RESUMO

Silica-based nanoparticles have been developed as powerful platforms for drug delivery and might also prevent undesired side effects of drugs. Here, a fast method to synthesize positively charged mesoporous silica nanoparticles (ζ = 20 ± 0.5 mV, surface area = 678 m2 g-1, and 2.3 nm of porous size) was reported. This nanomaterial was employed to anchor sodium nitroprusside (SNP), a vasodilator drug with undesired cyanide release. A remarkable incorporation of 323.9 ± 7.55 µmol of SNP per gram of nanoparticle was achieved, and a series of studies of NO release were conducted, showing efficient release of NO along with major cyanide retention (ca. 64% bound to nanoparticle). Biological assays with mammalian cells showed only a slight drop in cell viability (13%) at the highest concentration (1000 µM), while SNP exhibited an LC50 of 228 µM. Moreover, pharmacological studies demonstrated similar efficacy for vasodilation and sGC-PKG-VASP pathway activation when compared to SNP alone. Altogether, this new SNP silica nanoparticle has great potential as an alternative for wider and safer use of SNP in medicine with lower cyanide toxicity.

Congenital malformations are a highly diverse group of conditions reported in both humans and animals, characterized by defects in morphogenesis observed at birth. Although most cases are idiopathic, genetic and environmental factors may be involved. The frequency of such conditions varies with species, geographic regions, and the specific malformation involved. In polymelia, supernumerary limbs are attached to different parts of the body. Gastrointestinal duplications are described less frequently and can be associated with polymelia. Cloacal atresia is among the least-reported malformations in avian species, described only once in a kiwi. Here we describe a case with these 3 malformations in a single broiler chick (Gallus gallus domesticus) and provide a literature review about the occurrence of these malformations in birds. The 3-d-old chick also had an unidentified structure projecting from the pygostyle region. We performed clinical, radiographic, and postmortem examinations. The intestinal duplication was identified only during the postmortem evaluation. Detailed descriptions of avian congenital malformations are scarce. Although similar cases have been reported, we retrieved no cases of concurrent polymelia, intestinal duplication, and cloacal atresia in broiler chickens in our literature search, suggesting that the simultaneous occurrence of these conditions has not been reported previously in this species.

Elucidating the role of cyanobacteria in the biotransformation of arsenic (As) oxyanions is crucial to understand the biogeochemical cycle of this element and indicate species with potential for its bioremediation. In this study, we determined the EC50 for As(III) and As(V) and evaluated the biotransformation of As by Synechococcus sp. through high-performance liquid chromatography hyphenated to inductively coupled plasma mass spectrometry (HPLC-ICP-MS) and X-ray absorption fine structure spectroscopy (XAFS). Synechococcus sp. exhibited higher sensitivity to As(III) with an EC(50, 96 h) of 6.64 mg L(-1) that was approximately 400-fold lower than that for As(V). Even though the cells were exposed to concentrations of As(III) (6 mg L(-1)) approximately 67-fold lower than those of As(V) (400 mg L(-1)), similar intracellular concentrations of As (60.0 µg g(-1)) were observed after 30 days. As(V) was the predominant intracellular As species followed by As(III). Furthermore, organic As species such as monomethylarsonic acid (MMA) and dimethylarsinic acid (DMA) were observed in higher proportions after exposure to As(III). The differential toxicity among As oxyanions indicates that determining the redox state of As in the environment is fundamental to estimate toxicity risks to aquatic organisms. Synechococcus sp. demonstrated potential for its application in bioremediation due to the high accumulation of As and production of As organic compounds notably after exposure to As(III).
