RESUMO

Heat stress (HS) can cause damage to the organism, especially the intestinal tract. In this paper, we investigated the effects of the combined action of tea polyphenols (TP) and hydrogen-rich electrolyzed water (HRW) on HS in mice. The combination of HRW feeding and TP of intraperitoneal injection was screened by in vitro antioxidant activity assay. The results revealed that the combined treatment was more helpful in alleviating the effects of HS on the behavior, growth performance, oxidative damage, and intestinal tract of mice compared with the respective treatments of TP and HRW (P < 0.05). Additionally, the combined treatment could repair HS-induced intestinal dysbiosis in mice, augmenting the number and abundance of bacteria, increasing the number of beneficial genera (Lachnospiraceae_NK4A136_group and Lactobacillus), and decreasing the number of harmful genera (Desulfovibrio and Enterorhabdus), and the effect was significantly better than that of individual treatment (P < 0.05). In conclusion, the combined treatment of TP and HRW effectively mitigates the adverse effects of HS on mouse behavior, growth performance, oxidative damage, and intestinal dysbiosis, surpassing the efficacy of individual treatments with TP or HRW alone.

Phase difference sensing technology (PDST) is employed for topography measurement, and two interference structures are proposed to achieve upper-limit adjustment and high resolution in the measurement range: a dual-wavelength system with a single Fabry-Perot (FP) cavity and a single-wavelength system with dual FP cavities. The phase difference between the two interference signals is determined by an elliptic fitting algorithm (EFA), and this change in phase difference is utilized to characterize the step height. Experimental results indicate that the measurement upper-limit can be adjusted to either 410 µm, 187 µm, or 108 µm by varying the wavelength difference in the dual-wavelength system, which gives a measurement error of 2.96%. In contrast, while offering a measurement resolution of 3.47 nm, the single-wavelength system exhibits a measurement error of 5.38%. The proposed method is capable of satisfying the measurement requirements during micro-electromechanical system (MEMS) processing with proficiency.

Antibiotic resistant bacteria (ARB) and antibiotic resistance genes (ARGs) have drawn much more attention due to their high risk on human health and ecosystem. In this study, the performance of sulfidated nanoscale zero-valent iron (S-nZVI)/periodate (PI) system toward ARB inactivation and ARGs removal was systematically investigated. The S-nZVI/PI system could realize the complete inactivation of 1 × 108 CFU/mL kanamycin, ampicillin, and tetracycline-resistant E. coli HB101 within 40 min, meanwhile, possessed the ability to remove the intracellular ARGs (iARGs) (including aphA, tetA, and tnpA) carried by E. coli HB101. Specifically, the removal of aphA, tetA, and tnpA by S-nZVI/PI system after 40 min reaction was 0.31, 0.47, and 0.39 log10copies/mL, respectively. The reactive species attributed to the E. coli HB101 inactivation were HO• and O2•-, which could cause the destruction of E. coli HB101 morphology and enzyme system (such as superoxide dismutase and catalase), the loss of intracellular substances, and the damage of iARGs. Moreover, the influence of the dosage of PI and S-nZVI, the initial concentration of E. coli HB101, as well as the co-existing substance (such as HCO3-, NO3-, and humic acid (HA)) on the inactivation of E. coli HB101 and its corresponding iARGs removal was also conducted. It was found that the high dosage of PI and S-nZVI and the low concentration of E. coli HB101 could enhance the disinfection performance of S-nZVI/PI system. The presence of HCO3-, NO3-, and HA in S-nZVI/PI system showed inhibiting role on the inactivation of E. coli HB101 and its corresponding iARGs removal. Overall, this study demonstrates the superiority of S-nZVI/PI system toward ARB inactivation and ARGs removal.

