RESUMO

High-entropy oxides (HEOs), which contain five or more metal cations that are generally thought to be randomly mixed in a crystalline oxide lattice, can exhibit unique and enhanced properties, including improved catalytic performance, due to synergistic effects. Here, we show that band gap narrowing emerges in a high-entropy aluminate spinel oxide, (Fe0.2Co0.2Ni0.2Cu0.2Zn0.2)Al2O4 (A5Al2O4). The 0.9 eV band gap of A5Al2O4 is narrower than the band gaps of all parent spinel oxides. First-principles calculations for multicomponent AAl2O4 spinels indicate that the band gap narrowing arises from the broadening of the energy distribution of the 3d states due to variations in the electronegativities and crystal field splitting across the 3d transition-metal series. As a catalyst for the oxygen evolution reaction in an alkaline electrolyte, A5Al2O4 reaches a current density of 10 mA/cm2 at an overpotential of 400 mV, outperforming all of the single-metal end members at an applied potential of 1.7 V vs RHE. Catalyst deactivation occurs after 5 h at 10 mA/cm2 and is attributed, based on elemental analysis and grazing-incidence X-ray diffraction, to the formation of a passivating layer that blocks the high-entropy oxide surface. This result helps to validate that the HEO is the active catalyst. The observation of band gap narrowing in A5Al2O4 expands the scope of synergistic properties exhibited by high-entropy materials and offers insight into the question of how the electronic structure of multicomponent oxide materials can be engineered via a high-entropy approach to achieve enhanced catalytic properties.

We present a new design of a robust cavity-enhanced frequency comb-based spectrometer operating under the continuous-filtering Vernier principle. The spectrometer is based on a compact femtosecond Er-doped fiber laser, a medium finesse cavity, a diffraction grating, a custom-made moving aperture, and two photodetectors. The new design removes the requirement for high-bandwidth active stabilization present in the previous implementations of the technique, and allows scan rates up to 100 Hz. We demonstrate the spectrometer performance over a wide spectral range by detecting CO2 around 1575 nm (1.7 THz bandwidth and 6 GHz resolution) and CH4 around 1650 nm (2.7 THz bandwidth and 13 GHz resolution). We achieve absorption sensitivity of 5 × 10-9 cm-1 Hz-1/2 at 1575 nm, and 1 × 10-7 cm-1 Hz-1/2 cm-1 at 1650 nm. We discuss the influence of the scanning speed above the adiabatic limit on the amplitude of the absorption signal.

Therapeutic options for the treatment of leishmaniasis are insufficient and need improvements owing to their low efficiency and high toxicity as well as the emergence of resistant strains. The limited number of new drugs for neglected diseases and lack of innovation in your development are still challenges. In this context, the process of discovery and development of biological assays play a pivotal role for the identification of bioactive compounds. The assays currently used for screening of drugs with cytotoxic activity against Leishmania parasites, include different processes that utilize intact parasite (free or intracellular) or specific enzymes of metabolism as a target cell. These assays allow the screening of large numbers of samples followed by more detailed secondary confirmatory assays to confirm the observed activity and assess their toxicity. In the present study, we described the development of a new functional and more complete assay that enables simultaneous assessment of potential anti-Leishmania compounds through evaluation of internalization of fluorescein-labeled L. braziliensis promastigotes by human peripheral blood monocytes and their cytotoxicity by flow cytometry. We standardized the conditions for parasite labeling to achieve better phagocytosis analysis by setting the ratio of number of parasites per cell as 1 to 2, at incubation time of 6h. The cytotoxicity assessment was performed by the quantification of cells undergoing early/late apoptosis and necrosis using a double labelling platform employing 7AAD for late apoptosis and necrosis analysis and Annexin-V for early apoptosis evaluation. Hemolysis analysis was an additional parameter to test cytotoxicity. Two drugs used on clinic (Amphotericin B and Glucantime®) were used to validate the proposed methodology, and the assay was able to detect their known leishmanicidal activity and immunotoxicity properties. This new predictive assay will contribute to the development of translational medicine strategies in drug discovery for neglected diseases such as leishmaniasis.

