RESUMO

The plane grating double monochromator is the important part of the instrument for testing diffraction efficiency of plane grating, and in order to accomplish the research of the instrument, the optical system design and simulation of the plane grating double monochromator was investigated. The instrument mainly consists of light source, front monochromator, testing monochromator and detector, and combined with the practical requirement, the corresponding light source, detector and the structure of the optical path were selected. According to the design requirement, the design and simulation of the optical system of the front monochromator and testing monochromator were done respectively, and the spot diagram of the image surface and the results of real ray trace were given. The analysis showed that in the front monochromator the maximum width of the exit spot and the maximum height of the exit spot were 0.33 and 1.83 mm respectively. On the other hand, in the testing monochromator, the maximum width of the exit spot and the maximum height of the exit spot were 0.66 and 7.27 mm, respectively. Through selecting the size of the exit slit properly, the system can work without luminous flux lost, which guarantees the testing precision of the optical system of the instrument.

The wavelength calibration research on the automatic grating monochromator for orthogonal Czerny-Turner structure was investigated. Combined with the structure parameters and characteristic of this monochromator, the sinusoid that accords with the grating equation was proposed as the function of the wavelength calibration. Based on the principle of the least square method, the formula of the fitting residual error of the wavelength calibration was given. Using the Nelder-Mead simplex method, the undetermined coefficient of the fitting residual error was solved, which founded the precise formula between the wavelength and the grating turning angle. The accuracy of the method was verified through the experiment. The calibrated wavelength precision of the monochromator is less than 0.1 nm, which satisfies the application requirement. Applied in the wavelength calibration of the automatic grating monochromator with orthogonal Czerny-Turner structure, this method is simple to apply and easy for implementation. Using this method, the wavelength calibration can be realized quickly and real-timely, but it is only needed to modify the control program of the stepping motor slightly, which shows a better practicability.

Background: Hepatocellular carcinoma (HCC) is a solid tumor with high recurrence rate and high mortality. It is crucial to discover available biomarkers to achieve early diagnosis and improve the prognosis. The effect of LSM4 in HCC still remains unrevealed. Our study is dedicated to exploring the expression of LSM4 in HCC, demonstrating its clinical significance and potential molecular mechanisms. Methods: Clinical information and LSM4 expression values of HCC were obtained from Gene Expression Omnibus (GEO) and The Cancer Genome Atlas (TCGA) databases. Survival analysis and receiver operating characteristic (ROC) curve analysis were applied to evaluate the prognostic and diagnostic significance of LSM4. Calculating pooled standardized mean difference (SMD) and performing summary receiver operating characteristic (sROC) curve analysis to further determine its expression status and diagnostic significance. LSM4-related co-expressed genes (CEGs) were obtained and explored their clinical significance in HCC. LSM4-associated pathways were identified through Gene set enrichment analysis (GSEA). Results: Up-regulated LSM4 was detected in HCC tissues (SMD = 1.56, 95% CI: 1.29-1.84) and overexpressed LSM4 had excellent distinguishing ability (AUC = 0.91, 95% CI: 0.88-0.93). LSM4 was associated with clinical stage, tumor grade, and lymph node metastasis status (p < 0.05). Survival analysis showed that high LSM4 expression was related to poor overall survival (OS) of HCC patients. Cox regression analysis suggested that high LSM4 expression may be an independent risk factor for HCC. We obtained nine up-regulated CEGs of LSM4 in HCC tissues, and six CEGs had good prognostic and diagnostic significance. GSEA analysis showed that up-regulated LSM4 was closely related to the cell cycle, cell replication, focal adhesion, and several metabolism-associated pathways, including fatty acid metabolism. Conclusion: Overexpressed LSM4 may serve as a promising diagnostic and prognostic biomarker of HCC. Besides, LSM4 may play a synergistic effect with CEGs in promoting the growth and metastasis of HCC cells via regulating crucial pathways such as cell cycle, focal adhesion, and metabolism-associated pathways.

RESUMO

A new-style system that automatically measures the diffraction efficiency of plane reflection grating was designed. The continuous illuminant was adopted for illumination, the duplex grating spectrograph structure was applied, and the linear array NMOS was the receiving component. Wielding relevant principle of the grating spectrograph, theoretical analysis principle was carried out for the testing system. Integrating the aberration theory of geometrical optics, the image quality of this optics system was analyzed. Analysis indicated that the systematic device structure is compact, and electronics system is simplified. The system does not have the problem about wavelength sweep synchronization of the two grating spectrographs, and its wavelength repeatability is very good. So the precision is easy to guarantee. Compared with the former automated scheme, the production cost is reduced, moreover it is easy to operate, and the working efficiency is enhanced. The study showed that this automatic measurement instrument system features a spectral range of 190-1 100 nm and resolution is less than 3 nm, which entirely satisfies the design request. It is an economical and feasible plan.

OBJECTIVE: The abnormal expression of LARP1 and E-cadherin (E-cad) is related to tumor occurrence and metastasis. Our study analyzed the expression of LARP1 and E-cad in intrahepatic cholangiocarcinoma (ICC) and investigated the prognostic value of the two proteins. METHODS: Immunohistochemistry was performed to detect the expression of LARP1 and E-cad in 50 ICC clinical specimens with adjacent normal tissues and 20 normal bile duct tissues. In situ hybridization was performed to analyze the expression of LARP1 mRNA in all the specimens. RESULTS: LARP1 protein and mRNA expression levels in ICC tumor tissues were significantly higher compared with corresponding adjacent normal tissues and normal epithelial tissues (P<0.01). E-cad protein expression in ICC tumor tissues was remarkably lower than that of the adjacent normal tissues and benign bile duct tissues (P<0.01). Correlation analysis demonstrated that LARP1 and E-cad expression levels were significantly related with the tumor-node-metastasis staging and lymph node metastasis (P<0.01), while no correlation was observed with patient age, gender, and tumor size. Moreover, Spearman rank correlation test revealed that LARP1 expression was negatively related to E-cad (P<0.05). More importantly, the patients with higher LARP1 expression or lower E-cad expression had a shorter overall survival postoperatively than those with LARP1 lower expression or E-cad higher expression. Multivariate analysis demonstrated that LARP1 and E-cad were both considered as important prognostic factors for survival time. CONCLUSION: These findings suggest that the abnormal expression of LARP1 and E-cad showed a close relationship with the occurrence and metastasis of ICC, leading to poor prognosis indirectly.
