RESUMO

In order to investigate the effects of artificial diets on the population growth of root maggot Bradysia impatiens, its population growth parameters were assayed on eight artificial diets (Diet 1, D2, D3, D4, D5, D6, D7, and D8). Results showed that developmental duration from egg to pupa was successfully completed on all eight artificial diets. However, the egg to pupal duration was shortest, while the survival rate of four insect stages was lowest when B. impatiens was reared on D1. When B. impatiens was reared on D7 and D8, the survival rate, female longevity, and female oviposition were higher than those reared on other diets. When B. impatiens was reared on D7, the intrinsic rate of increase (rm = 0.19/d), net reproductive rate (R0 = 39.88 offspring per individual), and finite rate of increase (λ = 1.21/d) were higher for its population growth with shorter generation time (T = 19.49 d) and doubling time (Dt = 3.67 d). The findings indicate that the D7 artificial diet is more appropriate for the biological parameters of B. impatiens and can be used an indoor breeding food for population expansion as well as further research. We propose that vitamin C supplement added to the D7 is critical for the improvement of the B. impatiens growth.

The fall armyworm, Spodoptera frugiperda Smith (Lepidoptera: Noctuidae), a common agricultural pest known for its extensive migration and wide host ranges, causes considerable harm to maize (Zea mays L.). In this study, we utilized two molecular marker genes, COI and Tpi, to compare the genetic characteristics of the collected original samples. Additionally, through an interactive study between S. frugiperda larvae and six maize varieties aiming to understand the insect's adaptability and resistance mechanisms, our analysis revealed that both the COI and Tpi genes identified S. frugiperda as the corn strain. Further examination of the larvae showed significant differences in nutritional indices, digestive, and detoxification enzyme activities. Special maize varieties were found to offer higher efficiency in nutrient conversion and assimilation compared with common varieties. This study revealed adaptations in S. frugiperda's digestive and detoxification processes in response to the different maize varieties. For instance, larvae reared on common maize exhibited elevated amylase and lipase activities. Interestingly, detoxification enzyme activities exhibited different patterns of variation in different maize varieties. The Pearson correlation analysis between nutritional indices, enzyme activities, and the nutritional content and secondary metabolites of maize leaves provided deeper insights into the pest's adaptability. The results highlighted significant relationships between specific nutritional components in maize and the physiological responses of S. frugiperda. Overall, our findings contribute substantially to the understanding of S. frugiperda's host plant adaptability, offering critical insights for the development of sustainable pest management strategies.

Plant communities in the Loess Plateau's artificial afforestation forests play an important role in fragile ecosystem restoration. Therefore, the composition, coverage, biomass, diversity, and similarity of grassland plant communities in different years of artificial afforestation in cultivated land were investigated. The effects of years of artificial afforestation on grassland plant community succession in the Loess Plateau were also investigated. The results showed that as the number of years of artificial afforestation increased, grassland plant communities grew from scratch, constantly optimizing community components, improving community coverage, and increasing aboveground biomass. The community diversity index and similarity coefficient gradually approached those of a 10-year abandoned community that had recovered naturally. After 6 years of artificial afforestation, the dominant species of the grassland plant community changed from Agropyron cristatum to Kobresia myosuroides, and the main associated species changed from Compositae and Gramineae to Compositae, Gramineae, Rosaceae, and Leguminosae. The α-diversity index accelerated restoration, the richness index and diversity index increased, and the dominant index decreased. The evenness index had no significant difference from CK. The ß-diversity index decreased as the number of years of afforestation increased. The similarity coefficient between CK and grassland plant communities in various lands changed from medium dissimilarity to medium similarity at 6 years of afforestation. According to the analysis of various indicators of the grassland plant community, the grassland plant community had a positive succession within 10 years of artificial afforestation on the cultivated land of the Loess Plateau, and the threshold of the years from slow to fast was 6 years.