OBJECTIVE: At present, pancreatic cancer (PC) has a high morbidity and mortality rate and a poor prognosis. The aim of this article was to study the efficacy and safety of apatinib combined with radiotherapy in the treatment of advanced PC. METHODS: The PubMed, Cochrane Library, Embase, Wanfang, CNKI, VIP, and CBM databases were searched by computer to identify studies on the application of apatinib in patients with advanced PC. The patients in the included study were divided into an observation group (apatinib combined with radiotherapy) and a control group (radiotherapy only), and meta-analysis was performed for each outcome with Revman 5.4 software. This study was successfully registered on the PROSPERO website, and the registration number is CRD: 42,022,384,056 (available at https://www.crd.york.ac.uk/PROSPERO/display_record.php?RecordID=384056 ). RESULTS: A total of 7 randomized controlled trials (RCTs) with 428 patients were included, including 215 in the observation group and 213 in the control group. Compared with the control group, the observation group showed a greater objective response rate [OR = 3.26, 95% CI (2.18, 4.87), P < 0.0001], disease control rate [OR = 5.04, 95% CI (3.12, 8.12), P < 0.0001], complete response rate [OR = 3.87, 95% CI (1.51, 9.88), P = 0.005], and partial response rate [OR = 2.43, 95% CI (1.63, 3.61), P < 0.001], The 1-year survival rate [OR = 2.39, 95% CI (1.15, 4.96), P < 0.05], 2-year survival rate [OR = 2.41, 95% CI (1.03, 5.61), P < 0.05], progression-free survival time [MD = 1.17, 95% CI (0.37, 1.96), P < 0.05], overall survival time [MD = 1.47, 95% CI (0.13, 2.80), P < 0.05], while the stability rate [OR = 1.14, 95% CI (0.72, 1.81), P = 0.58] and various complications were not significantly different between the two groups. CONCLUSION: Apatinib combined with radiotherapy was more effective than radiotherapy alone in the treatment of advanced pancreatic cancer (PC), and apatinib had acceptable safety. However, since our study was limited by the quantity and quality of the included studies, we look forward to more large-sample, multicentre, and high-quality RCTs in the future to verify the conclusions.

To identify the function of the SpsNAC042 gene and its response to salt and drought stress, the SpsNAC042 gene was transformed into Populus hopeiensis by the Agrobacterium-mediated leaf disc method, and the phenotypic, physiological changes and related genes expression of transgenic lines were analyzed. The results showed that the number and length of roots of transgenic lines increased significantly. The leaves of transgenic lines curled inward. Under salt and simulated drought stress, the transgenic lines showed improved tolerance to salt and drought. The activities of SOD, POD, CAT and proline content in the transgenic lines were significantly increased, and the reduction rates of total chlorophyll and MDA content were significantly decreased, which indicated that the transgenic lines showed strong physiological responses under stress. Meanwhile, the gene expression of MPK6, SOS1, HKT1 and P5CS1 were significantly upregulated, and the gene expression of PRODH1 was significantly downregulated, which preliminarily verified the stress regulation mechanism that SpsNAC042 might activate. The above results showed that the SpsNAC042 gene could promote root development, make leaf morphology curl, and enhance P. hopeiensis tolerance to stress.

Laser Doppler velocimetry (LDV) based on a differential laser Doppler system has been widely used in fluid mechanics to measure particle velocity. However, the two outgoing lights must intersect strictly at the measurement position. In cross-interface applications, due to interface effects, two beams of light become easily disjointed. To address the issue, we present a laser velocimeter in a coaxial arrangement consisting of the following components: a single-frequency laser (wavelength λ = 532 nm) and a Twyman-Green interferometer. In contrast to previous LDV systems, a laser velocimeter based on the Twyman-Green interferometer has the advantage of realizing cross-interface measurement. At the same time, the sensitive direction of the instrument can be changed according to the direction of the measured speed. We have developed a 4000 m level laser hydrothermal flow velocity measurement prototype suitable for deep-sea in situ measurement. The system underwent a withstand voltage test at the Qingdao Deep Sea Base, and the signal obtained was normal under a high pressure of 40 MPa. The velocity contrast measurement was carried out at the China Institute of Water Resources and Hydropower Research. The maximum relative error of the measurement was 8.82% when compared with the acoustic Doppler velocimeter at the low-speed range of 0.1-1 m/s. The maximum relative error of the measurement was 1.98% when compared with the nozzle standard velocity system at the high-speed range of 1-7 m/s. Finally, the prototype system was successfully evaluated in the shallow sea in Lingshui, Hainan, with it demonstrating great potential for the in situ measurement of fluid velocity at marine hydrothermal vents.

