RESUMO

Chironomidae are aquatic insects that have become a model in laboratory tests to evaluate toxic effect of different pollutants. The use of chironomids as test organisms depends on standard protocols for the rearing under laboratory conditions and expanding knowledge on its biology. We standardize a culture of C. columbiensis and analyze its life cycle under laboratory conditions. The experiments were carried out with different treatments for water, temperature, and food. As a result, a protocol was proposed for the rearing of C. columbiensis under the following conditions: semi-soft and reconstituted water, shredded paper towels as a substrate, soft aeration, temperature of 22 ± 2°C, fed with 0.3 g TetraMin® twice a week and weekly renewal of 50% of the water. Under these conditions, the duration of the life cycle was 17-31 days, with a continuous growth of 1.16 to 14.05 mm in the larval stage and an average of 530 eggs per oviposition. In comparison with other species in the family, this study indicated that C. columbiensis is a species with a short life cycle, sensible to changing conditions (e.g., temperature and food), wide distribution, so is a good organism for being used in aquatic bioassays.

RESUMO

The aim of this study was to identify organochlorine (OC) and organophosphorus (OP) pesticides levels in water samples collected in secondary water bodies in agricultural area planted with coffee and plantain. A descriptive cross-sectional study was carried out. A validated method for microwave-assisted extraction and gas chromatography with electron microcapture detector (MAE-GC-µECD) was used to analyze pesticide residues in samples. The determinations were based on certified reference material, Organochlorine Pesticide Mix AB #3, Canadian Drinking Organophosphorus Pesticides Mix, and pentachloronitrobenzene (ISTD) Internal Standard Mix 508.1. Pesticide residues were found in 81.3% of the samples, including OCs: 4.4'-DDT (38%), endosulfan II (19.7%), endosulfan sulfate, and endrin (11.7% and 8.8%), and others identified as 4.4'-DDE, Delta-HCB, parathion, chlorpyrifos, endrin aldehyde, heptachlor, heptachlor epoxide, endrin ketone, and methoxychlor. Parathion and/or chlorpyrifos were found in 5.8-8% of samples; the water bodies most heavily affected were those in Filandia and Quimbaya in which 100% of samples were contaminated, followed by those in Calarcá, Córdoba, Pijao, and Génova, with contamination found in over 75% of samples. The results indicated that surface waters from Quindío municipalities are contaminated with pesticide residues hazardous to human health, which are still in use despite being either restricted or prohibited.

RESUMO

The use of imidacloprid is a common pest control practice in the Neotropical region. However, the imidacloprid unintended sublethal effects on Neotropical aquatic non-target arthropods and undesirable consequences for aquatic environments remain unclear. Here, we assessed the susceptibility of Chironomus columbiensis (Diptera: Chironomidae) larvae to the neonicotinoid imidacloprid and evaluated whether sublethal exposure types would trigger sex-dependent adaptive responses (e.g., emergence, body mass, reproduction, wing morphology). We conducted a concentration-mortality curve (96 h of exposure) and established chronic and acute sublethal exposure bioassays. While chronic sublethal exposures consisted of exposing individuals during their entire larval and pupal stages, the acute sublethal exposures represented a single short duration (24 h) exposure episode during either the first or fourth larval instar. Our results revealed that chronic sublethal exposure reduced the body mass of males, while acute sublethal exposures during the first instar resulted in heavier males than those that were not exposed to imidacloprid. Chronic exposure also reduced the reproduction of males and females, while the acute sublethal exposure only affected the reproduction of individuals that were imidacloprid-exposed on their later larval instar. Chronic and acute sublethal exposures did differentially affect the wing properties of C. columbiensis males (e.g., increased size when chronically exposed and highly asymmetric wings when acutely exposed in early larval phase) and females (e.g., highly asymmetric wings when chronically and acutely exposed). Collectively, our findings demonstrated that imidacloprid can cause unintended sublethal effects on C. columbiensis, and those effects are dependent on sex, exposure type, and developmental stage.

