, enlarged, and broken anther wall layers, degraded and irregularly shaped microspores, delayed tapetum degradation, less vacuolated microspores per locule, abnormal and aborted pollen grains; however, HPN improved the development of anthers under HT, particularly in LYPJ. A high rate of evapotranspiration resulted in an approximately 1°C decrease in panicle temperatures at HPN compared with that at LPN in both varieties under HT. Overall, these results demonstrate that the increased panicle N application favors normal anther development in LYPJ by decreasing the panicle temperature, which results in high pollen viability and spikelet fertility, and consequently less yield loss under HT.

Vein severing in plants caused by leaf damage is common in fields where crops are cultivated. It is hypothesized that leaves with complex reticulate venation can withstand hydraulic disturbances caused by vein severing, thereby preserving leaf carbon assimilation. However, limited research focuses on vein damage of leaves with parallel venation. We studied how vein-severing affected the photosynthetic traits of rice (Oryza sativa) leaves in seconds, minutes and days, under varying water-demand conditions and differing extents of water supply disruption. Rice leaves completely lost their photosynthetic capacity within 2.5 minutes after excision. Severing the midrib resulted in reduced light-saturated photosynthetic rate (A), stomatal conductance (gsw ) and transpiration rate (E) by 2.6, 6.8 and 5.9%, respectively, already after thirty minutes. We further investigated the photosynthetic trait responses to various extents of leaf width severing, while keeping the midrib functional. Surprisingly, A, gsw and E in the downstream area of the severed leaves largely remained stable, showing minimal variation across different leaf width severing ratios. These traits declined only slightly even under increased ambient light intensity and leaf-to-air vapor pressure deficit. This sustained photosynthesis post-severing is attributed to the efficient lateral water transport. Long-term leaf damage slightly but not significantly, impacted the downstream photosynthetic traits within five days post-severing. However, a more pronounced reduction in gas exchange during leaf senescence was observed nine days after severing. These findings suggested that rice leaves can tolerate hydraulic disturbances from vein severing and maintain functionality under various conditions, which is crucial for crop yield stability. However, long-term consequences require further investigation.

Panicle photosynthesis is crucial for grain yield in cereal crops; however, the limiting factors for panicle photosynthesis are poorly understood, greatly impeding improvement in this trait. In the present study, pot experiments were conducted to investigate the limiting factors for panicle photosynthesis at the anthesis stage in seven rice genotypes and to examine the temporal variations in photosynthesis during the grain filling stage in the Liangyou 287 genotype. At the anthesis stage, leaf and panicle photosynthesis was positively correlated with stomatal conductance and maximum carboxylation rate, which were in turn associated with hydraulic conductance and nitrogen content, respectively. Panicle hydraulic conductance was positively correlated with the area of bundle sheaths in the panicle neck. During grain filling, leaf and panicle photosynthesis remained constant at the early stage but dramatically decreased from 8 to 9 days after anthesis. The trends of variations in panicle photosynthesis were consistent with those in stomatal conductance but not with those in maximum carboxylation rate. At first, the maximum carboxylation rate and respiration rate in the panicle increased, through elevated panicle nitrogen content, but then drastically decreased, as a result of dehydration. The present study systematically investigated the limiting factors for panicle photosynthesis, which are vital for improving photosynthesis and crop yield.

Although mesophyll conductance (gm ) is known to be sensitive to temperature (T), the mechanisms underlying the temperature response of gm are not fully understood. In particular, it has yet to be established whether interspecific variation in gm -T relationships is associated with mesophyll anatomy and vein traits. In the present study, we measured the short-term response of gm in eight crop species, and leaf water potential (Ψleaf ) in five crop species over a temperature range of 15-35°C. The considered structural parameters are surface areas of mesophyll cells and chloroplasts facing intercellular airspaces per unit leaf area (Sm and Sc ), cell wall thickness (Tcw ), and vein length per area (VLA). We detected large interspecific variations in the temperature responses of gm and Ψleaf . The activation energy for gm (Ea,gm ) was found to be positively correlated with Sc , although it showed no correlation with Tcw . In contrast, VLA was positively correlated with the slope of the linear model of Ψleaf -T (a), whereas Ea,gm was marginally correlated with VLA and a. A two-component model was subsequently used to model gm -T relationships, and the mechanisms underlying the temperature response of gm are discussed. The data presented here indicate that leaf anatomy is a major determinant of the interspecific variation in gm -T relationships.

