RESUMO

To determine the correlation between the working environment and the health status of employees in solar greenhouse, 1171 employees were surveyed. The results show the 'Greenhouse diseases' are affected by many factors. Among general uncomforts, the morbidity of the bone and joint damage is the highest and closely related to labor time and age. Planting summer squash and wax gourd more easily cause skin pruritus. Asthma-related cough, eye disease, and skin pruritus are significantly correlated with the cultivation of wax gourd. The application of inorganic fertilizer and fertigation dramatically induce the bone and joint damage. The smell of covering film greatly influence skin pruritus. Personal protection is badly scanty and normative occupational health and safety need to be completed.

RESUMO

In order to explore the synergistic mechanisms of combinatorial treatment using curcumin and mitomycin C (MMC) for breast cancer, MCF-7 breast cancer xenografts were conducted to observe the synergistic effect of combinatorial treatment using curcumin and MMC at various dosages. The synergistic mechanisms of combinatorial treatment using curcumin and MMC on the inhibition of tumor growth were explored by differential gene expression profile, gene ontology (GO), ingenuity pathway analysis (IPA) and Signal-Net network analysis. The expression levels of selected genes identified by cDNA microarray expression profiling were validated by quantitative RT-PCR (qRT-PCR) and Western blot analysis. Effect of combinatorial treatment on the inhibition of cell growth was observed by MTT assay. Apoptosis was detected by flow cytometric analysis and Hoechst 33258 staining. The combinatorial treatment of 100 mg/kg curcumin and 1.5 mg/kg MMC revealed synergistic inhibition on tumor growth. Among 1501 differentially expressed genes, the expression of 25 genes exhibited an obvious change and a significant difference in 27 signal pathways was observed (p<0.05). In addition, Mapk1 (ERK) and Mapk14 (MAPK p38) had more cross-interactions with other genes and revealed an increase in expression by 8.14- and 11.84-fold, respectively during the combinatorial treatment by curcumin and MMC when compared with the control. Moreover, curcumin can synergistically improve tumoricidal effect of MMC in another human breast cancer MDA-MB-231 cells. Apoptosis was significantly induced by the combinatorial treatment (p<0.05) and significantly inhibited by ERK inhibitor (PD98059) in MCF-7 cells (p<0.05). The synergistic effect of combinatorial treatment by curcumin and MMC on the induction of apoptosis in breast cancer cells may be via the ERK pathway.

RESUMO

OBJECTIVE: To evaluate the efficacy of three categorized formulas for tonifying Shen yang, i.e. Shenqi Pill (SP), Yougui Pill (YP), and Yougui Drink (YD) based on rough set, thus exploring the law of compatibility between the core herbs and the edge herbs as well as the law of compatibility between the yang-tonifying herbs and the yin-tonifying herbs in the core herbs. METHODS: The rats were divided into the normal group, the Shen-yang deficiency model group, and the original prescription group, the core herbs group, and the edge herbs group of this three categorized formulas for tonifying Shen yang, 11 groups altogether. Thyroxine (T4), cortisol (CORT), and testosterone (T) were detected respectively. The decision rules model based on rough set was set up, the interactions between various herbs were analyzed according to the decision rules. RESULTS: There were synergies between yam and cinnamon, yam and aconite, as well as yam and wolfberry. Poria alisma, Cortex Moutan, and oriental waterplantain tuber had no effect themselves, but they had synergistic effects with the core herbs in SP. Angelica had a certain effect itself, but its functions were different to different core herbs in YP. Licorice had no effect itself, and it was antagonistic with the core herbs in YD. CONCLUSIONS: The compatibility of core herbs of categorized formula for tonifying Shen yang should focus on benefiting both yin and yang, as well as mutual rooting of yin-yang. Appropriate edge herbs should be chosen for compatibility according to different core herbs. The decision rules model based on rough set has a good prospect in exploring the law of compatibility of the categorized formulas.

