RESUMO

Pharmaceutical deltamethrin (Alpha Max), used as delousing treatments in aquaculture, has raised concerns due to possible negative impacts on the marine environment. A novel approach combining different scientific disciplines has addressed this topic. Acute (mortality) and sublethal effects (i.e., fitness, neurological, immunological, and oxidative responses) of exposure of northern shrimp (Pandalus borealis) were studied in laboratory experiments. Passive water sampling combined with sediment analyses revealed environmental concentrations. Finally, dispersal modeling was performed to predict environmental concentrations. Ecotoxicological analyses showed mortality in shrimp after 1 h of exposure to 2 ng L-1 (1000-fold dilution of treatment dose), revealing a high sensitivity to deltamethrin. Sublethal effects included induction of acetylcholinesterase and acyl CoA oxidase activities and oxidative impairment, which may be linked to neurotoxic responses. Environmental factors that can influence telomeres are diverse, but the association between telomeres and exposure to environmental contaminants is yet to be elucidated. To date, prior studies have focused on legacy persistent chlorinated pollutants (POPs), while the effects of poly- and perfluoroalkyl substances (PFAS) have been poorly documented. Here, we investigated the associations among PFAS congeners, absolute telomere length (cross-sectional approach), and telomere dynamics (rate of telomere length change over time, longitudinal approach) in one of the most contaminated arctic top predators, the glaucous gull Larus hyperboreus from Svalbard. We further estimated the effect of PFAS on apparent survival rates and re-sighting probabilities using a 10-year capture/recapture dataset (2010-2019). We found that birds exposed to higher concentrations of perfluorononadecanoate (PFNA) (median of 1565 pg/mL of ww in males and 1370 pg/mL of ww in females) and perfluorotetradecanoate (PFTeDA) (median of 370 pg/mL of ww in males and 210 pg/mL of ww in females) showed the slowest rate of telomere shortening. We also found that high blood concentrations of perfluorooctanoate (PFOA) (median of 120 pg/mL of ww in males and 150 pg/mL of ww in females) and perfluorohexanesulfonate (PFHxS) (median of 495 pg/mL of ww in males and 395 pg/mL of ww in females) were positively associated with higher re-sighting probabilities and apparent survival in males but not in females. Our work is the first to report an association between single PFAS compounds and telomeres, and the first to link PFAS exposure with survival probabilities, suggesting that the effect of PFAS exposure might be more tied to the type of compound rather than the total concentration of PFAS.

This work was designed to investigate biological impacts on blue mussels (Mytilus edulis spp) after being exposed to diesel spill. On December 2013, an 180,000-litre accidental acute diesel spill was reported in a small harbour of northern Norway (Skjervøy). In order to assess the biological effects on the wild population of blue mussels, bivalves were collected at three different locations: at the oil-spill spot, at the other side of the harbour (opposite the oil-spill area), and in an uncontaminated site. Body burden and seawater samples were collected from a few days up to five months after the diesel spill. Biomarkers of oxidative stress and immunological effects were assessed in the blue mussels digestive glands. Our findings reported significant modulation of GST (detoxification), SOD (antioxidant response) and MDA (lipid peroxidation) in bivalves exposed to diesel with a similar response at two and five months after the spill. Laccase-type enzyme also highlighted an important aspect in terms of biomarker response of the immune function. Overall, our study demonstrated that some biomarkers returned to basal levels a few months after the diesel spill. Consequently, it highlighted the usefulness of normalised tools and guidelines for biomonitoring strategies after a diesel spill.

