RESUMO

Follicular helper T (TFH) cells are a specialized subset of CD4+ T cells that essentially support germinal center responses where high-affinity and long-lived humoral immunity is generated. The regulation of TFH cell survival remains unclear. Here we report that TFH cells show intensified lipid peroxidation and altered mitochondrial morphology, resembling the features of ferroptosis, a form of programmed cell death that is driven by iron-dependent accumulation of lipid peroxidation. Glutathione peroxidase 4 (GPX4) is the major lipid peroxidation scavenger and is necessary for TFH cell survival. The deletion of GPX4 in T cells selectively abrogated TFH cells and germinal center responses in immunized mice. Selenium supplementation enhanced GPX4 expression in T cells, increased TFH cell numbers and promoted antibody responses in immunized mice and young adults after influenza vaccination. Our findings reveal the central role of the selenium-GPX4-ferroptosis axis in regulating TFH homeostasis, which can be targeted to enhance TFH cell function in infection and following vaccination.

BACKGROUND & AIMS: Immunotherapy for chronic hepatitis B virus (HBV) infection has not yet demonstrated sufficient efficacy. We developed a non-integrative lentiviral-vectored therapeutic vaccine for chronic hepatitis B and tested its antiviral effects in HBV-persistent mice and two inactive HBsAg carriers. METHODS: Lentiviral vectors (LVs) encoding the core, preS1, or large HBsAg (LHBs) proteins of HBV were evaluated for immunogenicity in HBV-naïve mice and therapeutic efficacy in a murine model of chronic HBV infection. In addition, two inactive HBsAg carriers each received two doses of 5×107 transduction units (TU) or 1×108 TU of lentiviral-vectored LHBs (LV-LHBs), respectively. The endpoints were safety, LHBs-specific T-cell responses, and serum HBsAg levels during a 24-week follow-up. RESULTS: In the mouse models, LV-LHBs was the most promising in eliciting robust antigen-specific T cells and in reducing the levels of serum HBsAg and viral load. By the end of the 34-week observation period, six out of ten (60%) HBV-persistent mice vaccinated with LV-LHBs achieved serum HBsAg loss and significant depletion of HBV-positive hepatocytes in the liver. In the two inactive HBsAg carriers, vaccination with LV-LHBs induced a considerable increase in the number of peripheral LHBs-specific T cells in one patient, and a weak but detectable response in the other, accompanied by a sustained reduction of HBsAg (-0.31 log10 IU/ml and -0.46 log10 IU/ml, respectively) from baseline to nadir. CONCLUSIONS: A lentiviral-vectored therapeutic vaccine for chronic HBV infection demonstrated the potential to improve HBV-specific T-cell responses and deplete HBV-positive hepatocytes, leading to a sustained loss or reduction of serum HBsAg. IMPACT AND IMPLICATIONS: Chronic HBV infection is characterized by an extremely low number and profound hypo-responsiveness of HBV-specific T cells. Therapeutic vaccines are designed to improve HBV-specific T-cell responses. We show that immunization with a lentiviral-vectored therapeutic HBV vaccine was able to expand HBV-specific T cells in vivo, leading to reductions of HBV-positive hepatocytes and serum HBsAg.

