RESUMO

Magnetic skyrmions in bulk materials are typically regarded as two-dimensional structures. However, they also exhibit three-dimensional configurations, known as skyrmion tubes, that elongate and extend in-depth. Understanding the configurations and stabilization mechanism of skyrmion tubes is crucial for the development of advanced spintronic devices. However, the generation and annihilation of skyrmion tubes in confined geometries are still rarely reported. Here, we present direct imaging of skyrmion tubes in nanostructured cuboids of a chiral magnet FeGe using Lorentz transmission electron microscopy (TEM), while applying an in-plane magnetic field. It is observed that skyrmion tubes stabilize in a narrow field-temperature region near the Curie temperature (Tc). Through a field cooling process, metastable skyrmion tubes can exist in a larger region of the field-temperature diagram. Combining these experimental findings with micromagnetic simulations, we attribute these phenomena to energy differences and thermal fluctuations. Our results could promote topological spintronic devices based on skyrmion tubes.

The identification of additional Echinococcus granulosus sensu lato (s.l.) complex species/genotypes in recent years raises the possibility that there might be more variation among this species in China than is currently understood. The aim of this study was to explore intra- and inter-species variation and population structure of Echinococcus species isolated from sheep in three areas of Western China. Of the isolates, 317, 322, and 326 were successfully amplified and sequenced for cox1, nad1, and nad5 genes, respectively. BLAST analysis revealed that the majority of the isolates were E. granulosus s.s., and using the cox1, nad1, and nad5 genes, respectively, 17, 14, and 11 isolates corresponded to Elodea canadensis (genotype G6/G7). In the three study areas, G1 genotypes were the most prevalent. There were 233 mutation sites along with 129 parsimony informative sites. A transition/transversion ratio of 7.5, 8, and 3.25, respectively, for cox1, nad1, and nad5 genes was obtained. Every mitochondrial gene had intraspecific variations, which were represented in a star-like network with a major haplotype with observable mutations from other distant and minor haplotypes. The Tajima's D value was significantly negative in all populations, indicating a substantial divergence from neutrality and supporting the demographic expansion of E. granulosus s.s. in the study areas. The phylogeny inferred by the maximum likelihood (ML) method using nucleotide sequences of cox1-nad1-nad5 further confirmed their identity. The nodes assigned to the G1, G3, and G6 clades as well as the reference sequences utilized had maximal posterior probability values (1.00). In conclusion, our study confirms the existence of a significant major haplotype of E. granulosus s.s. where G1 is the predominant genotype causing of CE in both livestock and humans in China.

The rapid start-up and advanced nutrient removal of simultaneous nitrification, endogenous denitrification, and phosphorus (P) removal aerobic granular sequence batch reactor (SNEDPR-AGSBR) is a challenge in the treatment of low carbon/nitrogen (C/N) domestic sewage. In this study, the feasibility of the SNEDPR-AGSBR process was examined in an exceedingly single-stage anaerobic/aerobic/anoxic sequencing batch reactor for treating low C/N ratio (3.3-5.0) domestic sewage. The initial results showed that accompanied by the rapid formation of the mature aerobic granular sludge based on the selection for slow-growing organisms, the rapid start-up (38 d) of the SNEDPR-AGSBR process was successfully realized. The formed mature aerobic granules had a dense structure with an average diameter of 667.7 µm and SVI30 of 30.0 mL/g. Two conditions for achieving the competitive balance between phosphorus-accumulating organisms/denitrifying phosphorus-accumulating organisms (PAOs/DPAOs) and glycogen accumulating organisms/denitrifying glycogen accumulating organisms (GAOs/DGAOs) were revealed by the long-term operation results. First, the dissolved oxygen (DO) concentration needed to be decreased to 3.0 mg/L in the aerobic phase, and then, the aerobic and anoxic phase hydraulic retention time (HRT) should be increased to 3.0 h. Notably, high removal efficiencies for NH4+-N (100%), total nitrogen (84.3%), and P (91.8%) of the SNEDPR-AGSBR process were stably obtained with a low C/N ratio of 3.9 domestic sewage. Simultaneous nitrification and endogenous denitrification (SNED) efficiency of 61.6% was achieved during a long-term operation of 142 days. Finally, microbial community analysis confirmed that GAOs (Defluviicoccus)/DGAOs (Candidatus_Competibacter) were responsible for the removal N, and PAOs (Acinetobacter, Candidatus_Accumulibacter, Hypomicrobinm)/DPAOs (Pseudomonas and Dechloromonas) ensured P removal.

