Early and immediate loading of dental implants has become a routine procedure in dental practices throughout the world, but the histological feature of peri-implant bone has not been fully understood. Therefore, we aimed to elucidate the histological response of peri-implant bone bearing the early occlusal loading using rat models. Four-week-old male Wistar rats were subjected to extraction of their maxillary left first molars and had titanium implants inserted immediately into the post-extraction sockets. In experimental groups at 1 week after placement, implants were loaded for 1 or 2 weeks by adding adhesive resin on the top of the screws. In control groups, no adhesive resin was added to the implants. After 1 or 2 weeks with loading, rats were fixed with an aldehyde solution for histochemical assessment. Newly-formed bone adhered broadly to the implant surface in both the control and experimental groups. The experimental group loaded for 2 weeks showed thicker trabeculae between the implant threads compared to those in the control group. Osteopontin- and osteocalcin-positive cement lines, which are histological hallmarks of bone remodeling, were narrow and smooth in the experimental groups, while featuring a complex meshwork with thick scalloped lines in the control groups. The index of sclerostin-positive osteocytes located close to implants loaded for 2 weeks was significantly lower than in controls, suggesting that osteoblast activity was preserved. Summarizing, our experimental model suggested that early implant loading increases trabecular thickness in the peri-implant bone tissue in a process that involves the regulation of bone remodeling.

Staphylococcus argenteus has been recently established as a novel species of Staphylococcus aureus complex. It is known to cause various human diseases, such as skin and soft-tissue infections, sepsis, and staphylococcal food poisoning, although the source of infection has not been clearly described. In food poisoning cases, the source of bacterial contamination in food is unknown. This study examined the prevalence of S. argenteus among retail fresh food and poultry slaughterhouses in Japan. Among 642 food samples examined, successful isolation of S. argenteus was achieved in 21 of 151 (13.9%) chicken samples. No isolations from pork, beef, fish, or vegetables in retail markets were confirmed. Multiple-locus sequence typing revealed that the 21 isolates were classified into four sequence types (ST) that were divided into 14 subtypes using spa-typing. All food isolates were susceptible to methicillin and did not show positivity for the Panton-Valentine leukocidin gene. When bacteria were isolated from two poultry slaughterhouses in the same region, 14 S. argenteus strains were successfully isolated from only one slaughterhouse. Thirteen of 14 strains were isolated from a poultry carcass and slaughterhouse environments during a certain sampling period and were all classified as ST5961 with identical spa-type. Also, the number of single nucleotide variants (SNVs) detected on the core genomes of the same 13 strains were between 0 and 17, suggesting a long-term inhabitation of an S. argenteus strain inside the facility. Furthermore, one isolate from chicken meat was also genetically linked with the same lineage of slaughterhouse isolates, with ≤15 SNVs being detected. Additionally, one slaughterhouse isolate from chiller water and three chicken isolates were classified into the same cluster by phylogenetic analysis, although the number of pairwise SNVs ranged from 62 to 128. To the best of the authors' knowledge, this is the first study that demonstrated S. argenteus in a food processing facility and the possible bacterial contamination on food during food processing.

An 11-year-old, spayed female mixed-breed dog showed clinical signs of right forelimb lameness and pain by palpation around the neck. Radiography and computed tomography (CT) revealed an extradural mass at the 6th and 7th cervical vertebrae, which compressed the spinal cord. The mass was surgically removed and histopathologically diagnosed as schwannoma. The dog recovered her normal gait after hemilaminectomy and removal of the mass. Ten months after the surgery, the tumor recurred with absolute erythrocytosis and was surgically removed again. This removal temporarily resolved the erythrocytosis with a decrease in plasma erythropoietin (EPO) concentration. EPO protein was detected immunohistochemically in the tumor cells. Erythrocytosis in this dog may be caused by ectopic EPO produced in the schwannoma tissues.

Caloric restriction (CR) slows the aging process and extends longevity, but the exact underlying mechanisms remain debatable. It has recently been suggested that the beneficial action of CR may be mediated in part by adipose tissue remodeling. Mammals have two types of adipose tissue: white adipose tissue (WAT) and brown adipose tissue (BAT). In this study, proteome analysis using two-dimensional gel electrophoresis combined with MALDI-TOF MS, and subsequent analyses were performed on both WAT and BAT from 9-month-old male rats fed ad libitum or subjected to CR for 6 months. Our findings suggest that CR activates mitochondrial energy metabolism and fatty acid biosynthesis in WAT. It is likely that in CR animals WAT functions as an energy transducer from glucose to energy-dense lipid. In contrast, in BAT CR either had no effect on, or down-regulated, the mitochondrial electron transport chain, but enhanced fatty acid biosynthesis. This suggests that in CR animals BAT may change its function from an energy consuming system to an energy reservoir system. Based on our findings, we conclude that WAT and BAT cooperate to use energy effectively via a differential response of mitochondrial function to CR.

BACKGROUND: Human papilloma virus (HPV)-related warts persist, evading host immune surveillance, but sometimes disappear with inflammation. OBJECTIVES: To elucidate the immune evasion mechanisms of HPV, we have examined the density, dynamics, and subsets of dendritic cell (DC) types in non-inflammatory or inflammatory HPV-related skin lesions such as warts and Bowen's disease (HPV-Bowen), and compared the epidermal expression levels of macrophage inflammatory protein (MIP)-3α and E-cadherin. METHODS: The expression of various DC markers, MIP-3α, and E-cadherin in the tissue samples obtained from patients with warts, HPV-Bowen and HPV-unrelated skin diseases was evaluated by immunohistochemistry. MIP-3α gene expression levels were examined in warts and HPV-Bowen by in situ hybridization (ISH) and real-time quantitative polymerase chain reaction (RT-qPCR). RESULTS: The numbers of Langerhans cells (LCs) and the expression levels of MIP-3α and E-cadherin were decreased in non-inflammatory warts and HPV-Bowen, as compared with normal skin. Both epidermal LCs and MIP-3α expression reappeared in inflammatory warts, associated with dermal infiltrates composed of many cytotoxic T cells and various subsets of DCs, while cellular infiltrates in HPV-Bowen contained many B cells and plasma cells with sparse infiltration of DCs. The upregulation of MIP-3α gene expression was confirmed in the inflammatory warts and HPV-Bowen by ISH and RT-qPCR. CONCLUSIONS: The depletion of LCs in the non-inflammatory warts and HPV-Bowen is associated with a down-regulation of expression levels of MIP-3α and E-cadherin in the lesional keratinocytes. MIP-3α expression is upregulated in lesional keratinocytes of inflammatory warts, with the subsequent recruitment of various DC subsets and cytotoxic T cells, whereas plasma cell-rich infiltration was induced in HPV-Bowen.