The glaucous gull (Larus hyperboreus) is an arctic top predator and scavenger exposed to high levels of mixtures of organohalogenated contaminants (OHCs) of which many interfere with the thyroid hormone (TH) system. In the present study, we applied statistical modeling to investigate the potential combined influence of the mixture of chlorinated, brominated and perfluorinated organic compounds in plasma of glaucous gulls on their plasma TH concentrations. In females, there were significant negative associations between several organochlorinated compounds (OCs) and free thyroxin (FT4) and triiodothyronine (FT3), indicating additive negative effects on FT4 and FT3. However, in these females there was also a significant positive association between perfluorooctane sulfonate (PFOS) and FT3. The inverse associations between several OCs and FT3 and the contrasting positive association between PFOS and FT3, indicate that these two groups of OHCs may have dissimilar and antagonistic effects on FT3 in female glaucous gulls. In males, there were no associations between any of the OHCs and the THs. That OHCs affect THs in a complex manner involving both additive and antagonistic effects add to the challenge of interpreting the overall functional effect of thyroid disruptive chemicals in wildlife. However, experimental studies are needed to confirm or disprove such effects.

Have Although persistent organic pollutants (POPs) may affect the immune system, few field studies actually examined this effect. There are indications that POP exert effects on the immune system; however, in the Arctic ecosystem data are scarce. The aim of this study was to examine immune functions in two medium trophic-positioned seabirds, the black-legged kittiwakes (Rissa tridactyla) and Atlantic puffins (Fratercula arctica). Overall POP concentrations were higher in kittiwakes than puffins and males had significantly higher concentrations than females. Mean concentrations of total polychlorinated biphenyls (ΣPCB9) were 4700 ± 200 and 9600 ± 1400 ng/g lipid weight and 2800 ± 180 and 3900 ± 200 ng/g lipid weight in female and male kittiwake and puffin blood, respectively. Levels of immunoglobulin-Y (IgY) in blood of kittiwakes were not markedly affected by concentrations of POP. Similarly, the primary IgY response to tetanus toxoid was not affected by POP concentrations in a subsample of immunized kittiwakes. In puffins, there were significant correlations between the IgY-response and some of the POPs, but with low explanatory values. These results suggest that POPs concentrations were lower than, or just at the threshold level for effects of the proposed IgY biomarker. It is also conceivable that the IgY levels are not a suitable endpoint for evaluating perturbation of the immune system in free-living seabirds.

Plastic pollution threatens many organisms around the world. In particular, the northern fulmar, Fulmarus glacialis, is known to ingest high quantities of plastics. Since data are sparse in the Eurasian Arctic, we investigated plastic burdens in the stomachs of fulmar fledglings from Kongsfjorden, Svalbard. Fifteen birds were collected and only particles larger than 1 mm were extracted, characterised and analysed with Fourier Transform InfraRed spectroscopy. All birds ingested plastic. In total, 683 plastic particles were found, with an average of 46 ± 40 SD items per bird. The most common shape, colour and polymer were hard fragment, white, and polyethylene, respectively. Microplastics (< 5 mm) were slightly more represented than mesoplastics (> 5 mm). This study confirms high numbers of ingested plastics in fulmar fledglings from Svalbard and suggests that fulmar fledglings may be suitable for temporal monitoring of plastic pollution, avoiding potential biases caused by age composition or breeding state.

This study tested whether the immune system of the glaucous gull (Larus hyperboreus) chicks became affected by existing environmental contaminants. An experimental group was given food that mimicked the natural contaminant mixture found in food from the North Atlantic marine environment, while the control group was given the equivalent of nearly clean food. All chicks were immunized with herpes virus (EHV), reovirus (REO), influenza virus (EIV), and tetanus toxoid (TET) in order to test their ability to respond to foreign specific antigens. At 8 wk, the experimental group had 3- to 13-fold higher concentrations of hexachlorobenzene (HCB), oxychlordane, p,p'-DDE, and total polychlorinated biphenyls (Sigma PCB) than did the control. The experimental group produced significantly lower antibody titer against EIV and had lower concentrations of immunoglobulin-G (IgG) and -M (IgM) in blood. Hematocrit percent and leukocyte numbers did not differ between the two groups. The ability of lymphocytes to proliferate in vitro was tested with three mitogens, phytohemagglutinin (PHA), concanavalin A (Con A), and pokeweed mitogen (PWM), and three antigens, keyhole limpet hemocyanin (KLH), TET, and Mycobacterium avium subsp. paratuberculosis tuberculin purified protein derivative (PPD). The experimental group had a significantly higher peripheral blood lymphocyte response to PHA and to spleen lymphocytes in vitro stimulated with Con A and PCB congeners 99 or 153, while the Con A, PWM, KLH, TET, PPD, and Con A plus PCB-156 or -126 showed nonsignificant differences between groups. Data indicate that the combined effect of multiple persistent organic pollution exposures occurring naturally in the Arctic negatively affect the immune system of the glaucous gull chick.