Nanoparticle self-assembly enables the generation of complex ordered nanostructures with enhanced properties or new functionalities. However, the ordering is often limited to the micrometer scale with chemical strategies due to the relative weak supramolecular interactions that govern the self-assembly process. Here a physical strategy via temperature-gradient-assisted self-assembly is reported to create three-dimensional (3D) macroscopic ordered nanocomposites with different gradient variations in grain size, constituent content, and crystal orientation. The resulting α-Fe/Pr2Fe14B ordered nanostructure with reverse gradients in both the grain size and α-Fe content exhibits a record-high energy density of about 25 MGOe for isotropic α-Fe/Pr2Fe14B systems, approximately 130% higher than that of its disordered counterpart. Both experiments and micromagnetic simulations demonstrate that creating ordered nanostructures is an alternative approach to develop high-performance permanent-magnet materials. Our findings make a significant step toward creating 3D macroscopic ordered nanostructures and will stimulate the development of ordered nanomaterials.

Resistance to inhibitors of cholinesterase 8 (Ric8) is a conserved guanine nucleotide exchange factor that is involved in the regulation of G-protein signalling in filamentous fungi. Here, we characterized an orthologous Ric8 (AoRic8) in Arthrobotrys oligospora by multi-omics analyses. The Aoric8 deletion (ΔAoric8) mutants lost an ability to produce traps essential for nematode predation, accompanied by a marked reduction in cAMP level. Yeast two-hybrid assay revealed that AoRic8 interacted with G-protein subunit Gα1. Moreover, the mutants were compromised in mycelia growth, conidiation, stress resistance, endocytosis, cellular components and intrahyphal hyphae. Revealed by transcriptomic analysis differentially upregulated genes in the absence of Aoric8 were involved in cell cycle, DNA replication and recombination during trap formation while downregulated genes were primarily involved in organelles, carbohydrate metabolism and amino acid metabolism. Metabolomic analysis showed that many compounds were markedly downregulated in ΔAoric8 mutants versus the wild-type strain. Our results demonstrated a crucial role for AoRic8 in the fungal growth, environmental adaption and nematode predation through control of cell cycle, organelle and secondary metabolism by G-protein signalling.

High security and convenient operation have been the aim of the anticounterfeiting field. An anticounterfeit system in which multiple information are written on an overlapping area with quantum dot (QD) inks of different fluorescence wavelengths that combines spectroscopy technology for information identification is proposed in this study. Because the emission spectra of QDs can be tuned by simply changing the size, ultraviolet (UV) glue mixed with different sizes of CdSe/ZnS QDs is used as the printing ink with different fluorescence wavelengths. Software-design labels with different information are printed on the overlapping area using inks with different luminescent QDs. The printed information can only be identified by the bandpass filter with the corresponding wavelength under UV light. Under natural light, the information cannot be identified by the naked eye. In the proposed anticounterfeiting system, the excitation light and filter are both indispensables. Our method makes the fabrication of anticounterfeit labels flexible in design, fast in production, and high in information concealment. Meanwhile, the proposed system is quick and convenient, which has huge application potential in the field of display and anticounterfeiting.

Bacterial antibiotic resistance in water environments is becoming increasingly severe, and new antibiotic resistant bacteria (ARB) and antibiotic resistance genes (ARGs) have also attracted the attention of researchers. The horizontal transfer of ARGs in water environments is considered one of the main sources of bacterial resistance in the natural environment. Horizontal gene transfer (HGT) mainly includes conjugation, natural transformation, and transduction, and conjugation has been investigated most. Several studies have shown that there are a large number of environmental factors that might affect the horizontal transfer of ARGs in water environments, such as nanomaterials, various oxidants, and light; however, there is still a lack of systematic and comprehensive reviews on the detection and the effects of the influence factors of on ARG horizontal transfer. Therefore, this study introduced three HGT modes, analysed the advantages and disadvantages of current methods for monitoring HGT, and then summarized the influence and mechanism of various factors on ARG horizontal transfer, and the possible reasons for the different effects caused by similar factors were mainly critically discussed. Finally, existing research deficiencies and future research directions of ARG horizontal transfer in water environments were discussed.

