RESUMO

As one of the main external factors affecting the fire extinguishing accuracy of sprinkler systems, it is necessary to analyze and study random wind. However, in practical applications, there is little research on the impact of random wind on sprinkler fire extinguishing points. To address this issue, a new random wind acquisition system was constructed in this paper, and a method for predicting jet trajectory falling points in Random Forest (RF) under the influence of random wind was proposed, and compared with the commonly used prediction model Support Vector Machine (SVM). The method in this article reduces the error in the x direction of the 50 m prediction result from 2.11 m to 1.53 m, the error in the y direction from 0.64 m to 0.6 m, and the total mean absolute error (MAE) from 31.3 to 23.5. Simultaneously, predict the falling points of jet trajectory at different distances under the influence of random wind, to demonstrate the feasibility of the proposed method in practical applications. The experimental results show that the system and method proposed in this article can effectively improve the influence of random wind on the falling points of a jet trajectory. In summary, the image acquisition system and error prediction method proposed in this article have many potential applications in fire extinguishing.

RESUMO

This research presents an innovative methodology aimed at monitoring jet trajectory during the jetting process using imagery captured by unmanned aerial vehicles (UAVs). This approach seamlessly integrates UAV imagery with an offline learnable prompt vector module (OPVM) to enhance trajectory monitoring accuracy and stability. By leveraging a high-resolution camera mounted on a UAV, image enhancement is proposed to solve the problem of geometric and photometric distortion in jet trajectory images, and the Faster R-CNN network is deployed to detect objects within the images and precisely identify the jet trajectory within the video stream. Subsequently, the offline learnable prompt vector module is incorporated to further refine trajectory predictions, thereby improving monitoring accuracy and stability. In particular, the offline learnable prompt vector module not only learns the visual characteristics of jet trajectory but also incorporates their textual features, thus adopting a bimodal approach to trajectory analysis. Additionally, OPVM is trained offline, thereby minimizing additional memory and computational resource requirements. Experimental findings underscore the method's remarkable precision of 95.4% and efficiency in monitoring jet trajectory, thereby laying a solid foundation for advancements in trajectory detection and tracking. This methodology holds significant potential for application in firefighting systems and industrial processes, offering a robust framework to address dynamic trajectory monitoring challenges and augment computer vision capabilities in practical scenarios.

microRNAs (miRNAs) are increasingly recognized as important regulators of many biological processes in mosquitoes, vectors of numerous devastating infectious diseases. Identification of bona fide targets remains the bottleneck for functional studies of miRNAs. In this study, we used CLEAR-CLIP assays to systematically analyze miRNA-mRNA interactions in adult female Anopheles gambiae mosquitoes. Thousands of miRNA-target pairs were captured after direct ligation of the miRNA and its cognate target transcript in endogenous Argonaute-miRNA-mRNA complexes. Using two interactions detected in this manner, miR-309-SIX4 and let-7-kr-h1, we demonstrated the reliability of this experimental approach in identifying in vivo gene regulation by miRNAs. The miRNA-mRNA interaction dataset provided an invaluable opportunity to decipher targeting rules of mosquito miRNAs. Enriched motifs in the diverse targets of each miRNA indicated that the majority of mosquito miRNAs rely on seed-based canonical target recognition, while noncanonical miRNA binding sites are widespread and often contain motifs complementary to the central or 3' ends of miRNAs. The time-lapse study of miRNA-target interactomes in adult female mosquitoes revealed dynamic miRNA regulation of gene expression in response to varying nutritional sources and physiological demands. Interestingly, some miRNAs exhibited flexibility to use distinct sequences at different stages for target recognition. Furthermore, many miRNA-mRNA interactions displayed stage-specific patterns, especially for those genes involved in metabolism, suggesting that miRNAs play critical roles in precise control of gene expression to cope with enormous physiological demands associated with egg production. The global mapping of miRNA-target interactions contributes to our understanding of miRNA targeting specificity in non-model organisms. It also provides a roadmap for additional studies focused on regulatory functions of miRNAs in Anopheles gambiae.

