RESUMO

3D printing technology is a tremendously powerful technology to fabricate electrochemical sensing devices. However, current conductive filaments are not aimed at electrochemical applications and therefore require intense activation protocols to unleash a suitable electrochemical performance. Current activation methods based on (electro)chemical activation (using strong alkaline solutions and organic solvents and/or electrochemical treatments) or combined approaches are time-consuming and require hazardous chemicals and dedicated operator intervention. Here, pioneering spark-discharge-activated 3D-printed electrodes were developed and characterized, and it was demonstrated that their electrochemical performance was greatly improved by the effective removal of the thermoplastic support polylactic acid (PLA) as well as the formation of sponge-like and low-dimensional carbon nanostructures. This reagent-free approach consists of a direct, fast, and automatized spark discharge between the 3D-electrode and the respective graphite pencil electrode tip using a high-voltage power supply. Activated electrodes were challenged toward the simultaneous voltammetric determination of dopamine (DP) and serotonin (5-HT) in cell culture media. Spark discharge has been demonstrated as a promising approach for conductive filament activation as it is a fast, green (0.94 GREEnness Metric Approach), and automatized procedure that can be integrated into the 3D printing pipeline.

The objective of the present study was to evaluate the relationship between body weight (BW) and hip width (HW) in dairy buffaloes (Bubalus bubalis). HW was measured in 215 Murrah buffaloes with a BW of 341 ± 161.6 kg, aged between three months and five years, and raised in southeastern Mexico. Linear and non-linear regressions were used to construct the prediction models. The goodness of fit of the models was evaluated using the Akaike information criterion (AIC), Bayesian information criterion (BIC), coefficient of determination (R2), mean squared error (MSE), and root MSE (RMSE). Additionally, the developed models were evaluated through internal and external cross-validation (k-folds) using independent data. The ability of the fitted models to predict the observed values was assessed based on the root mean square error of prediction (RMSEP), R2, and mean absolute error (MAE). The relationship between BW and HW showed a high correlation coefficient (r = 0.96, P < 0.001). The chosen fitted model to predict BW was: -176.33 (± 40.83***) + 8.74 (± 1.79***) × HW + 0.04 (± 0.01*) × HW2, because it presented the lowest MSE, RMSE, and AIC values, which were 1228.64, 35.05 and 1532.41, respectively. Therefore, with reasonable accuracy, the quadratic model using hip width may be suitable for predicting body weight in buffaloes.

Renal involvement is an important cause of morbidity and mortality in systemic lupus erythematosus (SLE). The present study included patients with recently diagnosed Class III and Class IV lupus nephritis (LN) treated by Rheumatology who, upon the detection of alterations in their kidney function, were referred to Nephrology for the joint management of both medical specialties. The purpose of this study was to compare the plasma expression of Toll-Like Receptor 7 (TLR7) and TLR9 in healthy control (HC) subjects and newly diagnosed Class III and Class IV LN patients with 12-month follow-ups. The plasma expression of TLR7 and TLR9 proteins was determined by the ELISA method. A significant increase in the expression of TLR7 protein was found in Class III LN in the basal determination compared to the expression in the HC (p = 0.002) and at 12 months of follow-up (p = 0.03) vs. HC. The expression of TLR9 showed a behavior opposite to that of TLR7. TLR9 showed decreased protein expression in LN Class III patients' baseline and final measurements. The result was similar in the basal and final determinations of LN Class IV compared to the expression in HC. A significant decrease in SLEDAI -2K was observed at 12 months of follow-up in patients in Class III (p = 0.01) and Class IV (p = 0.0001) of LN. Complement C3 levels improved significantly at 12-month follow-up in Class IV patients (p = 0.0001). Complement C4 levels decreased significantly at 12-month follow-up in LN Class III compared to baseline (p = 0.01). Anti-DNA antibodies decreased significantly at 12 months of follow-up in Class IV LN (p = 0.01). A significant increase in proteinuria was found at 12 months of follow-up in Class III LN, compared to the baseline determination (p = 0.02). In LN Class IV, proteinuria decreased at 12 months of follow-up compared to baseline (p = 0.0001). Albuminuria decreased at 12 months of follow-up in LN Class IV (p = 0.006). Class IV LN, albuminuria also decreased at 12 months of follow-up (p = 0.009). Hematuria persisted in all patients and the glomerular filtration rate did not change. Three Class IV patients died before 12 months of follow-up from various causes. In conclusion, although the rheumatologic data appeared to improve, the renal function data remained inconsistent. Decreased expression of TLR9 and increased expression of TLR7 could be useful in the early diagnosis of Class III and Class IV LN is correct.