INTRODUCTION: Dragendorff's reagent has low sensitivity and non-specificity for some alkaloids. A new alkaloid derivatisation reagent has been developed and optimised by using a Box-Behnken design method. This new reagent is applicable for structurally diverse natural alkaloids, and is proposed as a universal alkaloid staining reagent. OBJECTIVE: To establish an efficient and sensitive thin-layer chromatography (TLC) identification method for Leonuri Herba and Leonuri Fructus to characterise their differences and similarities. METHOD: Three key components (bismuth subnitrate, potassium iodide and iodine) in the derivatisation reaction were re-constructed and optimised using a response surface method. Different inorganic acids, essential additives of the staining reaction, were compared by a single-factor experiment design. RESULTS: This newly optimised reagent for alkaloids reported in this study, named the enhanced Dragendorff-Wagner's reagent, is composed of 0.82% bismuth subnitrate, 11.1% potassium iodide and 0.76% iodine in 38% phosphoric acid solution. Validation results indicate that the TLC spot of stachydrine stained with the enhanced Dragendorff-Wagner's reagent had a limit of detection of 2.0 µg, good intra- and inter-plate, and intra- and intra-day precisions with relative standard devition values less than 5.0%, and stability over 90 min. CONCLUSION: This enhanced Dragendorff-Wagner's reagent was applied for TLC identification of 16 reference alkaloids representing 11 structural skeletons, and two closely related herbs (Leonuri Herba and Leonuri Fructus). This newly enhanced Dragendorff-Wagner's reagent is a universal, effective, and sensitive staining reagent for alkaloids.

A unique avian-origin A/H7N9 influenza virus has so far caused 134 cases with 44 deaths. Probing the host factors contributing to disease severity, we found that lower levels of plasma inflammatory cytokines on hospital admission correlated with faster recovery in 18 patients with A/H7N9 influenza virus, whereas high concentrations of (in particular) IL-6, IL-8, and macrophage inflammatory protein-1ß were predictive of a less favorable or fatal outcome. Analysis of bronchoalveolar lavage samples showed up to 1,000-fold greater cytokine/chemokine levels relative to plasma. Furthermore, patients with the rs12252-C/C IFN-induced transmembrane protein-3 (IFITM3) genotype had more rapid disease progression and were less likely to survive. Compared with patients with the rs12252-T/T or rs12252-T/C genotype of IFITM3, patients with the C/C genotype had a shorter time from disease onset to the time point when they sought medical aid (hospital admission or antiviral therapy) and a shorter interval to development of the acute respiratory distress syndrome stage (reflected by shorter intervals between clinical onset and methylprednisolone treatments and higher rates of mechanical ventilator use), as well as experiencing elevated/prolonged lung virus titers and cytokine production and higher mortality. The present analysis provides reported data on the H7N9 influenza-induced "cytokine storm" at the site of infection in humans and identifies the rs12252-C genotype that compromises IFITM3 function as a primary genetic correlate of severe H7N9 pneumonia. Together with rs12252 sequencing, early monitoring of plasma cytokines is thus of prognostic value for the treatment and management of severe influenza pneumonia.

With the rapid development of artificial intelligence and data science, Dynamic Bayesian Network (DBN), as an effective probabilistic graphical model, has been widely used in many engineering fields. And swarm intelligence algorithm is an optimization algorithm based on natural selection with the characteristics of distributed, self-organization and robustness. By applying the high-performance swarm intelligence algorithm to DBN structure learning, we can fully utilize the algorithm's global search capability to effectively process time-based data, improve the efficiency of network generation and the accuracy of network structure. This study proposes an improved bacterial foraging optimization algorithm (IBFO-A) to solve the problems of random step size, limited group communication, and the inability to maintain a balance between global and local searching. The IBFO-A algorithm framework comprises four layers. First, population initialization is achieved using a logistics-sine chaotic mapping strategy as the basis for global optimization. Second, the activity strategy of a colony foraging trend is constructed by combining the exploration phase of the Osprey optimization algorithm. Subsequently, the strategy of bacterial colony propagation is improved using a "genetic" approach and the Multi-point crossover operator. Finally, the elimination-dispersal activity strategy is employed to escape the local optimal solution. To solve the problem of complex DBN learning structures due to the introduction of time information, a DBN structure learning method called IBFO-D, which is based on the IBFO-A algorithm framework, is proposed. IBFO-D determines the edge direction of the structure by combining the dynamic K2 scoring function, the designed V-structure orientation rule, and the trend activity strategy. Then, according to the improved reproductive activity strategy, the concept of "survival of the fittest" is applied to the network candidate solution while maintaining species diversity. Finally, the global optimal network structure with the highest score is obtained based on the elimination-dispersal activity strategy. Multiple tests and comparison experiments were conducted on 10 sets of benchmark test functions, two non-temporal and temporal data types, and six data samples of two benchmark 2T-BN networks to evaluate and analyze the optimization performance and structure learning ability of the proposed algorithm under various data types. The experimental results demonstrated that IBFO-A exhibits good convergence, stability, and accuracy, whereas IBFO-D is an effective approach for learning DBN structures from data and has practical value for engineering applications.