Cysticercosis caused by the metacestode larval stage of Taenia hydatigena formerly referred to as Cysticercus tenuicollis is a disease of veterinary importance that constitutes a significant threat to livestock production worldwide, especially in endemic regions due to condemnation of visceral organs and mortality rate of infected young animals. While the genetic diversity among parasites is found to be potentially useful in many areas of research including molecular diagnostics, epidemiology and control, that of T. hydatigena across the globe remains poorly understood. In this study, analysis of the mitochondrial DNA (mtDNA) of adult worms and larval stages of T. hydatigena isolated from dogs, sheep and a wild boar in China showed that the population structure consists of two major haplogroups with very high nucleotide substitutions involving synonymous and non-synonymous changes. Compared with other cestodes such as Echinococcus spp., the genetic variation observed between the haplogroups is sufficient for the assignment of major haplotype or genotype division as both groups showed a total of 166 point-mutation differences between the 12 mitochondrial protein-coding gene sequences. Preliminary analysis of a nuclear protein-coding gene (pepck) did not reveal any peculiar changes between both groups which suggests that these variants may only differ in their mitochondrial makeup.

Echinococcus shiquicus is currently limited to the Qinghai­Tibet plateau, a large mountainous region in China. Although the zoonotic potential remains unknown, progress is being made on the distribution and intermediate host range. In this study, we report E. shiquicus within Gansu and Qinghai provinces in regions located not only around the central areas but also the southeast edge of the plateau and describe their genetic relationship with previous isolates from the plateau. From 1879 plateau pikas examined, 2.39% (95% CI 1.79­3.18) were infected with E. shiquicus. The highest prevalence of 10.26% (4.06­23.58) was recorded in Makehe town, Qinghai province. Overall the prevalence was marginally higher in Qinghai (2.5%, CI 1.82­3.43) than in Gansu (2%, CI 1.02­3.89). The cox1 and nad1 genes demonstrated high and low haplotype and nucleotide diversities, respectively. The median-joining network constructed by the cox1­nad1 gene sequences demonstrated a star-like configuration with a median vector (unsampled haplotype) occupying the centre of the network. No peculiar distinction or common haplotype was observed in isolates originating from the different provinces. The presence of E. shiquicus in regions of the southeast and northeast edges of the Qinghai­Tibet plateau and high genetic variation warrants more investigation into the haplotype distribution and genetic polymorphism by exploring more informative DNA regions of the mitochondrial genome to provide epidemiologically useful insight into the population structure of E. shiquicus across the plateau and its axis.

Haemonchosis remains a significant problem in small ruminants. In this study, the assay of recombinase polymerase amplification (RPA) combined with the lateral flow strip (LFS-RPA) was established for the rapid detection of Haemonchus contortus in goat feces. The assay used primers and a probe targeting a specific sequence in the ITS-2 gene. We compared the performance of the LFS-RPA assay to a PCR assay. The LFS-RPA had a detection limit of 10 fg DNA, which was 10 times less compared to the lowest detection limit obtained by PCR. Out of 24 goat fecal samples, LFS-RPA assay detected H. contortus DNA with 95.8% sensitivity, compared to PCR, 79.1% sensitivity. LFS-RPA assay did not detect DNA from other related helminth species and demonstrated an adequate tolerance to inhibitors present in the goat feces. Taken together, our results suggest that LFS-RPA assay had a high diagnostic accuracy for the rapid detection of H. contortus and merits further evaluation.

Nonlinear interactions between light waves can exchange energy, linear momentum, and angular momentum. The direction of energy flow between frequency components is usually determined by the conventional phase-matching condition related to the linear momentum. However, the transfer law of orbital angular momentum (OAM) during frequency conversion remains to be elucidated. Here, we demonstrate experimentally that OAM transfer depends strongly on the phase-matching condition defined by both linear and orbital angular momenta. Under different phase-matching configurations, the second-harmonic wave exhibits variable OAM spectral characteristics such as the presence of just a single value or of odd orders only. Our results pave the way toward unveiling the underlying mechanism of nonlinear conversion of OAM states.