Three-dimensional (3D) printing technology has emerged as a powerful technology for the fabrication of low-cost microfluidics. Nevertheless, the fabrication of microfluidic devices integrating high-performance electrochemical sensors in practical applications is still an open challenge. Although automatic fabrication of the microfluidic device and the electrodes can be successfully carried out using a one-step multimaterial fused filament fabrication (FFF) approach, the as-printed electrochemical performance of these electrodes is not good enough for chemical (bio)sensing and their surface modification is challenging because after closing the channel there is no physical access to the electrode. Thus, here a pause-print-pause (PPP) microfabrication approach was implemented. The fabrication was paused before printing the microfluidics, and the filament-based electrodes were directly modified on the printing bed via stencil printing, drop casting, and electrodeposition. To exemplify this versatile workflow, the design of a microfluidic glucose sensor was proposed. To this end, first, the working and counter electrodes were stencil printed with graphite ink while the reference electrode was stencil printed with Ag|AgCl ink. Then, Prussian blue was formed on the working electrode either by drop casting or by electrodeposition, and glucose oxidase was drop cast on top. At this point, the microfabrication process was resumed, and the microfluidics were printed on top of the modified electrodes to complete the construction of hybrid electrochemical fluidic fused filament fabricated devices (h-eF4Ds). This print-pause-print approach is not limited to ink-based electrodes or glucose oxidase, and we envisage these results will pave the way for the effective integration of electrodes in microfluidic devices in a simple and clean-room-free approach, allowing the development of highly customized eF4Ds for a plethora of analytes with high significance.

High temperatures for extended periods, which do not allow animals to recover from heat stress, affect in particular those BLV-infected animals that carry a high proviral load. For this study, animals were discriminated between BLV (+) and BLV (-), and those belonging to the first group, were classified based on their proviral load. The expression of the inflammatory cytokine TNF-α and its receptors, which play an important role in disease progression, were quantified by qPCR in two different seasons. During the summer, average temperature was 19.8 °C, maximums higher than 30 °C were frequent. Instead, during the autumn, the average temperature was 12.63 °C, and temperatures never exceeded 27 °C. During this season, almost no periods of temperatures exceeded the comfort limit. Our results revealed that the expression levels of TNF-α and its receptors were downregulated in animals with high proviral load. This fact could affect their antiviral response and predispose to viral dissemination; over time, animals with a poorer immune system are prone to acquiring opportunistic diseases. Conversely, animals with LPL maintained their expression profile, with behavior comparable to non-infected animals. These findings should be considered by producers and researchers, given the problems that global warming is causing lately to the planet.

End-stage renal disease (ESRD) progression is closely related to oxidative stress (OS). The study objective was to determine the oxidant and antioxidant status in peritoneal dialysis (PD) patients with type 2 diabetes mellitus (DM). An analytical cross-sectional study from the PD program was carried out with 62 patients, 22 with and 40 without DM. Lipoperoxides (LPO) levels in patients with DM, 3.74 ± 1.09 mM/L, and without DM, 3.87 ± 0.84 mM/L were found to increase compared to healthy controls (HC) 3.05 ± 0.58 mM/L (p = 0.006). The levels of the oxidative DNA damage marker (8-OH-dG) were found to be significantly increased in patients with DM, 1.71 ng/mL (0.19-71.92) and without DM, 1.05 ng/mL (0.16-68.80) front to 0.15 ng/mL (0.15-0.1624) of HC (p = 0.001). The antioxidant enzyme superoxide dismutase (SOD) activity was found to be significantly increased in patients with DM, 0.37 ± 0.15 U/mL, and without DM, 0.37 ± 0.17 compared to HC, 0.23 ± 0.05 U/mL (p = 0.038). The activity of the enzyme glutathione peroxidase (GPx) showed a significant increase (p < 0.001) in patients with DM, 3.56 ± 2.18 nmol/min/mL, and without DM, 3.28 ± 1.46 nmol/min/mL, contrary to the activity obtained in HC, 1.55 ± 0.34 nmol/min/mL. In conclusion, we found an imbalance of oxidative status in patients undergoing PD with and without DM through the significant increase in LPO oxidants and the marker of oxidative damage in DNA. The activity of the antioxidant enzymes SOD and GPx were significantly increased in patients with and without DM undergoing PD, possibly in an attempt to compensate for the deregulation of oxidants. Antioxidant enzymes could be promising therapeutic strategies as a complement to the management of chronic kidney diseases.