RESUMO

OBJECTIVE: To evaluate the key indicators in the pituitary-target gland axes in the animal model of Shen-yang deficiency syndrome (SYDS). METHODS: The 8 biological indicators [thyroid stimulating hormone (TSH), 3, 3', 5-triiodothyronine (T3), thyroxine (T4), luteinizing hormone (LH), follicle-stimulating hormone (FSH), testosterone (T), adrenocorticotropic hormone (ACTH), and cortisol (CORT)] in the pituitary-target gland axes were grouped using factor analysis. Then the sensitivity of every indicator was calculated according to the sensitivity function defined in this paper, so as to find all the most sensitive indicators in every group as key indicators of SYDS. RESULTS: The key indicators in the early period of SYDS were T, LH, T4, and CORT. The key indicators in the middle period were LH,T, CORT, and ACTH. The key indicators in the late period were LH, T, CORT, and FSH. CONCLUSIONS: T, LH, and CORT were the common key indicators of the three periods, and other different key indicator of SYDS in the early, middle and late period were T4, ACTH, and FSH respectively, which changed from the thyroid axis to the adrenal axis and then to the gonadal axis as the period changed. The key indicators in the late period were mainly in the gonadal axis, showing gonadal dysfunction in the late period.

RESUMO

Jeon et al. (Reports, 29 January 2021, p. 498) reported reversible fluctuations between disordered and crystalline states during nucleation. We argue that the adoption of "magic numbers" for separation of the size range into three parts is misleading when using the sigmoid functions and that their figure 3B was wrongly illustrated. The crystal shape effect was neglected during theoretical analysis.

RESUMO

AIM: To investigate the efficacy of mitomycin C (MMC) in combination with curcumin in suppressing human breast cancer in vitro and in vivo. METHODS: Human breast cancer MCF-7 cells were used. Cell viability was measured using MTT assay. The cell cycle phase was detected with flow cytometric analysis. Cell cycle-associated proteins were examined using Western blot analysis. MCF-7 breast cancer xenografts were established to monitor tumor growth and cell cycle-associated protein expression. RESULTS: Curcumin inhibited MCF-7 breast cancer cell viability in a concentration-dependent manner (IC(50) value=40 µmol/L). Similarly, MMC inhibited the cell viability with an IC(50) value of 5 µmol/L. Combined treatment of MMC and curcumin showed a synergistic antiproliferative effect. In the presence of curcumin (40 µmol/L), the IC(50) value of MMC was reduced to 5 µmol/L. In MCF-7 xenografts, combined administration of curcumin (100 mg/kg) and MMC (1-2 mg/kg) for 4 weeks produced significantly greater inhibition on tumor growth than either treatment alone. The combined treatment resulted in significantly greater G(1) arrest than MMC or curcumin alone. Moreover, the cell cycle arrest was associated with inhibition of cyclin D1, cyclin E, cyclin A, cyclin-dependent kinase 2 (CDK2) and CDK4, along with the induction of the cell cycle inhibitor p21 and p27 both in MCF-7 cells and in MCF-7 xenografts. These proteins were regulated through p38 MAPK pathway. CONCLUSION: The results suggest that the combination of MMC and curcumin inhibits MCF-7 cell proliferation and cell cycle progression in vitro and in vivo via the p38 MAPK pathway.

