RESUMO

Objective: In order to understand the dust concentration in the workplace of dust exposure enterprises in Shenxian. To evaluate the degree of occupational hazard factors of dust exposure enterprises. And provide basis for the formulation of occupational protection standards and management system of dust exposure enterprises. Methods: In February 2022, the dust concentration monitoring data of 89 dust exposure enterprises from 2017 to 2020 by the Shenxian Center for Disease Control and Prevention were collected, and the qualified rates of dust concentration detection of dust exposure enterprises in different years, dust types and enterprise sizes were analyzed. Results: A total of 89 dust enterprises were monitored from 2017 to 2020, 2132 dust samples were collected, and 1818 qualified samples were taken, with a total qualified rate of 85.3%. From 2017 to 2020, the dust detection qualified rates showed a year-by-year increase trend, 78.7% (447/568), 84.1% (471/560), 88.6% (418/472) and 90.6% (482/532), respectively, with statistically significant differences (χ(2)=36.27, P=0.003). The differences in the qualified rates of dust detection samples of silicon dust (66.1%, 41/62), grain dust (86.7%, 1549/1786), cotton dust (84.1%, 106/126) and wood dust (77.2%, 122/158) were statistically significant (χ(2)=29.66, P=0.002). The qualified rate of dust samples in large and medium-sized enterprises (95.1%, 1194/1256) was higher than that of small-sized enterprises (71.2%, 624/876), and the difference was statistically significant (χ(2)=1584.40, P=0.001) . Conclusion: The qualified rate of dust concentration monitoring results of dust exposure enterprises in Shenxian showed an increase trend year by year, while the pualified rate of dust concentration monitoring in small-sized enterprises was low, and the occupational hazard of silica dust was still severe.

Objective: To study the phenotype of memory CD4(+) T cells in peripheral blood of asthmatics and its relationship with asthma severity. Methods: From Dec 2014 to Aug 2015, thirty-three asthmatics, twenty-six chronic obstructive pulmonary disease (COPD) patients and twenty-two healthy volunteers were enrolled in Respiratory Clinics of Zhongshan Hospital, Fudan University. Peripheral blood mononuclear cells (PBMCs) were isolated from peripheral blood. Cell surface markers (CD45RO, CRTH2, CD62L, and CCR7) and intracellular protein[interleukin (IL)-5, IL-17, interferon (IFN)-γ]staining was performed using flow-cytometric techniques. CD4(+) T cells were cultured under neutralization and then Th2, Th2+ Lipopolysaccharide (LPS), Th2+ Home dust mite (HDM) conditions for 6 days and then intracellular proteins were analyzed using flow cytometry. Correlation analysis between memory CD4(+) T cells, asthma severity and drug consumption were performed. Results: The percentage of memory CD4(+) T (CD4(+) Tm) cells in circulating white blood cells was higher in asthmatics, than that in healthy subjects (48.0%±5.7% vs 32.0%±4.1%, P<0.05). The cytokine profiles of CD4(+) Tm cells in asthma patients were substantially different from those of COPD and healthy subjects, with increased IL-5 and IL-17 production. For COPD patients, the predominant cytokines were IFN-γ instead. IL-17-producing CD4(+) Tm cells were associated with the severity of disease and the level of medication consumption in asthma patients (R(2)=0.829 6, P<0.05). Conclusions: The cytokine profile is IL-5 and IL-17 predominant in memory CD4(+) T cells from asthmatics. The amount of IL-17(+) CD4(+) memory T cells is positively correlated with asthma severity.

Objective: To analyze the efficacy of HLA-haploidentical peripheral hematopoietic stem cell transplantation (haplo-PBSCT) following reduced intensity conditioning (RIC) regimen to treat the patients with hematological malignancies who were older than 50 years old. Methods: Eighteen patients with hematological malignancies over 50 years were enrolled, including 8 male and 10 female patients. The median age of all patients was 52 (range: 50-66) years. Of them, 8 patients had acute myeloid leukemia (AML) , 2 chronic myelocytic leukemia (CML) , 5 myelodysplastic syndrome (MDS) , 2 acute lymphoblastic leukemia (ALL) , and 1 aggressive natural killer cell leukemia (ANKL) . All patients received fludarabine, cytarabine and melphalan with rabbit anti-human thymocyte globulin (FAB+rATG regimen) and transplanted with high dose non-T cell-depleted peripheral hematopoietic stem cells from donors. Enhanced graft versus host disease (GVHD) prophylaxis and infection prevention were administered. Results: Fifteen days after transplantation, 16 patients achieved complete donor chimerism. One of them rejected the donor graft completely at thirty days after transplantation, and the other 2 patients had mixed chimerism 15 days after transplantation and converted to complete recipient chimerism at 30 days after transplantation. The cumulative incidence of acute GVHD (aGVHD) was 61.1% (95%CI49.6%-72.6%) . The incidence of grade Ⅱ-Ⅳ aGVHD was 35.4% (95%CI 21.1%-49.7%) , whereas grade III-IV was 13.8% (95%CI 4.7%-22.9%) . The 2-year cumulative incidence of chronic GVHD (cGVHD) rate was estimated at 38.2% (95%CI 25.5%-50.9%) . Patients were followed-up for a median of 14.5 months (range, 3-44 months) . The Kaplan Meier estimates of 2-year overall survival (OS) and disease-free survival (DFS) was 72.6% (95%CI 60.1%-85.1%) and 63.7% (95%CI 49.2%-78.2%) , respectively. The 2-year cumulative incidence of relapse and non-relapse-mortality (NRM) was 31.2% (95%CI 16.5%-45.9%) and 12.5% (95%CI 4.2%-20.8%) , respectively. Conclusion: RIC-haplo-PBSCT protocol can achieve better results in patients with hematologic malignancies over 50 years old.