Botrytis cinerea causing tomato gray mold is a major cause of economic loss in tomato production. It is urgent and necessary to seek an effective and environmentally friendly control strategy to control tomato grey mold disease. In this study, Bacillus velezensis FX-6 isolated from the rhizosphere of plants displayed significant inhibitory ability against B. cinerea and could promote tomato plant growth. FX-6 could effectively inhibit the growth of Botrytis cinerea mycelium in vitro and in vivo, and the inhibitory rate in vitro could reach 78.63%. According to morphological observations and phylogenetic trees based on sequences of the 16S rDNA and gyrA (DNA gyrase subunit A) genes, the strain FX-6 was identified as Bacillus velezensis. In addition, B. velezensis FX-6 showed antagonistic activity against seven phytopathogens, this indicated that FX-6 had broad-spectrum biocontrol activity. We also found that FX-6 fermentation broth had the strongest antagonistic activity against B. cinerea when the culture time was 72 hours, and the inhibition rate was 76.27%. The growth promotion test revealed that strain FX-6 significantly promoted tomato seed germination and seedling growth. Further deeply study on growth-promoting mechanism indicated that the FX-6 produced IAA and siderophore, and had ACC deaminase activity. The trait of significant biological control activity and growth promoting effect on tomato imply that B. velezensis FX-6 has the potential to be used as a biocontrol agent for tomato gray mold management.

Tetranychus truncatus Ehara (Acari: Tetranychidae) has become one of the major phytophagous pests in China in recent years, and is found on a wide range of host plants. However, little information is available on the population performance of this arthropod pest on potatoes. In this study, we explored the population growth of T. truncatus on two drought-tolerant potato (Solanum tuberosum L.) cultivars under laboratory conditions using the age-stage, two-sex life table. Tetranychus truncatus completed its entire life history on both potato cultivars tested, Holland 15 and Longshu 10. There was no significant difference between two potato cultivars in developmental duration. Tetranychus truncatus had shorter adult longevity (20.61 days), adult female longevity (20.41 days), and total female longevity (33.66 days) on Longshu 10 than Holland 15 (21.16 days, 21.19 days, and 34.38 days, respectively). However, it exhibited a higher preadult survival rate, higher fecundity (F = 88.32 eggs per female), and relatively higher population parameters when reared on Longshu 10 than on Holland 15 (F = 75.70 eggs per female). Growth projection also showed that the population size of T. truncatus on Longshu 10 (expand 750-fold) was larger than that on Holland 15 (expand 273-fold) after 60 days. Our results demonstrate that the drought-sensitive potato variety, Holland 15, is relatively resistant to T. truncatus compared with the drought-tolerant variety, Longshu 10, and suggest that T. truncatus exhibited a trade-off between longevity and reproduction on both potato cultivars. Our findings provide information on population prediction, which may aid the management of this pest mite species of potatoes.

Host plants play an important role in the growth, development, and reproduction of insects. However, only a few studies have reported the effects of maize varieties on the growth and reproduction of S. frugiperda. In this study, a free-choice test was used to evaluate the oviposition preferences of female adults on ten common maize varieties and ten special maize varieties. The population fitness of S. frugiperda on six different maize varieties was also examined using the age-stage, two-sex life table method. The results showed that S. frugiperda oviposited and completed its life cycle across all maize cultivars. Moreover, the S. frugiperda females exhibited a significantly higher oviposition preference on the special maize varieties than on the common maize varieties. The highest number of eggs and egg masses occurred on Baitiannuo and the lowest on Zhengdan 958. The egg + larval stage, preadult, pupal stage, adult, APOP, TPOP, and total longevity of S. frugiperda were significantly shorter on the special maize varieties than on the common maize varieties. The fecundity, oviposition days, pupal weight, and hatching rate of S. frugiperda were significantly higher on the special maize varieties than on the common maize varieties. Specifically, S. frugiperda had the highest fecundity, female, and male pupal weight on Baitiannuo. Moreover, the net reproductive rate (R0), intrinsic rate of increase (r), and finite rate of increase (λ) of S. frugiperda were the greatest on Baitiannuo, whereas the shortest mean generation time (T) occurred on Zaocuiwang. The lowest R0, r, and λ, and longest T occurred on Zhengdan 958, suggesting that Zhengdan 958 is a non-preferred host plant compared to the other tested maize varieties. The findings of this study can provide a reference for the rational planting of maize and provide basic scientific information for the management of S. frugiperda.