RESUMO

OBJECTIVE: To study the clinicopathologic features and prognosis of primary lymphoma of breast. METHODS: Forty cases of primary breast lymphoma, diagnosed according to the 2008 World Health Organization classification of hematopoietic and lymphoid tumors, were retrospectively studied. Immunohistochemistry was performed by SP method. The follow-up data were analyzed. RESULTS: (1) All the patients were females and the median age was 47 years. Unilateral and bilateral breast involvement were noted in 36 and 4 patients, respectively. The number of tumor were 31 cases (77.5%, 31/40) less than 3, and 9 cases (22.5%, 9/40) were 3 and more than 3. According to Ann Arbor staging system, 33 cases (82.5%) were in stage I to II and 7 cases (17.5%) in stage III to IV. The level of LDH in 9 cases (24.3%, 9/37) went up. For ECOG scores, 34 cases (85.0%) were 0 to 1 score and 6 cases (15.0%) were more than 2 scores. With respect to international prognostic index, 83.8% (31/37) were of score 0 to 2 and 16.2% (6/37) were of score 3 and more than 3. The axillary lymph nodes of 21 patients (53.8%, 21/39) were involved by the malignancy. (2) Histologically, 38 cases (95.0%, 38/40) were classified as B-cell lymphoma [including 27 cases (67.5%) of diffuse large B-cell lymphoma, 8 cases (20.0%) of mucosa-associated lymphoid tissue lymphoma, 2 cases of follicular lymphoma and 1 case of lymphoplasmacytic lymphoma]. The remaining cases included one case of peripheral T-cell lymphoma and one case of lymphoblastic lymphoma. Immunohistochemically, expression of CD20+/- CD79a were demonstrated in the 38 cases (95.0%) of B-cell lymphoma. The staining for CK was negative in all cases. In 33 cases, the positive rates of MUM-1, bcl-6 and bcl-2 were 57.6% (19/33), 30.3% (10/33) and 72.7% (24/33), respectively. Three cases were germinal center B cell phenotype and 21 cases were non-germinal center B cell phenotype. (3) Follow-up information was available in 37 patients (92.5%, 37/40). Twenty-three patients (62.2%, 23/37) were still alive and fourteen ones (37.8%, 14/37) died. For the 27 cases with diffuse large B-cell lymphoma, the five-year and disease-free survival rates were 48.0% and 36.0%, respectively. CONCLUSIONS: Primary breast lymphoma is a rare disease entity. Diffuse large B-cell lymphoma is the commonest histologic type and the majority show a non-germinal center B cell phenotype. The level of LDH, number of tumor and international prognostic index are of prognostic significance.

RESUMO

Mitomycin C (MMC), a chemotherapeutic agent in breast cancer treatments, inhibits tumor growth through DNA cross-linking and breaking, but it has severe side effects. Here we examined whether and how curcumin reduced the side effects of MMC. We found that combination treatment with MMC and curcumin reduced tumor weight by 70% and 36% compared with saline and curcumin-treated groups, respectively. The combination treatment reduced weight loss and improved kidney function and bone marrow suppression compared with MMC treatment alone. Moreover, the combination treatment inhibited glucose regulatory protein (GRP58)-mediated DNA cross-linking. The combination treatment inhibited GRP58 through the ERK/p38 MAPK pathway. In conclusion, the current study provided evidence that MMC and curcumin combination treatment reduced MMC side effects by inhibiting GRP58-mediated DNA cross-linking through the ERK/p38 MAPK pathway.

RESUMO

AIM: To examine whether the cell growth inhibitory effect of the combination of baicalin and baicalein is related to apoptosis. Moreover, to determine whether the expression of some apoptosis-related proteins is regulated by the ERK/p38 MAPK pathway. METHODS: Cell viability was measured using a 3-(4,5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide (MTT) assay. Apoptosis was detected by acridine orange (AO) staining, DNA ladder assay and flow cytometric analysis. Apoptosis-related proteins were observed using Western blot analysis. RESULTS: Compared with baicalin or baicalein alone, the combination treatment of baicalin (50 micromol/L) and baicalein (25 micromol/L) had an anti-proliferative effect in a time-dependent manner. Isobologram analysis demonstrated that the combination treatment had a synergistic effect. Moreover, apoptosis in MCF-7 cells was increased by 12% and 20% with the combination treatment at 24 h and 48 h, respectively. With the combination treatment in MCF-7 cells, cleaved caspase-3 and caspase-9 were observed, and the level of bcl-2 expression was decreased approximately 20% and 40% at 24 h and 48 h, respectively. The expression of bax and p53 were increased about 25% and 15% at 48 h, respectively. Moreover, the activation of caspase-3, -9 and the regulation of bcl-2, bax and p53 were related to ERK /p38 MAPK activation. CONCLUSION: In this study, apoptosis was enhanced by the combination treatment of baicalin and baicalein, which activated caspases-3 and caspase-9, downregulated the level of bcl-2 and upregulated the level of bax or p53 via the ERK/p38 MAPK pathway.