With the increasing development of nanomaterials, the construction of multiscale nanostructured interphase has emerged as a viable technique to reinforce carbon fiber-reinforced polymer composites. Here, "flexible" aramid nanofibers (ANFs) were first introduced on the surface of carbon fibers (CF) by electrophoretic deposition (EPD), and then "rigid" MXene sheets were grafted by ultrasonic impregnation. This feasible two-step treatment introduces a hierarchical "rigid-flexible" structure at the CF/polyamide (PA) interface. Results showed that this "rigid-flexible" multilayer structure improved the roughness, chemical bonding, mechanical interlocking, and wettability of CF/PA composites. At the same time, the modulus variation between the fiber and the matrix is significantly smoothed due to the increased thickness of the interfacial layer, increasing the payload transfer from the PA matrix to the fiber and decreasing the stress concentration. Compared to the desized CF, the interlaminar shear strength (ILSS) and tensile strength of the modified CF-ANF@MX0.2/PA composite increased by 50.02 and 36.11%, respectively. This innovative interfacial design and feasible treatment method facilitate the construction of firmly interacting interfacial layers in CF/PA composites, offering broad prospects for the production of high-performance CF/PA composites.

Pile is a common foundation on the slope, which poses a serious threat to the construction and operation if the slope deformation and causes landslide. In this study, a model device of pile foundation on landslide was independently developed by relative displacement loading between pile and soil to explore the influence of landslide deformation on pile and analysis the soil failure rule and the deformation characteristics of pile in different stages of landslide deformation, a few model tests were completed including the relative displacement between soil and pile from 1 to 17 cm, and the pile diameter and the modulus of slide bed were also considered. The results indicated that: the evolution process of landslide deformation with pile foundation on could be divided into four stages including soil compaction, cracks growth, yield stage, and failure stage; ratios of the maximum soil pressure and bending moment growth from the soil compaction stage to the cracks growth stage to the total growth in these four stages are both exceeding 60%; the soil pressure increases with the increase of pile diameter and sliding bed modulus. Therefore, it is best to effectively monitor and control the landslide in the initial soil compression stage that in soil compaction stage and methods such as increasing pile foundations or reinforcing the sliding bed can be used for protection.

