BACKGROUND: The Kleihauer-Betke (KB) test allows the detection of fetal red blood cells (containing fetal hemoglobin, HbF) in the maternal blood to identify and quantify potential fetal-maternal hemorrhages. In certain cases, detecting fetal red blood cells with conventional staining is difficult. False-positive results or overestimation of the quantity of fetal red blood cells may occur in cases of maternal hemoglobinopathy. In this study, we developed a new staining protocol to facilitate the reading of difficult smears and improve the precision of the quantification of fetal red blood cells; we also analyzed the performance of this new method. This study assessed blood samples with and without hemoglobin abnormalities, which present difficulties when interpreting the KB test. METHODS: The new staining formula is based on an improved elution technique and the use of a different stain instead of hematoxylin. To test this staining method, 16 samples from patients with abnormal hemoglobin electrophoresis and 14 samples from patients with normal hemoglobin electrophoresis were analyzed using the KB test with the classical staining method and the new staining method. In addition, a second series was prepared using the same samples spiked with fetal red blood cells from newborn blood, to compare the accuracy of the two methods in identifying fetal red blood cells. RESULTS: In the 60 slides analyzed with both staining methods, we found that the new technique improved the accuracy from 78 to 85%; lowered the coefficient of variation between the operators, which decreased from 20.7% to 12.7%; increased the specificity in our population from 56 to 70%; and decreased the number of false-positive cases by 30%. CONCLUSIONS: We successfully developed a new staining technique that facilitates the reading of difficult slides and improves the specificity of the detection of fetal red blood cells. This technique is recommended as a secondary method to use before sending the sample for additional exploration.

One of the most important aspects of the detection of antioxidant compounds is developing a fast screening method. The screening of the overall relative antioxidant capacity (RAC) of several Romanian hydrosoluble plant extracts is the focus of this work. This is important because of the presence of increasing levels of reactive oxygen species (such as H2O2) generates oxidative stress in the human body. The consequences are a large number of medical conditions that can be helped by a larger consumption of plant extracts as food supplements, which do not necessarily contain the specified antioxidant contents. By exploiting the catalytic properties of gold nanoparticles, a specific and sensitive nanoparticle-based label-free electrochemical sensor was developed, where the working parameters were optimized for RAC screening of hydrosoluble plant extracts. First, electrochemical measurements (cyclic voltammetry and amperometry) were used to characterize different nanoparticle-based sensors, revealing the best performance of gold nanoparticle-based sensors, obtaining a RAC of 98% for lavender extracts. The sensing principle is based on the quenching effect of antioxidants for H2O2 amperometric detection, where the decrease in electrical signal suggests an increasing antioxidant capacity. The obtained results were expressed in terms of ascorbic acid and Trolox equivalents in order to be able to correlate our results with classical methods like chemiluminescence and UV-Vis spectrophotometry, where a correlation coefficient of 0.907 was achieved, suggesting a good correlation between electrochemistry and spectrophotometry. Considering these results, the optimized gold nanoparticle-based label-free sensor can be used as a simple, rapid alternative towards classical methods for relative antioxidant capacity detection of hydrosoluble plant extracts.

Acute myeloid leukemias (AMLs) are hematologic malignancies with varied molecular and immunophenotypic profiles, making them difficult to diagnose and classify. High-dimensional analysis algorithms might increase the utility of multicolor flow cytometry for AML diagnosis and follow-up. The objective of the present study was to assess whether a Compass database-guided analysis can be used to achieve rapid and accurate diagnoses. We conducted this study to determine whether this method could be employed to pilote the genetic and molecular tests and to objectively identify different-from-normal (DfN) patterns to improve measurable residual disease follow-up in AML. Three Compass databases were built using Infinicyt 2.0 software, including normal myeloid-committed hematopoietic precursors (n = 20) and AML blasts harboring the most frequent recurrent genetic abnormalities (n = 50). The diagnostic accuracy of the Compass database-guided analysis was evaluated in a prospective validation study (125 suspected AML patients). This method excluded AML associated with the following genetic abnormalities: t(8;21), t(15;17), inv(16), and KMT2A translocation, with 92% sensitivity [95% confidence interval (CI): 78.6%-98.3%] and a 98.5% negative predictive value (95% CI: 90.6%-99.8%). Our data showed that the Compass database-guided analysis could identify phenotypic differences between AML groups, representing a useful tool for the identification of DfN patterns.

The antioxidant properties of different plant extracts are usually claimed and used by food, medicine and cosmetic industry due to their health promoting capacities. In this study the presence of antioxidant compounds and the total antioxidant capacity of water-soluble extracts, prepared using two extraction methods and a variety of solvents, have been determined and a rapid screening method has been developed. Plant extracts characterisation, composition and antioxidant properties were confirmed by FTIR and Raman spectroscopies. Voltammetry, amperometry and electrochemical impedance were used to highlight the total antioxidant capacity of each extract using an electrochemical gold nanozyme-sensor based on the enzyme-like catalytic activity of gold nanoparticles. Both anodic area of cyclic voltammograms (10.31 µA V) and electrochemical index (153) calculated using differential potential voltammetry show the total content of antioxidant compounds, allowing to discriminate between different extracts. Amperometric total antioxidant capacity measurements were associated with those from classical chemiluminescence and good correlation has been found (Pearson's correlation coefficient of 0.958).

The aim of this study was to analyze the bone mineral content (BMC) of the skeleton and the body composition in lean and fat masses in a population of young overweight subjects, before diet or medical intervention. A total of 496 white youths (298 females, 198 males) were studied. Their body mass index (BMI) values were 29.2+/-5.5 kg/m2 in females and 28.3+/-5.1 kg/m2 in males, corresponding to relative weights (wts) [actual wt, W, divided by the mean wt normal for height (ht), Wi] of 1.55+/-0.24 and 1.53+/-0.24, respectively. Whole-body dual-energy x-ray absorptiometry (DXA) scans were performed (Norland XR36 DXA system) to obtain the total BMC (BMCt), lean tissue mass (LTMt) and fat mass (FMt), as well as the values of the corresponding parameters in arms (a), legs (l), and abdomen (ab). The measured data were compared with theoretical values calculated for Wi. The differences with reference values in LTMt and FMt were sex dependent, and the accretion in FMt, greater in males than in females, yielded a similar FMt/LTMt ratio in both sexes. The wt corresponding to the BMC of the bearing skeleton, lower than the actual wt by about 20%, seemed a reasonable first step to consider in the wt management of these youths.