Eggs of snow buntings (Plectrophenax nivealis) were applied as a bio-indicator to examine differences in exposure to legacy persistent organic pollutants (POPs) and perflouroalkyl subtances (PFAS) from the terrestrial environment surrounding the settlements of Longyearbyen, Barentsburg and Pyramiden on Svalbard, Norway. Significantly higher concentrations of summed polychlorinated biphenyls (sumPCB7) in eggs collected from Barentsburg (2980 ng/g lipid weight (lw)) and Pyramiden (3860 ng/g lw) compared to Longyearbyen (96 ng/g lw) are attributed to local sources of PCBs within these settlements. Similar findings were observed for p,p'-dichlorodiphenyldichloroethylene (p,p'-DDE) where higher median concentrations observed in Pyramiden (173 ng/g lw) and Barentsburg (75 ng/g lw) compared to Longyearbyen (48 ng/g lw) may be influenced by guano inputs from breeding seabird populations, although other point sources cannot be ruled out. Concentrations of perfluorooctane sulphonate (PFOS) and several perfluorinated carboxylic acids (PFCAs) in snow bunting eggs were found to be statistically higher in the populated settlements of Longyearbyen and Barentsburg compared to the abandoned Pyramiden. Narrow foraging ranges of snow buntings during breeding season was useful in assessing point sources of exposure for PCBs and PFAS at particular sites with extreme differences observed between nest locations. SumPCB7 concentrations ranged from 2 µg/g ww to below detection limits between nest sites located less than a kilometer from each other in Pyramiden. Similar findings were observed in Longyearbyen, where several PFCAs ranged from 2 to 55 times higher between nest sites with similar spatial distances. These findings indicate that snow buntings can be a useful bio-indicator offering high spatial resolution for contaminant source apportionment in terrestrial environments on Svalbard.

Due to retreating sea ice and predictions of undiscovered oil and gas resources, increased activity in Arctic shelf sea areas associated with shipping and oil and gas exploration is expected. Such activities may accidentally lead to oil spills in partly ice-covered ocean areas, which raises issues related to oil spill response. Net Environmental Benefit Analysis (NEBA) is the process that the response community uses to identify which combination of response strategies minimises the impact to environment and people. The vulnerability of Valued Ecosystem Components (VEC's) to oil pollution depends on their sensitivity to oil and the likelihood that they will be exposed to oil. As such, NEBA requires a good ecological knowledge base on biodiversity, species' distributions in time and space, and timing of ecological events. Biological resources found at interfaces (e.g., air/water, ice/water or water/coastline) are in general vulnerable because that is where oil can accumulate. Here, we summarize recent information about the seasonal, physical and ecological processes in Arctic waters and evaluate the importance these processes when considering in oil spill response decision making through NEBA. In spring-time, many boreal species conduct a lateral migration northwards in response to sea ice retraction and increased production associated with the spring bloom. However, many Arctic species, including fish, seabirds and marine mammals, are present in upper water layers in the Arctic throughout the year, and recent research has demonstrated that bioactivity during the Arctic winter is higher than previously assumed. Information on the seasonal presence/absence of less resilient VEC's such as marine mammals and sea birds in combination with the presence/absence of sea ice seems to be especially crucial to consider in a NEBA. In addition, quantification of the potential impact of different, realistic spill sizes on the energy cascade following the spring bloom at the ice-edge would provide important information for assessing ecosystem effects.