Icaritin, a small molecule currently being investigated in phase III clinical trials in China (NCT03236636 and NCT03236649) for treatment of advanced hepatocellular carcinoma (HCC), is a prenylflavonoid derivative obtained from the Epimedium genus. Previously, it was found that Icaritin decreased the expression of PD-L1, but its direct molecular targets and the underlying mechanisms have not been identified. In this study, we report the identification of IKK-α as the protein target of Icaritin by biotin-based affinity binding assay. The further mutagenesis assay has provided evidence that C46 and C178 in IKK-α were essential amino acids for Icaritin binding to IKK-α, revealing the binding sites of Icaritin to IKK-α for the first time. Functionally, Icaritin inhibited the NF-κB signalling pathway by blocking IKK complex formation, which led to decreased nuclear translocation of NF-κB p65, and subsequent downregulation of PD-L1 expression in a dose-dependent manner. More importantly, PD-L1-positive patients exhibited longer overall survival upon Icaritin therapy. Finally, Icaritin in combination with checkpoints antibodies, such as α-PD-1, has demonstrated much better efficacy than any single therapy in animal models. This is the first report that anticancer effects of Icaritin are mediated, at least in part, by impairing functions of IKK-α.

Malaria, caused by the protozoan parasite Plasmodium and transmitted by Anopheles mosquitoes, represents a major threat to human health. Plasmodium's infection cycle in the Anopheles vector is critical for transmission of the parasite between humans. The midgut-stage bottleneck of infection is largely imposed by the mosquito's innate immune system. microRNAs (miRNAs, small noncoding RNAs that bind to target RNAs to regulate gene expression) are also involved in regulating immunity and the anti-Plasmodium defense in mosquitoes. Here, we characterized the mosquito's miRNA responses to Plasmodium infection using an improved crosslinking and immunoprecipitation (CLIP) method, termed covalent ligation of endogenous Argonaute-bound RNAs (CLEAR)-CLIP. Three candidate miRNAs' influence on P. falciparum infection and midgut microbiota was studied through transgenically expressed miRNA sponges (miR-SPs) in midgut and fat body tissues. MiR-SPs mediated conditional depletion of aga-miR-14 or aga-miR-305, but not aga-miR-8, increased mosquito resistance to both P. falciparum and P. berghei infection, and enhanced the mosquitoes' antibacterial defenses. Transcriptome analysis revealed that depletion of aga-miR-14 or aga-miR-305 resulted in an increased expression of multiple immunity-related and anti-Plasmodium genes in mosquito midguts. The overall fitness cost of conditionally expressed miR-SPs was low, with only one of eight fitness parameters being adversely affected. Taken together, our results demonstrate that targeting mosquito miRNA by conditional expression of miR-SPs may have potential for the development of malaria control through genetically engineered mosquitoes.

An all-inorganic lead-free halides Cs-Cu-I system, represented by Cs3Cu2I5 and CsCu2I3, has attracted attention for their good photophysical characteristics recently. Successive works had reported their application potential in light-emitting devices. However, there is no report for CsCu2I3 in X-ray scintillation detectors so far. We notice that CsCu2I3 may be advantageous in such an application due to the one-dimensional crystal structure, the congruent-melting feature, and the high spectral matching to some photosensors. In this work, we explore the scintillation properties and imaging application of CsCu2I3 in X-ray scintillator detector. The oriented structure is designed to enhance the imaging performance of a CsCu2I3 detector. Close-space sublimation process and nanoscale seed screening strategy are employed to realize this design by producing a large-area (25 cm2) CsCu2I3 thick film layer with the oriented nanorod structure. This CsCu2I3 detector eventually achieves a high spatial resolution of 7.5 lp mm-1 in X-ray imaging.

