RESUMO

The ecotoxicological effect of after-usage released TiO2 nanoparticles in aquatic resources has been a major concern owing to their production and utilization in different applications. Addressing the issue, this study investigates the detailed in vivo molecular toxicity of TiO2 nanoparticles with Paramecium caudatum. TiO2 nanoparticles were synthesized at a lab scale using high energy ball milling technique; characterized for their physicochemical properties and investigated for their ecotoxicological impact on oxidative stress, steatosis, and apoptosis of cells through different biochemical analysis, flow cytometry, and fluorescent microscopy. TiO2 nanoparticles; TiO2 (N15); of size 36 ± 12 nm were synthesized with a zeta potential of - 20.2 ± 8.8 mV and bandgap of 4.6 ± 0.3 eV and exhibited a blue shift in UV-spectrum. Compared to the Bulk TiO2, the TiO2 (N15) exhibited higher cytotoxicity with a 24 h LC50 of 202.4 µg/ml with P. Caudatum. The mechanism was elucidated as the size and charge-dependent internalization of nanoparticles leading to abnormal physiological metabolism in oxidative stress, steatosis, and apoptosis because of their influential effect on the activity of metabolic proteins like SOD, GSH, MDA, and catalase. The study emphasized the controlled usage TiO2 nanoparticles in daily activity with a concern for ecological and biomedical aspects.

The effects of mono (single type) and hybrid (mixed types) fibres on the workability, compressive strength, flexural strength, and toughness parameters of fly ash geopolymer mortar were studied. The ratio of sand to geopolymer paste of the mortar was 2.75. It was found that workability of mortar decreased more with the use of PP fibres due to its higher dispersion into individual filaments in geopolymer mortar compared to the bundled ARG and PVA fibres. Compressive strength increased by 14% for using 1% steel with 0.5% PP fibres compared to that of the control mixture, which was 48 MPa. However, 25 to 30% decrease of compressive strength was observed in the mortars using the low-modulus fibres. Generally, flexural strength followed the trend of compressive strength. Deflection hardening behaviours in terms of the ASTM C1609 toughness indices, namely I5, I10 and I20 were exhibited by the mortars using 1% steel mono fibres, 0.5% ARG with 0.5% steel and 1% PVA with 0.5% steel hybrid fibres. The toughness indices and residual strength factors of the mortars using the other mono or hybrid fibres at 1 or 1.5% dosage were relatively low. Therefore, multiple cracking and deflection hardening behaviours could be achieved in fly ash geopolymer mortars of high sand to binder ratio by using steel fibres in mono or hybrid forms with ARG and PVA fibres.

OBJECTIVE: To determine disease characteristics, check the treatment status and compare outcome with other series. DESIGN AND SETTING: A retrospective study in a single unit of the Department of Surgery of Khulna Medical College Hospital, south west Bangladesh. PATIENTS AND METHODS: After diagnosis by clinical and radiological examination, 491 patients were treated during the period July 1992 to November 2002. Two options of treatment were carried out: simple closure and peritoneal lavage in 364 cases, and 127 patients managed by non-surgical methods. MAIN OUTCOME MEASURES: mortality and morbidities. RESULTS: Male:female ratio was 445:46, average age 43 years. Mortality in the surgical group was 6.8% and in the non-surgical group 0.02%. CONCLUSION: Perforated peptic ulcer is prevalent even after the wide use of H2 receptor antagonists. Strict case selection for surgical and non-surgical treatment has reduced mortality. These results support the case for non-operative treatment and for conservative surgery.

Sperm cells exhibit extremely high sensitivity in response to slight changes in temperature, osmotic pressure and/or presence of various chemical stimuli. In most cases throughout the evolution, these physico-chemical stimuli trigger Ca (2+)-signaling and subsequently alter structure, cellular function, motility and survival of the sperm cells. Few reports have recently demonstrated the presence of Transient Receptor Potential (TRP) channels in the sperm cells from higher eukaryotes, mainly from higher mammals. In this work, we have explored if the sperm cells from lower vertebrates can also have thermo-sensitive TRP channels. In this paper, we demonstrate the endogenous presence of one specific thermo-sensitive ion channel, namely Transient Receptor Potential Vanilloid family member sub type 1 (TRPV1) in the sperm cells collected from fresh water teleost fish, Labeo rohita. By using western blot analysis, fluorescence assisted cell sorting (FACS) and confocal microscopy; we confirm the presence of this non-selective cation channel. Activation of TRPV1 by an endogenous activator NADA significantly increases the quality as well as the duration of fish sperm movement. The sperm cell specific expression of TRPV1 matches well with our in silico sequence analysis. The results demonstrate that TRPV1 gene is conserved in various fishes, ranging from 1-3 in copy number, and it originated by fish-specific duplication events within the last 320 million years (MY). To the best of our knowledge, this is the first report demonstrating the presence of any thermo-sensitive TRP channels in the sperm cells of early vertebrates as well as of aquatic animals, which undergo external fertilization in fresh water. This observation may have implications in the aquaculture, breeding of several fresh water and marine fish species and cryopreservation of fish sperms.