Environmental contaminants are found throughout Arctic marine ecosystems, and their presence in seabirds has been associated with toxicological responses. However, there are few studies of genotoxicity in Arctic avian wildlife. The purpose of the present study was to quantify deoxyribonucleic acid (DNA) damage in lymphocytes of selected seabird species and to examine whether accumulation of organohalogen contaminants (ΣOHCs) affects DNA damage. Blood was sampled from common eider (Somateria mollissima), black guillemot (Cepphus grylle), black-legged kittiwake (Rissa tridactyla), glaucous gull (Larus hyperboreus), arctic skua (Stercorarius parasiticus), and great skua (Stercorarius skua) in Kongsfjorden, Svalbard (Norway). Contaminant concentrations found in the 6 species differed, presumably because of foraging ecology and biomagnification. Despite large differences in contaminant concentrations, ranging from ΣOHCs 3.3 ng/g wet weight in the common eider to ΣOHCs 895 ng/g wet weight in the great skua, there was no strong difference among the species in baseline DNA damage or sensitivity to a genotoxic stressor (hydrogen peroxide). Baseline levels of DNA damage were low, with median values ranging from 1.7% in the common eider to 8.6% in the great skua. There were no associations between DNA damage and contaminants in the investigated species, suggesting that contaminant concentrations in Kongsfjorden are too low to evoke genotoxic effects, or possibly that lymphocytes are resistant to strand breakage. Samples (blood or tissue fluid) from 594 arctic foxes (Alopex lagopus), 390 Svalbard reindeer (Rangifer tarandus platyrhynchus), 361 sibling voles (Microtus rossiaemeridionalis), 17 walruses (Odobenus rosmarus), 149 barnacle geese (Branta leucopsis), 58 kittiwakes (Rissa tridactyla), and 27 glaucous gulls (Larus hyperboreus) from Svalbard and nearby waters were assayed for antibodies against Toxoplasma gondii using a direct agglutination test. The proportion of seropositive animals was 43% in arctic foxes, 7% in barnacle geese, and 6% (1 of 17) in walruses. There were no seropositive Svalbard reindeer, sibling voles, glaucous gulls, or kittiwakes. The prevalence in the arctic fox was relatively high compared to previous reports from canid populations. There are no wild felids in Svalbard and domestic cats are prohibited, and the absence of antibodies against T. gondii among the herbivorous Svalbard reindeer and voles indicates that transmission of the parasite by oocysts is not likely to be an important mechanism in the Svalbard ecosystem. Our results suggest that migratory birds, such as the barnacle goose, may be the most important vectors bringing the parasite to Svalbard. In addition to transmission through infected prey and carrion, the age-seroprevalence profile in the fox population suggests that their infection levels are enhanced by vertical transmission.

The present study explored short-term temporal variations in circulating concentrations of 3 legacy organochlorines with different physicochemical properties (polychlorinated biphenyl 153 [PCB-153], p,p'-dichlorodiphenyldichloroethylene [DDE], and hexachlorobenzene [HCB]) in breeding kittiwakes (Rissa tridactyla) in a colony in Svalbard (78°N), Norwegian Arctic. Concentrations were measured in blood of a large number (n = 412-521 blood samples, depending on the data analyses) of prebreeding, incubating, and chick-rearing birds over a period of 5 yr (2007-2011). The PCB-153 concentrations were equal in male and female blood in the prebreeding period, whereas females had significantly lower concentrations during incubation and chick rearing, probably because of their ability to eliminate organochlorines through egg laying. A similar temporal pattern was observed with DDE, although the lower concentrations in incubating females were not significant. Males and females had similar concentrations of HCB over all reproductive stages. The concentrations of all 3 compounds varied greatly between years. The concentrations of PCB-153 tended to decline over the study period, whereas concentrations of HCB showed an increasing trend, especially among chick-rearing males late in the season. Concentrations of PCB-153 increased approximately 2.5 times from the prebreeding to the chick-rearing period, concurrent with mobilization of body lipids (reduced body mass). A similar, but less pronounced trend was found for HCB. For DDE, however, kittiwakes had the highest concentrations in the prebreeding period, suggesting relatively high exposure in their winter areas. The present study documented large variations in circulating concentrations of legacy organochlorines among and within breeding seasons in kittiwakes, but the alterations within seasons were relatively consistent from year to year. Shipping activities are expected to increase in the Arctic Seas. Today, the majority of vessels are using marine diesel oil (MDO) as propulsion fuel. However, there is a general lack of knowledge of how cold-water marine species respond to acute exposures to MDO. Arctic red king crabs (Paralithodes camtschaticus) were exposed to mechanically dispersed MDO in a flow-through exposure system for one week followed by three weeks of recovery. Observations of increased movements in exposed crabs were interpreted as avoidance behaviour. Further, glutathione peroxidase activity increased in high exposed crab, the catalase activity showed an insignificant increase with exposure, while no differences between groups were observed for lipid peroxidation and acetylcholinesterase activity. After three weeks of recovery in clean seawater, polycyclic aromatic hydrocarbons concentrations in the crabs were significantly reduced, with no specific biomarker responses in exposed groups compared to the control. The results suggest that effects from instantaneous MDO spill only will have short-term effects on the red king crab.