Low-light stress compromises photosynthetic and energy efficiency and leads to spikelet sterility; however, the effect of low-light stress on pollen tube elongation in the pistil remains poorly understood. The gene RGA1, which encodes a Gα-subunit of the heterotrimeric G-protein, enhanced low-light tolerance at anthesis by preventing the cessation of pollen tube elongation in the pistil of rice plants. In this process, marked increases in the activities of acid invertase (INV), sucrose synthase (SUS) and mitochondrial respiratory electron transport chain complexes, as well as the relative expression levels of SUTs (sucrose transporter), SWEETs (sugars will eventually be exported transporters), SUSs, INVs, CINs (cell-wall INV 1), SnRK1A (sucrose-nonfermenting 1-related kinase 1) and SnRK1B, were observed in OE-1 plants. Accordingly, notable increases in contents of ATP and ATPase were presented in OE-1 plants under low-light conditions, while they were decreased in d1 plants. Importantly, INV and ATPase activators (sucrose and Na2 SO3 , respectively) increased spikelet fertility by improving the energy status in the pistil under low-light conditions, and the ATPase inhibitor Na2 VO4 induced spikelet sterility and decreased ATPase activity. These results suggest that RGA1 could alleviate the low-light stress-induced impairment of pollen tube elongation to increase spikelet fertility by promoting sucrose unloading in the pistil and improving the metabolism and allocation of energy.

Leaf rolling is a physiological response to drought that may help to reduce water loss, but its significance as a contribution to drought tolerance is uncertain. We scored the leaf rolling of four rice genotypes along an experimental drought gradient using an improved cryo-microscopy method. Leaf water potential (Ψleaf), gas exchange, chlorophyll fluorescence, leaf hydraulic conductance, rehydration capacity, and the bulk turgor loss point were also analysed. During the drought process, stomatal conductance declined sharply to reduce water loss, and leaves rolled up before the stomata completely closed. The leaf water loss rate of rolled leaves was significantly reduced compared with artificially flattened leaves. The Ψleaf threshold of initial leaf rolling ranged from -1.95 to -1.04 MPa across genotypes. When a leaf rolled so that the leaf edges were touching, photosynthetic rate and stomatal conductance declined more than 80%. Across genotypes, leaf hydraulic conductance declined first, followed by gas exchange and chlorophyll fluorescence parameters. However, the Ψleaf threshold for a given functional trait decline differed significantly among genotypes, with the exception of leaf hydraulic conductance. Our results suggested that leaf rolling was mechanistically linked to drought avoidance and tolerance traits and might serve as a useful phenotypic trait for rice breeding in future drought scenarios.

Improvement of photosynthesis in non-foliar green tissues is beneficial for enhancing crop yield. Recently, we have demonstrated that panicle stomatal conductance is a major limiting factor for photosynthesis. However, mechanisms underlying the responses of panicle stomatal conductance (gs,panicle) and photosynthesis (Apanicle) to environmental stimuli remain unknown. In the present study, the responses of gs,panicle and leaf stomatal conductance (gs,leaf) to exogenous application of abscisic acid and step-changes in vapor pressure deficit were investigated at the anthesis stage in pot-grown rice plants. Furthermore, the effects of drought on Apanicle and leaf photosynthesis (Aleaf) were examined. Smearing and xylem feeding of abscisic acid significantly decreased gs,leaf. In contrast, while smearing of abscisic acid substantially increased gs,panicle, its xylem feeding dramatically decreased gs,panicle. In addition, both gs,leaf and gs,panicle effectively responded to step changes in vapor pressure deficit. Furthermore, both Aleaf and Apanicle were sensitive to plant dehydration; however, given the lower sensitivity of panicle water potential than leaf water potential to drought, Apanicle was less sensitive to soil drought than Aleaf. These findings indicate that gs,panicle is hydropassively regulated, while panicle photosynthesis is less sensitive to drought.

BACKGROUND AND AIMS: Plasticity of leaf growth and photosynthesis is an important strategy of plants to adapt to shading stress; however, their strategy of leaf development to achieve a simultaneous increase in leaf area and photosynthesis under shading remains unknown. METHODS: In the present study, a pot experiment was conducted using three rapeseed genotypes of Huayouza 50 (HYZ50), Zhongshuang 11 (ZS11) and Huayouza 62 (HYZ62), and the responses of plant growth, leaf morphoanatomical traits, cell wall composition and photosynthesis to shading were investigated. KEY RESULTS: Shading significantly increased leaf area per plant (LAplant) in all genotypes, but the increase in HYZ62 was greater than that in HYZ50 and ZS11. The greater increment of LAplant in HYZ62 was related to the larger decrease in leaf mass per area (LMA) and leaf density (LD), which were in turn related to less densely packed mesophyll cells and thinner cell walls (Tcw). Moreover, shading significantly increased photosynthesis in HYZ62 but significantly decreased it in HYZ50. The enhanced photosynthesis in HYZ62 was related to increased mesophyll conductance (gm) due primarily to thinner cell walls. CONCLUSIONS: The data presented indicate that the different plasticity of mesophyll cell density, cell wall thickness and cell wall composition in response to shading can dramatically affect leaf growth and photosynthesis.