RESUMO

Sublethal exposure to insecticides can trigger unintended responses in non-target insects that may disrupt reproductive and developmental performances of these organisms. Here, we assessed whether sublethal exposure to the pyrethroid insecticide deltamethrin in early life had sublethal and transgenerational effects on the reproduction (i.e., fecundity and fertility) and wing morphology of Chironomus columbiensis, an aquatic insect used as a water quality indicator. We first conducted concentration-response bioassays to evaluate the susceptibility of C. columbiensis larvae to deltamethrin. Our results revealed that deltamethrin toxicity was approximately 7-fold higher when C. columbiensis larvae where exposed to 96 h (LC50 = 0.17 [0.15-0.20] µg/L) than to 24 h (LC50 = 1.17 [0.97-1.43] µg/L). Furthermore, the sublethal exposures (at LC1 = 0.02 µg/L or LC10 = 0.05 µg/L) of immature C. columbiensis resulted in lower fecundity (e.g., reduced eggs production) and morphometric variation wing shapes. Further reduction in fertility rates (quantity of viable eggs) occurred at deltamethrin LC10 (0.05 µg/L). Almost 80% of the fecundity was recovered with only a single recovery generation; however, two subsequent recovery generations were not sufficient to fully recover fecundity in C. columbiensis. Specimens recovered from 98.5% of wing morphometric variation after two consecutive generations without deltamethrin exposure. Collectively, our findings demonstrates that sublethal exposure to synthetic pyrethroids such as deltamethrin detrimentally affect the reproduction and wing shape of C. columbiensis, but also indicate that proper management of these compounds (e.g., concentration and frequency of application) would suffice for these insects' population recovery.

RESUMO

Constantly, aquatic ecosystems are under pressure by complex mixtures of contaminants whose effects are not always easy to evaluate. Due to this, organisms are sought in which early warning signs may be detected upon the presence of potentially toxic xenobiotic substances. Thereby, the study evaluated the incidence of deformities and other morphometric variations in the mentum and wing of Chironomus columbiensis exposed to water from some of the Colombian Andes affected by mining, agriculture, and cattle raising. Populations of C. columbiensis were subjected throughout their life cycle (24 days) for two generations (F1 and F2). Five treatments were carried out in controlled laboratory conditions (water from the site without impact, site of mining mercury, mining mercury + cyanide, cattle raising, and agriculture) and the respective control (reconstituted water). Thereafter, the percentage of deformities in the mentum was calculated, and for the morphometric analysis 29 landmarks were digitized for the mentum and 12 for the wing. As a result, four types of deformities were registered in the C. columbiensis mentum, like absence of teeth, increased number of teeth, fusion and space between teeth, none of them detected in the individuals from the control. Additionally, the highest incidence of deformity in F1 occurred in the treatment of mining mercury, while for F2 this took place in the treatments of mining mercury + cyanide, cattle raising and agriculture. Differences were also found with respect to the morphometric variations of the mentum and wing of C. columbiensis among the control and the treatments with water from the creeks intervened. The treatments of mining mercury + cyanide and agriculture had the highest morphological variation in the mentum and wing of C. columbiensis. The results suggest that the anthropogenic impacts evaluated generate alterations in the oral apparatus of the larval state of C. columbiensis and in the adult state provoke alterations in the wing shape (increased width and reduced basal area). These deformities may be related to multiple stress factors, among them the xenobiotics metabolized by the organisms under conditions of environmental contamination.

RESUMO

Cancer cells modify lipid metabolism to proliferate, Passiflora edulis ( P. edulis ) fruit juice (ZuFru) has antitumor activity, but whether a mechanism is through modulation of cell lipids is unknown. T o establish if ZuFru modifies cholesterol and triglycerides in SW480 and SW620. ZuFru composition was studied by phytochemical march; antiproliferative activity by sulforhodamine B, cholesterol , and triglycerides by Folch method. Z ufru contains anthocyanins, flavonoids, alkaloids , and tannins. Cell lines showed differences in their growth rate ( p =0.049). At 39.6 µg/m L of ZuFru, cell viability was decreased: SW480 (45.6%) and SW620 (45.1%). In SW480, cholesterol (44.6%) and triglycerides (46.5%) decreased; In SW620, cholesterol decreased 14.8% and triglycerides increased 7%, with significant differences for both lines. A ntiproliferative activity of ZuFru could be associated with the inhibition of intracellular biosynthesis of cholesterol and triglycerides in SW480. Action mechanisms need to be further investigated.