Lupus nephritis (LN) is a severe complication of systemic lupus erythematosus (SLE) and is considered one of the leading causes of mortality. Multiple immunological pathways are involved in the pathogenesis of SLE, which makes it imperative to deepen our knowledge about this disease's immune-pathological complexity and explore new therapeutic targets. Since an altered redox state contributes to immune system dysregulation, this document briefly addresses the roles of oxidative stress (OS), oxidative DNA damage, antioxidant enzymes, mitochondrial function, and mitophagy in SLE and LN. Although adaptive immunity's participation in the development of autoimmunity is undeniable, increasing data emphasize the importance of innate immunity elements, particularly the Toll-like receptors (TLRs) that recognize nucleic acid ligands, in inflammatory and autoimmune diseases. Here, we discuss the intriguing roles of TLR7 and TLR9 in developing SLE and LN. Also included are the essential characteristics of conventional treatments and some other novel and little-explored alternatives that offer options to improve renal function in LN.

BACKGROUND: Food and nutrition were affected by the COVID-19 pandemic, increasing levels of food insecurity. University students were a risk group for food insecurity due to the closure of educational establishments where they received their main meals. AIM: To assess food insecurity among Chilean public university students before and during the COVID-19 pandemic. MATERIAL AND METHODS: The study had a non-probabilistic before-and-after design. Undergraduate students from all colleges at the university were invited to answer an online survey about food insecurity, devised based on FAO Food Insecurity Experience Scale. RESULTS: Nine hundred and one students answered the survey. The prevalence of moderate/severe food insecurity increased from 9.5% to 14.3% before and during the pandemic, respectively. Forty two percent (n = 196) of student households became food insecure during the sanitary crisis. Students commented on the positive and negative aspects of the pandemic in their eating behaviors. CONCLUSIONS: Undergraduate students are vulnerable to food insecurity. Mitigation actions should be carried out when educational establishments are closed.

BACKGROUND: The immigrant population in Chile is growing significantly, challenging the health care system's capacity to provide sensitive and effective care. AIM: To assess the cultural competence of Chilean primary health workers. MATERIAL AND METHODS: This mixed-methods study used a quantitative scale and semi-structured interviews to examine the cultural competence of a Chilean primary health care team in their care for the international migrant population. The study pointed to specific variables that were associated with increased cultural competence, including age, intercultural life experiences, and having received relevant training. RESULTS: The two approaches produced largely convergent results. The dimensions of cultural competence with the highest scores on the quantitative scale were also the theoretical dimensions with the greatest discursive density in the qualitative analysis. CONCLUSIONS: The analysis provides a starting point for policies aimed at the development of cultural competence in the national scenario, suggesting a general direction to foster transcultural competence in health, such as formal training and the promotion of informal spaces ofsensibilization.

The ubiquity and importance of ROS and RNS in cellular signaling, disease development, and death give rise to an outstanding interest in their detection and quantification. Among the analytical techniques available, electrochemical sensors stand out for the detection of ROS/RNS due to their high sensitivity and inherent miniaturization which allows the in situ and real-time detection together with a tunable selectivity due to the different electrochemical behavior of ROS/RNS. Nanomaterial-based enzyme-free electrochemical sensors possess improved sensitivity, selectivity, stability, and unique catalytic activities. In addition, their integration in nanoelectrodes, lab-on-chips, microfluidic systems, and stretchable electrodes allow the determination of ROS/RNS in individual cells, cell organelles, or cell populations, under different experimental conditions hardly accessible using classical detection methods.