MicroRNAs are important coordinators of circadian regulation that mediate the fine-tuning of gene expression. Although many studies have shown the effects of individual miRNAs on the circadian clock, the global functional miRNA-mRNA interaction network involved in the circadian system remains poorly understood. Here, we used CLEAR (Covalent Ligation of Endogenous Argonaute-bound RNAs)-CLIP (Cross-Linking and Immuno-Precipitation) to explore the regulatory functions of miRNAs in the circadian system by comparing the miRNA-mRNA interactions between Drosophila wild-type strain W1118 and a mutant of the key circadian transcriptional regulator Clock (Clkjrk ). This experimental approach unambiguously identified tens of thousands of miRNA-mRNA interactions in both the head and body. The miRNA-mRNA interactome showed dramatic changes in the Clkjrk flies. Particularly, among ~300 miRNA-mRNA circadian relevant interactions, multiple interactions involving core clock genes pdp1, tim, and vri displayed distinct changes as a result of the Clk mutation. Based on the CLEAR-CLIP analysis, we found a novel regulation of the circadian rhythm and sleep by the miR-375-timeless interaction. The results indicated that Clk disruption abolished normal rhythmic expression of miR-375 and the functional regulation occurred in the l-LNv neurons, where miR-375 modulated the circadian rhythm and sleep via targeting timeless. This work provides the first global view of miRNA regulation in the circadian rhythm.

Juvenile hormone (JH) regulates many aspects of insect development and reproduction. In some processes, JH plays a critical role in defining the action of the steroid hormone 20-hydroxyecdysone (20E). In Aedes aegypti mosquitoes, JH prepares newly emerged female adults to become competent to synthesize vitellogenin in response to 20E after blood ingestion. The molecular basis of this competence is still not well understood. Here, we report that JH regulates pre-mRNA splicing of the taiman gene, which encodes a key transcriptional regulator required for both JH- and 20E-controlled gene expression. JH stimulated the production of the Taiman isoforms A/B, while reducing the levels of the isoforms C/D, in the fat body after adult eclosion. The appearance of the A/B isoforms in maturing mosquitoes was accompanied by acquisition of the competence to respond to 20E. Depletion of the A/B isoforms, by inhibiting the alternative splicing or by isoform-specific RNA interference, considerably diminished the 20E-induced gene expression after a blood meal and substantially impaired oocyte development. In accordance with this observation, further studies indicated that in the presence of 20E, the Taiman A/B isoforms showed much stronger interactions with the 20E receptor complex than the Taiman C/D isoforms. In contrast, all four isoforms displayed similar capabilities of forming active JH receptor complexes with the methoprene-tolerant protein (Met). This study suggested that JH confers the competence to newly emerged female mosquitoes by regulating mRNA splicing to generate the Taiman isoforms that are essential for the vitellogenic 20E response.

Chemical modifications on histones and DNA/RNA constitute a fundamental mechanism for epigenetic regulation. These modifications often function as docking marks to recruit or stabilize cognate "reader" proteins. So far, a platform for quantitative and high-throughput profiling of the epigenetic interactome is urgently needed but still lacking. Here, we report a 3D-carbene chip-based surface plasmon resonance imaging (SPRi) technology for this purpose. The 3D-carbene chip is suitable for immobilizing versatile biomolecules (e.g., peptides, antibody, DNA/RNA) and features low nonspecific binding, random yet function-retaining immobilization, and robustness for reuses. We systematically profiled binding kinetics of 1,000 histone "reader-mark" pairs on a single 3D-carbene chip and validated two recognition events by calorimetric and structural studies. Notably, a discovery on H3K4me3 recognition by the DNA mismatch repair protein MSH6 in Capsella rubella suggests a mechanism of H3K4me3-mediated DNA damage repair in plant.

