RESUMO

At present, increasing the output power of a single fiber laser is a problem worthy of attention, and a fiber signal combiner which can carry high power and high beam quality is a solution to this problem. In this paper, a high beam quality 4×1 fiber signal combiner was developed. Firstly, the simulation model of a 4×1 fiber signal combiner was established, and the factors affecting the beam quality and the transmission efficiency of combiner were simulated. Secondly, based on corrosion technology, a high beam quality 4×1 fiber signal combiner with an output fiber of 50/400 µm (NA=0.12) was fabricated by using the taper-fused fiber bundles technology according to simulation results. Finally, four Yb-doped fiber lasers with a central wavelength of 1080 nm were used to test the output power and beam quality of combiner. The test result showed that the total output power of the signal combiner was 12.03 kW, the overall transmission efficiency was more than 96%, and the M2 factor was measured to be 4.03.

During the aging process, the reduced osteogenic differentiation of bone marrow mesenchymal stem cells (BM-MSCs) results in decreased bone formation, which contributes to senile osteoporosis. Previous studies have confirmed that interrupted circadian rhythm plays an indispensable role in age-related disease. However, the mechanism underlying the impaired osteogenic differentiation of BM-MSCs during aging and its relationship with interrupted circadian rhythm remains unclear. In this study, we confirmed that the circadian rhythm was interrupted in aging mouse skeletal systems. The level of the core rhythm component BMAL1 but not that of CLOCK in the osteoblast lineage was decreased in senile osteoporotic specimens from both human and mouse. BMAL1 targeted TTK as a circadian-controlled gene to phosphorylate MDM2 and regulate H2Bub1 level, while H2Bub1 in turn regulated the expression of BMAL1. The osteogenic capacity of BM-MSCs was maintained by a positive loop formed by BMAL1-TTK-MDM2-H2Bub1. Furthermore, we demonstrated that using bone-targeting recombinant adeno-associated virus 9 (rAAV9) to enhance Bmal1 or Ttk might have a therapeutic effect on senile osteoporosis and delays bone repair in aging mice. In summary, our study indicated that targeting the BMAL1-TTK-MDM2-H2Bub1 axis via bone-targeting rAAV9 might be a promising strategy for the treatment of senile osteoporosis.

The combination effect of multiwavelength active coherent beam combination (CBC) is investigated theoretically. The dependence of the combination effect on the optical path control precision, spectral width, wavelength number, and channel number is revealed. In the case of small optical path variance, the combination effect approximately decreases in quadratic form with wavelength number N, spectral width Δν, and optical path variance σ increasing. In the case of large optical path variance, the combination effect is independent of the optical path variance and the spectral width. The larger the wavelength number is, the smaller the Strehl ratio expectation is, and it finally degenerates to the incoherent combination. The necessity of optical path control is discussed. This study is helpful for multiwavelength CBC system design and the combination effect estimation.

In this paper we determine the optimum propagation distance between measurement planes and the plane of the lens in a wavefront curvature sensor with the diffraction optics approach. From the diffraction viewpoint, the measured wavefront aberration can be decomposed into Fourier harmonics at various frequencies. The curvature signal produced by a single harmonic is analyzed with the wave propagation transfer function approach, which is the frequency analysis of wavefront curvature sensing. The intensity of the curvature signal is a sine function of the product of the propagation distance and the squared frequency. To maximize the curvature signal, the optimum propagation distance is proposed as one quarter of the Talbot length at the critical frequency (average power point at which the power spectrum density is the average power spectrum density). Following the determination of the propagation distance, the intensity of the curvature signal varies sinusoidally with the squared frequencies, vanishing at some higher frequency bands just like a comb filter. To cover these insensitive bands, wavefront curvature sensing with dual propagation distances or with multi-propagation distances is proposed.

The evaluation of the toxicity of magnetic nanoparticles (MNPs) has attracted much attention in recent years. The current study aimed to investigate the cytotoxic effects of Fe3O4, oleic acid-coated Fe3O4 (OA-Fe3O4), and carbon-coated Fe (C-Fe) nanoparticles on human hepatoma BEL-7402 cells and the mechanisms. WST-1 assay demonstrated that the cytotoxicity of three types of MNPs was in a dose-dependent manner. G1 (Fe3O4 and OA-Fe3O4) phase and G2 (C-Fe) phase cell arrests and apoptosis induced by MNPs were detected by flow cytometry analysis. The increase in apoptosis was accompanied with the Bax over-expression, mitochondrial membrane potential decrease, and the release of cytochrome C from mitochondria into cytosol. Moreover, apoptosis was further confirmed by morphological and biochemical hallmarks, such as swollen mitochondria with lysing cristae and caspase-3 activation. Our results revealed that certain concentrations of the three types of MNPs affect BEL-7402 cells viability via cell arrest and inducing apoptosis, and the MNPs-induced apoptosis is mediated through the mitochondrial-dependent pathway. The influence potency of MNPs observed in all experiments would be: C-Fe > Fe3O4 > OA-Fe3O4.

RESUMO

The optimum phase defocus grating for wavefront curvature sensing is proposed. It features an equidistantly quantized, binary-phase-step defocus grating with a phase-step height of pi. The diffractive efficiency of the phase defocus grating is theoretically computed. The optical transfer function is obtained. The optimum phase defocus grating is fabricated. The high diffractive efficiencies of the +/-1 diffraction orders are verified experimentally, the average values of which are 38.08% and 40.36%, respectively.