OBJECTIVE: To investigate oxidative stress markers and antioxidants in bipolar disorder (BD). METHODS: Electronic MEDLINE/PubMed/Cochrane-Library/Scopus/TripDatabase search until 06/30/2019 for studies comparing antioxidant or oxidative stress markers between BD and healthy controls (HCs). Standardized mean differences (SMD) and 95% confidence intervals (CIs) were calculated for ≥3 studies. RESULTS: Forty-four studies (n = 3,767: BD = 1,979; HCs = 1,788) reported on oxidative stress markers malondialdehyde (MDA), thiobarbituric acid reactive substances (TBARS), and total nitrites; antioxidants glutathione (GSH), uric acid, and zinc; or antioxidantenhancing enzymes superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPX), and GSH-transferase (GST). Compared with HCs, BD was associated with higher GST (P = .01), CAT (P = .02), nitrites (P < .0001), TBARS (P < .0001), MDA (P = .01), uric acid (P < .0001), and lower GSH (P = .006), without differences in SOD, GPX, and zinc. Compared to HCs, levels were higher in BD-mania for TBARS (P < .0001) and uric acid (P < .0001); in BD-depression for TBARS (P = .02); and BD-euthymia for uric acid (P = .03). Uric acid levels were higher in BD-mania vs BD-depression (P = .002), but not vs BD euthymia. TBARS did not differ between BD-mania and BD-depression. Medication-free BD-mania patients had higher SOD (P = .02) and lower GPX (P < .0001) than HCs. After treatment, BD did not differ from HCs regarding SOD and GPX. CONCLUSIONS: Beyond a single biomarker of oxidative stress, the combination of several parameters appears to be more informative for BD in general and taking into account illness polarity. BD is associated with an imbalance in oxidative stress with some phase-specificity for uric acid and TBARS and possible treatment benefits for SOD and GPX. Future studies should take into account confounding factors that can modify oxidative stress status and simultaneously measure oxidative stress markers and antioxidants including different blood sources.

A new green and effective sonochemical liquid-phase exfoliation (LPE) is proposed wherein a flavonoid compound, catechin (CT), promotes the formation of conductive, redox-active, water-phase stable graphene nanoflakes (GF). To maximize the GF-CT redox activity, the CT concentration and sonication time have been studied, and the best performing nanomaterial-fraction selected. Physicochemical and electrochemical methods have been employed to characterize the morphological, structural, and electrochemical features of the GF-CT nanoflakes. The obtained GF intercalated with CT exhibits fully reversible electrochemistry (ΔEp = 28 mV, ipa/ipc = ⁓1) because of the catecholic adducts. GF-CT-integrated electrochemistry was generated directly during LPE of graphite, with no need of graphene oxide production, nor activation steps, electropolymerization, or ex-post functionalization. The GF-CT electro-mediator ability has been proven towards hydrazine (HY) and ß-nicotinamide adenine dinucleotide (NADH) by simply drop-casting the redox-material onto screen-printed electrodes. GF-CT-based electrodes by using amperometry exhibited high sensitivity and extended linear ranges (HY: LOD = 0.1 µM, L.R. 0.5-150 µM; NADH: LOD = 0.6 µM, L.R. 2.5-200 µM) at low overpotential (+ 0.15 V) with no electrode fouling. The GF-CT electrodes are performing significantly better than commercial graphite electrodes and graphene nanoflakes exfoliated with a conventional surfactant, such as sodium cholate. Recoveries of 94-107% with RSD ≤ 8% (n = 3) for determination of HY and NADH in environmental and biological samples were achieved, proving the material functionality also in challenging analytical media. The presented GF-CT is a new functional redox-active material obtainable with a single-pot sustainable strategy, exhibiting standout properties particularly prone to (bio)sensors and cutting-edge device development.

RESUMO

Phosphonate-directed ortho C-H borylation of aromatic phosphonates is reported. Using simple starting materials and commercially accessible catalysts, this method provides steady access to o-phosphonate arylboronic esters bearing pendant functionality and flexible substitution patterns. These products serve as flexible precursors for a variety of highly substituted phosphoarenes, and in situ downstream functionalization of the products is described.