Mango fruit has a high nutritional value and health benefits due to important components. The present manuscript is a comprehensive update on the composition of mango fruit, including nutritional and phytochemical compounds, and the changes of these during development and postharvest. Mango components can be grouped into macronutrients (carbohydrates, proteins, amino acids, lipids, fatty, and organic acids), micronutrients (vitamins and minerals), and phytochemicals (phenolic, polyphenol, pigments, and volatile constituents). Mango fruit also contains structural carbohydrates such as pectins and cellulose. The major amino acids include lysine, leucine, cysteine, valine, arginine, phenylalanine, and methionine. The lipid composition increases during ripening, particularly the omega-3 and omega-6 fatty acids. The most important pigments of mango fruit include chlorophylls (a and b) and carotenoids. The most important organic acids include malic and citric acids, and they confer the fruit acidity. The volatile constituents are a heterogeneous group with different chemical functions that contribute to the aromatic profile of the fruit. During development and maturity stages occur important biochemical, physiological, and structural changes affecting mainly the nutritional and phytochemical composition, producing softening, and modifying aroma, flavor, and antioxidant capacity. In addition, postharvest handling practices influence total content of carotenoids, phenolic compounds, vitamin C, antioxidant capacity, and organoleptic properties.

RESUMO

OBJECTIVES: To establish the correlation between hypothyroidism and blood pesticide levels. MATERIALS AND METHODS: Cross-sectional study in agricultural workers and their permanent partners in plantain and coffee producing municipalities as reference population. A representative sample was estimated and thyroid function tests were performed using ELISA Stat Fax 303/Plus reader, at a wavelength of 450 nm. Organochlorine pesticide residuality was determined, a dispersive liquid-liquid microextraction (DLLME) assisted by sonication was implemented, and a gas chromatography-micro-electron capture detector (GC-pECD) was used for the analysis. RESULTS: 819 participants, 58.7% men and 41.3% women were included; their average age was 48.1 years. Prevalence of symptomatic hypothyroidism (1.2%) and subclinical hypothyroidism (6.7%) was observed, with a higher prevalence in people older than 60 years (2.6% and 8.9%, respectively). Non-causal association was found between subclinical hypothyroidism and the organochlorine pesticides 4,4'-DDE (sig.0,006), Heptachlor (sig.0,04), and Endosulfan I (sig.0,02). Antiperoxidase (Anti TPO) antibodies ≥60 lU/ml were associated with subclinical hypothyroidism (OR 2.6). CONCLUSIONS: The prevalence of hypothyroidism in the studied population is similar to that reported in the literature, and lower than in urban areas. In turn, the prevalence of subclinical hypothyroidism is higher and positive anti-TPO values are related to risk of progression to frank hypothyroidism, which is why follow-up is required in these patients. Three organochlorine pesticides were associated with subclinical hypothyroidism. TSH screening is recommended in people aged 40 and over, especially if they are exposed to the aforementioned agrochemicals.