An improved Near-Field Computer Vision (NFCV) system for intelligent fire robot was proposed that was based on our previous works in this paper, whose aims are to realize falling position prediction of jet trajectory in fire extinguishing. Firstly, previous studies respecting the NFCV system were briefly reviewed and several issues during application testing were analyzed and summarized. The improved work mainly focuses on the segmentation and discrimination of jet trajectory adapted to complex lighting environment and interference scenes. It mainly includes parameters adjustment on the variance threshold and background update rate of the mixed Gaussian background method, jet trajectory discrimination based on length and area proportion parameters, parameterization, and feature extraction of jet trajectory based on superimposed radial centroid method. When compared with previous works, the proposed method reduces the average error of prediction results from 1.36 m to 0.1 m, and the error variance from 1.58 m to 0.13 m. The experimental results suggest that every part plays an important role in improving the functionality and reliability of the NFCV system, especially the background subtraction and radial centroid methods. In general, the improved NFCV system for jet trajectory falling position prediction has great potential for intelligent fire extinguishing by fire-fighting robots.

OBJECTIVE: To evaluate the clinical effect and safety of transurethral 180 W front-firing GreenLight laser vaporization of the prostate (PVP) in the treatment of benign prostatic hyperplasia (BPH). METHODS: A total of 61 BPH patients underwent 180W front-firing GreenLight laser PVP (n = 30, the PVP group) or transurethral plasmakinetic resection of the prostate (n = 31, the control group) from March to December 2019. We collected the pre-, intra- and post-operative clinical data and compared them between the two groups of patients. RESULTS: Operations were successfully completed in all the cases with no blood transfusion or serious complications. Compared with the controls, the patients of the PVP group showed remarkably less intra-operative blood loss (ï¼»62.3 ± 15.9ï¼½ vs ï¼»48.8 ± 9.6ï¼½ ml, P < 0.05), shorter operation time (ï¼»75.0 ± 9.9ï¼½ vs ï¼»57.5 ± 19.0ï¼½ min, P < 0.05), postoperative bladder lavage time (ï¼»64.4 ± 10.5ï¼½ vs ï¼»25.2 ± 11.5ï¼½ h, P < 0.05), catheter-indwelling time (ï¼»5.1 ± 0.5ï¼½ vs ï¼»2.5 ± 0.5ï¼½ d, P < 0.05) and hospitalization time (ï¼»7.3 ± 1.7ï¼½ vs ï¼»4.1 ± 0.6ï¼½ d, P < 0.05), and a lower incidence of postoperative hematuria (12.9% ï¼»4/31ï¼½ vs 0% ï¼»0/30ï¼½, P < 0.05). No statistically significant differences, however, were found between the two groups in the incidence rates of capsular perforation, transurethral resection syndrome (TURS), urinary incontinence, urethral stricture and post-extubation urinary retention. Significant improvement was observed in IPSS, QOL, Qmax and PVR in both groups post-operatively (P < 0.05). CONCLUSIONS: Compared with transurethral plasmakinetic resection of the prostate, 180W front-firing GreenLight laser PVP, with the advantages of less bleeding, shorter catheter-indwelling time and faster recovery, is safer and more effective for the treatment of BPH, with no need for drug withdrawal for those taking anticoagulants, and especially applicable to the elderly and high-risk patients.

A novel method of near-field computer vision (NFCV) was developed to monitor the jet trajectory during the jetting process, which was used to precisely predict the falling point position of the jet trajectory. By means of a high-resolution webcam, the NFCV sensor device collected near-field images of the jet trajectory. Preprocessing of collected images was carried out, which included squint image correction, noise elimination, and jet trajectory extraction. The features of the jet trajectory in the processed image were extracted, including: start-point slope (SPS), end-point slope (EPS), and overall trajectory slope (OTS) based on the proposed mean position method. A multiple regression jet trajectory range prediction model was established based on these trajectory characteristics and the reliability of the model was verified. The results show that the accuracy of the prediction model is not less than 94% and the processing time is less than 0.88s, which satisfy the requirements of real-time online jet trajectory monitoring.