A novel benchtop approach to fabricate xurography-enabled thermally transferred (XTT) carbon nanomaterial-based electrochemical sensors is proposed. Filtered nanomaterial (NM) films were transferred from Teflon filters to polyethylene terephthalate-ethylene vinyl acetate (PET-EVA) substrates by a temperature-driven approach. Customized PET-EVA components were xurographically patterned by a cutting plotter. The smart design of PET-EVA films enabled us to selectively transfer the nanomaterial to the exposed EVA side of the substrate. Hence, the substrate played an active role in selectively controlling where nanomaterial transfer occurred allowing us to design different working electrode geometries. Counter and reference electrodes were integrated by a stencil-printing approach, and the whole device was assembled by thermal lamination. To prove the versatility of the technology, XTT materials were exclusively made of carbon black (XTT-CB), multiwalled carbon nanotubes (XTT-MWCNTs), and single-walled carbon nanotubes (XTT-SWCNTs). Their electrochemical behavior was carefully studied and was found to be highly dependent on the amount and type of NM employed. XTT-SWCNTs were demonstrated to be the best-performing sensors, and they were employed for the determination of l-tyrosine (l-Tyr) in human plasma from tyrosinemia-diagnosed patients. High analytical performance toward l-Tyr (linear range of 0.5-100 µM, LOD = 0.1 µM), interelectrode precision (RSD ip,a = 3%, n = 10; RSD calibration slope = 4%, n = 3), and accurate l-Tyr quantification in plasma samples with low relative errors (≤7%) compared to the clinical declared values were obtained. The proposed benchtop approach is cost-effective and straightforward, does not require sophisticated facilities, and can be potentially employed to develop pure or hybrid nanomaterial-based electrodes.

A high-performance screen-printed electrode (SPE) based nanocomposite sensor integrating tungsten disulfide (WS2) flakes decorated with catechin-capped gold nanoparticles (AuNP-CT) and carbon black (CB) has been developed. The excellent antifouling properties of WS2 decorated with AuNP-CT into a high conductivity network of CB results in high selectivity, sensitivity, and reproducibility for the simultaneous determination of hydroxycinnamic acid (hCN) structural analogs: caffeic (CF), sinapic (SP), and p-coumaric acids (CM). Using differential pulse voltammetry (DPV), the target hCNs resulted in three well-resolved oxidation peaks at SPE-CB-WS2/AuNP-CT sensor. Excellent antifouling performance (RSD ip,a ≤ 3%, n = 15 for three analytes' simultaneous measure) and low detection limits (CF 0.10 µmol L-1; SP, 0.40 µmol L-1; CM, 0.40 µmol L-1) are obtained despite the analyzed compounds having a high passivation tendency towards carbon-based sensors. The SPE-CB-WS2/AuNP-CT sensor was successfully applied to determine CF, SP, and CM in food samples with good precision (RSD ≤ 4%, n = 3) and recoveries (86-109%; RSD ≤ 5%, n = 3). The proposed sensor is the first example exploiting the simultaneous determination of these compounds in food samples. Given its excellent electrochemical performance, low cost, disposability, and ease of use, this SPE-CB-WS2/AuNP-CT nanocomposite sensor represents a powerful candidate for the realization of electrochemical devices for the determination of (bio)compounds with high passivation tendency. One of the main functions of infrared thermography (IRT) consists in detecting temperature changes in organisms caused by variations in surface blood circulation. IRT is a useful tool that has been used mainly as a diagnostic method for various stress-causing pathologies, though recent suggestions indicate that it can be used to assess the block quality of certain body regions. In the field of anaesthesiology, IRT has been applied to brachial and epidural blocks, while in algology, changes in surface blood circulation associated with sympathetic activity have been investigated. Thermography has also been employed to complement pain level scales based on the facial expressions of patients in critical condition, or after surgery. In addition, it has been used as a tool in research designed to evaluate different surgical procedures in human medicine, as in the case of surgical burrs for placing dental implants, where IRT helps assess the degree of heating associated with bone devascularisation, reduction in vascular perfusion as a consequence of stroke, and changes in the autonomous nervous system, or the degree of vascular changes in flaps applied to burn patients. In veterinary medicine, thermography has brought several benefits for animals in terms of evaluating lesions, diseases, and surgical procedures. The aim of this review is to evaluate how IRT can be used as a tool in surgical procedures, cases of vascular change, and pain monitoring in veterinary medicine with an emphasis on small animals.