In modern air combat, collaborative detection and engagement among multiple aircraft have gradually become a predominant combat approach. In response to the challenges posed by modern stealth aircraft, although their external factors such as coatings significantly reduce the chances of enemy detection, once these stealth aircraft activate their radar systems, they become susceptible to detection. Therefore, an application model has been proposed to mitigate enemy detection of our stealth aircraft through a collaborative approach. The underlying principle involves employing the concept of multi-aircraft collaboration, where the aircraft are divided into transmitters and receivers. The transmitters emit radar waves while the receivers are responsible for receiving these waves. This approach effectively mitigates the increased probability of enemy detection resulting from the activation of our receivers' radar systems. The optimization problem we aim to address is determining the optimal formation configuration for cooperative flight, specifically a formation with a specific configuration that maximizes the detectable range. This optimization problem is known as the configuration optimization problem for Airborne Radar Network with Separate Transmitting and Receiving (ARN-STAR). Existing methods for this problem typically suffer from limitations in either effectiveness or efficiency. To overcome these limitations, we propose an optimized configuration method based on an improved Artificial Fish Swarm Algorithm (IFSA) for ARN-STAR. Firstly, leveraging the distribution characteristics of the target radar wave's spatial scattering and the concept of dual-radar spatial diversity, we establish a mathematical model and an optimization objective function for ARN-STAR. Secondly, to address efficiency concerns, we optimize the computational process using the IAFS, successfully improving the speed of computation. To address the issue of effectiveness, we introduce adaptive adjustments to the movement step size of the artificial fish and improve the implementation of the three behavioral modes, thereby avoiding local optima and enhancing the accuracy of finding the optimal configuration. Finally, using our self-developed multi-aircraft collaborative simulation platform, we apply the improved AFSA to obtain the optimal formation configuration scheme and compare it with other methods. Simulation results demonstrate that our proposed method effectively solves the problem of finding the optimal formation configuration in multi-aircraft collaborative detection scenarios with "one transmission and multiple receptions." It overcomes the low computational efficiency associated with traditional methods while maintaining good accuracy. This approach enables the enhancement of overall combat capabilities while ensuring the safety of our aircraft to the greatest extent possible. It should be noted that the scenarios discussed in this study are at the configurational configuration level between UAVs, rather than involving the design of the UAVs combat control system itself.

Objectives: Sex-specific differences in internet gaming disorder (IGD) neurophysiology remain underexplored. Here we investigated sex-related variability in regional homogeneity (ReHo) and functional connectivity (FC) in IGD and their correlations with sleep quality. Methods: Resting-state functional magnetic resonance imaging (fMRI) scans were performed on 52 subjects with IGD and 50 healthy controls (HCs). Two-way ANOVA was used to examine sex and diagnosis interactions in ReHo and FC, followed by post-hoc analyses to explore FC biomarkers for different sexes. Results: In ReHo analysis, the four groups showed significant sex and diagnosis interactions in the right middle frontal gyrus (rMFG). FC analysis with rMFG as the seed region revealed a significant sex and diagnosis interaction effect in FC of the rMFG with the bilateral postcentral gyrus (PoCG). In male IGD group, FC between the rMFG and the bilateral PoCG correlates strongly with daytime dysfunction score and the Pittsburgh sleep quality inventory (PSQI) total score. Conclusion: These findings emphasize the importance of considering sexual dimorphism in the neurobiology of IGD, which might influence subsequent treatment strategies.

The Shroom (Shrm) family of actin-binding proteins has a unique and highly conserved Apx/Shrm Domain 2 (ASD2) motif. Shroom protein directs the subcellular localization of Rho-associated kinase (ROCK), which remodels the actomyosin cytoskeleton and changes cellular morphology via its ability to phosphorylate and activate non-muscle myosin II. Therefore, the Shrm-ROCK complex is critical for the cellular shape and the development of many tissues, including the neural tube, eye, intestines, heart, and vasculature system. Importantly, the structure and expression of Shrm proteins are also associated with neural tube defects, chronic kidney disease, metastasis of carcinoma, and X-link mental retardation. Therefore, a better understanding of Shrm-mediated signaling transduction pathways is essential for the development of new therapeutic strategies to minimize damage resulting in abnormal Shrm proteins. This paper provides a comprehensive overview of the various Shrm proteins and their roles in morphogenesis and disease.

