BACKGROUND: Saliva is supersaturated with respect to calcium and phosphate ions. Salivary ions may well play a role in the subsequent adsorption of proteins and consequently in the formation of the acquired enamel pellicle. Among several biological functions, the enamel pellicle forms a selectively permeable barrier that regulates demineralization processes. AIM: The aim of this study was to evaluate the importance of salivary proteins when adsorbed on enamel surface and the resultant protective effect against demineralization without the presence of salivary ions. METHODS: Enamel surfaces were coated with whole saliva, parotid saliva, dialyzed whole saliva or dialyzed parotid saliva (molecular weight cutoff 1 kDa). Adsorption was allowed to proceed for a period of 2 h. Enamel specimens were then washed with deionized water and immersed into a demineralization solution of pH 4.5 for 12 days. This solution was used to measure the amount of calcium and phosphate released from enamel specimens after the demineralization period. RESULTS: All coated specimen groups showed a significantly higher protection than those not coated with any type of saliva. In addition, undialyzed saliva (whole saliva and parotid saliva) was more effective in protecting the enamel against demineralization than dialyzed saliva. CONCLUSION: The present investigation indicates that the ionic composition of saliva can amplify the demineralization protection effect by reducing acid-induced enamel demineralization. Moreover, a protective effect of salivary proteins without presence of ions was demonstrated in this study.

The dental profession has endured unprecedented disruption amid COVID-19. Novel stressors have included a high risk of occupational exposure to COVID-19, financial losses, and stricter infection prevention and control requirements. The present study investigated the longitudinal impact of COVID-19 on the stress and anxiety levels of a cohort of Canadian dentists (N = 222) between September 2020 and October 2021. Salivary cortisol was selected as a biomarker of mental stress, and 10 sets of monthly saliva samples (2,131 in total) were self-collected, sent to our laboratory in prepaid courier envelopes, and analyzed by enzyme-linked immunosorbent assay. To assess COVID-19 anxiety, 9 monthly online questionnaires were administered, comprising a general COVID-19 anxiety instrument and 3 items regarding the impact of dentistry-related factors. Bayesian log-normal mixed effect models were fitted to estimate the longitudinal trajectory of salivary cortisol levels and their association with the disease burden of COVID-19 in Canada. After accounting for age, sex, vaccination status, and the diurnal rhythm of cortisol secretion, a modest positive association was found between dentists' salivary cortisol levels and the count of COVID-19 cases in Canada (96% posterior probability). Similarly, the self-reported impact of dentistry-related factors, such as fear of getting COVID-19 from a patient or coworker, was greatest during peaks of COVID-19 waves in Canada; however, general COVID-19 anxiety decreased consistently throughout the study period. Interestingly, at all collection points, the majority of participants were not concerned about personal protective equipment. Overall, participants reported relatively low rates of psychological distress symptoms in relation to COVID-19, a result that should be reassuring for the dental community. Our findings strongly suggest a link between self-reported and biochemical measurements of stress and anxiety in Canadian dentists during the COVID-19 pandemic.

With the aim of improving phototherapy for neonatal jaundice (hyperbilirubinemia), this study investigates the degradation of unconjugated bilirubin under irradiance by conventional light and by white, red, green, and blue LED sources in vitro. The absorption spectra of bilirubin under these different light sources are compared. The results demonstrate that white LED phototherapy promotes more efficient bilirubin degradation than conventional blue-light therapy. This study provides a basis for the design of novel phototherapy devices for the treatment of hyperbilirubinemia.

AIMS/HYPOTHESIS: The role of TNF-alpha in impaired wound healing in diabetes was examined by focusing on fibroblasts. METHODS: Small excisional wounds were created in the db/db mice model of type 2 diabetes and normoglycaemic littermates, and in a streptozotocin-induced type 1 diabetes mouse model and control mice. Fibroblast apoptosis was measured by the TUNEL assay, proliferation by detection of proliferating cell nuclear antigen, and forkhead box O1 (FOXO1) activity by DNA binding and nuclear translocation. TNF-alpha was specifically inhibited by pegsunercept. RESULTS: Diabetic wounds had increased TNF-alpha, fibroblast apoptosis, caspase-3/7 activity and activation of the pro-apoptotic transcription factor FOXO1, and decreased proliferating cell nuclear antigen positive fibroblasts (p < 0.05). TNF-alpha inhibition improved healing in the diabetic mice and increased fibroblast density. This may be explained by a decrease in fibroblast apoptosis and increased proliferation when TNF-alpha was blocked (p < 0.05). Although decreased fibroblast proliferation and enhanced FOXO1 activity were investigated in type 2 diabetes, they may also be implicated in type 1 diabetes. In vitro, TNF-alpha enhanced mRNA levels of gene sets related to apoptosis and Akt and p53 but not mitochondrial or cell-cycle pathways. FOXO1 small interfering RNA reduced gene sets that regulate apoptosis, Akt, mitochondrial and cell-cycle pathways. TNF-alpha also increased genes involved in inflammation, cytokine, Toll-like receptor and nuclear factor-kB pathways, which were significantly reduced by FOXO1 knockdown. CONCLUSIONS/INTERPRETATION: These studies indicate that TNF-alpha dysregulation in diabetic wounds impairs healing, which may involve enhanced fibroblast apoptosis and decreased proliferation. In vitro, TNF-alpha induced gene sets through FOXO1 that regulate a number of pathways that could influence inflammation and apoptosis.