A pilot-scale fluidised pellet bed (FPB) bioreactor, which combines chemical coagulation, biological degradation, particle pelletisation and separation in one unit, was applied for onsite wastewater treatment and reuse. As a result of rational use of inorganic coagulant and organic polymer and moderate mechanical agitation, spherical particles were generated in the upflow column and a well-fluidised bed was formed. With a continuous supply of dissolved oxygen through a recycling loop, an aerobic condition was kept in the bottom section of the FPB column. Under such conditions the pellets in the FPB column showed the following characteristics: (1) compact structure and high density; (2) rich in microorganisms; and (3) high MLSS and MLVSS concentrations. Therefore, the FPB bioreactor achieved more than 90% removal of SS, COD, BOD and TP from raw domestic wastewater within a total hydraulic retention time (HRT) of only about 30 minutes. It also showed nitrification and denitrification ability and the TN removal could be about 50% as the recycling ratio was increased to 1:1. The treated water quality is generally competitive with the secondary effluent from a conventional activated sludge process. With these advantages the FPB bioreactor is recommendable as a compact system for onsite wastewater treatment and reuse.

Penicillium sp. P6, isolated from coal mine soil at the Qiantong colliery Liaoning Province, Northwest China, can degrade Chinese lignite in 36 h on a plate colony and in 48 h using a 4-day cultured cell-free filtrate. Results of elemental analysis and IR spectrometry indicated that solubilized products exhibited some alterations in comparison to the original lignite. The amount of fulvic acid extracted from the biodegraded lignite was high, and the molecular distribution of the humic acids from biodegraded lignite changed distinctively in comparison to which extracted from the control lignite, possibly due to the depolymerization associated with fungal biodegradation.

OBJECTIVE: To compare occurrence and curative effect of interstitial pneumonia (IP) of patients with malignant hematologic disease after related HLA-haploidentical peripheral blood stem cell transplantation without T-cell depletion (RHNT-PBSCT) and non T cell-depleted HLA-matched sibling peripheral blood stem cell transplantation (MSNT-PBSCT). METHODS: 109 patients with malignant hematologic disease as the research cases received RHNT-PBSCT from January 2006 to December 2014, which were compared with 125 patients treated with MSNT-PBSCT during the same period to determine the occurrence of IP and curative effect produced by ganciclovir and joint adrenal glucocorticoids. RESULTS: The incidences of IP in RHNT-PBSCT and MSNT-PBSCT groups were 15.60%(17/109) and 13.60%(17/125) (P=0.150), respectively; both the effective rates were 76.47%(13/17) (P=0.536), the difference was not statistically significant. Single factor analysis showed that acute graft versus host disease was a risk factor for the occurrence of IP after RHNT-PBSCT (P=0.001). CONCLUSION: The incidence of IP in patients with malignant hematologic disease after RHNT-PBSCT didn' t increase when compared with patients after MSNT-PBSCT. The curative effects of the two groups were equivalent after priority to ganciclovir and joint adrenal glucocorticoids.

A fluidized-pellet-bed separator with movable sludge hoppers was applied in pilot scale for the separation and thickening of activated sludge mixture liquid. Under the condition of suspension SS around 4,000 mg/L, polymer (CJX103, cationic, MW 5 x 10(6)) dose at a dry solid ratio of 0.003 and upward flow rate at 5.4 m/hr, the fluidized pellet bed performed solid/liquid separation and sludge thickening well. The SS concentration of the treated water was about 5 mg/L on average and the moisture content of the sludge after screening for 5 min was less than 94%, which is much lower than that after conventional settling and thickening and easy to be finally disposed. At a higher upward flow rate of 7.2 m/hr, similar results could also be obtained but higher polymer dose (solid ratio of 0.004) was required. The morphological characteristics and density-size relationship of the granular particles formed in the fluidized pellet bed were also investigated by image analysis and settling velocity measurement of individual particles. The two-dimensional fractal dimension was evaluated to be 1.6-1.8, showing a good quasi-spherical morphology of the granular particles with their density much higher than the conventional flocs. The results of the pilot study indicate a possible way to innovate the conventional secondary settling and gravitational thickening processes for solid/liquid separation and sludge handling, especially for small scale wastewater treatment plants to reach the goal of space saving and higher treatment efficiency.

Nine cis-beta-diketonato titanium (IV) complexes were synthesized and their chemical structures were confirmed by IR, UV spectra and elemental analyses. Among them 6 complexes have not been reported before. Preliminary pharmacological experiments showed that this kind of complexes were active against tumors.

Aurora kinases have emerged as attractive targets for the development of novel anti-cancer agents. A combined study of molecular docking, pharmacophore modelling and 3D-QSAR was performed on a series of imidazo [1, 2-a] pyrazines as novel Aurora kinase inhibitors to gain insights into the structural determinants and their structure-activity relationship. An ensemble of conformations based on molecular docking was used for PHASE pharmacophore studies. The developed best-fitted pharmacophore model was validated by diverse chemotypes of Aurora A kinase inhibitors and was consistent with the structural requirements for the docked binding mechanism. Subsequently, the pharmacophore-based alignment was used to develop PHASE and comparative molecular similarity indices analysis (CoMSIA) 3D-QSAR models. The best CoMSIA model showed good statistics (q (2 )= 0.567, r (2 )= 0.992), and the predictive ability of the model was validated using an external test set of 13 compounds giving a satisfactory prediction ([Formula: see text]). The 3D contour maps provided insight into the binding mechanism and highlighted key structural features that are essential to the inhibitory activity. Based on the PHASE and CoMSIA 3D-QSAR results, a set of novel Aurora A inhibitors were designed that showed excellent potencies.
