Quantum computers are expected to outperform conventional computers in several important applications, from molecular simulation to search algorithms, once they can be scaled up to large numbers-typically millions-of quantum bits (qubits)1-3. For most solid-state qubit technologies-for example, those using superconducting circuits or semiconductor spins-scaling poses a considerable challenge because every additional qubit increases the heat generated, whereas the cooling power of dilution refrigerators is severely limited at their operating temperature (less than 100 millikelvin)4-6. Here we demonstrate the operation of a scalable silicon quantum processor unit cell comprising two qubits confined to quantum dots at about 1.5 kelvin. We achieve this by isolating the quantum dots from the electron reservoir, and then initializing and reading the qubits solely via tunnelling of electrons between the two quantum dots7-9. We coherently control the qubits using electrically driven spin resonance10,11 in isotopically enriched silicon12 28Si, attaining single-qubit gate fidelities of 98.6 per cent and a coherence time of 2 microseconds during 'hot' operation, comparable to those of spin qubits in natural silicon at millikelvin temperatures13-16. Furthermore, we show that the unit cell can be operated at magnetic fields as low as 0.1 tesla, corresponding to a qubit control frequency of 3.5 gigahertz, where the qubit energy is well below the thermal energy. The unit cell constitutes the core building block of a full-scale silicon quantum computer and satisfies layout constraints required by error-correction architectures8,17. Our work indicates that a spin-based quantum computer could be operated at increased temperatures in a simple pumped 4He system (which provides cooling power orders of magnitude higher than that of dilution refrigerators), thus potentially enabling the integration of classical control electronics with the qubit array18,19.

In the Mediterranean region, water scarcity has already prompted concern in the wine sector due to the strong impact it has on vineyard productivity and wine quality. Water footprint is an indicator that takes account of all the water involved in the creation of a product and may help producers to identify hotspots, and reduce water consumption and the corresponding production costs. In recent years several studies have been reported on wine water footprint determination, but mostly focused on the viticulture phase or assuming no grey water footprint at the winery since it has a treatment system. In the framework of the WineWaterFootprint project a medium-size winery was monitored, with direct measurements, regarding determination of the blue and grey components of water footprint. The determined winery water footprint ranged from 9.6 to 12.7 L of water per wine bottle of 0.75 L, the wastewater produced being responsible for about 98%, which means that the grey component cannot be disregarded. The developed scenarios show that a potential reduction of 87% in winery water footprint can be obtained with almost no investment. The challenge of reducing the grey footprint is not in technology development, but rather in the proper maintenance and monitoring of treatment systems.

Interest in life-cycle assessment (LCA) for foodstuffs has increased over recent years. In the same period, there could also be noticed an increased focus on food losses and waste (FLW); hence the need for guidance or a method that assesses the environmental impacts of FLW when analyzing agrifood systems with the use of LCA. This study introduces a method of assessing FLW in the context of supply chain LCA through: i) calculating the impacts of the food supply chain with the associated FLW, in order to account for the overall environmental impacts; and ii) assessing the benefits of FLW prevention strategies. After the method is presented, a practical application follows. The results of this study show that this method is able to aid LCA practitioners to include FLW on their LCA for food studies so as not to underestimate the impacts. Furthermore, the method is able to aid decision makers to assess the benefits of implementing a FLW prevention action in comparison to a baseline scenario. This method has some limitations: the attributional approach, lack of guidelines on how to estimate prevention potentials, as well as lack of guidelines to estimate additional impacts due to prevention actions.

Source separation systems for the management of domestic wastewater and food waste has been suggested as more sustainable sanitation systems for urban areas. The present study used an attributional life cycle assessment to investigate the carbon footprint and potential for nutrient recovery of two sanitation systems for a hypothetical urban area in Southern Sweden. The systems represented a typical Swedish conventional system and a possible source separation system with increased nutrient recovery. The assessment included the management chain from household collection, transport, treatment and final return of nutrients to agriculture or disposal of the residuals. The results for carbon footprint and nutrient recovery (phosphorus and nitrogen) concluded that the source separation system could increase nutrient recovery (0.30-0.38 kg P capita-1 year-1 and 3.10-3.28 kg N capita-1 year-1), while decreasing the carbon footprint (-24 to -58 kg CO2-eq. capita-1 year-1), compared to the conventional system. The nutrient recovery was increased by the use of struvite precipitation and ammonium stripping at the wastewater treatment plant. The carbon footprint decreased, mainly due to the increased biogas production, increased replacement of mineral fertilizer in agriculture and less emissions of nitrous oxide from wastewater treatment. In conclusion, the study showed that source separation systems could potentially be used to increase nutrient recovery from urban areas, while decreasing the climate impact.