We use broadband near-infrared continuous-filtering Vernier spectroscopy (CF-VS) for time-resolved detection of H2O and OH radical in a premixed CH4/air flat flame. The CF-VS spectrometer is based on a femtosecond Er:fiber laser, an external cavity that contains the flame, and a detection system comprising a rotating diffraction grating and photodetectors. Spectra of H2O and OH radical around 1570 nm are continuously recorded with 6.6 GHz spectral resolution, 4.0 × 10-7 cm-1 absorption sensitivity, and 25 ms time resolution, while the fuel-air equivalence ratio is periodically modulated with a square wave. The concentrations of the two analytes are retrieved with percent level precision by a fit of a Vernier model to each spectrum spanning 13 nm. The temporal profiles of both concentrations in each modulation cycle are repeatable and the steady-state concentration levels are in good agreement with predictions based on one-dimensional simulations of a static flat flame. The robust CF-VS spectrometer opens up for quantitative monitoring of multiple products of time-varying combustion processes with relatively simple data acquisition procedures.

We report a photoacoustic spectroscopy setup with a high-power mid-infrared frequency comb as the light source. The setup is used in broadband spectroscopy of radiocarbon methane. Owing to the high sensitivity of a cantilever-enhanced photoacoustic cell and the high-power light source, we can reach a detection limit below 100 ppb in a broadband measurement with a sample volume of only a few milliliters. The first infrared spectrum of CH414 is reported and given a preliminary assignment. The results lay a foundation for the development of optical detection systems for radiocarbon methane.

This work presents an analytical method based on terahertz-time domain spectroscopy (THz-TDS) and partial least squares (PLS) regression models to quantify mebendazole (MBZ) polymorphs (forms A, B and C) in pharmaceutical raw material. Mebendazole polymorphs A, B and C were quantified with RMSEP values of 1.5% w/w, 1.2% w/w and 1.8% w/w, respectively. The limits of detection (LOD) ranges obtained with the best PLS regression models were 2.7-4.3% w/w, 2.9-4.0% w/w and 2.4-3.1% w/w, for polymorphs A, B and C, respectively. This analytical performance is better than those for the methods described in the literature using near (NIR) and middle (MIR) infrared spectroscopies. The main advantage of THz spectroscopy is its ability to access directly information related to crystal lattices. According to the results, the developed method is a powerful technique for the quantification of MBZ polymorphs in raw material. This methodology can be implemented as a Process Analytical Technology (PAT) tool for quality control of pharmaceutical feedstock.

Terahertz dual frequency comb spectroscopy (THz-DFCS) yields high spectral resolution without compromising bandwidth. Nonetheless, the resolution of THz-DFCS is usually limited by the laser repetition rate, which is typically between 80 MHz and 1 GHz. In this paper, we demonstrate a new method to achieve sub-repetition rate resolution in THz-DFCS by adaptively modifying the effective laser repetition rate using integrated Mach-Zehnder electro-optic modulators (MZ-EOMs). Our results demonstrate that it is possible to improve the 100 MHz resolution of a terahertz frequency comb by at least 20x (down to 5 MHz) across the terahertz spectrum without compromising the average output power, and to a large extent, its bandwidth. Our approach can augment a wide range of existing THz-DFCS systems to provide a significant and easily adaptable resolution improvement.

AIM: To determine the ultrasonographic findings that predict death in fetal ascites. METHODS: This was a retrospective cohort study involving pregnancies with ultrasonographic findings related to fetal ascites. The inclusion criteria were as follows: single pregnancy with a live fetus; ultrasound findings of ascites; ascites unrelated to maternal fetal alloimmunization; and pregnancy follow-up at our institution. The χ2-test was used to evaluate the association of ultrasound findings and death. Multiple logistic regression analysis was performed to determine the ultrasound findings that are predictive of death prior to hospital discharge. RESULTS: A total of 154 pregnancies were included in the study. In 8 (5.19%) cases, ascites was an isolated finding, and in 146 cases, other alterations were observed during the ultrasound evaluation. Death before hospital discharge occurred in 117 cases (76.00%). The following ultrasonographic findings were significantly associated with death: gestational age at diagnosis <24 weeks (P<0.0001); stable/progressive ascites evolution (P=0.004); the presence of hydrops (P<0.0001); and the presence of cystic hygroma (P<0.0001). The presence of hydrops, the presence of respiratory tract malformations, and stable/progressive ascites evolution were significantly associated with the prediction of death. CONCLUSIONS: Based on ultrasound examination, the presence of hydrops, malformation of the respiratory tract, and stable/progressive evolution of ascites increase the chances of death in cases of fetal ascites.