RESUMO

The effects of red/blue=3/1 [R/B=3/1(9/3), as control, CK1], white light (W, as control, CK2), and combined light of red, blue and white [R/B/W=3/1/1 (9/3/3), T1; R/B/W=9/3/8, T2; R/B/W=3/1/6 (9/3/18), T3; R/B/W=3/1/16 (9/3/48), T4] on the growth, root development, quality and yield of eggplants were examined to provide theoretical and technical support for intensive and high-efficient light supplement and eggplant seedlings cultivation, using LED to accurately regulate light environment and 'Gailiangdalong' eggplant, based on the previous findings of the benefits of R/B=3/1 to the cultivation of eggplant seedlings. The results showed that R/B/W=9/3/8 treatment significantly increased plant height, stem diameter, seedling index, shoot dry weight, root dry weight, root development, anthocyanins and flavonoids content in eggplant peels and early yield. Root/shoot, total phenolic content in eggplant peels and soluble sugar content in pulps under R/B/W=3/1/1 treatment were significantly higher than those of other treatments. Total leaf area and free amino acid content in pulps were higher under R/B/W=3/1/6 treatment. The soluble protein content in pulps was higher under R/B/W=3/1/16 treatment. In summary, combined LED light of red, blue and white could improve the quality of eggplant seedlings and the yield and quality of eggplant fruits, with R/B/W=9/3/8 treatment being the best one in our study.

RESUMO

To investigate the effects of purified humic acid (PHA) on the growth and nitrogen metabolism of cucumber under different levels of nitrogen stress and to explore the mechanism of PHA's alleviation on abiotic stress, we examined the effects of PHA on 'Xintaimici' cucumber growth and nitrogen metabolism under low nitrogen (1.0 mmol·L-1 NO3-) stress and high nitrogen (101 mmol·L-1 NO3-) stress in hydroponics compared with normal level (11 mmol·L-1 NO3-). The results showed that the growth of cucumber seedlings was inhibited under nitrogen stress treatments. Plant height, stem diameter, leaf area and dry matter accumulation under nitrogen stress were lower than that at normal nitrogen level. Dry matter accumulation under the normal nitrogen level and low nitrogen stress were significantly increased by PHA, but there was no significant difference under high nitrogen stress. The absorption of NO3- in cucumber seedlings was affected by PHA, showing increased nitrate content in cucumber seedlings under low nitrogen stress and decreased under high nitrogen stress. PHA significantly decreased ammonium content in roots and leaves under low and high nitrogen stress. Compared with normal level of nitrogen (CK), NR, GS, GOGAT, GDH activity in roots and leaves and NiR activity in roots significantly decreased under low and high nitrogen stress. PHA increased the activities of NR, NiR, GS, GOGAT, and GDH to different extents. PHA increased amino acid and soluble protein content in cucumber seedling roots and leaves. In summary, PHA addition alleviated the inhibitory effect of nitrogen stress on the growth of cucumber seedlings.

RESUMO

To get optimal nitrogen (N), phosphorus (P2O5) and potassium (K2O) concentrations range for high quality and yield of eggplant with vermiculite as cultivation substrate, three factors and quadratic saturation D-optimal regression (310) were used to establish a ternary quadratic mathematical model with N, P2O5 and K2O concentrations as independent variables and eggplant yield and quality as objective functions. The results showed that yield and quality of eggplants were significantly influenced by N, P2O5 and K2O concentrations. The yield was most influenced by K2O concentration, and followed by N and then P2O5. The quality was also most influenced by K2O concentration, and followed by P2O5 and then N. There were significant interactive effects of N+P2O5, N+K2O and P2O5+K2O on yield, and significant interaction of N+K2O on eggplant quality. Under lower concentrations, the yield and quality of eggplants enhanced with the increasing N, P2O5 and K2O concentrations. When the nutrient concentrations exceeded a threshold, both yield and quality decreased. According to computer simulations, the yield could reach to 3600 g·plant-1 when the fertilization schemes were N 16.0-20.0 mmol·L-1, P2O5 2.2-2.6 mmol·L-1 and K2O 9.9-12.9 mmol·L-1; the fertilization scheme needed to obtain a comprehensive quality score of higher than 90 were N 18.0-21.1 mmol·L-1, P2O5 1.9-2.6 mmol·L-1 and K2O 10.6-13.3 mmol·L-1. In summary, fertilization scheme of high yield (43.2 kg·plot-1) and high quality(comprehensive score of higher than 90) of eggplants were N 18.0-20.0 mmol·L-1, P2O5 2.2-2.6 mmol·L-1, K2O 10.6-12.9 mmol·L-1, with an appropriate N, P2O5 and K2O proportion of 1:0.13:0.62.