The photonic spin Hall effect (SHE) manifests itself as the spin-dependent spatial and angular shifts. There are some ways for controlling the spatial shifts, however, lacks an effective method for manipulating angular shifts. In this work, we propose a simple and effective way for manipulating the spin angular shifts in photonic SHE by considering the light beam reflected at the air-layered structure interface. We theoretically derive the general expressions of the in-plane and transverse spin angular shifts in this layered structure. It is found that the in-plane and transverse spin angular shifts can be effectively regulated by adjusting the structure parameters of layered model, including amplifying or suppressing the magnitude of the angular shifts and switching their signs. Interestingly, the in-plane angular shifts can be adjusted from spin-independent to spin-dependent or vice versa when the polarization state of the incident beam varies. Importantly, as for the incident beam with vertical polarization, a near-zero reflection angle similar to Brewster angle appears. In the vicinity of this point, the large spin angular shifts can be explored.

RESUMO

We report nonlinear Cherenkov radiations (NCRs) in a Ti in-diffused LiNbO3 planar waveguide. The radiations were modulated exploiting different polarizations and orders of the guided modes, the fundamental wavelengths and the working temperatures. Some characteristics related to NCRs, such as radiation angles and relative intensities were investigated in detail. The experimental results matched well with theoretical calculations.

Manipulating photon's orbital angular momentum (OAM) through nonlinear interactions has drawn increasing research interests in recent years. In this work, we propose a scheme to control the OAM of the third harmonic wave through two cascaded second-order nonlinear processes. A Gaussian beam was frequency doubled at the first stage. Subsequent sum frequency mixing of the Gaussian second harmonic wave and an orthogonal-polarized Laguerre-Gaussian-like fundamental wave generate the third harmonic wave, which carries the same OAM as that of the Laguerre-Gaussian-like fundamental wave. In this experiment, we demonstrated controlling the OAM of the third harmonic wave in a tandem periodically-poled LiTaO3 optical superlattice, and the results are in accordance with theoretical predictions.

We report generation of tunable vortex beams in the blue spectral range, with a 3.3 nm spectral tuning range, by frequency tripling of the near-infrared (IR) wave at around 1.34 um in a LiTaO3 optical superlattice. The nonlinear crystal used in this work has a chirped dual-periodical structure which can provide two expanded reciprocal vectors for tunable performance of the cascaded third harmonic generation (THG). The maximum THG efficiency reaches about 1.4%.

This work concerns interfacial adsorption and attachment of swollen microgel with low- to medium-level cross-linking density. Compared to colloids that form a second, dispersed phase, the suspended swollen microgel particles are ultrahigh molecular weight molecules, which are dissolved like a linear polymer, so that solvent and solute constitute only one phase. In contrast to recent literature in which microgels are treated as particles with a distinct surface, we consider solvent-solute interaction as well as interfacial adsorption based on the chain segments that can form trains of adsorbed segments and loops protruding from the surface into the solvent. We point out experimental results that support this discrimination between particles and microgels. The time needed for swollen microgels to adsorb at the air/water interface can be 3 orders of magnitude shorter than that for dispersed particles and decreases with decreasing cross-linking density. Detailed analysis of the microgels deformation, in the dry state, at a solid surface enabled discrimination particle like microgel in which case spreading was controlled predominantly by the elasticity and molecule like adsorption characterized by a significant overstreching, ultimately leading to chain scission of microgel strands. Dissipative particle dynamics simulations confirms the experimental findings on the interfacial activity and spreading of microgel at liquid/air interface.

Cryptosporidium is a widespread protozoan parasite that infects a large number of vertebrate animals, resulting in varying degrees of diarrhea or even death. As dairy cattle feces is an important source of Cryptosporidium spp. infection, development of a handy and accurate detection method via its oocysts in dairy cattle feces would be interesting and necessary. We herein developed a quick detecting method using recombinase polymerase amplification (RPA) combined with lateral flow (LF) strip to detect DNA of Cryptosporidium oocysts in dairy cattle feces. The DNA was released by boiled water with 0.1 % N-lauroylsarcosine sodium salt (LSS). The established method was proven to be of higher sensitivity than normal polymerase chain reaction (PCR) amplification with the lowest detection of 0.5 oocyst per reaction, and specificity with no cross reactivity to other common protozoan species in the intestine of dairy cattle. The diagnostic method established herein is simple, rapid, and cost-effective, and has potential for further development as a diagnostic kit for the diagnosis of cryptosporidiosis of dairy cattle.