Terahertz-time domain spectroscopy (THz-TDS) has the ability to probe the crystallinity of several materials, due to the interaction of THz radiation with optical phonons in crystal lattices. In this work, THz-TDS has been used to quantify the degree of crystallinity of microcrystalline cellulose (MCC) samples. The THz spectra of cellulose present absorption features which could be directly correlated with the crystallinity index (CI) obtained by means of the well-established powder X-ray diffraction (PXRD) technique. The effect of THz time-domain signal processing was investigated, and both univariate and multivariate, based on partial least-squares (PLS), regressions were carried out with the signal in the frequency domain to correlate the THz spectra with CI. Results show that the multivariate regression models based on spectral data, collected with the sample displaced from the focal plane of the THz optics to improve representativeness and measurement repeatability, present the best performance with external validation achieving an absolute root-mean-square error of prediction (RMSEP) of 4% for CI. This result compares well with the PXRD technique.

Dirac and Weyl semimetals are a central topic of contemporary condensed matter physics, and the discovery of new compounds with Dirac/Weyl electronic states is crucial to the advancement of topological materials and quantum technologies. Here we show a widely applicable strategy that uses high configuration entropy to engineer relativistic electronic states. We take the AMnSb2 (A = Ba, Sr, Ca, Eu, and Yb) Dirac material family as an example and demonstrate that mixing of Ba, Sr, Ca, Eu and Yb at the A site generates the compound (Ba0.38Sr0.14Ca0.16Eu0.16Yb0.16)MnSb2 (denoted as A5MnSb2), giving access to a polar structure with a space group that is not present in any of the parent compounds. A5MnSb2 is an entropy-stabilized phase that preserves its linear band dispersion despite considerable lattice disorder. Although both A5MnSb2 and AMnSb2 have quasi-two-dimensional crystal structures, the two-dimensional Dirac states in the pristine AMnSb2 evolve into a highly anisotropic quasi-three-dimensional Dirac state triggered by local structure distortions in the high-entropy phase, which is revealed by Shubnikov-de Haas oscillations measurements.

OBJECTIVES: Changes in placental features, such as maternal and fetal vascular malperfusion, are associated with SARS-CoV-2 infection. The anatomopathologic study of the placenta is crucial for understanding pregnancy and fetal complications. To that end, this study aimed to describe placental features and analyze the association between placental findings and perinatal outcomes in a cohort of pregnant women with severe COVID-19. METHODS: This nested study within a prospective cohort study consisted of 121 singleton pregnant women with a diagnosis of severe COVID-19. Placental pathologic findings were described, and the associations between severe COVID-19 and clinical parameters and perinatal outcomes were assessed. RESULTS: The prevalence of maternal vascular malperfusion was 52.1%, followed by fetal vascular malperfusion at 21.5%, ascending intrauterine infections at 11.6%, and inflammatory lesions at 11.6%. Other lesions were observed in 39.7% of the placentas examined. Inflammatory lesions were an independent factor (P = .042) in 5-minute Apgar scores below 7. Ascending infection was associated with fetal death (P = .027). CONCLUSIONS: Maternal vascular malperfusion was the most prevalent placental feature in patients with severe COVID-19. Chorangiosis is associated with poor perinatal outcomes.

BACKGROUND: Myelomeningocele (MMC) is a neural tube defect disease. Antenatal repair of fetal MMC is an alternative to postnatal repair. Many agents can be used as tocolytics during the in utero fetal repair such as ß2-agonists and oxytocin receptor antagonists, with possible maternal and fetal repercussions. This study aims to compare maternal arterial blood gas analysis between terbutaline or atosiban, as tocolytic agents, during intrauterine MMC repair. METHODS: Retrospective cohort study. Patients were divided into two groups depending on the main tocolytic agent used during intrauterine MMC repair: atosiban (16) or terbutaline (9). Maternal arterial blood gas samples were analyzed on three moments: post induction (baseline, before the start of tocolysis), before extubation, and two hours after the end of the surgery. RESULTS: Twenty-five patients were included and assessed. Before extubation, the terbutaline group showed lower arterial pH (7.347 ± 0.05 vs. 7.396 ± 0.02 for atosiban, p = 0.006) and higher arterial lactate (28.33 ± 12.76 mg.dL-1 vs. 13.06 ± 6.35 mg.dL-1, for atosiban, p = 0.001) levels. CONCLUSIONS: Patients who received terbutaline had more acidosis and higher levels of lactate, compared to those who received atosiban, during intrauterine fetal MMC repair.