BACKGROUND/AIMS: Unlike other organs, which only have one set of capillary network, the renal microvasculature consists of two sets of capillary network series connected by efferent arterioles. Angiotensin II constricts the efferent glomerular artery. Hence, renal tumor blood flow (BF) distribution may be different from tumors in other organs. This study aims to investigate the effects of angiotensin II on the hemodynamics of intrarenal VX2 tumors using perfusion computed tomography(CT). METHODS: Twenty-four male New Zealand white rabbits were randomly divided into three groups: groups A (blank controls), group B (negative controls), and group C (angiotensin II-treated animals). Group B and C were established to the model of intrarenal VX2 tumors. Furthermore, perfusion CT of the kidney was performed in each group. Prior to perfusion CT scan in group C, the mean arterial blood was elevated to 150-160 mmHg by angiotensin II. The BF, blood volume (BV), mean transit time (MTT), capillary permeability-surface area product (PS), and relative permeability-surface area product (RPS) of tumors and renal tissues were calculated. RESULTS: Compared with normal renal cortex tissues in group A, the BF, BV and PS values of tumors in group B were significantly lower, MTT was prolonged and RPS increased. Compared with group B, only the RPS of these tumors increased from 83.23 ± 29.17% to 120.94 ± 31.84% by angiotensin II infusion. Angiotensin II significantly increased the RPS value of the renal cortex distant from the tumor (CDT) and the right renal cortex (RRC). CONCLUSIONS: Perfusion CT can accurately observe the influence of angiotensin II on normal and tumor BF in kidneys. This clarifies the effect of angiotensin II on intrarenal tumor hemodynamics.

Juvenile hormone (JH) is a key regulator of a wide diversity of developmental and physiological events in insects. Although the intracellular JH receptor methoprene-tolerant protein (MET) functions in the nucleus as a transcriptional activator for specific JH-regulated genes, some JH responses are mediated by signaling pathways that are initiated by proteins associated with plasma membrane. It is unknown whether the JH-regulated gene expression depends on the membrane-mediated signal transduction. In Aedes aegypti mosquitoes, we found that JH activated the phospholipase C (PLC) pathway and quickly increased the levels of inositol 1,4,5-trisphosphate, diacylglycerol, and intracellular calcium, leading to activation and autophosphorylation of calcium/calmodulin-dependent protein kinase II (CaMKII). When abdomens from newly emerged mosquitoes were cultured in vitro, the JH-activated gene expression was repressed substantially if specific inhibitors of PLC or CaMKII were added to the medium together with JH. In newly emerged female mosquitoes, RNAi-mediated depletion of PLC or CaMKII considerably reduced the expression of JH-responsive genes, including the Krüppel homolog 1 gene (AaKr-h1) and the early trypsin gene (AaET). JH-induced loading of MET to the promoters of AaKr-h1 and AaET was weakened drastically when either PLC or CaMKII was inactivated in the cultured tissues. Therefore, the results suggest that the membrane-initiated signaling pathway modifies the DNA-binding activity of MET via phosphorylation and thus facilitates the genomic responses to JH. In summary, this study reveals an interplay of genomic and nongenomic signaling mechanisms of JH.

The structural, magnetic and magnetoelectric properties were investigated for sol-gel prepared BaFe12O19 nanorods and plate-like nanoparticles. Based on comparative experiments with bulk ceramics, it is found that larger structural distortion is present in nanostructures, which could cause the enhancement of magnetocrystalline anisotropy and the off-center displacement of Fe3+ ions, and thus result in improved magnetic and magnetoelectric properties in BaFe12O19 plate-like nanoparticles. Meanwhile, the local (Fe2+-Fe3+) dipoles, which usually appear during a high temperature sintering process, can also contribute to the negative magnetoelectric effect of BaFe12O19 nanorods and a large room temperature magnetodielectric coefficient of about -13% is observed at 104 Hz and 9 kOe.

The relationship among dielectric anomaly, ferroelectric response, defects, and microstructures was established for (K0.48(1+x)Na0.52)0.95Li0.05Nb0.95Sb0.05O3 (x = 0.04, 0.00, -0.02, -0.04 and -0.08) ceramics. For x = -0.02 and -0.04, larger coercive fields and lower remnant polarizations were obtained; besides, an additional dielectric relaxation behavior was observed with the activation energy Ea being about 2.19 eV and 1.92 eV, respectively. Furthermore, the grain and grain boundary contributions to the capacitance were separated using impedance spectroscopy, which, combined with back-scattering characterization, firmly indicates the core-shell structure of K-deficient samples (x = -0.02 and -0.04). Unlike the cores, the shells possess a large amount of K+ vacancies (). This work paves a way for regulating the fine structure and more on the electrical properties of KNN-based materials.