RESUMO

BACKGROUND: Policosanol, commonly derived from purified sugar cane wax, has been reported to exert lipid-lowering effects. Policosanol is available in the United States as a nutritional supplement despite no US research clinical experience. This trial was designed to rigorously establish the lipid-lowering efficacy of policosanol as monotherapy and its potential additive and possibly synergistic effects when added to statin therapy. METHODS: A randomized, parallel, double-blind, double-dummy, placebo-controlled design was used. Patients with low-density lipoprotein cholesterol (LDL-C) levels from 140 to 189 mg/dL were assigned into 1 of 4 groups to receive policosanol 20 mg, atorvastatin 10 mg, combination therapy, or placebo for 12 weeks. RESULTS: A total of 99 patients were examined. Baseline characteristics were similar among all treatment groups. Policosanol (20 mg/d for 12 weeks) did not significantly change plasma total cholesterol, LDL-C, high-density lipoprotein cholesterol, or triglyceride levels when compared with baseline values or with values of placebo-treated patients. Atorvastatin (10 mg/d for 12 weeks) reduced total cholesterol by 27% and LDL-C by 35%. Addition of policosanol to atorvastatin failed to produce any further reduction in lipid levels above that of atorvastatin alone. Policosanol was safe and did not affect liver enzyme or creatinine phosphokinase levels. CONCLUSIONS: Policosanol did not reduce LDL-C or total cholesterol levels either alone or in combination with atorvastatin. This observation supports the need for systematic evaluation of available products containing policosanol to determine their clinical lipid-lowering efficacy under rigorous experimental conditions. We propose that policosanol should be added to the list of nutritional supplements lacking scientific validity to support their use.

RESUMO

INTRODUCTION: Nutritional support generates complications that must be detected and treated on time. OBJECTIVE: To estimate the incidence of some complications of nutritional support in patients admitted to general hospital wards who received nutritional support in six high-complexity institutions. METHODS: Prospective, descriptive and multicentric study in patients with nutritional support; the variables studied were medical diagnosis, nutritional condition, nutritional support duration, approach, kind of formula, and eight complications. RESULTS: A total of 277 patients were evaluated; 83% received enteral nutrition and 17% received parenteral nutrition. Some 69.3% presented risk of malnourishment or severe malnourishment at admittance. About 35.4% of those receiving enteral nutrition and 39.6% of the ones who received parenteral nutrition had complications; no significant difference per support was found (p= 0.363). For the enteral nutrition, the most significant complication was the removal of the catheter (14%), followed by diarrhea (8.3%); an association between the duration of the enteral support with diarrhea, constipation and removal of the catheter was found (p < 0.05). For parenteral nutrition, hyperglycemia was the complication of highest incidence (22.9%), followed by hypophosphatemia (12.5%); all complications were associated with the duration of the support (p < 0.05). Nutritional support was suspended in 24.2% of the patients. CONCLUSIONS: Complications with nutritional support in hospital-ward patients were frequent, with the removal of the catheter and hyperglycemia showing the highest incidence. Duration of the support was the variable that revealed an association with complications. Strict application of protocols could decrease the risk for complications and boost nutritional support benefits.

We attempted to characterize the 5-hydroxytryptamine (5-HT) receptor type/subtype and mediator mechanisms involved in the contractile effects of 5-HT in the in situ autoperfused kidney of long-term diabetic rats. Diabetes was induced in male Wistar rats by a single subcutaneous injection of alloxan. Intra-arterial (i.a.) bolus injection of 5-HT (0.00000125 to 0.1µg/kg) increased renal perfusion pressure in a dose dependent way but did not affect the systemic blood pressure in long-term diabetic rats. The selective 5-HT2 receptor agonist, α-methyl-5-HT, caused a local vasoconstrictor effect in the in situ autoperfused rat kidney similar to 5-HT. However, BW723C86, a selective 5-HT2B receptor agonist, m-CPP (1-(3-chlorophenyl)piperazine), a selective 5-HT2B/2C receptor agonist, the 5-HT1 receptor agonist, 5-carboxamidotryptamine (5-CT), and the selective 5-HT3 receptor agonist, 1-phenylbiguanide did not modify the renal perfusion pressure. In long-term diabetic rats, vasoconstriction elicited by 5-HT and α-methyl-5-HT was significantly decreased by ritanserin (a 5-HT2 receptor antagonist), spiperone (a 5-HT2A receptor antagonist), and the cyclooxygenase (COX) inhibitors, indomethacin (non-selective COX inhibitor), FR 122047 or nimesulide (selective COX-1 and COX-2 inhibitors, respectively), but was not modified by pretreatment with SB 206553 (3,5-dihydro-5-methyl-N-3-pyridinylbenzo[1,2.b:4,5-b']dipyrrole(1H)-carboxamide hydrochloride), a non-selective 5-HT2C receptor antagonist, prazosin, propranolol, enalapril or losartan. The results of protein expression support these results: COX-1 and COX-2 are expressed in renal tissue. Inducible COX (COX-2) is overexpressed in long-term diabetes. Our data suggest that, in the in-situ autoperfused kidney of long-term diabetic rats, 5-HT vasoconstrictor action is mediated, through cyclooxygenase pathway, by local activation of 5-HT2A receptors.