RESUMO

The effects of iron deficiency and NO3- stress on the growth and the iron absorption cha-racteristic of tomato seedlings were investigated under suboptimal temperature (18 ℃/12 ℃ (day/night)). The results indicated that the growth was significantly inhibited under suboptimal temperature compared with appropriate temperature treatment; the plant heights, leaf areas and dry matter accumulations were markedly decreased. The effects of iron deficiency under suboptimal temperature on the growth of tomato seedlings were larger than that of under appropriate temperature treatments. Under suboptimal temperature, the plant heights of tomato seedlings in iron deficiency and/or NO3- stress treatments were not significantly different from no stress treatment, but leaf areas and leaf chlorophyll content of tomato seedlings were decreased obviously. The leaf electrolytic leakage, root activity and Fe3+ reductase activity of tomato seedlings were markedly increased under suboptimal temperature, but chlorophyll content, total root length, root surface area, root tip number and root volume were obviously reduced. The iron contents in roots, stems and leaves of tomato seedlings were also remarkably decreased. The NO3- stress and the combined stress of iron deficiency and NO3- stress under suboptimal temperature aggravated the reduction of dry matter, leaf electrolytic leakage and the inhibition of iron ion absorption in tomato seedlings. The iron ion absorption showed the antagonistic impact on the absorption of potassium and calcium ion, but showed different effects among different organs. The iron deficiency symptom of tomato seedlings was aggravated by reducing the concentration of iron ion in the nutrient solution.

RESUMO

The solution culture method was used to study the effects of sodium nitroprusside (SNP) on plant growth, nutrient uptake and antioxidant enzyme activities of tomato seedlings under iron deficiency and NO3- stress. The results indicated that after 7 d of treatment, iron deficiency inhibited the tomato seedling growth, decreased chlorophyll (a and b) and carotenoid contents, and led to obvious chlorosis; iron deficiency also reduced the activity of SOD, POD and CAT, resulting in significant accumulation of MDA contents and higher electrolytic leakage; proline and soluble sugar contents were not significantly changed; contents of N, P, K, Ca, Mg and Fe in both leaves and roots of tomato seedlings were decreased under iron deficiency in varying degrees. The combined stress of NO3- stress and iron deficiency promoted the inhibition of plant growth, decreased chlorophyll (a and b), carotenoid contents and the activities of SOD, POD and CAT; electrolytic leakage MDA, proline and soluble sugar contents of N, P, Mg, Fe in both leaves and roots of tomato seedlings were much lower, but the contents of K and Ca were significantly increased. Compared to the treatment without SNP, adding 0.1 mmol·L-1 SNP could alleviate the growth inhibition of stressed tomato seedlings. Adding 0.1 mmol·L-1 SF (sodium ferrocyanide) also showed alleviating or promoting effect on some above indexes including the activity of SOD, POD and CAT to some degree, but the other physiological indexes were not significantly changed because iron was contained in SF.

RESUMO

Effects of high concentration manganese on active oxygen production and antioxidant enzymes in cucumber leaves under natural irradiation and 1/2 natural irradiation were studied. High concentration manganese increased the H2O2 content and O2*- producing rate of cucumber leaves (Fig.1A, B) and caused lipid peroxidation (Fig.1C). Compare to natural irradiation, 1/2 natural irradiation significantly decreased active oxygen production and lipid peroxidation (Fig.1). CAT activities in cytosols and chloroplasts were inhibited by high concentration manganese (Fig.2B), and other antioxidant enzyme activities were enhanced by high concentration manganese (Fig.2A, C-F). Especially activities of APX, DHAR and GR in chloroplasts were greatly stimulated by high concentration manganese under natural irradiation compared to 1/2 irradiation, which may play important roles in scavenging active oxygen species. Antioxidant enzymes in mitochondria showed higher activities under high concentration manganese under natural irradiation, but the differences were not significant under 1/2 natural irradiation (Fig.2A, C-F).

