RESUMO

Serosurvey results for Crimean-Congo hemorrhagic fever virus antibodies in dromedary camels in Algeria indicate that the pathogen is circulating endemically in desertic areas, despite the hostile environment. Thus, dromedaries are suitable sentinels for detecting human risk for Crimean-Congo hemorrhagic fever in desertic areas.

This study was designed to investigate, at a laboratory scale, the possibility of valorizing the leftover carob fruits to produce the eco-friendly biopolymer polyhydroxybutyrate (PHB) by using the bacterial strain Bacillus paramycoides, which has been isolated from the botanical garden of Skikda University in Algeria. The PHB production was tested under various conditions: a pH of 3-8, temperature range of 30-44 °C, carob extracted molasses concentration of 2-8% v/v, an incubation time of 24-96 h and an agitation speed of 150-300 rpm. The effects of different nitrogen sources and carob extracted molasses treatment types were also investigated. The PHB concentration was determined quantitatively as crotonic acid by measuring the absorbance at 300 nm. Cell growth was quantified by measuring the density of the culture at 600 nm. The presence of PHB was confirmed by applying high-performance liquid chromatography (HPLC) using an Aminex HPX-87H and implementing gas chromatography analysis. The best yield of PHB synthesis was obtained by using 6% v/v of 5 M H2SO4 treated with carob molasses as a carbon source, with peptone as a nitrogen source; incubation was conducted at 37 °C for 96 h at an agitation speed of 300 rpm (114.95 mg/L). The HPLC analysis confirmed the synthesis of PHB by B. paramycoides to have a chromatogram retention time of 22.5 min. Carob waste was successfully valorized to PHB.

Vector-borne diseases represent a real threats worldwide, in reason of the lack of vaccine and cure for some diseases. Among arthropod vectors, mosquitoes are described to be the most dangerous animal on earth, resulting in an estimated 725,000 deaths per year due to their borne diseases. Geographical position of Algeria makes this country a high risk area for emerging and re-emerging diseases, such as dengue coming from north (Europe) and malaria from south (Africa). To prevent these threats, rapid and continuous surveillance of mosquito vectors is essential. For this purpose we aimed in this study to create a mosquito vectors locale database using MALDI-TOF mass spectrometry technology for rapid identification of these arthropods. This methodology was validated by testing 211 mosquitoes, including four species (Aedes albopictus, Culex pipiens, Culex quinquefasciatus, and Culiseta longiareolata), in two northern wilayahs of Algeria (Algiers and Bejaia). Species determination by MALDI TOF MS was highly concordant with reference phenotypic and genetic methods. Using this MALDI-TOF MS tool will allow better surveillance of mosquito species able to transmit mosquito borne diseases in Algeria.

Vitamin E is a potent molecule, especially when loaded in cyclodextrin, in modulating oxidative stress during the freeze-thawing process. The present study aimed to investigate the effect of different concentrations of cyclodextrin-vitamin E complex (CD-Vit E) on cryopreserved ram sperm. Ejaculates collected from five adult rams were pooled and divided into four aliquots. All aliquots were treated in Tris-extender (Tris-glucose-citric acid) containing 2 mg cholesterol-loaded methyl-ß- cyclodextrin/120 × 106 spermatozoa and either 0 (Control), 2, 4 or 6 mg CD-Vit E/120 × 106 spermatozoa, corresponding to 0, 0.5, 1 or 1.5 of pure vitamin E, respectively. After incubation at 22 °C for 15 min and the addition of Tris-extender containing glycerol and egg yolk (v/v), all aliquots were frozen in liquid nitrogen. After thawing, motility (computer aided sperm analysis), viability (eosin staining), membrane integrity (HOST), acrosome integrity (Coomassie G-250 staining) and lipid peroxidation (Thiobarbituric acid assay) were evaluated. Compared to control, 2 mg CD-Vit E had a significant positive effect on total motility, progressive motility, movement linearity (LIN%), viability and lipid peroxidation. At 4 mg, however, CD-Vit E had a significant negative effect on total motility, progressive motility, membrane functionality and acrosome integrity. At a greater concentration (6 mg), the negative effects were greater as compared with inclusion of 4 mg in the cryoprotectant and the percentage of rapidly and moderately motile gametes and viability were also altered. In conclusion, the effect of CD-Vit E on cryopreserved ram sperm was concentration-dependent with the 2 mg amount having a beneficial effect while greater concentrations (4 and 6 mg) had a harmful effect on sperm motility and gamete integrity but without affecting oxidative stress status.