This paper is the product of the advice given to a research on this topic. It reviews the notion of phases of psychosexual development by taking into account Sigmund Freud's correction to the theory on sexuality in his paper The Infantile Genital Organization. Likewise, it is guided by Jacques Lacan's theorization on this topic, who introduces it into the logic of signifier and takes it away from the geneticist stances in order to give it a place in the subject's structure.

We analyzed the modulation of serotonin on the bradycardia induced in vivo by vagal electrical stimulation in alloxan-induced long-term diabetic rats. Bolus intravenous administration of serotonin had a dual effect on the bradycardia induced either by vagal stimulation or exogenous Ach, increasing it at low doses and decreasing it at high doses of 5-hydroxytryptamine (5-HT), effect reproduced by 5-carboxamidotryptamine maleate (5-CT), a 5-HT(1/7) agonist. The enhancement of the bradycardia at low doses of 5-CT was reproduced by 5-HT(1A) agonist 8-hydroxy-2-dipropylaminotetralin hydrobromide (8-OH-DPAT) and abolished by WAY-100,635, 5-HT(1A) antagonist. Pretreatment with 5-HT(1) antagonist methiothepin blocked the stimulatory and inhibitory effect of 5-CT, whereas pimozide, 5-HT(7) antagonist, only abolished 5-CT inhibitory action. In conclusion, long-term diabetes elicits changes in the subtype of the 5-HT receptor involved in modulation of vagally induced bradycardia. Activation of the 5-HT(1A) receptors induces enhancement, whereas attenuation is due to 5-HT(7) receptor activation. This 5-HT dual effect occurs at pre- and postjunctional levels.

RESUMO

We analysed the type and/or subtype of 5-hydroxytryptamine (5-HT) receptors involved in the inhibitory mechanisms of 5-HT on the pressor responses induced by stimulation of sympathetic vasopressor outflow in long-term diabetic pithed rats. Diabetes was induced in male Wistar rats by a single subcutaneous injection of alloxan. Eight weeks later, rats were anaesthetized, pre-treated with atropine, and pithed. The effect of 5-HT on the pressor responses elicited by stimulation of the sympathetic outflow was analysed in eight-week alloxan-induced diabetic pithed rats. 5-HT (20 microg/kg/min) reduced the pressor action obtained by electrical stimulation of the sympathetic outflow. However, there was no effect on exogenous noradrenaline-induced pressor responses. 5-CT (5 microg/kg/min), 8-OH-DPAT (5 microg/kg/min), and alpha-methyl-5-HT (5 microg/kg/min), selective 5-HT(1), 5-HT(1A) and 5-HT(2) receptor agonists, respectively, reproduced the 5-HT inhibitory action. Nevertheless, infusion of 5 microg/kg/min of 1-phenylbiguanide, CGS-12066B, L-694,247, BW273C86 or MK212 (5-HT(3), 5-HT(1B), 5-HT(1D), 5-HT(2B) and 5-HT(2C) receptor agonists, respectively) had no effect on the pressor responses elicited by stimulation of the sympathetic outflow. Methiothepin (100 microg/kg) and a cocktail of WAY-100,635 (100 microg/kg) and spiperone (125 microg/kg) blocked the 5-HT inhibitory effect on the pressor action obtained by sympathetic stimulation. Moreover, WAY-100, 635 abolished the 8-OH-DPAT inhibitory effect and spiperone blocked alpha-methyl-5-HT action. In conclusion, this study revealed that long-term experimental diabetes induces changes in the receptor type/subtype involved in the 5-HT inhibitory action on the sympathetic pressor responses produced by electrical stimulation. This is mainly mediated by pre-junctional 5-HT(1A) and 5-HT(2A) receptors.