Human epidermal growth factor receptor type 2 (c-erbB-2), an oncoprotein with potential prognostic marker and therapeutic use, is overexpressed in several human and animal tumours. But information regarding this molecule in feline tumours is scarce. This study aimed to assess the changes in the immunohistochemical expression of c-erbB-2 in feline endometrial adenocarcinomas (FEA) compared to normal endometrium. An immunohistochemistry assay using a specific antibody against c-erbB-2 was performed in FEA samples (n = 34) and in normal endometrium in the follicular (FS; n = 12) and luteal (LS; n = 11) stages. In FEA, the c-erbB-2 immunoexpression was assessed in neoplastic epithelial cells whilst in normal endometria it was individually evaluated in the surface and the superficial and deep glandular epithelia (SE, SGE and DGE, respectively). In FS and in LS, all the epithelia were positive for c-erbB-2; positivity was higher in the SE and the SGE than in DGE. Twenty of the 34 FEA samples (58.8%) were positive for c-erbB-2 immunolabelling. Nevertheless, its expression was higher in all the epithelia in the FS compared to FEA (p ≤ 0.0001) or the LS (p = 0.016). The results presented herein suggest that c-erbB-2 molecule is differently expressed in the feline endometrium through the oestrous cycle and though it may also be involved in feline endometrial carcinogenesis, a question remains unanswered on the importance of additional pathways of epithelial proliferation in the neoplastic changes in feline endometrium.

We report the dispersive readout of the spin state of a double quantum dot formed at the corner states of a silicon nanowire field-effect transistor. Two face-to-face top-gate electrodes allow us to independently tune the charge occupation of the quantum dot system down to the few-electron limit. We measure the charge stability of the double quantum dot in DC transport as well as dispersively via in situ gate-based radio frequency reflectometry, where one top-gate electrode is connected to a resonator. The latter removes the need for external charge sensors in quantum computing architectures and provides a compact way to readout the dispersive shift caused by changes in the quantum capacitance during inter-dot charge transitions. Here, we observe Pauli spin-blockade in the high-frequency response of the circuit at finite magnetic fields between singlet and triplet states. The blockade is lifted at higher magnetic fields when intra-dot triplet states become the ground state configuration. A line shape analysis of the dispersive phase shift reveals furthermore an intra-dot valley-orbit splitting Δvo of 145 µeV. Our results open up the possibility to operate compact complementary metal-oxide semiconductor (CMOS) technology as a singlet-triplet qubit and make split-gate silicon nanowire architectures an ideal candidate for the study of spin dynamics.

The anaerobic co-digestion process in a continuous stirred-tank reactor (CSTR) was carried out under mesophilic conditions (37 ± 0.2 °C). All the trials were performed at a hydraulic retention time (HRT) of 15 days and the AD reactor was daily fed with a mixture of sewage sludge (SS) and elephant grass hydrolyzate (EGH). In this study, three different trials were assessed, with different mixture proportions of SSSS and EGH: F0 (100:0,v/v), F1 (75:25, v/v) and F2 (50:50, v/v), during 90 days each trial, keeping the organic loading rate (OLR) in a range of 0.94-1.16 g VS L(-1) day(-1). The experimental results obtained showed that the soluble chemical oxygen demand (SCOD) removal efficiency was around 77% and 86% for trials F1 and F2, respectively. SS co-digestion with EGH enhanced methane yield, leading to an increment between 23% and 38%, in comparison with the reference scenario (F0).

Blue jack mackerel, Trachurus picturatus, is a carangid fish which constitutes an important commercial resource in the north-east Atlantic. Its metazoan parasite community from Portuguese mainland waters was investigated here for the first time. Nine parasite taxa were found, most of which are common parasites of Trachurus spp. The parasite community was broadly similar to that of the Atlantic horse mackerel, T. trachurus, from the same region, but two digenean species were detected in blue jack mackerel, Monascus filiformis and Tergestia sp., which did not occur in horse mackerel from this region. A comparison with the two previous studies of T. picturatus parasite communities shows that continental-shelf regions are characterized by higher prevalences of digenean trematodes and an absence of trypanorhynch cestodes, in contrast with oceanic regions.