Eight arctic species, including fish, birds and mammals, from diverse habitats (marine and terrestrial) within the Svalbard Archipelago, Norway, were screened for 14 organophosphorus flame retardant (PFR) compounds. Ten PFRs were detected: tris(2-chloroethyl)phosphate (TCEP), tris(2-chloroisopropyl)phosphate (TCIPP), tris(1,3-dichloro-2-propyl)phosphate (TDCIPP), triphenyl phosphate (TPHP); 2-ethylhexyl diphenyl phosphate (EHDPP); tris(2-butoxyethyl)phosphate (TBOEP); tritolyl phosphate (TCrP); triisobutyl phosphate (TIBP); tris(2-ethylhexyl)phosphate (TEHP); and butyl diphenyl phosphate (DPhBP). The greatest number of different PFR compounds, and the highest detection frequency were measured in capelin (Mallotus villotus), and the lowest in Brünnich's guillemot (Uria lomvia). The highest concentrations of ΣPFR, as well as the highest concentration of a single PFR compound, TBOEP, were measured in arctic fox (Vulpes lagopus). The presence of PFR compounds in arctic biota indicates that these compounds can undergo long-range transport and are, to some degree, persistent and bioaccumulated. The potential for biomagnification from fish to higher trophic levels seems to be limited.

Chronic exposure to persistent organic pollutants (POPs) in wildlife might alter the response to environmental changes through interference with the regulation of stress hormones. Here, we examined the relationship between blood concentrations of several POPs and baseline plasma corticosterone levels in the black-legged kittiwake (Rissa tridactyla) during three distinct periods in the breeding season. The concentrations of POPs and corticosterone increased, whereas body mass decreased progressively from the pre-laying period to the incubation and the chick rearing period. ∑PCB (polychlorinated biphenyls) correlated positively with the baseline corticosterone levels during the pre-laying period, which might suggest that PCBs affect the regulation of corticosterone. However, this relationship was not found during the incubation or the chick rearing period. Possible explanations are discussed with emphasis on how total stress/allostatic load is handled during different periods and conditions.

Female common eiders (Somateria mollissima) starve during the nesting stage and may loose 30-45% of their initial body mass, mostly through lipid mobilization. In this study, the effects of fasting on the blood concentrations of three lipid-soluble organochlorines (OCs: polychlorinated biphenyl [PCB]-153; 1-dichloro-2,2-bis (p-chlorophenyl) ethylene [p,p'-DDE]; and hexachlorobenzene [HCB]) were examined in eiders breeding in the high Arctic. Blood samples were taken from females (n=47) at day 5 and day 20 of the incubation period. The mean wet weight concentrations of PCB-153 and p,p'-DDE increased strongly between day 5 and day 20 (3.6 and 8.2-fold, respectively), while HCB increased less (1.7-fold). There was a strong negative association between daily increase in PCB-153 and clutch size, and a weaker relationship for p,p'-DDE, suggesting that maternal transfer to the eggs is a significant pathway of elimination of OCs in eiders. Moreover, poor body condition (body mass controlled for body size) late in the incubation period was associated with strong daily increase of both p,p'-DDE and PCB-153, which may suggest that the release of these compounds increases when lipid reserves become depleted. For HCB, the increase was mainly associated with increase in blood lipid concentrations, and weakly to the amount of burned lipids. The causes for the differences between the compounds are, however, poorly understood. Although the absolute levels of OCs in eiders were relatively low, their rapid build up during incubation is worrying as it coincides with poor body condition and weakened immune systems.