Real-time dynamic displacement and acceleration responses of the main span section of the Tianjin Fumin Bridge in China under ambient excitation were tested using a Global Navigation Satellite System (GNSS) dynamic deformation monitoring system and an acceleration sensor vibration test system. Considering the close relationship between the GNSS multipath errors and measurement environment in combination with the noise reduction characteristics of different filtering algorithms, the researchers proposed an AFEC mixed filtering algorithm, which is an combination of autocorrelation function-based empirical mode decomposition (EMD) and Chebyshev mixed filtering to extract the real vibration displacement of the bridge structure after system error correction and filtering de-noising of signals collected by the GNSS. The proposed AFEC mixed filtering algorithm had high accuracy (1 mm) of real displacement at the elevation direction. Next, the traditional random decrement technique (used mainly for stationary random processes) was expanded to non-stationary random processes. Combining the expanded random decrement technique (RDT) and autoregressive moving average model (ARMA), the modal frequency of the bridge structural system was extracted using an expanded ARMA_RDT modal identification method, which was compared with the power spectrum analysis results of the acceleration signal and finite element analysis results. Identification results demonstrated that the proposed algorithm is applicable to analyze the dynamic displacement monitoring data of real bridge structures under ambient excitation and could identify the first five orders of the inherent frequencies of the structural system accurately. The identification error of the inherent frequency was smaller than 6%, indicating the high identification accuracy of the proposed algorithm. Furthermore, the GNSS dynamic deformation monitoring method can be used to monitor dynamic displacement and identify the modal parameters of bridge structures. The GNSS can monitor the working state of bridges effectively and accurately. Research results can provide references to evaluate the bearing capacity, safety performance, and durability of bridge structures during operation.

The application scope of surface plasmon resonance (SPR) and SPR imaging (SPRi) is rapidly growing, and tools such as high-performance and low-cost slides could enable more rapid growth of the field. We describe herein a novel silver slide, addressing the inherent instability of plain silver structure by improving adhesion between the glass substrate and the silver layer with a thin buffer layer of gold. Covered by a self-assembled monolayer (SAM) only, SPR characteristics of the slide remain steady for more than 3 months under regular storage. In a bioassay, the slide substantiates the predicted nearly 100% sensitivity improvement over gold slides and exhibits exceptional performance stability as determined by sensitivity and resolution measurements during the extended 40,000 s multicycle experiment. We demonstrate the suitability of this new slide for large-area SPRi, describing analysis results from a 1 296-ligand protein microarray. We predict this slide structure will provide a stable, high-sensitivity solution for high-throughput SPRi applications and other surface analysis platforms.

Self-assembled monolayers designed to immobilize capture antibodies are usually prepared using a mixture of functional and inactive linkers. Here, using low molar ratios (1:1 to 1:100) of the two linkers resulted in loss of binding capability of the anti-EGFR (epidermal growth factor receptor) antibody nimotuzumab, as assessed by surface plasmon resonance imaging. We then developed a simple theoretical model to predict the optimal surface density of the functional linker, taking into account the antibody size and linker diameter. A high (1:1000) dilution of the functional linker yielded the best results. As an advantage, this approach does not require chemical modification of the protein.

From the perspective of portable monitoring devices,we use an analog front-end AFE4490 design a module of Non-invasive blood oxygen measurement, used to collect human pulse wave signal and peak (valley) value detection and then use the principles of non-invasive oximetry calculated oxygen saturation (SPO2). This design of noninvasive oximetry module has the characteristics of small size, low power consumption, and the results of test show that the measurement of oxygen saturation are correct.