Spatial distribution studies in pest management identify the locations where pest attacks on crops are most severe, enabling us to understand and predict the movement of such pests. Studies on the spatial distribution of two mite species, however, are rather scarce. The mites Polyphagotarsonemus latus and Tetranychus bastosi are the major pests affecting physic nut plantations (Jatropha curcas). Therefore, the objective of this study was to measure the spatial distributions of P. latus and T. bastosi in the physic nut plantations. Mite densities were monitored over 2 years in two different plantations. Sample locations were georeferenced. The experimental data were analyzed using geostatistical analyses. The total mite density was found to be higher when only one species was present (T. bastosi). When both the mite species were found in the same plantation, their peak densities occurred at different times. These mites, however, exhibited uniform spatial distribution when found at extreme densities (low or high). However, the mites showed an aggregated distribution in intermediate densities. Mite spatial distribution models were isotropic. Mite colonization commenced at the periphery of the areas under study, whereas the high-density patches extended until they reached 30 m in diameter. This has not been reported for J. curcas plants before.

Cyclooxygenase-2 (COX-2) is overexpressed in several human and animal neoplasms, including the human endometrial carcinoma. It has been suggested as a prognostic marker and a potential therapeutic target. This study aimed to (i) clarify histological aspects of feline endometrial adenocarcinomas (FEA) of the papillary serous type and (ii) characterize COX-2 immunohistochemical expression in normal, hyperplastic and neoplastic endometrium in this species. Archived paraffin-embedded tissue samples of 33 FEA, eight cystic endometrial hyperplasias (CEHs) and 21 samples of normal, healthy endometrium in the follicular (FS; n = 10) and luteal (LS; n = 11) stages were evaluated. Histological evaluation of haematoxylin and eosin-stained sections of the FEA revealed a papillary proliferation of neoplastic cells of serous type, accompanied by clear and multinucleated cells. Other architectural arrangements mainly included solid and tubular growth. Randomly distributed areas of necrosis within the tumours were commonly observed. Invasion of the myometrium, of the serosa and of the vascular and/or lymphatic vessels was not constant features. The mean number of mitoses was higher in FEA compared to non-neoplastic endometrium. COX-2 scores were lower in FEA (p = 0.003) and CEH (p = 0.05) when compared to normal epithelium (NE). The loss of the membrane apical reinforcement in epithelial cells was observed in FEA samples, which was accompanied by the dislocation of COX-2 labelling into the cytoplasm and the perinuclear area; in contrast, in epithelial cells in the healthy and hyperplastic endometria, the immunoreaction showed the characteristic pattern of apical membrane reinforcement, suggestive of the membrane polarization. COX-2 epithelial scores were higher in the FS than in the LS. No differences were found in stromal COX-2 expression between normal, CEH and FEA groups, but it was higher in the LS than in the FS. In summary, loss of COX-2 compartmentalization in neoplastic epithelial cells might be one of the molecular events underlying endometrial carcinogenesis.

Food production is challenged by changes in climate and land use and expanding human population growth. Proper pollination can increase the production and quality of fruit, nut, oil, and fiber crops. We reviewed crop dependence on pollinators and estimated the economic value of pollination per year for each crop in Brazil. We analyzed 141 crops and found that 85 depend on pollinators. Almost one-third of these crops have an essential or great dependence on pollinators. The economic contribution of pollinators totals ∼30% (∼US$12 billion) of the total annual agricultural income of the dependent crops (totalizing almost US$45 billion). However, half of these figures includes soybean crop (US$5.7 billion of pollinators' contribution and US$22 billion of annual income). Soybean was considered as having a modest dependence on pollinators, but this remains controversial because pollinator dependence might vary among different varieties cultivated in Brazil. Moreover, we have no information about pollinator dependence regarding some important crops, showing the urgent need for basic research into reproductive biology and pollination ecology.