The prediction of a higher parasite infection as a consequence of an impaired immune system with increasing persistent organic pollution (POP) and heavy metal levels were investigated in adult glaucous gull (Larus hyperboreus) from Svalbard. The levels of chlorinated pesticides, polychlorinated biphenyls (PCBs), toxaphenes and polybrominated diphenyl ethers (PBDEs) were measured in liver. Copper, cadmium, lead, mercury, selenium and zinc were measured in kidney samples. An elevated ratio of PCB-118 was found, suggesting that local contamination from the settlement was detectable in the glaucous gull. Eight cestodes, four nematodes, two acanthocephalan and three trematode helminth species were found in the intestine. A positive correlation was found between cestode intensities and selenium levels and between acanthocephalan intensities and mercury levels. No correlation was found between parasite intensities and POP concentrations. It is concluded that the contaminant levels found in glaucous gulls do not cause immune suppression severe enough to affect parasite intensity.

Dead and dying glaucous gulls (Larus hyperboreus) were collected on Bjørnøya in the Barents Sea in 2003, 2004 and 2005. Autopsies of the seabirds only explained a clear cause of death for three (14%) of the 21 birds. A total of 71% of the birds were emaciated. Liver and brain samples were analysed for organochlorine pesticides (OCPs), polychlorinated biphenyls (PCBs), polybrominated diphenyl ether (PBDEs), hexabromocyclododecanes (HBCDs) and mercury (Hg). High levels of SigmaOCPs, SigmaPCBs, SigmaPBDEs and alpha-HBCD were found in liver and brain. Compared to the dead and dying glaucous gulls found 1989, the congeners' composition tended to change toward more persistent compounds in the 2003-2005 samples. The brain levels of OCPs and PCBs did not differ between 1989 and 2003-2005, while the liver levels were significantly lower. The brain/liver ratio for PCB and PBDE significantly decreased with halogenations of the molecule, indicating a clear discrimination of highly halogenated PCBs and PBDEs entering the brain. There was further a clear negative correlation between contaminant concentrations and body condition. The brain levels were not as high as earlier published lethal levels of p,p'-DDE or PCB. However, more recent studies reported a range of sub-lethal OCP- and PCB-related effects in randomly sampled glaucous gulls. An additional elevation of pollutants due to emaciation may increase the stress of the already affected birds. The high brain levels of OCP, PCB and PBDE of present study might therefore have contributed to the death of weakened individuals of glaucous gull.

Biomagnification contributes to high concentrations of persistent organochlorines (OC) in some Arctic vertebrates. Glaucous gulls (Larus hyperboreus) on Bjørnøya in the western Barents Sea were studied to compare the intraspecific variation in OC concentration with variation in trophic feeding levels, estimated from ratios of nitrogen isotopes. Liver tissue samples from 40 adult glaucous gulls were analysed for hexachlorobenzene (HCB), oxychlordane, p,p'-dichlorodiphenyltrichloroethane (DDT) and its metabolite p,p'-DDE, Mirex, and nine congeners of polychlorinated biphenyl (PCB). The ratios of the heavier to lighter isotope of carbon (13C/12C) and nitrogen (15N/14N), expressed as delta13C and delta15N, were measured in liver and muscle. Hepatic concentrations of the nine PCB congeners (Sigma 9 PCB) ranged from 16 microg/g lipid weight to 292 microg/g lipid weight. The delta15N ranged from 14.0/1000 to 15.3/1000 in muscle. Seven of the 14 OC measured, sigma DDT, and sigma 9 PCB were positively correlated to delta15N from muscle tissue. No correlations were found between OC and delta13C. The present results indicate that OC concentrations are partly dependent on the foraging strategy of the gull. The r2 of the linear regressions suggests that up to 18% of the variation in the OC concentrations could be explained by variation in food preference.