Temperature crosstalk has always been a critical problem for fiber intermodal sensors. In this work, we have proposed a novel method based on the special temperature response of photosensitive fiber to control the temperature sensitivity of the fiber intermodal sensor. The control of temperature sensitivity has been realized via adjusting the proportion of photosensitive fiber to single-mode fiber in the sensing part. The temperature sensitivity as high as -192 pm/°C, and as low as -2.6 pm/°C can be obtained, satisfying the demand in both research and application. The torsion sensor is taken as an example to illustrate feasibility of this method, showing no evident interference in the measurement of torsion parameters. The proposed method outstrips the conventional one by simple structure, facile manufacture, multiple use and low cost, which brings great promise for further employment in laboratory and industry.

A phase-shifted demodulation technique with a 3×3 coupler and ellipse fitting algorithm (EFA) for the interrogation of interferometric sensors is proposed. To reduce the error of the EFA as to measure small phase signals, additional phase modulation is introduced. The additional modulation provides a walk of the operating point along the Lissajous ellipse large enough to permit calculation of the ellipse parameters at every moment. Experimental result shows that this technique demonstrates high accuracy and stability for measuring small phase signals. The setting of this technology expands the application of the EFA in fiber-optic phase demodulation technology.

Phase-difference sensing technology (PDST) has been applied to strain measurement, but its completeness is destroyed by the phase-difference measurement range. A scheme that can realize the completeness of the PDST for low-frequency strain interrogation is proposed. It is built on dual-interferometers and the elliptic-fitting algorithm. To break the measurement range limitation (0, π), a phase compensation setting is applied. The experimental results demonstrate that the method can obtain low-frequency strain signals, and the low-frequency signal whose phase amplitude is greater than π is recovered. The scheme is an efficient and complete method for measuring the strain of low-frequency optical fiber length, which could be applied to low-frequency seismic wave monitoring and rock deformation detection.

In this study, spent Auricularia auricula substrate (AS)-derived biochar (ASBCs) and activated biochar with NaOH (A-ASBC) were evaluated for the adsorption of cationic azo dyes, including methylene blue (MB), rhodamine B (RB), and crystal violet (CV), from single and binary adsorptive systems. A-ASBC showed a higher maximum adsorption capacity for these dyes (MB: 53.62 mg·g-1, RB: 32.33 mg·g-1, CV: 735.73 mg·g-1) than ASBCs in a single system because it had a greater specific surface area and more oxygen containing-functional groups on the surface. The adsorption process of the three dyes onto the adsorbents was in good agreement with the Freundlich adsorption isotherm and fit the pseudo-second-order kinetic model, which revealed sorbate polymolecular layer formation over the adsorbent surface and the involvement of chemisorption. The adsorption mechanism showed that the adsorption of three dyes on adsorbents could be postulated as a multistep process with extraordinary affinity-induced adsorption in terms of both physisorption and chemisorption. In the binary adsorptive system, the results showed that all MB, RB, and CV had antagonistic/competitive effects on each other's adsorption (QBinary/QSingle < 1). Furthermore, a phytotoxic assay affirmed the effectiveness of the adsorbent in adsorbing dye species from aqueous solutions using Brassica pekinensis L. seeds as the model. Therefore, activated biochar prepared from AS can be used as a potentially economical and effective adsorbent for treating printing and dyeing wastewater.