Xylella fastidiosa is a xylem-limited, Gram-negative phytopathogen responsible for economically relevant crop diseases. Its genome was thus sequenced in an effort to characterize and understand its metabolism and pathogenic mechanisms. However, the assignment of the proper functions to the identified open reading frames (ORFs) of this pathogen was impaired due to a lack of sequence similarity in the databases. In the present work, we used small-angle X-ray scattering and in silico modeling approaches to characterize and assign a function to a predicted LysR-type transcriptional regulator in the X. fastidiosa (XfLysRL) genome. XfLysRL was predicted to be a homologue of BenM, which is a transcriptional regulator involved in the degradation pathway of aromatic compounds. Further functional assays confirmed the structural prediction because we observed that XfLysRL interacts with benzoate and cis,cis-muconic acid (also known as 2E,4E-hexa-2,4-dienedioic acid; hereafter named muconate), both of which are co-factors of BenM. In addition, we showed that the XfLysRL protein is differentially expressed during the different stages of X. fastidiosa biofilm formation and planktonic cell growth, which indicates that its expression responds to a cellular signal that is likely related to the aromatic compound degradation pathway. The assignment of the proper function to a protein is a key step toward understanding the cellular metabolic pathways and pathogenic mechanisms. In the context of X. fastidiosa, the characterization of the predicted ORFs may lead to a better understanding of the cellular pathways that are linked to its bacterial pathogenicity.

A study was carried out on the gastrointestinal helminth communities of blackspot seabream, Pagellus bogaraveo, from the Portuguese mainland and island waters. Fifteen helminth parasite species were detected, including the following new records in this host: the digeneans Accacladocoelium petasiporum, Brachyenteron helicoleni, Glomericirrus macrouri, Lecithocladium excisum, Lepocreadium album and Pachycreadium carnosum; the nematode Camallanus sp. and the acanthocephalan Rhadinorhynchus pristis. Infection levels were generally low. Samples from mainland waters had a significantly higher prevalence of digeneans than those from the islands, probably due to the greater availability of invertebrate intermediate hosts on the continental slope. The helminth community of P. bogaraveo from Azores was diverse but mostly composed of species that infected no more than a single host, whereas in Madeira the helminth community was very depauperate. Most helminth species exhibited niche preferences within the gastrointestinal tract of the host. Higher functional richness and evenness were obtained in helminth communities of P. bogaraveo from mainland waters, indicating a balanced use of available resources, which is suggestive of more stable gastrointestinal helminth communities in these waters.

OBJECTIVE: Susceptibility to and severity of periodontal disease is influenced by gene polymorphisms related to the immune response. Co-stimulatory molecules, such as CD28 and CTLA-4, are critical in the development of such responses. Our hypothesis is that polymorphisms in genes that code for these molecules may be associated with periodontitis. The aim of the study was to investigate the association between +17 (T/C) CD28 and +49 (A/G) CTLA-4 gene polymorphisms and periodontitis in Brazilians. MATERIALS AND METHODS: Genomic DNA was obtained from oral swabs of 424 individuals categorized into three groups (control group, aggressive, and chronic periodontitis) considering clinical parameters such as probing depth and clinical attachment loss. The genotyping was performed by polymerase chain reaction-restriction fragment length polymorphism. RESULTS: There was an association between the T(-) genotype of the CD28 polymorphism and aggressive periodontitis (P = 0.04). Moreover, the A(+) genotype for CTLA-4 was associated with greater clinical attachment loss in non-smokers with aggressive periodontitis (P = 0.006, OR = 16.25, CI = 2.25-117.11). CONCLUSIONS: These findings show that T(-) in CD28 + 17 (T/C) and the A(+) in CTLA-4 +49 (A/G) genotypes are associated with susceptibility to aggressive periodontal disease. Thus, our study highlights these polymorphisms as potential genetic susceptibility markers of periodontitis in Brazilians.

Uterine neoplasms of epithelial origin are rare in cats and most often are described in older females. Yet, in less than 2 years, four ovariohysterectomy specimens were submitted from different practices to the Laboratory of Histology and Anatomical Pathology, at UTAD (Vila Real, Portugal), that emitted a diagnosis of feline endometrial adenocarcinoma. Untypically, all the females were aged <1 year old at the surgery. Access to the clinical files was requested to document the clinical features of the four cases, including any complementary data available, to construct the present case reports. The clinical situation developed with discrete signs, but vulvar discharge was present in three cases, ranging from bloody to brownish or colourless, and from purulent to mucous. The females were in dioestrus, although the oestrus remained unperceived in most cases. In this study, the four clinical situations are described and discussed on the basis of available literature, highlighting the aspects that may impair an early diagnosis and that may favour the progression of the disease and also that age should not be an excluding criteria when analysing the differential diagnosis list.

