Storage and transportation of coal, as well as operation of coal-fired power plants, produce amounts of metallic exhaust that may lead to different atmospheric environment in the overlapped areas of farmland and coal resource (OAFCR) environment. To investigate the effects of different atmospheric environment in the OAFCR region (north of Xuzhou) on wheat seedlings (AK-58), a box experiment was conducted and compared to an area far from the OAFCR (south of Xuzhou). The study revealed that (1) compared to the southern suburb of Xuzhou, the fresh and dry weight, activities of photosynthetic enzymes and POD of wheat seedlings in the OAFCR reduced obviously. (2) Significantly higher levels of Cr, Cd, Pb, Zn, and Cu were found in the shoots and roots of wheat seedlings in the OAFCR, with lower transfer factor for heavy metals (except Cd and As) in comparison to those in the southern suburb. And the bioconcentration factors of heavy metals (except As) in wheat seedlings in the OAFCR were significantly higher. (3) Nearly 90% of heavy metals (Pb, Cu, Cd, Zn, and Cr) absorbed by wheat were stored in cell walls and soluble fractions, with significantly higher contents of Cu and Cr in wheat seedlings' cell walls and higher contents of Pb, Zn, and Cd in soluble components found in the OAFCR. Our results showed that atmospheric deposition in the mining area has a certain toxic effect on wheat seedlings, and this study provides a theoretical basis for OAFCR crop toxicity management.

This study aimed to better understand environmental heat stress and physiological heat strain indicators in lactating dairy cows. Sixteen heat stress indicators were derived using microenvironmental parameters that were measured at the surrounding of cows and at usual fixed locations in the barn by using handheld and fixed subarea sensors, respectively. Twenty high-producing Holstein-Friesian dairy cows (> 30.0 kg/day) from an intensive dairy farm were chosen to measure respiration rate (RR), vaginal temperature (VT), and body surface temperature of forehead (FT), eye (ET), and muzzle (MT). Our results show that microenvironments measured by the handheld sensor were slightly warmer and drier than those measured by the fixed subarea sensor; however, their derived heat stress indicators correlated equally well with physiological indicators. Interestingly, ambient temperature (Ta) had the highest correlations with physiological indicators and the best classification performance in recognizing actual heat strain state. Using segmented mixed models, the determined Ta thresholds for maximum FT, mean FT, RR, maximum ET, mean ET, VT, mean MT, and maximum MT were 24.1 °C, 24.2 °C, 24.4 °C, 24.6 °C, 24.6 °C, 25.3 °C, 25.4 °C, and 25.4 °C, respectively. Thus, we concluded that the fixed subarea sensor is a reliable tool for measuring cows' microenvironments; Ta is an appropriate heat stress indicator; FT, RR, and ET are good early heat strain indicators. The results of this study could be helpful for dairy practitioners in a similar intensive setting to detect and respond to heat strain with more appropriate indicators.

5-Aza-2'-deoxycytidine (5-Aza-dC), an inhibitor of DNA methyltransferases, is an effective treatment for various cancers and has improved the development rate of cloned embryos. Previous studies have reported the effect of 5-Aza-dC on fibroblasts; however, the mechanism whereby 5-Aza-dC affects sika deer granulosa cells and hormone secretion is presently unknown. Here, we showed that the cell cycle after treatment with different doses of 5-Aza-dC was significantly altered. The number of cells in the S phase was significantly increased in response to a concentration of 0.1 µM 5-Aza-dC. The rate of apoptosis was increased when cells were treated with 0.1 µM and 5 µM 5-Aza-dC. We showed that the protein level of H3K9me2 was significantly decreased in response to 5-Aza-dC. The activity levels of DNA methyltransferase were reduced by a moderate dose of 5-Aza-dC. Furthermore, the secretion of E2 and P4 was influenced by different doses of 5-Aza-dC. Our study suggested that 5-Aza-dC affected hormone secretion in sika deer granulosa cells through cell development and epigenetic regulation. The findings of this study lay the foundation for further epigenetic studies in sika deer.

Heavy metals (As, Cu, Cd, Cr, Pb, Zn, Hg) in soil of a typical overlapped area of farmland and coal resources (OAFCR) in Xuzhou were investigated, meanwhile the pollution levels and risks of there were discussed. The results are: Pollution Load Index (PLI) showed no heavy metals (HMs) pollution; 3.74% of soil samples were above moderately accumulated with Hg; the ecological risk (Er) values followed the order: Hg > Cd > Cu > As > Pb > Cr > Zn, and there has a moderate degree of potential ecological risk (PER) with the mean PER of 184.26; the non-carcinogenic risk of all seven HMs to human beings is acceptable, and the carcinogenic risk caused by As, Cd and Cr can be tolerated; the exposure ways both of carcinogenic and non-carcinogenic is: oral > dermal > inhalation, while children are suffering higher health risks than adults.

The oocyte-specific protein JY-1 was reported as an important regulator of both function of ovarian granulosa cells and early embryogenesis in cattle. Here, we found that the transcripts of JY-1 were also present in sika deer granulosa cells (GCs) through in situ hybridization and qRT-PCR. The complete sika deer JY-1 coding sequence was identified, which had three exons separated by two introns. It was detected that JY-1 knockdown caused apoptosis and abnormal cell cycle progression in GCs of sika deer cultured in vitro. Taken together, these data suggest that JY-1 is involved in the regulation of proliferation in sika deer ovarian GCs.