The utilization of carbon-based fibers as a fundamental constituent holds strong appeal for diverse materials and devices. However, the poor fiber graphitic structure resulting from the heat treatment of atactic polyacrylonitrile (PAN) precursors often leads to a modest performance of carbon-based fibers. This paper takes electrospun carbon nanofibers (CNFs) as the research object and provides a seed-assisted graphitization strategy to improve the fiber graphitic structures. The typical melamine/cyanuric acid self-assembly precursor of graphitic carbon nitride is applied as supramolecular seeds in CNFs and demonstrates significant promotion of fiber graphitization, while it decomposes at elevated temperatures. Further studies show that the higher carbon content contributes to the better heat resistance of seeds; thus, nanoscale 2,6-diaminopyridine/cyanuric acid and 2,4,6-triaminopyrimidine/barbituric acid supramolecular seeds are developed. Both systems can be uniformly distributed in PAN precursors through in situ self-assembly and withstand high-temperature carbonization without severe pyrolysis. The dispersed seeds contribute to the formation of fibrillar PAN crystals and promote their conversion to ordered graphitic domains through nucleation and templating roles. The obtained CNFs exhibit increased crystallinity and graphitization degree with improved orientation and refined size of fiber crystals. As a result, the strength, modulus, and elongation at break of CNFs are comprehensively enhanced.

Objectives: Major Depressive Disorder (MDD) is significantly influenced by childhood trauma (CT), affecting brain anatomy and functionality. Despite the unique disease trajectory in MDD patients with CT, the underlying neurobiological mechanisms remain unclear. Our objective is to investigate CT's impact on the white matter structure of the brain in patients with MDD. Methods: This research employed tract-based spatial statistics (TBSS) to detect variations between groups in Fractional Anisotropy (FA) throughout the whole brain in 71 medication-free MDD patients and 97 HCs. Participants filled out the Childhood Trauma Questionnaire (CTQ) and assessments for depression and anxiety symptoms. The relationship between FA and CTQ scores was explored with partial correlation analysis, adjusting for factors such as age, gender, educational background, and length of illness. Results: Compared to HCs, the MDD group showed decreased FA values in the right posterior limb of the internal capsule (PLIC), the inferior fronto-occipital fasciculus (IFOF), and bilateral superior longitudinal fasciculus (SLF). Simple effects analysis revealed that compared to HC-CT, the MDD-CT group demonstrated decreased FA values in right PLIC, IFOF, and bilateral SLF. The MDD-nCT group showed decreased FA values in right PLIC and IFOF compared to HC-nCT. The total scores and subscale scores of CTQ were negatively correlated with the FA in the right SLF. Conclusion: The right SLF may potentially be influenced by CT during the brain development of individuals with MDD. These results enhance our knowledge of the role of the SLF in the pathophysiology of MDD and the neurobiological mechanisms by which CT influences MDD.

Epoxy and epoxide composites have a wide range of outdoor applications wherein they are affected by ageing. In this study, epoxy casting plates and epoxy-based composite rods for use in overhead conductors were prepared. A concurrent investigation concerning the ageing of epoxy resins and their carbon fibre composites was carried out via artificially accelerated experiments under hygrothermal and salt mist conditions. The moisture penetration along the depth, water absorption, appearance, hardness, density of the epoxy resins, and variation patterns of the impact strength and tensile strength of the epoxy-based composites were investigated. The ageing mechanisms were explored using Fourier transform infrared spectroscopy (FTIR) and scanning electron microscopy (SEM). Both ageing modes had essentially similar influences on the properties of the resins and their composites; moreover, they did not significantly affect the chemical structure and microstructure of the epoxy resin, with the physical adsorption of water primarily observed during the ageing process. The moisture absorption behaviour of the epoxy obeyed Fick's law. Although the water penetration rate in the salt mist ageing mode was slightly higher than that in the hygrothermal ageing mode during the early ageing stage, it was essentially the same during the later stage. The final moisture absorption rate at saturation was approximately 1.1% under both modes. The flexural strengths and impact strengths of the composites in both ageing modes followed a similar trend. They decreased gradually with the ageing time and then stabilized at almost the same value. The flexural strength was reduced from 803 MPa to 760 MPa and the impact strength from 383 J/m2 to 310 J/m2, indicating a decrease of approximately 5.4% and 19%, respectively. The absorbed water during the ageing process caused micro-cracks at the interface between the fibres and resin, weakening the interfacial strength and reducing the mechanical properties of the composites.