RESUMO

INTRODUCTION: Apolipoprotein E (apoE) polymorphism is a genetic determinant of total cholesterol and LDL-c levels. Several studies have examined the relationship between variations at the APO E and cardiovascular disease. It is important to determine this relationship in the Colombian population. OBJECTIVES: The relationship of apoE polymorphisms was associated with lipid profile by age and sex. MATERIALS AND METHODS: A sample of 500 children aged 8 to 18 years, were selected from school populations in Quindio Province, Colombia. ApoE genotypes were determined by polymerase chain reaction and restriction enzyme. Lipid profiles were obtained by commercial kits. RESULTS: The apoE allele frequencies were as follows: E3 - 91.6%, E2 - 5.3, and E4 - 3.1%. Genotypic frequencies were as follows: E3/E3 - 90.8%, E2/E3 - 5.6%, and E3/E4 - 3.2%. No homozygotes for E2/E2 and E4/E4 were recovered. Similar genotypic and allelic distribution was found for each gender. ApoE genotype and gender had a significant effect on total serum cholesterol (CT) and low density lipoprotein cholesterol (c-LDL). In boys these variables were related as follows: E4>E3>E2 (p<0.05); in girls the relationships were somewhat changed (E3>E4>E2). High density lipoprotein cholesterol (c-HDL) levels in boys were related as follows: E2>E4>E3; for girls, E4>E2>E3. In boys, for all variables, the protector effect was in E2; but in girls this allele was only a protector for CT and c-LDL, For triacylglycerol (TAG), very low density lipoprotein cholesterol (c-VLDL) and c-HDL, this the protector effect was associated with the E4 allele, (p<0.05). CONCLUSION: The results suggested that modulation of apoE genotype on plasma lipids was influenced by gender and age, especially in girls older than 10 years.

RESUMO

OBJECTIVE: To determine the occurrence of association between homocysteine, folate, or vitamin B12 plasma levels and acute coronary syndrome in Colombian patients. METHODS: Case control study: cases were 50 patients with acute coronary syndrome and controls were 50 outpatients without coronary syndrome. Homocysteine, folate and vitamin B12 levels were determined by means of chemiluminescence immunoassay. Cholesterol and lipoproteins, triglycerides, BUN, creatinine, hemoglobin and hematocrit were also measured. RESULTS: Mean homocysteine plasma concentrations were significantly different between cases (12.4 micromol/l +/- 6.0) and controls (9.7 micromol/l +/- 2.4), p=0.01. The folic acid levels of the cases were lower than those of the control patients (10.5 ng/ml +/- 3.5 vs 12.6 ng/ml +/- 3.6, respectively, p=0.01). An inverse relationship was found between folate and homocysteine levels. No relationship was observed between vitamin B12 levels and homocysteine levels. There was a significant difference in triglyceride levels between case and control groups (136.91 +/- 67.27 vs 174.3 +/- 77.6, respectively, p=0.01). The odds ratio for hyperhomocysteinemia in acute coronary syndrome was 4.45 (95% confidence interval: 1.5 - 13.3). CONCLUSION: The present study found a significant association between homocysteine levels and acute coronary syndrome in Colombian patients, similarly to the European and North American populations. There was a negative correlation between homocysteine plasma levels and folate levels. No association between plasmatic levels of homocysteine and those of vitamin B12 was observed.

About 35.4% of those receiving enteral nutrition and 39.6% of the ones who received parenteral nutrition had complications; no significant difference per support was found (p = 0.363). Strict application of protocols could decrease the risk for complications and boost nutritional support benefits
