RESUMO

OBJECTIVES: Bisphenol A (BPA) is widely used in epoxy resins in China. There are few reports on the adverse health effects of occupational exposure to BPA. This study examined associations between urinary BPA concentrations in workers and laboratory parameters for health status. METHODS: Spot urine checks at the end shift on Friday were used for cross-sectional analysis of BPA concentrations, and blood or urinary markers of liver function, glucose homeostasis, thyroid function and cardiovascular diseases were measured. The 28 participants were workers in two semiautomatic epoxy resin factories. RESULTS: The average urinary BPA concentration was 55.73±5.48 ng/ml (geometric mean ± geometric SD) (range 5.56-1934.85 ng/ml). After adjusting for urine creatinine (Cr), it was 31.96±4.42 µg/g Cr (geometric mean ± geometric SD) (range 4.61-1253.69 µg/g Cr). BPA feeding operators showed the highest concentrations, over 10 times those of the crushing and packing and office workers. Higher BPA concentrations were associated with clinically abnormal concentrations of FT3, FT4, TT3, TT4, thyroid-stimulating hormone, glutamic-oxaloacetic transaminase and γ-glutamyl transferase. Workers with higher BPA concentrations showed higher FT3 concentrations (linear trend: p<0.001). Bivariate correlation tests for laboratory analytes within normal limits showed FT3 to be positively associated with logged BPA concentrations, r=0.57, p=0.002. FT4 was positively associated with lactate dehydrogenase, r=0.45, p=0.020, and insulin was positively associated with thyroid-stimulating hormone with r=0.57, p=0.009. CONCLUSIONS: Higher occupational BPA exposure, reflected in urinary concentrations of BPA, may be associated with thyroid hormone disruption.

OBJECTIVES: : To investigate the exposure to terephthalic acid (TPA), and to evaluate it's effects on organ function including the potential risk factors for uroliths and bladder tumor to TPA. METHODS: : Exposure-response modeling was carried out in a cohort of 141 TPA exposure workers and three subgroups were classified according to their urine TPA concentration. The control group consisted of 77 workers with no exposure to TPA dust. The inhalatory exposure of the application workers was estimated from biological monitoring data. Urine and blood samples were collected from all workers before and after work shift to monitor variables of liver, kidney, and lung. Haematological variables and serum biochemistry were valued, pulmonary functions were tested, and ion changes in both serum and urine were measured. RESULTS: : Increased urinary excretion of TPA (0-5mmol/molCr after the shift) reflected occupational workers TPA exposure. We also observed the exposure-response relations for the intensity of TPA exposure and the urine variables. Increased serum aspartate aminotransferase (AST) and lactate dehydrogenase (LDH) but within normal range is not increased. The slightly increased serum angiotensin-converting enzyme activity (SACE) was considered to be related to particulate of airborne TPA dust inhalation. No difference between referents and workers exposed to TPA was found for haematological variables. CONCLUSIONS: : No clinical organ dysfunctions were found in this investigation working with TPA. However, special precautions are still necessarily taken to avoid excessive or prolonged contact.

OBJECTIVE: To investigate the insulin-like growth factor-I (IGF-I) gene and polypeptide expression in cultured rat osteoblast (ROB) and the role of IGF-I in mediating the cell-to-cell communication by mimicking the pharmacokinetics of parathyroid hormone (PTH). METHODS: The ROB was cultured with three kinds of treatment: (1) Control (Ctr), the cells were cultured without PTH during the first 6 hours and the subsequent 42 hours in a 48-hour cycle; (2) Intermittent exposure to PTH (Itm), the cells were cultured with PTH during the first 6 hours, but without PTH in the subsequent 42 hours; and (3) Continuous exposure to PTH (Ctu), the cells were cultured with PTH during the first 6 hours and the subsequent 42 hours. RESULTS: The bone-forming activities of ROB were increased in Itm and inhibited in Ctu. The IGF-I mRNA content in Itm cells was elevated only during the first 6 hours and that in Ctu cells was elevated at any time during an incubation cycle. The free IGF-I concentration in the medium of Itm cells was generally higher and that of the Ctu cells was generally lower compared with those of the Ctr cells. The IGF-I antibody significantly reduced the alkaline phosphatase activity within the cells of Ctr and Itm. CONCLUSIONS: PTH rapidly and constantly stimulates the IGF-I gene transcription of osteoblast. There was an obvious discrepancy between the IGF-I mRNA content within the osteoblast and the free IGF-I level around the osteoblast in either mode of PTH action. The IGF-I might be important for osteoblast-osteoblast communication and bone-forming activity, not only in intermittent PTH administration, but also in the physiological functioning of osteoblasts.