Developing universal CARs with improved flexible targeting and controllable activities is urgently needed. While several studies have suggested the potential of CD16a in tandem with monoclonal antibodies to construct universal CAR-T cells, the weak affinity between them is one of the limiting factors for efficacy. Herein, we systematically investigated the impact of Fcγ receptor (FcγR) affinity on CAR-T cells properties by constructing universal CARs using Fcγ receptors with different affinities for IgG1 antibodies, namely CD16a, CD32a, and CD64. We demonstrated that the activities of these universal CAR-T cells on tumor cells could be redirected and regulated by IgG1 antibodies. In xenografted mice, 64CAR chimeric Jurkat cells with the highest affinity showed significant antitumor effects in combination with herceptin in the HER2 low expression U251 MG model. However, in the CD20 high expression Raji model, 64CAR caused excessive activation of CAR-T cells, which resulted in cytokine release syndrome (CRS) and the decline of antitumor activity, and 32CAR with a moderate affinity brought the best efficacy. Our work extended the knowledge about FcγR-based universal CAR-T cells and suggested that only the FcγRCAR with an appropriate affinity can offer the optimal antitumor advantages of CAR-T cells.

Objectives: Childhood trauma (CT) is a known risk factor for major depressive disorder (MDD), but the mechanisms linking CT and MDD remain unknown. The purpose of this study was to examine the influence of CT and depression diagnosis on the subregions of the anterior cingulate cortex (ACC) in MDD patients. Methods: The functional connectivity (FC) of ACC subregions was evaluated in 60 first-episode, drug-naïve MDD patients (40 with moderate-to-severe and 20 with no or low CT), and 78 healthy controls (HC) (19 with moderate-to-severe and 59 with no or low CT). The correlations between the anomalous FC of ACC subregions and the severity of depressive symptoms and CT were investigated. Results: Individuals with moderate-to severe CT exhibited increased FC between the caudal ACC and the middle frontal gyrus (MFG) than individuals with no or low CT, regardless of MDD diagnosis. MDD patients showed lower FC between the dorsal ACC and the superior frontal gyrus (SFG) and MFG. They also showed lower FC between the subgenual/perigenual ACC and the middle temporal gyrus (MTG) and angular gyrus (ANG) than the HCs, regardless of CT severity. The FC between the left caudal ACC and the left MFG mediated the correlation between the Childhood Trauma Questionnaire (CTQ) total score and HAMD-cognitive factor score in MDD patients. Conclusion: Functional changes of caudal ACC mediated the correlation between CT and MDD. These findings contribute to our understanding of the neuroimaging mechanisms of CT in MDD.

BACKGROUND AND AIMS: Chronic HBV infection evolves through different phases. Interactions between viral replication and the host immune response in the liver underlie the pathogenesis of this disease. The aim of this study was to directly visualize the HBV replication intermediates at a single-cell resolution inscribed on morphological changes corresponding to disease activity. METHODS: A set of archived formalin-fixed paraffin-embedded liver needle biopsies from treatment-naïve patients were collected and categorized into phases according to the American Association for the Study of the Liver Diseases (AASLD) guidelines. HBV RNA and DNA were detected using in situ hybridization assays. RESULTS: The hepatocytes were ubiquitously infected in subjects with immune tolerance, and their percentage was gradually decreased in immune-active and inactive chronic hepatitis B phases. HBV-infected hepatocytes were prone to localize close to fibrous septa. The subcellular distribution of signals was able to distinguish hepatocytes with productive infection from those harboring HBV integrants and transcriptionally inactive covalently closed circular DNAs. A smaller number of hepatocytes with productive infection and more harboring transcriptionally inactive covalently closed circular DNA or HBV integrants became apparent in the inactive chronic hepatitis B phase. CONCLUSION: An atlas of in situ characteristics of viral-host interactions for each phase is described, which sheds light on the nature of viral replication and disease pathogenesis among the phases of chronic HBV infection.