This study was conducted to determine the aphicidal effect of a leaf extract of the Atlantic potato cultivar on the performance of green peach aphids. Three concentrations of the leaf extract (100, 75, and 50% potato extract), synthetic pesticide (Beta cypermethrin 4.5%), and distilled water (control) treatments were applied in a greenhouse experiment. The results showed that the synthetic pesticide, which was used as a standard check, caused the maximum aphid mortality, followed by the 100% potato leaf extract. Compared with the other botanical treatments, the 100% extract produced low mean rates of survival, aphids' average daily reproduction, the number of nymphs per plant, and the number of nymphs per adult. This treatment also increased the accumulation of hydrogen Peroxide (H2O2) and malondialdehyde (MDA), glutathione-s-transferase, mixed-function oxidase, and carboxylesterase content in the green peach aphid. Moreover, the 100% extract also protected the host plants against green peach aphid attacks by demonstrating higher chlorophyll content, net photosynthesis, above-ground fresh weight, and above-ground dry weight of the host plant. This study demonstrates that the highest concentration of potato (Atlantic cultivar) leaf extract (100% extract) could be used as the appropriate dosage for the control of green peach aphids on potatoes, which could greatly reduce the use of synthetic insecticides and promote ecosystem sustainability.

Botanical pesticide is highly recommended for integrated pest management (IPM), due to its merits such as environmental friendliness, safe to non-target organisms, operators, animals, and food consumers. The experiment was conducted to determine the lethal and sub-lethal effects of allyl isothiocyanate (AITC) on eggs, third instar larvae, pupae, and females and males of Bradysia impatiens Johannsen (B. impatiens). Different concentrations of AITC under ambient CO2 by the conical flask sealed fumigation method were used for the experiment. The results showed that there was a significant linear relationship between different concentrations of AITC and the toxicity regression equation of B. impatiens. The sub-lethal concentrations of AITC had significant effects on the larval stage, pupal stage, pupation rate, pupal weight, adult emergence rate, and oviposition. The pupation rate, pupal weight, and adult emergency rate were significantly (p < 0.05) affected by AITC fumigation. The pupation rate was the lowest after fumigation treatment of AITC at LC50 (36.67%), followed by LC25 (41.94%), compared with the CK (81.39%). Female longevity was significantly (p < 0.05) shortened by fumigation at LC25 (1.75 d) and LC50 (1.64 d), compared with that of CK (2.94 d). Male longevity was shorter at LC25 (1.56 d) than at LC50 (1.25 d) and had no significant difference between these two treatments. The fumigation efficiency of AITC was significantly increased under high CO2 condition. Furthermore, detoxification enzyme activities and antioxidant enzyme activities were accumulated under high CO2 condition. The fumigation method in the application of AITC can be useful in areas where B. impatiens is a major concern.

Insect chitinases play a crucial part to digest chitin in the exoskeleton during the molting process. However, research on insect chitinase related to the environmental stress response is very limited. This study was the first conducted to expression analysis of chitinase- related genes in A. pisum under abiotic stresses. Here, we identified five chitinase-like proteins (ApIDGF, ApCht3, ApCht7, ApCht10 and ApENGase), and clustered them into five groups (group II, III, V, Ⅹ, and ENGase). Developmental expression analysis revealed that the five A. pisum chitinase-related genes were expressed at whole developmental stages with different relative expression patterns. When aphids were exposed to various abiotic stresses including temperature, insecticide and the stress 20-hydroxyecdysone (20E), all five chitinase genes were differentially expressed in A. pisum. The results showed that insecticide such as imidacloprid down-regulated the expression of these five Cht-related genes. Analysis of temperature stress of A. pisum chitinase suggested that ApCht7 expression was high at 10°C, which demonstrates its important role in pea aphids under low temperature. Conversely, ApCht10 was more active under high temperature stress, as it was significantly up-regulated at 30°C. Besides, 20E enhanced ApCht3 and ApCht10 expression in A. pisum, but reduced ApCht7 expression. These findings provide basic information and insights for the study of the role of these genes under abiotic stress, which advances our knowledge in the management of pea aphids under multiple stresses.