We devise a platform for noise-resistant quantum computing using the valley degree of freedom of Si quantum dots. The qubit is encoded in two polarized (1,1) spin-triplet states with different valley compositions in a double quantum dot, with a Zeeman field enabling unambiguous initialization. A top gate gives a difference in the valley splitting between the dots, allowing controllable interdot tunneling between opposite valley eigenstates, which enables one-qubit rotations. Two-qubit operations rely on a stripline resonator, and readout on charge sensing. Sensitivity to charge and spin fluctuations is determined by intervalley processes and is greatly reduced as compared to conventional spin and charge qubits. BACKGROUND: The aim of this work was to evaluate the interactions between aminoglycosides and the ethyl-acetate fraction of the fern Lygodium venustum SW (EAFLV) METHODS: The ethyl-acetate fraction was obtained from the ethanol extract of L. venustum and was assayed via the checkerboard method associated with aminoglycosides against two bacterial strains multiresistant to antibiotics. RESULTS: The antibiotic activity of all drugs, when associated with the ethyl-acetate fraction, was enhanced in an additive manner, except for the association between EAFLV and amikacin, which showed a synergistic interaction against the Escherichia coli strain. CONCLUSIONS: The results indicated that L. venustum can be a source of secondary metabolites to be used in association with antibiotics like aminoglycosides in antibiotic chemotherapy against resistant bacteria.

A synopsis of the species of Chloromyxum Mingazinni, 1890 (Myxozoa: Myxosporea: Chloromyxidae) is presented, including 140 nominal species. For each species the most relevant morphological and morphometric characteristics are indicated. Included are data on the site of infection within the host, the original host and the host locality, plus a full bibliography of the original records for these species. A diagrammatic illustration of a spore of each species is also provided.

The OxyR oxidative stress transcriptional regulator is a DNA-binding protein that belongs to the LysR-type transcriptional regulators (LTTR) family. It has the ability to sense oxidative species inside the cell and to trigger the cell's response, activating the transcription of genes involved in scavenging oxidative species. In the present study, we have overexpressed, purified and characterized the predicted OxyR homologue (orf xf1273) of the phytopathogen Xylella fastidiosa. This bacterium is the causal agent of citrus variegated chlorosis (CVC) disease caused by the 9a5c strain, resulting in economic and social losses. The secondary structure of the recombinant protein was analyzed by circular dichroism. Gel filtration showed that XfoxyR is a dimer in solution. Gel shift assays indicated that it does bind to its own predicted promoter under in vitro conditions. However, considering our control experiment we cannot state that this interaction occurs in vivo. Functional complementation assays indicated that xfoxyR is able to restore the oxidative stress response in an oxyr knockout Escherichia coli strain. These results show that the predicted orfxf1273 codes for a transcriptional regulator, homologous to E. coli OxyR, involved in the oxidative stress response. This may be important for X. fastidiosa to overcome the defense mechanisms of its host during the infection and colonization processes.

Canine renal cell carcinomas (RCCs) are uncommon aggressive tumors that occur mainly in middle-aged male dogs. Their histologic classification bears no relationship with prognosis, and little information is available concerning their immunohistochemical properties. In this retrospective study, formalin-fixed, paraffin-embedded tissues from 13 canine RCCs were retrieved from the archives, classified histologically, and evaluated immunohistochemically. The dogs were 7 males and 6 females (1 spayed) of 10 different breeds, averaging 8 years in age. The tumors were classified as papillary, tubulopapillary, papillary-cystic, solid, or sarcomatoid. All 13 tumors were immunohistochemically positive for uromodulin, 12 for c-KIT, 11 for vimentin, 9 for wide-spectrum-screening cytokeratins, 7 for cytokeratins AE1/AE3 and carcinoembryonic antigen, 4 for cytokeratins CAM 5.2, and 3 for CD10. All 3 solid RCCs expressed vimentin, c-KIT, and carcinoembryonic antigen and were negative for cytokeratins. All 7 papillary and tubulopapillary tumors expressed vimentin; 6 (86%), cytokeratins; and 6 (86%), c-KIT. Both papillary-cystic RCCs were positive for cytokeratins and c-KIT and negative for vimentin. These results indicate that the different histologic types of RCC have characteristic immunohistochemical profiles and that c-KIT may be involved in the pathogenesis of canine RCC.