RESUMO

Recombinase polymerase amplification (RPA) is a recently -developed isothermal nucleic-acid-amplification technology that is based on the nucleic acid replication mechanism in T4 bacteriophage. With this technique, nucleic-acid templates can be amplified to measurable levels within 20 min at 37-42 °C. The. RPA process has high sensitivity and specificity, and is simple to operate, thus nucleic acids can be detected rapidly in non-laboratory conditions. Since its development in 2006, the RPA technique has been applied in agriculture, food safety, medicine, transgene detection, etc. In this review, we will give an overview on the research progress of RPA and its application in parasite detection.

Herein, we investigate the interfacial behavior of temperature-sensitive aqueous microgels on the toluene/water interface. Copolymer microgels based on N-vinylcaprolactam (VCL) and two acrylamides, N-isopropylacrylamide (NIPAm) and N-isopropylmethacrylamide (NIPMAm), with various copolymer compositions were used in this study. It is revealed that these copolymer microgels have the similar internal structure, regardless of the chemical composition. A classic kinetics of interfacial tension with three distinct regimes is found in the dynamic interfacial tension plots of copolymer microgels, which is similar to inorganic nanoparticles and proteins. The influences of the copolymer composition and the temperature on the interfacial behavior of microgels are investigated. The results show that the interfacial behavior of copolymer microgels at the toluene/water interface follows exactly the trend of the volume phase behavior of microgels but, on the other hand, strongly depends upon the chemical compositions of copolymer microgels. In contrast, with respect to the size range of microgels studied here (50-500 nm), the size of the microgel has no influence on the interfacial tension. Below the volume phase transition temperature (VPTT), the equilibrium interfacial tensions of all microgel systems decrease as the temperature increases. Above VPTT, the equilibrium interfacial tension remains at a certain level for poly(N-vinylcaprolactam) (PVCL)- and poly(N-isopropylmethacrylamide) (PNIPMAm)-rich microgel systems and increases slightly for poly(N-isopropylacrylamide) (PNIPAm)-rich microgel systems. The evolution of dynamic interfacial tension for microgel solutions against toluene at T < VPTT is faster than that at T > VPTT, because of the reduced deformability of the microgel with the increase of the temperature. The softer microgels with lower cross-linking degrees exhibit faster kinetics of reduction of interfacial tension compared to those with more cross-linked degrees, which strongly supports the deformation-controlled interfacial behavior of microgels.

The combination of a set of complementary techniques allows us to construct an unprecedented and comprehensive picture of the internal structure, temperature dependent swelling behavior, and the dependence of these properties on the cross-linker concentration of microgel particles based on N-vinylcaprolactam (VCL). The microgels were synthesized by precipitation polymerization using different amounts of cross-linking agent. Characterization was performed by small-angle neutron scattering (SANS) using two complementary neutron instruments to cover a uniquely broad Q-range with one probe. Additionally we used dynamic light scattering (DLS), atomic force microscopy (AFM), and differential scanning calorimetry (DSC). Previously obtained nuclear magnetic resonance spectroscopy (NMR) results on the same PVCL particles are utilized to round the picture off. Our study shows that both the particle radius and the cross-link density and therefore also the stiffness of the microgels rises with increasing cross-linker content. Hence, more cross-linker reduces the swelling capability distinctly. These findings are supported by SANS and AFM measurements. Independent DLS experiments also found the increase in particle size but suggest an unchanged cross-link density. The reason for the apparent contradiction is the indirect extraction of the parameters via a model in the evaluation of DLS measurements. The more direct approach in AFM by evaluating the cross section profiles of observed microgel particles gives evidence of significantly softer and more deformable particles at lower cross-linker concentrations and therefore verifies the change in cross-link density. DSC data indicate a minor but unexpected shift of the volume phase transition temperature (VPTT) to higher temperatures and exposes a more heterogeneous internal structure of the microgels with increasing cross-link density. Moreover, a change in the total energy transfer during the VPT gives evidence that the strength of hydrogen bonds is significantly affected by the cross-link density. A strong and reproducible deviation of the material density of the cross-linked microgel polymer chains toward a higher value compared to the respective linear chains has yet to be explained.