Resistant variety screening is widely recommended for the management of Sitobion avenae. The purpose of this study was to assess responses of six wheat varieties (lines) to S. avenae. The aphid quantity ratio (AQR) was used to assess S. avenae resistance. Pearson's correlation coefficient was used to perform a correlation analysis between AQR, biological parameters, and the accumulation of total phenolic and flavonoid content. When compared to the other cultivars, the results showed that two cultivars, Yongliang No.15 and Ganchun No.18, had high resistance against S. avenae. The correlation analysis revealed a positive relationship between total phenol and flavonoid content accumulation and developmental duration (DD), and a negative relationship between accumulation and weight gain (WG) and mean relative growth rate (MRGR). The correlation between flavonoid and biological parameters was statistically stronger than the correlation between total phenol and biological parameters. This research provides critical cues for screening and improving aphid-resistant wheat varieties in the field and will aid in our understanding of the resistance mechanism of wheat varieties against S. avenae.

This study was conducted to determine the root system architecture and biochemical responses of three potato (Solanum tuberosum L.) cultivars to drought and aphid (Myzus persicae Sulzer) infestation under greenhouse conditions. A factorial experiment comprising three potato cultivars (Qingshu 9, Longshu 3, and Atlantic), two levels of water (Well watered and drought) application and aphid infestation (Aphids and no aphids) was conducted. The results show that drought stress and aphid infestation significantly increased the root-projected area, root surface area, number of root tips, and number of root forks of all cultivars, relative to their corresponding control plants. The least root projected area, root surface area, number of root tips, and number of root forks occurred on DXY under both drought and aphid infestation. Nevertheless, the greatest root projected area, root surface area, number of root tips and number of root forks occurred on QS9 plants. Moreover, increased SOD, CAT, and POD activities were observed across all cultivars, under drought and aphid stress. The highest SOD, POD, and CAT activities occurred in QS9; under drought and aphid stress, while the least SOD, POD, and CAT activities was observed in DXY. The Atlantic cultivar, which possesses a root system sensitive to water deficit, demonstrated greater resistance to aphid infestation under well-watered and drought-stressed conditions. Conversely, Qingshu 9, which possesses a root system tolerant to water deficit, was highly susceptible to aphids. This study shows that the root architectural and biochemical traits that enhance potato tolerance to drought do not necessarily correlate to a plant's tolerance to aphids.

Trehalose serves multifarious roles in growth and development of insects. In this study, we demonstrated that the high trehalose diet increased the glucose content, and high glucose diet increased the glucose content but decreased the trehalose content of Acyrthosiphon pisum. RNA interference (RNAi) of trehalose-6-phosphate synthase gene (ApTPS) decreased while RNAi of trehalase gene (ApTRE) increased the trehalose and glucose contents. In the electrical penetration graph experiment, RNAi of ApTPS increased the percentage of E2 waveform and decreased the percentage of F and G waveforms. The high trehalose and glucose diets increased the percentage of E2 waveform of A. pisum red biotype. The correlation between feeding behavior and sugar contents indicated that the percentage of E1 and E2 waveforms were increased but np, C, F and G waveforms were decreased in low trehalose and glucose contents. The percentage of np, E1 and E2 waveforms were reduced but C, F and G waveforms were elevated in high trehalose and glucose contents. The results suggest that the A. pisum with high trehalose and glucose contents spent less feeding time during non-probing phase and phloem feeding phase, but had an increased feeding time during probing phase, stylet work phase and xylem feeding phase.