The histone deacetylase inhibitor (HDACi) and tumor suppressor play an important role in genome reorganization and epigenetic regulation. In this study, granulosa cells (GCs) isolated from sika deer ovaries were cultured and treated with different concentrations of trichostatin A (TSA) for 48 h. It was found that TSA inhibited GCs proliferation and induced GCs apoptosis by upregulating expression of BAX, meanwhile, downregulating expression of GLUT3, GLUT8, BCL-XL. In addition, TSA caused cell cycle arrest at the G1 and G2/M phase accompanied by reducing expression of Cyclin D2 and CDK4. TSA pretreatment increased DNMT3a, DNMT1, HDAC1, and HAT1 expression, and attenuated them when TAS higher than 50 nM. The protein levels of H3K9ac and H4K8ac in GCs were increased at 48 h after TSA treatment. TSA stimulated the secretion of estradiol and progesterone at a moderate dose. Our data suggest that TSA is important as a regulator of steroid hormone synthesis in granulosa cells during follicular development in the sika deer ovary.

Endoplasmic reticulum stress (ERS) is closely associated with the occurrence and development of many liver diseases. ERS models mostly include experimental animals such as rats and mice. However, pigs are more similar to humans with regards to digestion and metabolism, especially liver construction, yet few reports on ERS in pigs exist. In order to explore changes in the liver under ERS, we used tunicamycin (TM), which can cause liver jaundice and damage liver function, to establish acute ERS models in piglets using a low TM dosage (LD, 0.1 mg/kg body weight (bw)), high TM dosage (HD, 0.3 mg/kg bw), or vehicle for 48 h. We found that both LD- and HD-induced ERS, as verified by the ERS-linked proteins. Furthermore, the concentrations of the proinflammatory cytokines, namely, TNF-α and IL-6 were elevated in TM-treated piglet livers, and the plasma levels of IL-6 and CRP were also higher, indicating the occurrence of inflammation in TM-treated piglets. The complement system was activated in TM-treated piglets, as indicated by increased levels of complement factors and activation products C3, C5a, and AP50. In order to gain insights into the global changes in liver proteins under ERS, we performed an isobaric tag for relative and absolute quantitation (iTRAQ)-based proteomic analysis on the livers of HD- and vehicle-treated piglets. Proteomic analysis identified 311 differentially expressed proteins (DEPs) between the two groups, and a Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis suggested that the DEPs were mainly enriched in signaling pathways such as metabolic pathways, protein processing in the endoplasmic reticulum, and complement and coagulation cascades. Many proteins involved in protein folding, lipid transport, and oxidation were upregulated. Proteins involved in lipid synthesis were downregulated to alleviate liver steatosis, and most complement factors were upregulated to protect the body, and Pearson correlation analysis found that most of the DEPs in the complement and coagulation pathway were significantly correlated with plasma CRP, IL6 and AP50. Our results revealed that TM can activate ERS, marked by liver injury and steatosis, inflammatory reactions, and complement activation in piglets.

Eye temperature (ET) has long been used for predicting or indicating heat stress in dairy cows. However, the region of interest (ROI) and temperature parameter of the eye have not been standardized and various options were adopted by previous studies. The aim of this study was to determine the best ROI for measuring ET as the predictor of heat stress in dairy cows in consideration of repeatability and validity. The ET of 40 lactating Holstein dairy cows was measured using infrared thermography. The mean and maximum temperature of five ROIs-medial canthus (MC), lateral canthus, eyeball, whole eye (WE), and lacrimal sac (LS)-were manually captured. The results show that the ET of left eyes was slightly higher than that of right eyes. The ET taken in MC, WE, and LS within 2 min had a moderate to substantial repeatability. The maximum temperature obtained at the LS had the highest correlation coefficients with respiration rate and core body temperature (all p < 0.001). Therefore, the maximum temperature of LS should be considered by future studies that want to use ET as the predictor or indicator of heat stress in dairy cows.

Oxidative stress refers to the dramatic increase in the production of free radicals in human and animal bodies or the decrease in the ability to scavenging free radicals, thus breaking the antioxidation-oxidation balance. Various factors can induce oxidative stress in pig production. Oxidative stress has an important effect on pig performance and healthy growth, and has become one of the important factors restricting pig production. Based on the overview of the generation of oxidative stress, its effects on pigs, and signal transduction pathways, this paper discussed the nutritional measures to alleviate oxidative stress in pigs, in order to provide ideas for the nutritional research of anti-oxidative stress in pigs.

Ice-free cryopreservation, referred to as vitrification, is receiving increased attention in the human and animal assisted reproduction. However, it introduces the detrimental osmotic stress by adding and removing high contents of cryoprotectants. In this study, we evaluated the effects of normalizing cell volume regulation by adding glycine, an organic osmolyte, during vitrification of mouse germinal vesicle stage oocyte and/or subsequent maturation on its development. The data showed that glycine supplementation in either vitrification/thawing or maturation medium significantly improved the cytoplasmic maturation of MII oocytes manifested by spindle assembly, chromosomal alignment, mitochondrial distribution, euploidy rate, and blastocyst development following fertilization in vitro, compared to the control without glycine treatment. Furthermore, glycine addition during both vitrification/thawing and maturation further enhanced the oocyte quality demonstrated by various markers, including ATP contents and embryo development. Lastly, the effect of anti-apoptosis was also observed when glycine was added during vitrification. Our result suggests that reducing osmotic stress induced by vitrification could improve the development of vitrified mouse oocyte.