The plasma membrane subfamily of aquaporins [plasma membrane intrinsic proteins (PIPs)], which facilitates the CO2 diffusion across membranes, is proposed to play an important role in mesophyll conductance to CO2 (gm ), a major limiting factor of photosynthesis. However, recent studies implied no causal relationship between gm and PIPs because they failed to repeat the previous observed differences in gm between PIP knockout lines and their wild-type. The contrasting results on the role of PIPs in gm were interpreted as the different growth conditions among studies, which has never been tested. Here, we assessed whether the differences in gm between wild-type and PIP knockout lines, Ospip1;1, Ospip1;2 and Ospip2;1, varied with growth condition (field versus pot condition) and growth stages in rice. Under field conditions, no differences were observed in plant performance, photosynthetic rate (A) and gm between PIP knockout lines and the wild-type. However, in agreement with previous studies, two out of three knockout lines showed significant declines in tiller number, plant height, A and gm under pot conditions. Moreover, we found that the differences in A and gm between PIP knockout lines and the wild-type varied with the growth stage of the plants. Our results showed that the differences in gm between PIP knockout lines and wild-type were depending on the growth environments and stage of the plants, and further efforts are required to reveal the underlying mechanisms.

BACKGROUND: Walnuts (Juglans regia L.) are known for their nutrient-rich nuts and are one of the important economic tree species in the world. However, due to global warming and soil salinization, walnuts suffer from various abiotic stresses. TIFY (TIF[F/Y]XG) proteins play an essential role in the growth and development of plants, signal transduction, and stress response in plants. At present, although the TIFY gene family of a number of plants has been identified and studied, how TIFY takes part in stress tolerance remains obscure and many functions of TIFY require further investigation. RESULT: In this study, twenty-one TIFY transcription factors were identified in the walnut genome database, and they were divided into four subfamilies (TIFY, JAZ, ZML, and PPD) by bioinformatics analysis. Chromosome location revealed tandem duplication of some genes. Phylogenetic tree analysis showed JrTIFYs were closely related to the TIFY gene family of Arabidopsis thaliana (A. thaliana). qRT-PCR (quantitative real-time PCR) analysis revealed the TIFY genes have different expression patterns in 'Qingxiang' and 'Xiangling' walnut varieties under drought, heat, and salt stress. JAZ subfamily was more expressed in different abiotic stress than other subfamilies. The expressions of JrTIFY14 under heat and salt stress were significantly higher than those under drought stress. However, the expression of JrTIFYs was not significant in 'Xiangling'. CONCLUSION: This study reveals the TIFY gene family plays an important role in walnuts facing abiotic stresses and provides a theoretical basis for walnut breeding.

Phloem unloading plays an important role in photoassimilate partitioning and grain yield improvements in cereal crops. The phloem unloading strategy and its effects on photoassimilate translocation and yield formation remain unclear in rice. In this study, plasmodesmata were observed at the interface between the sieve elements (SEs) and companion cells (CCs), and between the SE-CC complex and surrounding parenchyma cells (PCs) in phloem of the dorsal vascular bundle in developing caryopses. Carboxyfluorescein (CF) signal was detected in the phloem of caryopses, which showed that CF was unloaded into caryopses. These results indicated that the SE-CC complex was symplasmically connected with adjacent PCs by plasmodesmata. Gene expression for sucrose transporter (SUT) and cell wall invertase (CWI), and OsSUT1 and OsCIN1 proteins were detected in developing caryopses, indicating that rice plants might actively unload sucrose into caryopses by the apoplasmic pathway. Among three rice recombinant inbred lines, R201 exhibited lower plasmodesmal densities at the boundaries between cell types (SE-CC, SE-PC and CC-PC) in developing caryopses than R91 and R156. R201 also had lower expression of SUT and CWI genes and lower protein levels of OsSUT1 and OsCIN1, as well as CWI activity, than R91 and R156. These data agreed with stem non-structural carbohydrate (NSC) translocation and grain yields for the three lines. The nitrogen application rate had no significant effect on plasmodesmal densities at the interfaces between different cells types, and did not affect CF unloading in the phloem of developing caryopses. Low nitrogen treatment enhanced expression levels of OsSUT and OsCIN genes in the three lines. These results suggested that nitrogen application had no substantial effect on symplasmic unloading but affected apoplasmic unloading. Therefore, we concluded that poor symplasmic and apoplasmic unloading in developing caryopses might result in low stem NSC translocation and poor grain yield formation of R201.