Interference energy for C-H and C-C bonds of a set of saturated hydrocarbons is calculated by the generalized product function energy partitioning (GPF-EP) method in order to investigate its sensitivity to the type of chain and also its contribution to the bond dissociation energy. All GPF groups corresponding to chemical bonds are calculated by use of GVB-PP wave functions to ensure the correct description of bond dissociation. The results show that the interference energies are practically the same for all the C-H bonds, presenting only small variations (0.5 kcal.mol(-1)) due to the structural changes in going from linear to branched and cyclic chains. A similar trend is verified for the C-C bonds, the sole exception being the cyclopropane molecule, for which only the C-C bond exhibits a more significant variation. On the other hand, although the interference energy is quantitatively the most important contribution to the bond dissociation energy (DE), one cannot predict DE only from the bond interference energy. Differences in the dissociation energies of C-C and C-H bonds due to structural changes in the saturated hydrocarbons can be mainly attributed to quasi-classical effects.

OBJECTIVES: To determine SARS-CoV-2 seroprevalence over time and risk factors among pregnant women at delivery in São Paulo, Brazil; and to evaluate the suitability of pregnant women as a sentinel population for SARS-CoV-2 serosurveillance. METHODS: Unselected consecutive pregnant women presenting at the labor ward of a single large hospital between July 20th 2020 to February 21st 2021 were enrolled and tested for SARS-CoV-2 serology using two assays: the rapid chromatic Wondfo One Step (for total IgA and IgG detection) and Roche Elecsys assay (detecting anti-nucleoprotein [N] IgG). SARS-CoV-2 seroprevalence was computed as smooth spline function over time with 95% confidence intervals (CI). Risk factors were evaluated for positivity by each assay. We compared timepoint seroprevalence by the two assays with four concomitant community household surveys (HHS), in which the Roche assay was used, to determine the sensitivity and relevance of the pregnant women population as sentinel population. RESULTS: Overall SARS-CoV-2 seroprevalence was 28.9% (221/763) by Roche and 17.9% (137/763) by Wondfo. Reported symptoms experienced during pregnancy were all significantly correlated with being SARS-CoV-2 seropositive at delivery with any assay (with odds-ratios ranging from 3.0 [95% CI: 2.1-4.3] for coryza to 22.8 [95% CI: 12.3-46.6] for ageusia). Seropositivity by either assay was high in women at delivery in the early period of the pandemic (June 2020), compared with seropositivity in women from the concomitant HHS: 44.1% (95% CI: 21.8-66.4) for Roche, 54.1% (30.9-78.5) for Wondfo, versus 11.4% (95% CI: 9.2-13.6) for HHS. For later periods (October 2020 and January 2021), the seropositivity in women at delivery measured by Roche corresponded well with the prevalence found among women in the HHS using the same assay, whilst prevalence measured by Wondfo dropped. CONCLUSIONS: Women at delivery represent a highly exposed and readily accessible population for sentinel surveillance of emerging infections such as SARS-CoV-2.

OBJECTIVES: We sought to determine the short-term and long-term overall mortality rates in obese and non-obese patients after tracheotomy and to evaluate which factors, including the Charlson Comorbidity Index (CCI), predict mortality rates among obese patients. METHODS: We performed a retrospective chart review of patients who underwent open tracheotomy in the operating room at a single hospital from 2005 to 2010. RESULTS: Of 200 patients reviewed, 146 were non-obese and 54 were obese. The rate of mortality was higher at 30 days (p = 0.02) and at 1 year (p = 0.04) in obese patients (35.1% and 59.2%, respectively) than in non-obese patients (19.2% and 42.5%, respectively). The need for tracheotomy due to ventilator-dependent respiratory failure (VDRF) was much higher (p < 0.001) in obese patients (83.3%) than in non-obese patients (56.8%), and the rate of mortality was significantly higher (p < 0.001) in those who required tracheotomy for VDRF (32.8% at 30 days and 57% at 1 year) than in those who required tracheotomy for all other indications (4.2% at 30 days and 25% at 1 year). The mortality risk increased with higher CCI scores at both 30 days (p = 0.08) and 1 year (p = 0.009). CONCLUSIONS: The overall mortality rate is higher in obese patients after tracheotomy than in non-obese control subjects in the short and long terms. This increased rate of mortality is due to the heightened incidence of tracheotomy for VDRF among obese patients. The mortality rates after tracheotomy correlate well with the CCI.