OBJECTIVE: To explore the perspectives of the decision makers and community members in primary health care (PHC) around the conceptualization of social participation (PS). DESIGN: An exploratory cross-sectional study with qualitative methodology. LOCATION: Health Centers of the Metropolitan Region (RM), Santiago, Chile. PARTICIPANTS: Eight informants from the management level (group 1), 13 from execution level in PHC (group 2), 28 community members and four community agents of health (group 3). METHOD: Interviews and discussion groups were conducted, which were recorded and transcribed. The organization and analysis of the data was done with Atlas.ti 8.1. The narratives were systematized using a thematic analysis. All the documents were codified, and we hold periodic meetings to review the existing codes, as well as discussing the inclusion of new codes. RESULTS: Group 1 refers to a more theoretical conception of PS. Group 2 expresses more concrete and operative dimensions. Group 3 indicates that PS is embodied in particular personal experiences. Groups 1 and 3 have more than one notion of social participation in health. CONCLUSIONS: An institutional conception of participation prevails transversally, rooted since the 1990s. At the community level, the narratives take the form of collective practices lived around the improvement of the quality of community life mediated by the level of execution.

A new hybrid nanomaterial is used in a screen-printed electrode (SPE) for sensing of the ortho-diphenols oleuropein (OLEU) and hydroxytyrosol (HYT) in extra virgin olive oil (EVOO) and related samples. The hybrid material consists of carbon black (CB) and molybdenum disulfide (MoS2). In comparison with individual nanomaterials, CB-MoS2 exhibits improved charge-transfer ability, low charge-transfer resistance, high electrical conductivity and enhanced electrocatalysis. The sensor is also characterized by (a) high sensitivity that avoids the need for adsorptive voltammetry, (b) reduced analysis time, and (c) high anti-fouling ability (electrode RSDOLEU < 8%, for n = 10). OLEU can be detected in the 0.3 to 30 µM concentration range with a 0.1 µM LOD, and HYT in the 2-100 µM range with a 1 µM LOD. A comparison of the data obtained by this sensor and by HPLC-UV exhibited high correlation (r = 0.995, p < 0.05). These data revealed the reliability of CB-MoS2 for analysis of complex EVOO and related samples. Diabetic peripheral neuropathy is a major debilitating late complication of diabetes, which significantly reduces the quality of life in patients. Diabetic peripheral neuropathy is associated with a wide spectrum of sensory abnormalities, where in loss of sensation or hypoalgesia to applied external stimuli is paradoxically accompanied by debilitating tonic spontaneous pain. In numerous studies on animal models of diabetic peripheral neuropathy, behavioural measurements have been largely confined to analysis of evoked withdrawal to mechanical and thermal stimuli applied to dermatomes, whereas spontaneous, on-going pain has not been widely studied. In the Streptozotocin model of type 1 diabetes, we employed the Conditioned Place Preference test to assess tonic pain. Our results indicate that both phases, that is, early evoked hypersensitivity (i.e. 5-7 weeks post-Streptozotocin) as well as late stage hypoalgesia (i.e. 17-20 weeks post-Streptozotocin) are accompanied by significant tonic pain in mice with diabetic peripheral neuropathy. We also report on the temporal relation between on-going pain and neuropathological changes in the dorsal root ganglia of mice with diabetic peripheral neuropathy up to 6 months post-Streptozotocin. Neither early hypersensitivity nor late hypoalgesia were associated with markers of cellular stress in the dorsal root ganglia. Whereas significant neutrophil infiltration was observed in the dorsal root ganglia over both early and late stages post-Streptozotocin, T-cell infiltration in the dorsal root ganglia was prominent at late stages post-Streptozotocin. Thus, longitudinal analyses reveal that similar to patients with chronic diabetic peripheral neuropathy, mice show tonic pain despite sensory loss after several months in the Streptozotocin model, which is accompanied by neuroimmune interactions in the dorsal root ganglia.