BACKGROUND: Juglans regia L. is an important nut tree that has a wide range of distribution in temperate regions of the world. In some walnut orchards, walnut blight can become a problematic disease that affects the growth of walnut trees. To explore the correlation between biochemical response and walnut resistance, we inoculated four walnut cultivars with Xanthomonas arboricola pv. juglandis (Xaj). The walnut cultivars were, namely, 'Xiangling', 'Xiluo 2', 'Yuanfeng' and 'Xifu 2'. Total phenol content (TPC) and total flavonoid content (TFC) were measured, whereby nine major phenolic compounds and several relevant enzymes were identified. RESULTS: The results showed that the most resistant and susceptible walnut varieties were 'Xiluo 2' and 'Xifu 2' respectively. The reaction of walnut to Xaj was characterized by the early accumulation of phenolic compounds in the infected site. After inoculation with Xaj, we found that the resistant variety 'Xiluo 2' show the significant differences with other varieties at different time points through the determination of related antioxidant enzymes such as catalase (CAT) and peroxidase (POD). Meanwhile, the phenylalanine ammonia lyase (PAL) of 'Xiluo 2' increased significantly at 8 day post infection (dpi) and made differences from the control samples, while other varieties changed little. And the polyphenol oxidase (PPO) was significantly higher than in the control at 16 dpi, maintaining the highest and the lowest activity in 'Xiluo 2' and 'Xifu 2' respectively. It was also found that the content of protocatechuic acid in all cultivars increased significantly at 4 dpi, and 'Xiluo 2' was significantly higher than that of the control. In the early stage of the disease, ferulic acid content increased significantly in 'Xiluo 2'. CONCLUSION: Our findings confirmed that the metabolism of phenolic compounds and related defense enzymes are of great significance in the response of walnut to Xaj.

MAIN CONCLUSION: This study suggests that stomatal and leaf structures are highly correlated, and mesophyll cell size is an important anatomical trait determining the coordination between stomatal size and mesophyll porosity. A comprehensive study of the correlations between the structural traits and on their relationships with gas exchange parameters may provide some useful information into leaf development and improvement in efficiencies of photosynthetic CO2 fixation and transpirational water loss. In the present study, nine plant materials from eight crop species were pot grown in a growth chamber. Leaf structural traits, gas exchange, and leaf nitrogen content were measured. We found that stomatal size, mesophyll cell size (MCS), and mesophyll porosity were positively correlated and that the surface areas of mesophyll cells and chloroplasts facing intercellular air spaces were positively correlated with both stomatal density and stomatal area per leaf area (SA). These results suggested that the developments of stomata and mesophyll cells are highly correlated among different crop species. Additionally, MCS was positively correlated with leaf thickness and negatively correlated with leaf density and leaf mass per area, which indicated that MCS might play an important role in leaf structural investments and physiological functions among species. In summary, this study illustrates the correlations between stomatal and mesophyll structures, and it highlights the importance of considering the covariations among leaf traits with the intent of improving photosynthesis and iWUE.

Studies investigating the mechanisms underlying the variation of photosynthesis along plant phylogeny and especially during domestication are of great importance, and may provide new insights to further improve crop photosynthesis. In the present study, we compiled a database including 542 sets of data of leaf gas exchange parameters and leaf structural and chemical traits in ferns and fern allies, gymnosperms, non-crop angiosperms, and crops. We found that photosynthesis was dramatically improved from ferns and fern allies to non-crop angiosperms, and further increased in crops. The improvement of photosynthesis during phylogeny and domestication was related to increases in carbon dioxide diffusional capacities and, to a lesser extent, biochemical capacity. Cell wall thickness rather than chloroplast surface area facing intercellular airspaces drives the variation of mesophyll conductance. The variation of the maximum carboxylation rate was not related to leaf nitrogen content. The slope of the relationship between mass-based photosynthesis and nitrogen was lower in crops than in non-crop angiosperms. These findings suggest that the manipulation of cell wall thickness is the most promising approach to further improve crop photosynthesis, and that an increase of leaf nitrogen will be less efficient in improving photosynthesis in crops than in non-crop angiosperms.