The coronavirus disease 2019 (COVID-19) pandemic has had deleterious effects among the obstetric population. Pregnant and postpartum women constitute a high-risk group for severe COVID-19. Vaccination reduces the risk of infection, but it is not known whether women who become infected despite vaccination have a milder course of disease than those who had not been vaccinated. This retrospective cohort study evaluated whether vaccination reduces the severity of COVID-19 infection, as measured by severe maternal morbidity and mortality among hospitalized pregnant and postpartum individuals. A total of 2284 pregnant and postpartum women hospitalized with severe COVID-19 were included. Those who did and who did not receive COVID-19 vaccination were compared. The rates of intensive care unit admission, intubation, and mortality were significantly lower among subjects in the vaccinated group (p < 0.001, p < 0.001 and p < 0.001, respectively). The numbers of patients who needed to be vaccinated to avoid one case of intensive care unit admission, intubation, or death due to COVID-19 were 7, 7, and 9, respectively. The COVID-19 vaccine offers protective effects against intensive care unit admission, intubation, and death in hospitalized pregnant and postpartum women with severe SARS-CoV-2-induced SARS.

BACKGROUND: Combined spinal-epidural technique (CSE) for labour analgesia has been associated with fetal bradycardia and uterine hypertonia when compared with epidural analgesia (EA), possibly due to a decrease in epinephrine levels following neuraxial anaesthesia. However, there are no recent studies comparing plasmatic catecholamines levels between those two techniques. This study aimed to compare CSE versus EA regarding pre- and post-analgesia catecholamines levels, uterine tone and fetal heart rate. PATIENTS AND METHODS: Randomised clinical trial with 47 labouring patients divided in two groups (CSE and EA). Primary outcome was plasmatic catecholamine measurements before and after neuraxial block. Secondary outcomes were fetal heart rate changes, uterine hypertonia, hypotension episodes, pain relief and fetal outcomes. RESULTS: For CSE group, the median decrease of plasmatic epinephrine was 0 pg/mL [(-) 480-(+) 41] and for norepinephrine was -21 pg/mL [(-) 2507-(+) 94]. For EA group, the median decrease for epinephrine was 0 pg/mL [(-) 326-(+) 15] and for norepinephrine was -5 pg/mL [(-) 190-(+76)]. There were no differences between groups (p = 0.96 and p = 0.63 for epinephrine and norepinephrine, respectively). There were no differences for secondary outcomes. CONCLUSIONS: There was no evidence of a more significant decrease of catecholamines with CSE when compared with EA. Catecholamines decrease theory may not be valid for modern labour analgesia techniques.

SARS-CoV-2 Omicron subvariants have generated a worldwide health crisis due to resistance to most approved SARS-CoV-2 neutralizing antibodies and evasion of vaccination-induced antibodies. To manage Omicron subvariants and prepare for new ones, additional means of isolating broad and potent humanized SARS-CoV-2 neutralizing antibodies are desirable. Here, we describe a mouse model in which the primary B cell receptor (BCR) repertoire is generated solely through V(D)J recombination of a human VH1-2 heavy chain (HC) and, substantially, a human Vκ1-33 light chain (LC). Thus, primary humanized BCR repertoire diversity in these mice derives from immensely diverse HC and LC antigen-contact CDR3 sequences generated by nontemplated junctional modifications during V(D)J recombination. Immunizing this mouse model with SARS-CoV-2 (Wuhan-Hu-1) spike protein immunogens elicited several VH1-2/Vκ1-33-based neutralizing antibodies that bound RBD in a different mode from each other and from those of many prior patient-derived VH1-2-based neutralizing antibodies. Of these, SP1-77 potently and broadly neutralized all SARS-CoV-2 variants through BA.5. Cryo-EM studies revealed that SP1-77 bound RBD away from the receptor-binding motif via a CDR3-dominated recognition mode. Lattice light-sheet microscopy-based studies showed that SP1-77 did not block ACE2-mediated viral attachment or endocytosis but rather blocked viral-host membrane fusion. The broad and potent SP1-77 neutralization activity and nontraditional mechanism of action suggest that it might have therapeutic potential. Likewise, the SP1-77 binding epitope may inform vaccine strategies. Last, the type of humanized mouse models that we have described may contribute to identifying therapeutic antibodies against future SARS-CoV-2 variants and other pathogens.