Potato production is adversely affected by aphid infestation across the globe. Understanding the mechanism of host plant defense against aphids under drought stress is paramount for insect pest management. This study was conducted to examine the cross-talk of phytohormones in potato glycoalkaloids' defense against green peach aphids under greenhouse conditions. A 3 × 2 × 2 factorial experiment comprising three potato cultivars (Qingshu 9, Longshu 3, and Atlantic) and two levels each of water availability and aphid infestation was conducted. The results show that under drought stress, green peach aphids thrive well on host plants, which contain a relatively high water content. The resistant cultivar DXY, which exhibited a higher level of phytohormones, also demonstrated higher α-chaconine and α-solanine contents in both leaf and root, under drought and aphid stress. Conversely, the susceptible cultivar QS9, which exhibited a lower level of phytohormones, also demonstrated low α-chaconine and α-solanine contents in both leaf and root, under drought and aphid stress. The DXY cultivar, which possessed high resistant traits such as α-chaconine and α-solanine, can be used in areas where green peach aphid infestation is a major setback.

The chive maggot Bradysia cellarum and the fungus gnat B. impatiens are two primary root pests of plants, which can coexist on the same host plants and are the devastating pests on liliaceous crops and edible fungi. Their growth and development are affected by the nutrient contents of their host plants. In this study, we assessed the effects of different host plant nutrients on the nutrient contents of these two Bradysia species. The nutrients of the chive (Allium tuberosum Rottl. ex Spreng.), board bean (Vicia faba L.), lettuce (Lactuca sativa L. var. ramosa Hort.), cabbage (Brassica oleracea L.), wild cabbage (Brassica oleracea var. capitata rubra) and pepper (Capsicum annuum L.) roots were determined, and their effect on nutrient content of the two Bradysia species after feeding on the host plant for three continuous generations were evaluated. The results show that chive and B-bean contained higher levels of protein, free amino acid, soluble sugar and starch than others. As a result, the soluble sugar, fat and protein contents were significantly higher in both Bradysia species reared on chive and B-bean than on cabbage, lettuce, W-cabbage and pepper, suggesting nutritional preference of these insects. Based on our results, we concluded that the two Bradysia species displayed nutrient preference toward chive and B-bean, which provides a reference for understanding their host plant range and for control of the insect species via field crop rotations.

The fungus gnat Bradysia difformis Frey not only attacks edible and medicinal fungi, it also causes the damages to leek, green onion, garlic and other vegetable crops. To confirm the effects of temperature and sex ratio on adult fecundity, longevity and egg hatchability of B. difformis, we assayed the adults survival longevity and numbers of oviposition, as well as egg hatching rate under female-male ratios of 1:1, 1:2, 1:3, 2:1 and 3:1 at 10, 15, 20, 25 and 30°C. Female fecundity and egg hatchability were increased with temperature and peaked at 25°C, which, however, were adverse to adult longevity. Furthermore, female-male ratio of 1:1, 1:2 and 1:3 seemed suitable for female fecundity, of which the sex ratio of 1:1 was the most suitable ratio for its oviposition behavior. These results provide an insight for predicting the population density of B. difformis and offer a literature reference in the effective treatment of harmful insects by controlling and changing the sex ratio artificially.

Drought stress on plants can cause cellular water deficits and influence the physiology of host plants, which alter the performance of insect pests. This study was conducted to determine the effect of drought and aphid (Myzus persicae Sulzer) infestation on three potato (Solanum tuberosum L) genotypes under greenhouse conditions. A factorial experiment involving three potato genotypes, two levels of drought, and two levels of aphid infestation was conducted. The potato genotypes possessed different levels of tolerance to drought and are described as tolerant (Qingshu 9), moderately tolerant (Longshu 3), and sensitive (Atlantic). Sixty-day-old potato plants were infested with aphid nymphs and monitored for 20 d. There was a significant variety × drought × aphid interaction effect on the parameters measured. The genotype Atlantic, which is sensitive to drought, exhibited greater tolerance to aphid infestation under drought or no drought conditions than the other genotypes. This genotype also exhibited poor host acceptance and the aphid survival rate, colonization success, and average daily reproduction were low. Qingshu 9, which is tolerant to drought, was highly susceptible to aphid infestation and exhibited high host acceptance and greater aphid survival rate, colonization success, and average daily reproduction compared to the other genotypes. This study demonstrates that the biochemical and morphological traits that confer drought tolerance in potato do not necessarily confer aphid tolerance.