Plant functional traits, including leaf and root economic traits, are important for understanding the composition and function of ecosystems. However, plant functional traits of crop species and the relationships between them, and their responses to environmental variations are not fully understood. In the present study, the traits in the leaf and root economics spectrum (LES and RES) and plant biomass were investigated in 14 wheat genotypes grown with sufficient or limited nitrogen (N) supply. We found that N had significant impacts on the LES and RES traits and on the relationships among them. Our results generally supported the hypothesized LES, but did not support the RES or plant economics spectrum concept among wheat plants regardless of N treatment. More importantly, we found that more conservative leaf and root economic traits are beneficial for shoot biomass accumulation in wheat plants grown with limited N supply, and for the improvement in the tolerance of wheat to N stress. The data presented suggest that growth conditions should be accounted for when studying trait-to-trait relationships, and that more conservative resource use strategies could be used as promising targets for wheat breeding programs with limited N input.

BACKGROUND AND AIMS: Leaf structure is an important determinant of leaf photosynthesis; however, the impacts of leaf structural traits on gas exchange parameters are still not fully understood. In the present study, 11 rice genotypes were grown in pots to investigate the influence of leaf structural traits on leaf photosynthesis and hydraulic conductance (Kleaf). METHODS: In this study, leaf photosynthetic rate (A), stomatal conductance (gs), mesophyll conductance and Kleaf were measured. In addition, leaf structural traits including leaf thickness (LT), leaf mass per area and leaf xylem and phloem sizes were also measured to investigate their impacts on rice photosynthesis. KEY RESULTS: We found that the total area of xylem conduits per major vein (Xmajor), leaf phloem area per minor vein (Pminor) and LT were positively correlated with Kleaf, gs and A. The path analysis suggested that, however, only Pminor had a direct impact on A; Xmajor had an indirect impact on A via gs and Pminor, while LT did not show any direct or indirect impact on A. CONCLUSION: This study highlighted the importance of manipulations in Xmajor and Pminor, two previously overlooked leaf traits, to improve leaf photosynthesis in rice plants.

Phloem unloading and loading are associated with stem non-structural carbohydrates (NSCs) accumulation and remobilization in rice (Oryza sativa L.). Four rice recombinant inbred lines (R032, R191, R046, and R146) derived from a cross between Zhenshan 97 and Minghui 63 were used to investigate the contributions of stem large and small vascular bundles (SVBs) to NSCs accumulation and translocation. Before heading, the parenchyma cells in stem cortex tissues (PCs) surrounding SVBs had higher starch density than those surrounding large vascular bundles (LVBs). Moreover, the protein levels of sucrose transporters (SUTs), cell wall invertase, sucrose synthase, and adenosine diphosphate glucose pyrophosphorylase, as well as the phloem plasmodesma densities were higher in SVBs than those in LVBs. After heading, starch density decreased more in PCs surrounding SVBs than in LVBs. Also, the protein levels of SUTs, α-amylase, sucrose phosphate synthase and sucrose synthase, the phloem plasmodesma densities in SVBs were higher than those in LVBs. The correlations of the number and total cross-sectional area of SVBs with mass and contribution to yield of transferred NSCs were higher than those of LVBs. Our results suggest that SVBs may have higher contributions to pre-anthesis stem NSCs accumulation and post-anthesis translocation than LVBs, which is potentially attributed to the high level of protein and enzyme involved in stem unloading and loading via apoplastic and symplastic pathways.

Nitrogen (N) is one of the most important nutrients for crop growth and yield formation, as it is an important constituent in a large amount of proteins, cell walls, and membranes related to photosynthesis. Recently, increasing studies have suggested the important roles of photosynthetic induction and stomatal movement under fluctuating light in regulating plant carbon assimilation and water use efficiency. How leaf N content affects photosynthetic induction remains uncertain. Here, we observed a significantly faster photosynthetic induction with the increasing supply of N under fluctuating light conditions. Photosynthetic induction was mainly limited by biochemical processes but not stomatal opening after a stepwise increase in irradiance across different N supplies. Higher N supply enhanced photosynthetic efficiency under constant and fluctuating light conditions but reduced leaf intrinsic water use efficiency (WUEi ). This study is mainly focused on clarifying the crucial limitation of photosynthetic induction under different N treatments, which may facilitate the improvement of photosynthetic efficiency under complicated environments in the future.