RESUMO

Enteropathy-associated T-cell lymphoma (EATL) is a rare gastrointestinal non-Hodgkin's lymphoma, originating from intraepithelial T-lymphocyte, which is specifically associated with celiac disease. EATL most commonly presents in the sixth and seventh decades of life. We report a unique case of type I EATL in the colon with liver metastasis, which was presented with nonspecific radiological findings and at a very young age (29 years old) compared with previously published data. We suggest that EATL should be regarded as part of differential diagnosis in any patient presenting with abdominal pain, diarrhea, weight loss, and malabsorption because delay in treatment can result in an irreversible clinical outcome.

RESUMO

Taking 'Xintaimici' and 'Jinyou 1' as experimental cucumber cultivars, this paper studied the effects of watering purified humic acid (PHA) at different levels (0, 50, 100, 150, 200 mg·L-1) on growth and nutrient absorption of cucumber seedlings under low nitrogen stress (1 mmol·L-1 NO3-) in sand culture pots. The results showed that, under low N condition, PHA could significantly increase the total root length, root surface area, root tip number and root volume of cucumber seedlings, and promote plant height, stem diameter and leaf area. Proline and soluble sugar contents in cucumber leaves were significantly increased by PHA. The absorption of N, P, K, Ca, Mg, Fe in cucumber seedlings was promoted. According to the response of the two cucumber cultivars to PHA under low N stress, some traits of two cultivars showed different sensitivity to PHA level. The comprehensive analysis of the above results inferred that the PHA level from 100 mg·L-1 to 150 mg·L-1 could significantly enhance the growth and nutrient absorption of cucumber seedlings.

RESUMO

The effects of white light, red light, blue light, yellow light, red+blue light and red+blue+yellow light on the growth and quality of Gynura bicolor were investigated under the same light intensity (350±5 µmol·m-2·s-1) by using light-emitting-diodes (LEDs) which could accurately regulate light quality and light intensity. The results indicated that compared with white light, red light could significantly promote the growth of G. bicolor and induce the accumulation of dry substance and soluble sugar content, but blue light inhibited the plant growth. The content of chlorophyll significantly decreased under the colored LEDs. Although the combination of red, blue and yellow light did not increase the dry substance accumulation, it resulted in significant elevation of total phenols, flavonoids and anthocyanins. The accumulation of these reduced substances could increase the tolerance to oxidative stress and the nutrient value in G. bicolor. This study provided a theoretical basis for G. bicolor diverse production regulated by light quality.

RESUMO

To investigate the effect of calcium and ABA on photosynthesis and the activities of antioxidant enzymes in cucumber seedlings under drought stress, the cucumber was used as the expe-riment materials, normal nutrient solution culture was considered as the control, and PEG-6000 application in the nutrient solution simulated the drought stress. There were five different treatments which were spraying water, ABA, CaCl2+ABA, LaCl3(calcium channel inhibitor)+ABA and EGTA (calcium ion chelating agent)+ABA under drought stress. The results showed that drought stress inhibited the growth of cucumber seedlings, and reduced the activities of antioxidant enzymes, nitrate reductase, net photosynthetic rate and fluorescence parameters of the cucumber seedlings leaves. The application of ABA reduced the inhibition of activities of superoxide dismutase (SOD), peroxidase (POD), catalase (CAT) and ascorbate peroxidase (APX), photosynthesis (Pn, gs) and the fluorescence parameters (Fv'/Fm', qP and ETR), and decreased the damage of drought stress on plant. Spraying CaCl2+ABAsignificantly promoted the positive effect of ABA, while EGTA+ABA and LaCl3 +ABA didn't show the promoting effect.