Karst is a common engineering environment in the process of tunnel construction, which poses a serious threat to the construction and operation, and the theory on calculating the settlement without the assumption of semi-infinite half-space is lack. Meanwhile, due to the limitation of test conditions or field measurement, the settlement of high-speed railway tunnel in Karst region is difficult to control and predict effectively. In this study, a novel intelligent displacement prediction model, following the machine learning (ML) incorporated with the finite difference method, is developed to evaluate the settlement of the tunnel floor. A back propagation neural network (BPNN) algorithm and a random forest (RF) algorithm are used herein, while the Bayesian regularization is applied to improve the BPNN and the Bayesian optimization is adopted for tuning the hyperparameters of RF. The newly proposed model is employed to predict the settlement of Changqingpo tunnel floor, located in the southeast of Yunnan Guizhou Plateau, China. Numerical simulations have been performed on the Changqingpo tunnel in terms of variety of karst size, and locations. Validations of the numerical simulations have been validated by the field data. A data set of 456 samples based on the numerical results is constructed to evaluate the accuracy of models' predictions. The correlation coefficients of the optimum BPNN and BR model in testing set are 0.987 and 0.925, respectively, indicating that the proposed BPNN model has more great potential to predict the settlement of tunnels located in karst areas. The case study of Changqingpo tunnel in karst region has demonstrated capability of the intelligent displacement prediction model to well predict the settlement of tunnel floor in Karst region.

Variants of severe acute respiratory syndrome coronavirus 2 frequently arise within infected individuals. Here, we explored the level and pattern of intra-host viral diversity in association with disease severity. Then, we analyzed information underlying these nucleotide changes to infer the impetus including mutational signatures and immune selection from neutralizing antibody or T-cell recognition. From 23 January to 31 March 2020, a set of cross-sectional samples were collected from individuals with homogeneous founder virus regardless of disease severity. Intra-host single-nucleotide variants (iSNVs) were enumerated using deep sequencing. Human leukocyte antigen (HLA) alleles were genotyped by Sanger sequencing. Medical records were collected and reviewed by attending physicians. A total of 836 iSNVs (3-106 per sample) were identified and distributed in a highly individualized pattern. The number of iSNVs paced with infection duration peaked within days and declined thereafter. These iSNVs did not stochastically arise due to a strong bias toward C > U/G > A and U > C/A > G substitutions in reciprocal proportion with escalating disease severity. Eight nonsynonymous iSNVs in the receptor-binding domain could escape from neutralization, and eighteen iSNVs were significantly associated with specific HLA alleles. The level and pattern of iSNVs reflect the in vivo viral-host interaction and the disease pathogenesis.

SARS-CoV-2 vaccine booster dose can induce a robust humoral immune response, however, its cellular mechanisms remain elusive. Here, we investigated the durability of antibody responses and single-cell immune profiles following booster dose immunization, longitudinally over 6 months, in recipients of a homologous BBIBP-CorV/BBIBP-CorV or a heterologous BBIBP-CorV/ZF2001 regimen. The production of neutralizing antibodies was dramatically enhanced by both booster regimens, and the antibodies could last at least six months. The heterologous booster induced a faster and more robust plasmablast response, characterized by activation of plasma cells than the homologous booster. The response was attributed to recall of memory B cells and the de novo activation of B cells. Expanded B cell clones upon booster dose vaccination could persist for months, and their B cell receptors displayed accumulated mutations. The production of antibody was positively correlated with antigen presentation by conventional dendritic cells (cDCs), which provides support for B cell maturation through activation and development of follicular helper T (Tfh) cells. The proper activation of cDC/Tfh/B cells was likely fueled by active energy metabolism, and glutaminolysis might also play a general role in promoting humoral immunity. Our study unveils the cellular mechanisms of booster-induced memory/adaptive humoral immunity and suggests potential strategies to optimize vaccine efficacy and durability in future iterations.