PURPOSE: This study investigated whether the report of mothers about the amount of dentifrice applied on a toothbrush could be a reliable method to estimate the inadvertent use of dentifrices. METHODS: A cross-sectional study was conducted in a randomly selected sample of 334 mothers of children enrolled in public day care centres of a city in the Brazilian northeast. Initially, the mothers chose the figure corresponding to the amount of dentifrice used during their children's daily toothbrushing. Then, they simulated real-life conditions with the toothbrush routinely used to brush the children's teeth. The amount of dentifrice on toothbrush was weighted using an analytical scale. Data were analysed by the Kruskal-Wallis followed by the Dunn test (α = 0.05). RESULTS: The weight of dentifrice on the toothbrush was high (> 0.3 g), mainly considering the categories 'covered only' and 'completely covered' that showed no statistical difference (p > 0.05). The proportion of agreement between the mothers' reports and the weight of dentifrice on the toothbrush was 0.23 (CI 0.18-0.28). CONCLUSION: The mothers' report regarding the amount of dentifrice on the toothbrush by demonstrative figures was not a reliable method to estimate the inadvertent ingestion of fluoride from dentifrices.

Predatory mites that belong to the Phytoseiidae family are one of the main natural enemies of phytophagous mites, thus allowing for their use as a biological control. Phytoseiulus macropilis (Banks, 1904) (Acari: Phytoseiidae) is among the main species of predatory mites used for this purpose. Tetranychus urticae Koch, 1836 (Acari: Tetranychidae) is considered to be one of the most important species of mite pests and has been described as attacking over 1,100 species of plants in 140 families with economic value. The objective of the present study was to investigate, in the laboratory, the reproductive parameters of the predatory mite P. macropilis when fed T. urticae. Experiments were conducted under laboratory conditions at 25 ± 2 °C of temperature, 70 ± 10% RH and 14 hours of photophase. In addition, biological aspects were evaluated and a fertility life table was established. The results of these experiments demonstrated that the longevity of adult female was 27.5 days and adult male was 29.0 days. The population was estimated to increase approximately 27 times (Ro) in mean generation time (T), which was 17.7 days. Lastly, the mite population grew 1.2 times/day (λ) and doubled every 3.7 days (TD).

Resumo Os ácaros predadores pertencentes à família Phytoseiidae constituem um dos principais inimigos naturais de ácaros fitófagos, o que possibilita o seu uso em controle biológico. Entre as principais espécies destaca-se Phytoseiulus macropilis (Banks, 1904) (Acari: Phytoseiidae). O ácaro Tetranychus urticae Koch, 1836 (Acari: Tetranychidae) é considerado uma das espécies de ácaros-praga mais importantes, atacando mais de 1.100 espécies de plantas em 140 famílias de valor econômico. Este trabalho foi realizado com o objetivo de estudar, em laboratório, os parâmetros reprodutivos do ácaro predador P. macropilis, quando alimentado com T. urticae. Os estudos foram conduzidos em laboratório à temperatura de 25 ± 2 °C, 70 ± 10% de UR e 14 horas de fotofase e foram avaliados os aspectos biológicos e confeccionada a tabela de vida de fertilidade. Foi constatada uma longevidade de 27,5 dias para fêmeas adultas e 29,0 dias para machos. A população foi estimada em aumentar aproximadamente 27 vezes (Ro) no período médio de duração de geração (T) que foi de 17,7 dias. A população do ácaro cresceu 1,2vezes/dia (λ) e dobrou a cada 3,7 dias (TD).

OBJECTIVE: To focus attention on a rare pathology of the childhood which presents premature aging of the skin and can be lethal. METHODS: The authors present a case of cutis laxa, syndrome of premature aging, in an eight year-old child, and discuss the classification, diagnosis and prognosis of the disease. RESULTS: The child presented signs of premature aging when he was four years-old. The diagnosis of cutis laxa was confirmed by skin biopsy. The patient presented heart failure, a systemic complication different from those previously described, and died at eight years of age. CONCLUSIONS: The importance of the diagnosis of cutis laxa resides in the fact that besides characteristic dermatological findings, there are frequent systemic complications that can be the focus of preventive measures, since there is no specific treatment for this disease. Genetic counseling is another important issue in this condition.