BACKGROUND: DNA methylation is implicated in coronary heart disease (CHD), but current evidence is based on small, cross-sectional studies. We examined blood DNA methylation in relation to incident CHD across multiple prospective cohorts. METHODS: Nine population-based cohorts from the United States and Europe profiled epigenome-wide blood leukocyte DNA methylation using the Illumina Infinium 450k microarray, and prospectively ascertained CHD events including coronary insufficiency/unstable angina, recognized myocardial infarction, coronary revascularization, and coronary death. Cohorts conducted race-specific analyses adjusted for age, sex, smoking, education, body mass index, blood cell type proportions, and technical variables. We conducted fixed-effect meta-analyses across cohorts. RESULTS: Among 11 461 individuals (mean age 64 years, 67% women, 35% African American) free of CHD at baseline, 1895 developed CHD during a mean follow-up of 11.2 years. Methylation levels at 52 CpG (cytosine-phosphate-guanine) sites were associated with incident CHD or myocardial infarction (false discovery rate<0.05). These CpGs map to genes with key roles in calcium regulation (ATP2B2, CASR, GUCA1B, HPCAL1), and genes identified in genome- and epigenome-wide studies of serum calcium (CASR), serum calcium-related risk of CHD (CASR), coronary artery calcified plaque (PTPRN2), and kidney function (CDH23, HPCAL1), among others. Mendelian randomization analyses supported a causal effect of DNA methylation on incident CHD; these CpGs map to active regulatory regions proximal to long non-coding RNA transcripts. CONCLUSION: Methylation of blood-derived DNA is associated with risk of future CHD across diverse populations and may serve as an informative tool for gaining further insight on the development of CHD.

BACKGROUND: Domain of unknown function (DUF) proteins represent a number of gene families that encode functionally uncharacterized proteins in eukaryotes. The DUF4228 gene family is one of these families in plants that has not been described previously. RESULTS: In this study, we performed an extensive comparative analysis of DUF4228 proteins and determined their phylogeny in the plant lineage. A total of 489 high-confidence DUF4228 family members were identified from 14 land plant species, which sub-divided into three distinct phylogenetic groups: group I, group II and group III. A highly conserved DUF4228 domain and motif distribution existed in each group, implying their functional conservation. To reveal the possible biological functions of these DUF4228 genes, 25 ATDUF4228 sequences from Arabidopsis thaliana were selected for further analysis of characteristics such as their chromosomal position, gene duplications and gene structures. Ka/Ks analysis identified seven segmental duplication events, while no tandemly duplication gene pairs were found in A. thaliana. Some cis-elements responding to abiotic stress and phytohormones were identified in the upstream sequences of the ATDUF4228 genes. Expression profiling of the ATDUF4228 genes under abiotic stresses (mainly osmotic, salt and cold) and protein-protein interaction prediction suggested that some ATDUF4228 genes are may be involved in the pathways of plant resistance to abiotic stresses. CONCLUSION: These results expand our knowledge of the evolution of the DUF4228 gene family in plants and will contribute to the elucidation of the biological functions of DUF4228 genes in the future.

Genome-wide association studies have identified hundreds of genetic variants associated with blood pressure (BP), but sequence variation accounts for a small fraction of the phenotypic variance. Epigenetic changes may alter the expression of genes involved in BP regulation and explain part of the missing heritability. We therefore conducted a two-stage meta-analysis of the cross-sectional associations of systolic and diastolic BP with blood-derived genome-wide DNA methylation measured on the Infinium HumanMethylation450 BeadChip in 17,010 individuals of European, African American, and Hispanic ancestry. Of 31 discovery-stage cytosine-phosphate-guanine (CpG) dinucleotides, 13 replicated after Bonferroni correction (discovery: N = 9,828, p < 1.0 × 10-7; replication: N = 7,182, p < 1.6 × 10-3). The replicated methylation sites are heritable (h2 > 30%) and independent of known BP genetic variants, explaining an additional 1.4% and 2.0% of the interindividual variation in systolic and diastolic BP, respectively. Bidirectional Mendelian randomization among up to 4,513 individuals of European ancestry from 4 cohorts suggested that methylation at cg08035323 (TAF1B-YWHAQ) influences BP, while BP influences methylation at cg00533891 (ZMIZ1), cg00574958 (CPT1A), and cg02711608 (SLC1A5). Gene expression analyses further identified six genes (TSPAN2, SLC7A11, UNC93B1, CPT1A, PTMS, and LPCAT3) with evidence of triangular associations between methylation, gene expression, and BP. Additional integrative Mendelian randomization analyses of gene expression and DNA methylation suggested that the expression of TSPAN2 is a putative mediator of association between DNA methylation at cg23999170 and BP. These findings suggest that heritable DNA methylation plays a role in regulating BP independently of previously known genetic variants.