Topological spin textures are characterized by magnetic topological charges, Q, which govern their electromagnetic properties. Recent studies have achieved skyrmion bundles with arbitrary integer values of Q, opening possibilities for exploring topological spintronics based on Q. However, the realization of stable skyrmion bundles in chiral magnets at room temperature and zero magnetic field - the prerequisite for realistic device applications - has remained elusive. Here, through the combination of pulsed currents and reversed magnetic fields, we experimentally achieve skyrmion bundles with different integer Q values - reaching a maximum of 24 at above room temperature and zero magnetic field - in the chiral magnet Co8Zn10Mn2. We demonstrate the field-driven annihilation of high-Q bundles and present a phase diagram as a function of temperature and field. Our experimental findings are consistently corroborated by micromagnetic simulations, which reveal the nature of the skyrmion bundle as that of skyrmion tubes encircled by a fractional Hopfion.

Carbon sources are critical factors influencing bacterial bioaugmentation, however, the underlying mechanisms, particularly the metabolic characteristics of bioaugmented bacteria remain poorly understood. The bioaugmented bacterium Rhodococcus sp. BH4 secretes the quorum quenching (QQ) enzyme QsdA to disrupt the quorum sensing (QS) in the activated sludge (AS) process, reducing AS yield in-situ. This study investigated the carbon metabolic characteristics of BH4 and explored the effects on bioaugmentation with different influent carbon sources. Because of the absence of glucose-specific phosphoenol phosphotransferase system (PTS), BH4 prefers sodium acetate to glucose. However, the lactones produced during extracellular glucose metabolism enhance BH4 qsdA expression. Moreover, BH4 possess carbon catabolite repression (CCR), acetate inhibits glucose utilization. BH4 microbeads were added to reactors with different carbon sources (R1: sodium acetate; R2: glucose; R3: a mixture of sodium acetate and glucose) for in-situ AS yield reduction. During operation, AS reduction efficiency decreased in the following order: R1 > R3 > R2. R2 and R3 microbeads exhibited similar QQ activity to R1, with less BH4 biomass at 5 d. 13C labeling and Michaelis-Menten equation showed that, due to differences in the competitiveness of carbon sources, R1 BH4 obtained the most carbon, whereas R2 BH4 obtained the least carbon. Moreover, acetate inhibited glucose utilization of R3 BH4. Transcriptome analysis showed that R1 BH4 qsdA expression was the lowest, R2 BH4 was the most serious form of programmed cell death, and the R3 BH4 PTS pathway was inhibited. At 10 d, R1 BH4 biomass and microbead QQ activity were higher than that in R3, and the R2 BH4 lost viability and QQ activity. This study provides new insights into bioaugmentation from the perspectives of carbon source competitiveness, carbon metabolism pathways, and CCR.

Widespread use of nanomaterials raises concerns. The underlying mechanism by which graphene oxide (GO) nanoparticles causes poor settleability of activated sludge remains unclear. To explore this mechanism, three reactors with different GO concentrations were established. Extended Derjaguin-Landau-Verwey-Overbeek theory indicated that GO destroyed the property of extracellular polymeric substances (EPS), increasing the energy barrier between bacteria. Low levels of uronic acid and hydrogen bonding in exopolysaccharide weakened the EPS gelation increasing aggregation repulsion. Lager amounts of hydrophilic amino acid and looser structure of extracellular proteins for exposing inner hydrophilic groups significantly contributed to the hydrophilicity of EPS. Both changes implied deterioration in EPS structure under GO stress. Metagenome demonstrated a decrease in genes responsible for capsular polysaccharide colonization and genes regulated the translocation of loose proteins were increased, which increased repulsion between bacteria. This study elucidated that changes in EPS secretion under GO exposure are the underlying causes of poor settleability.

Existing phosphorus (P) resources are becoming increasingly scarce, so it is necessary to recover P from potential sources. This paper is based on thermal hydrolysis process (THP) at 140-180 °C, coupled with low-temperature pyrolysis at 300 °C, to study its effect on the recovery and conversion of P from sewage sludge. Most significant change was observed in apatite P, which increased from 3.43 ± 0.48 mg/g in raw sludge to 30.17 ± 1.17 mg/g in biochar (BTHP-180-4-300) during optimal process (THP condition: 180 °C, 4 h; pyrolysis condition: 300 °C). Reactions between phosphates and metal ions became more complete during this combined process. Unstable forms of P were converted into more stable forms, with transformations from Al-P and Fe-P toward Ca-P compounds like Ca3(PO4)2, Ca3Mg3(PO4)4, Ca2P2O7, and Ca(H2PO4)2, making P less degradable and more suitable as slow-release fertilizers. Additionally, P characteristics of actual THP in a sewage treatment plant were similar to those of laboratory THP.