Small GTPases of the ADP-ribosylation factor (Arf) family and their activating proteins (Arf-GAPs) regulate mycelial development and pathogenicity in yeast and filamentous fungi; however, little is known about their roles in nematode-trapping (NT) fungi. In this study, an ortholog of Arf-GAP Glo3 (AoGlo3) in Saccharomyces cerevisiae was characterized in the NT fungus Arthrobotrys oligospora. Deletion of the Aoglo3 gene resulted in growth defects and an increase in hyphal septum. Meanwhile, the sporulation capacity of the ΔAoglo3 mutant was decreased by 98%, and 67.1-71.2% spores became gourd or claviform in shape (from obovoid), which was accompanied by a significant decrease in the spore germination rate. This reduced sporulation capacity correlated with the transcriptional repression of several sporulation-related genes including fluG, rodA, abaA, medA, and lreA. The ΔAoglo3 mutant was also sensitive to several chemical stressors such as Congo red, NaCl, and sorbitol. Additionally, AoGlo3 was found to be involved in endocytosis, and more myelin figures were observed in the ΔAoglo3 mutant than in the wild-type strain, which was consistent with the presence of more autophagosomes observed in the mutant. Importantly, AoGlo3 affected the production of mycelial traps and serine proteases for nematode predation. In summary, AoGlo3 is involved in the regulation of multiple cellular processes such as mycelial growth, conidiation, environmental adaption, endocytosis, and pathogenicity in A. oligospora.

Ca2+/calmodulin-dependent protein kinases (CaMKs) are unique second-messenger molecules that impact almost all cellular processes in eukaryotes. In this study, five genes encoding different CaMKs were characterized in the nematode-trapping fungus Arthrobotrys oligospora. These CaMKs, which were retrieved from the A. oligospora genome according to their orthologs in fungi such as Aspergillus nidulans and Neurospora crassa, were expressed at a low level in vitro during mycelial growth stages. Five deletion mutants corresponding to these CaMKs led to growth defects in different media and increased sensitivity to several environmental stresses, including H2O2, menadione, SDS, and Congo red; they also reduced the ability to produce conidia and traps, thus causing a deficiency in nematicidal ability as well. In addition, the transcriptional levels of several typical sporulation-related genes, such as MedA, VelB, and VeA, were down-regulated in all ΔCaMK mutants compared with the wild-type (WT) strain. Moreover, these mutants exhibited hypersensitivity to heat shock and ultraviolet-radiation stresses compared with the WT strain. These results suggest that the five CaMKs in A. oligospora are involved in regulating multiple cellular processes, such as growth, environmental stress tolerance, conidiation, trap formation, and virulence.

The critical swimming speed (U crit, cm s-1) of juvenile tiger puffer Takifugu rubripes was determined under different temperatures (15, 21, 25 and 30 °C), salinities (5, 10, 20, 32 and 40), body lengths (3.32, 4.08, 5.06 and 5.74 cm) and starvation days (1, 3, 6 and 9 days). Acute temperature change, body length and starvation significantly influenced the U crit of tiger puffers, whereas acute salinity change had no significant effect. The U crit increased as the temperature increased from 15 to 30 °C. The U crit increased as the body length increased from 3.32 to 5.74 cm, whereas relative critical swimming speed (U crit', body length s-1) decreased. The relationship between the body length (l, cm) and U crit or U crit' can be described by the quadratic model as U crit = - 1.4088 l 2 + 16.976 l - 11.64, R 2 = 0.9698 (P < 0.01) or U crit' = - 0.1937 l 2 + 0.9504 l + 7.7666, R 2 = 0.9493 (P < 0.01). The U crit decreased as starvation days increased from 1 to 9 days. Low temperature and starvation can reduce the swimming ability of juvenile tiger puffers. Results can be of value in evaluating the swimming ability of juvenile tiger puffers, understanding ecological processes and improving the population enhancement of tiger puffers.
